Cascade Multicomponent Synthesis of Indoles,Pyrazoles, and Pyridazinones by Functionalization of Alkenes

The development of multicomponent reactions for indole synthesis is demanding and has hardly been explored. The present study describes the development of a novel multicomponent, cascade approach for indole synthesis. Various substituted indole derivatives were obtained from simple reagents, such as unfunctionalized alkenes, diazonium salts, and sodium triflinate, by using an established straightforward and regioselective method. The method is based on the radical trifluoromethylation of alkenes as an entry into Fischer indole synthesis. Besides indole synthesis, the application of the multicomponent cascade reaction to the synthesis of pyrazoles and pyridazinones is described.

Machine-Driven Chemoenzymatic Synthesis of Glycopeptide

Historically, researchers have put considerable effort into developing automation systems to prepare natural biopolymers such as peptides and oligonucleotides. The availability of such mature systems has significantly advanced the development of natural science. Over the past twenty years, breakthroughs in automated synthesis of oligosaccharides have also been achieved. A machine-driven platform for glycopeptide synthesis by a reconstructed peptide synthesizer is described. The designed platform is based on the use of an amine-functionalized silica resin to facilitate the chemical synthesis of peptides in organic solvent as well as the enzymatic synthesis of glycan epitopes in the aqueous phase in a single reaction vessel. Both syntheses were performed by a peptide synthesizer in a semiautomated manner.     

Total synthesis of antitumor depsipeptide (-)-doliculide

[reaction: see text] (-)-Doliculide, a potent antitumor agent, is synthesized stereoselectively in a convergent manner. The key strategy involves a stereoselective synthesis of the polyketide unit and synthesis of the D-tyrosine derivative, followed by assembly of the fragments by an esterification and cycloamidation reaction sequence. The synthesis of the polyketide fragment was achieved by an iterative asymmetric synthesis to install stereoselectively both 1,3-dimethyl groups and the 1,3-diol unit by utilizing asymmetric cyclopropanations and Sharpless asymmetric epoxidations as the key steps.     

常压催化合成N-乙基间甲氧基苯胺

以间氨苯酚为原料,用溴乙烷做烷基化试剂,硫酸二甲酯作甲基化试剂,通过改变溶剂及加入合适的相转移催化剂等方法优化工艺,经两步反应合成N-乙基间甲氧基苯胺,产率达到48.8%。该反应步骤少,原料廉价易得,反应条件极其温和,方法简单,为实际生产提供了依据。     

倍半硅氧烷合成进展

对以苯基三氯硅烷、苯基三烷氧基硅烷、甲基三氯硅烷、甲基三烷氧基硅烷、甲基三乙酸基硅烷分别为原料采用碱催化平衡水解三步法合成可溶性高分子量聚倍半硅氧烷及其共聚物进行了综述,并对以苯基三氯硅烷为原料采取一步法合成热塑性聚苯基倍半硅氧烷予以介绍,总结了以不同有机基为原料采用碱催化平衡法合成不溶性立方体聚倍半硅氧烷的研究结果,最后对卤盐催化非水解sol-gel法制备有机倍半硅氧烷凝胶作了阐述,以期对合成不同结构的聚倍半硅氧烷提供帮助。     

分子氧催化氧化苯直接合成苯酚

苯酚是重要的有机合成中间体,当前主要通过异丙苯法合成苯酚的技术路线存在制备流程长、消耗丙烯、副产丙酮等不足。以分子氧为氧化剂由苯氧化直接合成苯酚则具有潜在重大的经济效益、社会效益和环境效益,已成为催化与有机合成等研究领域中极具挑战性的热点课题之一。本文较为系统地总结了分子氧氧化苯通过一步法合成苯酚的研究工作,着重综述了用于该反应的催化剂如Pd、Cu、V等金属或其化合物,也归纳了影响此反应的主要因素,并介绍相应的反应机理。最后,对分子氧催化氧化苯合成苯酚反应的研究提供了一些建议和展望。     

