应用自主研发的具有分离-富集模式的制备色谱工厂,建立了分离制备五味子木脂素有效部位及其单体化合物的方法。该方法首先以C18(250 mm×4.6 mm,5 μm)为色谱柱,水和甲醇为流动相,分离度和保留时间为考察指标,采集4个不同梯度的色谱信息,通过XTool色谱专家系统软件模拟,确定五味子木脂素第一、二维色谱条件;然后采用线性放大的方法,以C18(250 mm×30 mm,10 μm)为第一、二维分离柱,C18(80 mm×30 mm,10 μm)为富集柱,水为富集稀释液,对五味子木脂素进行二维色谱分离纯化;最后第一维分离得到9个可重复组分,第二维分离得到20个高纯度化合物,其中有6个单体化合物。结果表明该法重现性良好,可以实现五味子木脂素的系统性分离,对五味子化学成分的研究具有重要意义。  相似文献

以化妆品中23种防腐剂检测方法为例,探讨色谱柱选择对液相色谱方法测定结果的影响。参照《化妆品安全技术规范》甲基异噻唑啉酮等23个组分的检验方法,在2台不同的高效液相色谱仪上用15款不同品牌、型号的C18色谱柱检测23种防腐剂,计算色谱峰的理论塔板数和分离度,对23种组分的分离效果进行分析,并应用USP (United States Pharmacopeia)数据库和PQRI (Product Quality Research Institute)数据库等2种等效色谱柱选择方法,对不同色谱柱的分离效果及等效性进行评价和预测。实验结果表明,15款色谱柱对23种防腐剂的分离效果差异显著,仅有2款色谱柱可以实现23种组分的完全分离。USP和PQRI数据库中2种等效色谱柱选择方法均无法预测出合适的等效色谱柱,对23种防腐剂的液相色谱分析参考价值均较小。色谱柱是影响23种防腐剂液相色谱法测定结果准确性的关键因素,有关实验室在应用该方法时,应考虑色谱柱选择性差异。化妆品基质复杂,如何在现有研究成果的基础上,开发色谱柱的筛选和预测评价体系,进而指导实际样品的分离是下一步研究的重点、难点。建议有关部门在制修订检测方法时,注重色谱柱的耐用性考察,完善系统适应性指标,细化色谱柱分类和增加描述信息,指导色谱柱的合理选择,从而规避由于色谱柱使用过程中选择依据缺失而导致测定结果不准确的风险。  相似文献

Employing isocratic and gradient-elution high-performance liquid chromatography (HPLC) a number of straight-chain fatty acid esters (decanoate, laurate, myristate, palmitate) of violaxanthin, auroxanthin, lutein, zeaxanthin, isozeaxanthin, and beta-cryptoxanthin, prepared by partial synthesis, have been separated on a C18 reversed-phase column. Several chromatographic conditions were developed that separated a mixture of di-fatty acid esters (dimyristate, myristate palmitate mixed ester, dipalmitate) of violaxanthin, auroxanthin, lutein, and zeaxanthin in a single chromatographic run. Hydroxycarotenoids such as lutein, zeaxanthin, and isozeaxanthin that are not easily separated by HPLC on C18 reversed-phase columns, can be readily separated after derivatization with fatty acids and chromatography of their esters. Chromatographic conditions for optimum separation of carotenoids from various classes are discussed.  相似文献

