Three new cyclopentapeptides, versicoloritides A-C (1-3), a new orcinol tetramer, tetraorcinol A (4), and two new lactones, versicolactones A and B (5 and 6) together with three known metabolites, diorcinol, glyantrypine, and cordyol C were isolated from the fermentation broth of the coral-associated fungus Aspergillus versicolor LCJ-5-4. Their structures were elucidated by spectroscopic and chemical methods. The new compounds 1-4 were evaluated for their radical-scavenging activity and antimicrobial activity against Staphylococcus aureus, Escherichia coli, Enterobacter aerogenes, Bacillus subtilis, Pseudomonas aeruginosa, and Candida albicans and cytotoxicity against P388 and Hela cell lines. Compound 4 showed weak radical-scavenging activity against the DPPH radical with an IC50 value of 67 μM.  相似文献

A new lovastatin analogue versicorin (1), together with three related compounds, decumbenones A (2) and B (3) and versiol (4), were isolated from mycelial solid cultures of Aspergillus versicolor SC0156. Their structures were elucidated on the basis of MS data and NMR spectroscopic analysis. The new compound versicorin (1) possesses a hexahydro-2H-naphtho[1,8-bc]furan moiety, which is a rare type of the lovastatin-analogous compounds. A hypothetical biosynthetic pathway for compounds 14 was proposed.  相似文献

Abstract

Two new β-carboline alkaloids, aspergillspins A-B (12), three new quinolone alkaloids, aspergillspins C-E (35), and two new isocoumarins, aspergillspins F-G (67), together with four known alkaloids were isolated from the marine gorgonian-derived fungus Aspergillus sp. SCSIO 41501. Their structures were identified by spectroscopic analysis, and the absolute configurations of several chiral carbons in 2 and 3 were further established by quantum chemical calculations of the electronic circular dichroism (ECD) spectra. Their cytotoxic and antibacterial activities were also evaluated.  相似文献

Known diketopiperazine WIN 64821 and its asymmetric stereoisomers (13) had been isolated from the culture broth of a marine gut fungus Aspergillus sp. DX4H. The planar and stereochemistry for new compounds were determined by a suite of techniques including mass, NMR and CD spectra together with Marfey’s method. (4S,5S)-4-Formyl-5-vinyl-2-oxazolidinone (4b), which is readily obtained via a zinc-silver-mediated reductive elimination of alpha-d-lyxofuranosyl phenyl sulfone (3b), is successfully converted to the naturally occurring, nonproteinogenic amino acid (2S,3R)-3-amino-2-hydroxydecanoic acid (2). Also in this study, a facile "oxazolidinone rearrangement" reaction is uncovered during the attempted formation of the (methylthio)thiocarbonate derivative of the oxazolidinone alcohol 7.  相似文献

A novel polycyclic aspochalasin, aspochalazine A (1), was isolated along with three related known compounds 24 from the culture broth of Aspergillus sp., which was found in the gut of a marine isopod Ligia oceanica. The structures were determined on the basis of ESI-MS, NMR spectrum data and single-crystal X-ray diffraction. This is the first report about aspochalasin derivative with an azabicyclo. Cytotoxicity against the prostate cancer PC3 cell line was assayed by MTT method. Aspochalazine A showed weak activity. A plausible biosynthetic pathway for aspochalazine A was proposed.  相似文献

A new methyl 2-(4-((2-hydroxy-3-methylbut-3-en-1-yl)oxy)phenyl) acetate 1, together with five known compounds 26, was isolated from the culture of the deep sea-derived fungus Aspergillus westerdijkiae SCSIO 05233. The new structure was determined by NMR (1H and 13C NMR, HSQC, HMBC and MS) and optical rotation analysis. Compound 5 displayed weak inhibitory activities towards K562 and promyelocytic HL-60 with IC50 values of 25.8 and 44.9 μM, and compound 6 showed strong antifouling activity with EC50 value 8.81 μg/mL.  相似文献

A novel diketopiperazine (DKP) dimer, aspergilazine A (1), dimerized by two DKP units via a rare N-1 to C-6 linkage, was isolated from the marine-derived fungus Aspergillus taichungensis ZHN-7-07. The structure was determined by the combination of spectroscopic and Marfey’s methods. Biological evaluation indicated that aspergilazine A (1) had a weak activity against influenza A (H1N1) virus.  相似文献

An enantiospecific synthesis of the tetrahydrofuran amino acid (2S,5S)-5-(aminomethyl)-tetrahydrofuran-2-carboxylic acid 1 is reported. The sugar enone 2-(S)-octyl 6-O-acetyl-3,4-dideoxy-α-d-glycero-hex-3-enopyranosid-2-ulose 2a, derived from galactose, was employed as a chiral precursor. The enone 2a was converted by chemical manipulation of the functional groups into the 6-azido-2-O-tosyl-3,4,6-trideoxy-d-erythro-hexono-1,5-lactone 9 as key intermediate. Methanolysis of 9 induced the opening of the lactone and the attack of the hydroxyl group at C-5 to C-2 with the displacement of the tosylate. This reaction led to the formation of the tetrahydrofuran ring of methyl (2S,5S)-5-(azidomethyl)-tetrahydrofuran-2-carboxylate 10, which was readily converted into 1. The overall yield of the sequence was 35%, and all the intermediates and the final product have been fully characterized. In addition, the preferential conformations in solution of lactone 9 and target molecule 1 have been established.  相似文献

A new 12-membered macrolide, aspergillolide (1), along with nine known compounds (210), were isolated from the fungus Aspergillus sp. S-3-75 associated with the sea cucumber Holothuria nobilis Selenka. The structure and absolute stereochemistry of 1 were elucidated on the basis of extensive spectroscopic methods and single crystal X-ray diffraction analysis.  相似文献

It is found that 2, 4-dimethyl-5-nitrofuran-3-carboxylic acid and its methyl ester can give comparatively stable anionic groups, which are prepared electrochemically and investigated by EPR. Their lives (80–100 sec) are 4- 5 times greater than the lives of other anionic groups of the 5-nitrofuran series previously studied. Starting from 2, 4-dimethyl-5-nitrofuran-3-carboxylic acid and 2, 4-dimethylfuran-3-carboxylic acid, two new semisynthetic penicillins are prepared, with activities basically extending to Gram-positive microorganisms, including forms of staphylcocci resistant to benzylpenicillin. Introduction of the nitro group into the furan ring increases the stability of penicillin to acid 79-fold. Low toxicity penicillins are synthesized (LD50 1000–1500 mg/kg).  相似文献

Conditions for the selective preparation of 5-carbethoxy-4-methyl-2-substituted pyrimidines or 5-acetyl-4-hydroxy-2-substituted pyrimidines by condensation of ethoxymethyleneacetoacetic ester with 1,3-binucleophiles are proposed. Six new sesquiterpenoid quinones with an amino acid residue, metachromins L-Q (1-6), have been isolated from an Okinawan marine sponge Spongia sp. The structures and stereochemistry of 1-6 were elucidated on the basis of the spectroscopic data and chemical correlations. Metachromins L (1) and M (2) showed modest cytotoxicity.  相似文献