与癌症相关的糖类抗原Globo-H六糖的全合成进展

Globo-H作为一种和乳腺癌、前列腺癌相关的复杂糖类抗原,其发现为糖类疫苗开发和癌症免疫治疗带来了机遇,但如何高效、高纯地获得合成糖类抗原,以供研究和临床应用,也向寡糖合成方法学提出了挑战.综述了1995年Danishefsky首次以糖烯组装策略全合成Globo-H以来的各种新方法,如:Schmidt的三氯乙酰亚胺酯法、Boons的双向糖苷化法、Wong的基于糖基给体活性差异的一锅煮策略、Seeberger的液相线性合成和固相自动组装法、Huang的多组份反复预活化一锅煮法和最新报道的酶法.就糖合成方法学而言,硫苷法依旧可称为"明星方法",糖烯、三氯乙酰亚胺酯和氟代糖也普遍采用,磷酸酯糖基给体在固相合成中的应用正显示出其新的活力.这些方法代表了当今糖化学的水平和发展趋势.

以反应类型分类综述了近年来介孔分子筛MCM-41的修饰及在非均相催化有机合成, 特别是在手性不对称合成、烯烃环氧化等领域的研究进展, 并对其发展趋势进行了展望.     

Synthesis of 8‐Desmethoxy Psymberin: A Putative Biosynthetic Intermediate Towards the Marine Polyketide Psymberin

The synthesis of a putative biosynthetic precursor of psymberin including a formal synthesis of the natural product is described. The key step towards the densely functionalized tetrahydropyran core was an enantioselective catalytic Mukaiyama aldol reaction using a titanium(IV)–BINOL catalyst system. syn‐Selective reduction followed by ozonolysis led to a rapid assembly of the tetrahydropyran ring. This flexible approach also allows the synthesis of similar fragments of other complex molecules such as bryostatins and pederins. The syn‐selective coupling between the tetrahydropyran and the aromatic aldehyde was achieved using a boron‐mediated aldol reaction which was followed by further transformations to complete the synthesis of the precursor as well as the formal synthesis of the natural product.     

First total synthesis of phenylpyridine analogues of the antimitotic rhazinilam

The first synthesis of phenylpyridine analogues of rhazinilam and evaluation of these new structures as inhibitors of microtubule disassembly by interaction with tubulin are described. The synthesis is based on such key steps as picolinic metalation, hetero-ring cross-coupling and reduction of an acetyl group to an ethyl group. Elaboration of a quaternary picolinic carbon is one of the challenges of the synthesis. Biological evaluation of compounds bearing a quaternary picolinic carbon showed interactions with tubulin similar to (-)-rhazinilam but at a lower level.     

Fmoc solid-phase synthesis of peptide thioesters by masking as trithioortho esters

[reaction: see text] Total chemical synthesis of proteins by chemoselective ligation relies on C-terminal peptide thioesters as building blocks. Their preparation by standard Fmoc solid-phase peptide synthesis is made difficult by the lability of thioesters to aminolysis by the secondary amines used for removal of the Fmoc group. Here we present a novel backbone amide linker (BAL) strategy for their synthesis in which the thioester functionality is masked as a trithioortho ester throughout the synthesis.     

Palladium-Catalyzed Allylic C-OH Functionalization for Efficient Synthesis of Functionalized Allylsilanes

A new method is described for palladium-catalyzed allylic silylation using allylic alcohols and disilanes as precursors. The reactions proceed smoothly under mild and neutral conditions, and this method is suitable for synthesis of regio- and stereodefined allylsilanes. The presented silylation reaction can be easily extended to include synthesis of allylboronates by change of the dimetallic reagent. The presented synthetic procedure offers a broad platform for the selective synthesis of functionalized allyl metal reagents, which are useful precursors in advanced organic chemistry and natural product synthesis.     

