Organotrifluoroborates serve as coupling partners during transmetalation in the Suzuki–Miyaura reaction but require hydrolysis prior to the coupling reaction. Their anionic nature allows study of their hydrolysis by electrospray ionization mass spectrometry (ESI-MS) through real-time monitoring, complemented by pH analysis. The induction period varied according to the borates employed, and a dynamic series of equilibria for numerous ions was observed during hydrolysis. We found that the induction periods and reaction rates were sensitive to the R group of the borates, the shape of the reaction vessel, and stir rate.  相似文献

Searching for functional polyesters with stability and degradability is important due to their potential applications in biomedical supplies, biomass fuel, and environmental protection. Recently, a cyclobutane-fused lactone (CBL) polymer was experimentally found to have superior stability and controllable degradability through hydrolysis reactions after activation by mechanical force. In order to provide a theoretical basis for developing new functional degradable polyesters, in this work, we performed a detailed quantum chemical study of the alkaline and acidic hydrolysis of CBL using dispersion-corrected density functional theory (DFT-D3) and mixed implicit/explicit solvent models. Various possible hydrolysis mechanisms were found: BAC2 and BAL2 in the alkaline condition and AAC2, AAL2, and AAL1 in the acidic condition. Our calculations indicated that CBL favors the BAC2 and AAC2 mechanisms in alkaline and acidic conditions, respectively. In addition, we found that incorporating explicit water solvent molecules is highly necessary because of their strong hydrogen-bonding with reactant/intermediate/product molecules.  相似文献

为解决生物酶在战场应用环境中易失活且环境耐受性差的问题,我们合成了大环多胺双锌模拟酶配合物-2,6-双{[双(2-羟乙基)氨基]甲基}-4-甲基酚(Zn L2).Zn L2对GD(Soman,梭曼)具有较高的催化活性,能够实现多次催化循环,并且在Zn L2与GD化学计量比为(0.56∶1)的条件下水解率为100%.使用高效液相色谱-质谱与核磁共振确定催化产物为甲氟膦酸酯、甲氟膦酸、甲基膦酸频哪酯与甲基膦酸,说明Zn L2水解GD为双平行反应,反应途径分别为GD的P—F与P—O键的断裂.建立在实验的基础上,使用密度泛函理论(DFT)的B3LYP方法,从P—O与P—F键断裂这两个平行反应方向对催化过程进行计算.计算结果表明:Zn L2上Ot末端醇盐作为一个碱试剂去激活水分子,使其成为羟基并作为亲核试剂进攻中心磷原子,形成一个五配位的磷酰中间体,通过进一步亲核进攻水解中间体形成最终P—O与P—F键断裂产物.Zn L2催化GD水解的活化能为分别为5.6 kcal/mol(P—F键断裂)和11.5 kcal/mol(P—O键断裂),说明P—O键断裂过程是GD酶促水解反应的速控步骤.  相似文献

This work provides new insights from our team regarding advances in targeting canonical and non-canonical nucleic acid structures. This modality of medical treatment is used as a form of molecular medicine specifically against the growth of cancer cells. Nevertheless, because of increasing concerns about bacterial antibiotic resistance, this medical strategy is also being explored in this field. Up to three strategies for the use of DNA as target have been studied in our research lines during the last few years: (1) the intercalation of phenanthroline derivatives with duplex DNA; (2) the interaction of metal complexes containing phenanthroline with G-quadruplexes; and (3) the activity of Mo polyoxometalates and other Mo-oxo species as artificial phosphoesterases to catalyze the hydrolysis of phosphoester bonds in DNA. We demonstrate some promising computational results concerning the favorable interaction of these small molecules with DNA that could correspond to cytotoxic effects against tumoral cells and microorganisms. Therefore, our results open the door for the pharmaceutical and medical applications of the compounds we propose.  相似文献