With a recently developed liquid chromatographic (LC) method, using a phosphate buffer, several unknown impurities present in dirithromycin samples were separated. In this paper, a reversed-phase liquid chromatography-tandem mass spectrometry method is described for the investigation of dirithromycin and related substances. The method employed uses a Zorbax Extend C18 column (250 mm x 4.6 mm I.D.), 5 microm, and a mobile phase consisting of acetonitrile, 2-propanol, water and ammonium acetate solution pH 8.5. Mass spectral data are acquired on an LCQ ion trap mass spectrometer equipped with an electrospray ion (ESI) source operated in the positive ion mode. The LCQ is ideally suited for the identification of related substances because it provides on-line LC/MS(n) capability, which allows efficient identification without time-consuming isolation and purification procedures. Using this method, the fragmentation behavior of dirithromycin and its related substances was studied and the unknown impurities occurring in commercial samples were investigated. In total the structures of nine impurities were elucidated, among which three were different analogues with a modification in the side chain on the oxazine ring. Two impurities showed a different alkyl group in position C13. In two impurities the desosamine sugar was involved with changes in the degrees of methylation of the amino group. One unknown impurity was identified as dirithromycin F and another unknown was characterized as dirithromycin N-oxide.  相似文献

Direct and indirect reversed-phase (RP) high-performance liquid chromatographic methods were developed for the separation of enantiomers of 18 unnatural beta-amino acids, including several beta-3-homo amino acids. The direct separations of the underivatized analytes were performed on chiral stationary phases (CSPs) containing macrocyclic glycopeptide antibiotic teicoplanin (Chirobiotic T column) and teicoplanin aglycone (Chirobiotic TAG column). The indirect method involved pre-column derivatization with a new chiral derivatizing agent (CDA), (S)-N-(4-nitrophenoxycarbonyl)phenylalanine methoxyethyl ester ((S)-NIFE), and subsequent separation of diastereomers on Discovery C18 and Hyperpep 300 C18 columns. The different methods were compared in systematic chromatographic examinations. The effects of organic modifier, mobile phase composition, pH and flow rate on the separation were investigated.  相似文献

构建了一种在线阵列式二维常规柱液相色谱系统,并将其应用于分离血浆中的完整蛋白质。该系统以1根强阴离子交换柱作为第一维分离柱,8根阵列式反相色谱柱作为第二维分离柱。强阴离子交换柱分离的馏分通过十通阀被依次转移到第二维预柱上并得到保留富集,随后第二维流动相通过分流器同时将预柱上的蛋白质反冲至分析柱上进行分离。二维之间以及第二维阵列色谱柱之间均相互独立,从而可以提高系统分离的通量和总峰容量。采用该系统对血浆中的蛋白质进行了完整蛋白质水平上的分离。该系统具有高通量和高分辨率的特点,为血浆样品中高丰度蛋白质的去除以及血浆样品的深入研究提供了一种有效的手段。  相似文献

A two-dimensional high-performance liquid chromatographic method for the determination of creatinine with direct serum injection without sample pretreatment has been developed. The column-switching technique allowed a switch from columns packed with internal surface reversed-phase (ISRP) material to columns of almost any other material, even if the eluents necessary in a particular case do not appear to be directly compatible. A Pinkerton ISRP column, which stands out because of its very good stability when loaded with undiluted serum samples, was used as precolumn. The creatinine-containing fraction was switched to a reversed-phase Shandon RP18 column and was focused there by alteration of the eluent from pH 6.5 to phosphoric acid-ion-pair reagent. The separation occurs via a pH gradient, with ultraviolet detection at 234 nm. This method stands out particularly for its good long-term stability, simple sample handling without pretreatment, high selectivity, a broad linearity (0.3-30 mg/dl creatinine), good reproducibility (inter-assay coefficient of variation less than 3%) and high recovery (97-100%) relative to values obtained with gas chromatography-mass spectrometry.  相似文献

Three commercially-available high-performance liquid chromatographic columns packed with restricted access media were evaluated for suitability in multi-residue direct injection analysis at the ng/ml level. The internal surface reversed-phase and shielded hydrophobic phase columns were not sufficiently retentive to separate all analytes from the tail of the matrix peak. Coelution of some of the analytes was also observed with these columns. The semi-permeable surface column was significantly more retentive and selective, providing good separation of analyte and matrix peaks. With this column, an analytical protocol requiring no organic solvents was developed for the assay of six sulfonamides at a detection limit of 25 ng/ml.  相似文献