酸性条件下纯硅六方介孔分子筛的合成(Ⅱ)合成温度、时间的影响及其与碱性合成的比较

本文以十六烷基三甲基溴化铵为模板剂,正硅酸乙酯为硅源,考察了反应温度和时间对酸性条件下合成纯硅六方介孔分子筛的影响,并与碱性合成路径相比较。结果表明,由于无机物种与表面活性剂之间的相互作用不同,六方介孔分子筛的酸性合成经历了与碱性合成完全不同的机理。对要到性合成来说,由于六方结构的形成取决于稳定的模板胶束的存在,并不特别依赖于硅物种的缩合,故高温、长反应时间等有利于缩合的因素对提高产品质量几乎没有促进作用,较高的反应温度甚至起反作用。因此,酸性合成宜采用室温条件。     

以天然矿物为全部硅铝源低成本绿色合成ZSM-5分子筛的研究

ZSM-5分子筛被广泛地应用于催化、吸附分离、离子交换和绿色化工等领域,但目前其合成主要是以无机化学品为硅铝源,从源头来看是非绿色化的过程.我们以热活化的硅藻土和亚熔盐活化的累托土为全部硅铝源,通过调变两种矿物的加入比例调变合成体系的硅铝比,在水热合成体系中制备ZSM-5分子筛.系统地考察模板剂用量、碱度、投料硅铝比、晶化时间等合成条件对产物结构和性质的影响,并对在最优条件下合成的ZSM-5分子筛的物化性质进行了详细地表征,结果表明：以天然矿物为全部硅铝源可以合成出具有较高结晶度、晶粒尺寸约为4 μm的六面体挛晶形貌的ZSM-5分子筛,该分子筛比商业ZSM-5分子筛具有更高的水热稳定性和更优的加氢异构性能.     

Total synthesis of (+)-nakadomarin A

The total synthesis of (+)-nakadomarin A is described. A three-component cycloaddition of a hydroxylamine, aldehyde, and cyclopropane to form a highly functionalized tetrahydro-1,2-oxazine serves as the foundation for this synthesis. The resulting oxazine is formed as a single diastereomer with the absolute configuration being dictated by the chirality of the cyclopropane. Other key steps include: desymmetrization of a malonate by reduction, Heck cyclization and pyrrolidine formation, and ring-closing metathesis to form both cycloalkenes. Overall, the synthesis required 23 linear steps from the cyclopropane, which in turn is available (six steps) in optically pure form from commercially available d-mannitol.     

Nonthermal Plasma Synthesis of Nanocrystals: Fundamentals,Applications, and Future Research Needs

Nonthermal plasma synthesis has emerged as a viable alternative to nanocrystal synthesis in the liquid phase or by other gas phase based methods. The nonequilibrium environment containing free charge carriers enables the synthesis of nanocrystals with excellent crystallinity and narrow size distributions. This paper reviews the fundamental mechanisms involved in the synthesis of nanocrystals with nonthermal plasmas. It discusses the luminescent properties of plasma-produced silicon nanocrystals and their application in devices such as light emitting diodes. The ability of plasma synthesis to generate doped nanocrystals is a particularly appealing attribute. We present boron and phosphorous doped silicon nanocrystals and review their applications as near infrared plasmonic materials. Finally, the author presents his view of some important research needs in the area of nonthermal plasma synthesis of nanocrystals.     

Synthesis of a soluble fullerene-rotaxane incorporating a furamide template

The synthesis of a fullerene-rotaxane is described. The thread is constituted by a C₆₀ unit, which acts as a stopper, functionalized with a solubilizing side chain by 1,3 dipolar cycloaddition. The rotaxane is assembled by hydrogen bond-assisted synthesis using a fumaramide template.     

甘油脂与Germatranes缀合物的合成

报道了甘油脂与Ｇｅｒｍａｔｒａｎｅｓ缀合物的合成，合成中以ＤＣＣ为缩合剂，以吡啶为催化剂，经缩合反应得到目标产物，对缩合方法和反应条件进行了探讨，化合物结构经ＨＮＭＲ、ＩＲ和元素分析确证。     

Synthesis of Zn/Al mixed-oxide catalyst for carbonylation of glycerol with urea

Zn/Al mixed oxide was prepared by the coprecipitation or the hydrothermal method under different conditions and used as catalyst for synthesis of glycerol carbonate by carbonylation of glycerol with urea. The physical properties of the prepared Zn/Al mixed oxide particles were investigated, as well as their activity as catalyst in the mentioned synthesis. The dried Zn/Al mixed-oxide particles prepared by the coprecipitation method showed higher activity in synthesis of glycerol carbonate than those prepared by the hydrothermal method. The Zn/Al mixed oxide prepared by the coprecipitation method without NaNO₃ showed the highest catalytic activity in synthesis of glycerol carbonate.