The self‐assembly of peptides onto the surface of gold nanoparticles has emerged as a promising strategy towards the creation of artificial enzymes. The resulting high local peptide density surrounding the nanoparticle leads to cooperative and synergistic effects, which result in rate accelerations and distinct catalytic properties compared to the unconjugated peptide. This Minireview summarizes contributions to and progress made in the field of catalytically active peptide–gold nanoparticle conjugates. A N-phosphoryl branched peptide (DIPP-Leu) 2 -Lys-OCH 3 was found to efficiently cleave double-strand DNA such as PUC19 and Lambda DNA at pH 8.5 in 100 mM Tris-HCl buffer. The T4 ligase experiment implied that the DNA cleavage occurs via a hydrolytic path.  相似文献

The molecular interactions between the CeIV‐substituted Keggin anion [PW11O39Ce(OH2)4]3? ( CeK ) and hen egg‐white lysozyme (HEWL) were investigated by molecular dynamics simulations. The analysis of CeK was compared with the CeIV‐substituted Keggin dimer [(PW11O39)2Ce]10? ( CeK2 ) and the ZrIV‐substituted Lindqvist anion [W5O18Zr(OH2)(OH)]3? ( ZrL ) to understand how POM features such as shape, size, charge, or type of incorporated metal ion influence the POM???protein interactions. Simulations revealed two regions of the protein in which the CeK anion interacts strongly: cationic sites formed by Arg21 and by Arg45 and Arg68. The POMs chiefly interact with the side chains of the positively charged (arginines, lysines) and the polar uncharged residues (tyrosines, serines, aspargines) via electrostatic attraction and hydrogen bonding with the oxygen atoms of the POM framework. The CeK anion shows higher protein affinity than the CeK2 and ZrL anions, because it is less hydrophilic and it has the right size and shape for establishing interactions with several residues simultaneously. The larger, more negatively charged CeK2 anion has a high solvent‐accessible surface, which is sub‐optimal for the interaction, while the smaller ZrL anion is highly hydrophilic and cannot efficiently interact with several residues simultaneously.  相似文献

Optically active dialkoxyisopropylsulfonium salts were obtained by methylation (ethylation) of optically active alkyl isopropanesulfinates using methyl (ethyl) trifluoromethanesulfonate. Alkaline hydrolysis of a series of methoxy(alkoxy)sulfonium salts afforded the two sulfinate products methyl isopropanesulfinate and alkyl isopropanesulfinate, both formed with a slightly prevailing inversion of configuration at the sulfur atom. DFT calculations revealed that this substitution reaction proceeded stepwise according to an addition-elimination (A–E) mechanism involving the formation of high tetracoordinate SIV sulfurane intermediates. In addition, the DFT calculations showed that recombination of the hydroxy anion with the methoxy(alkoxy)sulfonium cation—leading to the parallel formation of the two most stable primary sulfuranes, with the hydroxy and alkoxy groups in apical positions and their direct decomposition—is the most energetically favorable pathway.  相似文献

The performance of MOFs in catalysis is largely derived from structural features, and much work has focused on introducing structural changes such as defects or ligand functionalisation to boost the reactivity of the MOF. However, the effects of different parameters chosen for the synthesis on the catalytic reactivity of the resulting MOF remains poorly understood. Here, we evaluate the role of metal precursor on the reactivity of Zr-based MOF-808 towards hydrolysis of the peptide bond in the glycylglycine model substrate. In addition, the effect of synthesis temperature and duration has been investigated. Surprisingly, the metal precursor was found to have a large influence on the reactivity of the MOF, surpassing the effect of particle size or number of defects. Additionally, we show that by careful selection of the Zr-salt precursor and temperature used in MOF syntheses, equally active MOF catalysts could be obtained after a 20 minute synthesis compared to 24 h synthesis.  相似文献