The separation of n = 2-5 n-butyloligostyrenes has been illustrated by reversed-phase reversed-phase (RP-RP) coupled-column liquid chromatography. The coupled-column separation has been achieved by use of a C18 column with methanol as the mobile phase followed by a DiamondBond C18 column with acetonitrile (ACN) mobile phase. The DiamondBond C18 is a hybrid carbon clad zirconia (CCZ)-C18 stationary phase. Unlike a C18-carbon clad zirconia two-dimensional liquid chromatographic system, which is orthogonal, the C18 and DiamondBond C18 columns combination exhibit correlations based upon the molecular weight of n-butyloligostyrenes. Using an alternative strategy to two-dimensional liquid chromatography, the molecular weight dependence displayed by both the C18 column and DiamondBond C18 has been used to increase throughput or decrease analysis time in the analysis of the n-butyloligostyrenes. However, this is at the expense of a portion of the two-dimensional peak capacity displayed by the C18-carbon clad zirconia system.  相似文献

膜脂作为细胞质膜的主要组成部分,在生命活动中扮演着重要的作用,其涉及多种重要疾病的发生和发展过程。发展适用于膜脂分离分析的新型色谱材料对于其后续结构和生物学功能研究具有重要的意义。该文选用具有潜在生物相容性的离子液体溴化1-乙烯基-3-十二烷基咪唑(1-vinyl-3-dodecylimidazole bromide,VDI)为功能单体,通过一步法点击反应将其接枝到巯基功能化硅球表面,制备得到了新型溴化1-乙烯基-3-十二烷基咪唑硅胶键合固定相(Sil-VDI)。利用傅里叶变换红外光谱仪和热重分析仪对Sil-VDI固定相材料的结构进行表征,结果证明Sil-VDI色谱固定相已被成功制备。保留机制研究显示填充Sil-VDI色谱柱具有典型的反相/离子交换混合模式保留特性。基于此,采用不同疏水性物质烷基苯、多环芳烃、苯胺、苯衍生物和无机阴离子BrO3 - 、NO3 - 和IO3 - 为测试物,对所制备固定相的色谱性能进行了研究。结果表明,该固定相对4类疏水性物质和无机阴离子均有较好的分离选择性和良好的峰对称性。进一步研究了所制备的Sil-VDI色谱柱对鸡蛋黄磷脂和肺腺癌细胞提取膜脂的分离效果,结果显示Sil-VDI色谱柱对2种磷脂样品均显示出了良好的分离能力。该文所制备的Sil-VDI色谱固定相合成方法简便,具有良好的分离分析应用潜能,后续工作会进一步研究该固定相在生物样品中的分离分析性能。  相似文献

The performance of a monolithic silica capillary column coated with poly(octadecyl methacrylate) (ODM column) for the reversed-phase liquid chromatographic separation of some polar and non-polar compounds was studied, and the results were compared to those obtained by using a monolithic silica capillary column modified with octadecylsilyl-(N,N-diethylamino)silane (ODS column). Benzene and naphthalene derivatives, polycyclic aromatic hydrocarbons (PAHs), steroids, alkyl phthalates, and tocopherol homologues were used as test samples. In general, compounds with aromatic character, rigid and planar structures, and lower length-to-breadth ratios (more compacted structures) seem to have more preference for the polymer coated stationary phase (ODM). Compounds with acidic character have also a higher retention on ODM columns because of the presence of ester groups in the stationary phase. The polymer coated column allowed the separation of some PAHs, alkyl phthalates, steroids, and of beta- and gamma-tocopherol isomers which cannot be separated under the same conditions on ODS columns, while keeping similar column efficiency. These results allowed to suggest ODM columns as a good alternative to conventional ODS columns for reversed-phase liquid chromatography.  相似文献