In the present work, the hydrolysis process of non-classical transplatin(Ⅱ) with two same planar heterocycle amines has been studied using hybrid density functional theory (B3LYP) and IEF-PCM solvation models. Optimizations were performed at the B3LYP level using a combined basis set of (LanL2DZ+6-31 +G(d,p)) with single-point energy evaluations using the B3LYP/6-31 + +G(3df,2pd) approach in vacuo and in aqueous solution. For the obtained structures of reactants, intermediates, transition states, and products, both thermodynamic (reaction energies and Gibbs energies) and kinetic (reaction barriers) characteristics were estimated. In comparison with cisplatin, decreased activation energies were obtained. The result implies that the non-classical transplatin with two same planar heterocycle amines increases the equatorial steric effect and lowers reaction barriers, which may assist in designing novel Pt-based anticancer drugs.  相似文献

Anthraquinone (AQ) derivatives, including members of the anthraquinone imide (AQI) family, have been synthesized to afford good candidates for electron-transfer studies in DNA. Electron-withdrawing groups on the AQ ring give a less negative reduction potential, as desired. As expected, the AQI derivatives have less negative reduction potentials than AQ derivatives. The AQI ring system has a half-life for hydrolysis of about 75 min in a 3:7 MeCN and 0.005 M K2CO3 in MeOH.  相似文献

On a subnanometer scale, an only one‐atom difference in a metal cluster may cause significant transitions in the catalytic activity due to the electronic and geometric configurations. We now report the atomicity‐specific catalytic activity of platinum clusters with significantly small atomicity, especially below 20. The atomic coordination structure is completely different from that of the larger face‐centered cubic (fcc) nanocrystals. Here, an electrochemical study on such small clusters, in which the atomicity ranged between 12 and 20, revealed Pt19 as the most catalytically active species. In combination with a theoretical study, a common structure that leads to a high catalytic performance is proposed.  相似文献

报道了三种新的含醇羟基、胺基或咪唑基长链配体与铜锌离子构成的金属胶束对几种羧酸酯与磷酸酯底物催化水解动力学。结果表明,设计的金属胶束N-十四烷基-2-(N-2'-羟乙基氨基甲基)咪唑锌铜(2a,2b)在共胶束CTAB(溴化十六烷基三甲铵)存在下,对强配位羧酸酯对硝基苯酚-2-吡啶甲酸酯具有很强的催化水解能力,对磷酸酯对硫磷具有很高的反应性,2a,2b的醇羟基参与了对弱配位底物羧酸酯及磷酸酯的进攻,对不同电荷性质的羧酸酯底物具有较强的静电选择性。  相似文献

蛋白质酶解反应动力学行为的复杂性在于体系中底物与产物的多样性,以及由此决定的动力学常数的可变性.基于此,以酪蛋白(case in)—胰蛋白酶(trypsin)为模式体系,拟合求得动力学常数(Km和kcat)随水解度(DH)值变化的函数表达式,其规律为:随DH值增加,Km增大,kcat减小,kcat/Km减小,证明:酶与底物的亲和力随肽链缩短而减小,即高分子量多肽为蛋白酶的适宜底物,而酶解效率与酶解专一性随反应进行逐渐降低.进一步,根据分子水平的蛋白质酶解作用机制,关联水解实验数据,得到case in-trypsin酶解反应的经验修饰动力学方程(模型平均相对误差<5%),为定量表征复杂酶解历程以及高效制备活性多肽提供了理论基础.  相似文献

The catalytic hydrolysis of bis(4-nitrophenyl)phosphate (BNPP) by lanthanum(Ⅲ) ion in the presence of amino-alcoholic ligands: diethanolamine (DEA) and triethanolamine (TEA), was investigated kinetically at 30 ℃. The results indicated that the dinuclear dihydroxo complexes formed by lanthanum(Ⅲ) ion with aminoalcoholic ligands might be the catalytically active species which catalyze the hydrolysis of BNPP to different extents and the catalytic mechanism was believed to involve the synergism of double Lewis acid activation of the substrate and an intramolecular nucleophilic attack of a bridging oxo ligand.  相似文献