The current method prescribed in official monographs for the purity control of vancomycin is inappropriate in that several components are not separated from each other and other components are coeluted with the main component vancomycin B. The method uses an ODS column at pH 3.2. In this study, several changes were introduced in order to improve the separation. The optimization of the separation method at low pH indicated that pH 1.7 was optimum and that the use of dioxane as organic modifier drastically improved the separation. These conditions were used to test a set of more than 40 reversed-phase columns for their selectivity towards vancomycin components. The selection of the most suitable columns was performed by means of principal component analysis. Most of these columns did not allow the separation of didechlorovancomycin from monodechlorovancomycin 1. It was found that neutral to slightly alkaline mobile phases allowed better separation. Further optimization of the separation method and a robustness study were performed by means of experimental design. This optimization indicated that pH 7.7 was optimum and gradient elution was also used to effect complete analysis. The final method uses a Kromasil column and the mobile phase comprises dioxane, water and ammonium formate solution pH 7.7. The separation of monodechlorovancomycin 2 and of some unknown impurities from the main component vancomycin B is described for the first time. The method shows good repeatability, linearity and sensitivity.  相似文献

 采用反相高效液相法测定苁蓉酒中松果菊苷的含量。该法能有效地排除复方中其他组分的干扰,使峰得到完全分离。条件:固定相为C18;流动相为乙腈 1%的乙酸水溶液(体积比为14∶86);柱温25℃;紫外检测波长为330nm。结果表明,在0 52μg~2 60μg内松果菊苷的进样量与其峰面积呈良好的线性关系;样品的加标回收率为99 7%。该方法快速、简便、结果稳定,重现性好,是一种有效的质量检验方法。  相似文献

Several methods for the separation of vitamins on HPLC columns were already validated in the last 20 years. However, most of the techniques focus on separating either fat- or water-soluble vitamins and only few methods are intended to separate lipophilic and hydrophilic vitamins simultaneously. A mixed-mode reversed-phase weak anion exchange (RP-WAX) stationary phase was developed in our laboratory in order to address such mixture of analytes with different chemical characteristics, which are difficult to separate on standard columns. The high versatility in usage of the RP-WAX chromatographic material allowed a baseline separation of ten vitamins within a single run, seven water-soluble and three fat-soluble, using three different chromatographic modes: some positively charged vitamins are eluted in ion exclusion and ion repulsion modes whereas the negatively charged molecules are eluted in the ion exchange mechanism. The non-charged molecules are eluted in a classical reversed-phase mode, regarding their polarities. The method was validated for the vitamin analysis in tablets, evaluating selectivity, robustness, linearity, accuracy, and precision. The validated method was finally employed for the analysis of the vitamin content of some commercially available supplement tablets.  相似文献

Improved and simplified reversed-phase liquid chromatographic conditions for the separation and simultaneous profiling of both steroidal glycoalkaloids and their aglycones, having solanidane- or spirosolane-type structures, are described. The most reproducible retention behavior for these ionizable compounds on C18 columns was achieved under isocratic and gradient elution conditions using acetonitrile in combination with triethylammonium phosphate buffer at pH 3.0, when basic functional groups of solutes and silanol groups on the silica are fully protonated minimizing ionic interactions. Gradient elution was the only feasible approach for the simultaneous separation of steroidal glycoalkaloids and their aglycones. A Zorbax SB C18 column, specially designed for low-pH separations, showed good performance in critical separations. The impurities of the commercial tomatine and tomatidine standards were studied and confirmed using mass spectrometric, liquid chromatographic and thin-layer chromatographic methods.  相似文献

建立了反相离子对色谱-直接电导检测六氟磷酸根(PF6-)离子液体阴离子的分析方法。用DiamonsilC18反相色谱柱为分离柱,以离子对试剂-柠檬酸-乙腈混合水溶液为流动相,考察了离子对试剂、乙腈含量、pH值及色谱柱温度对六氟磷酸根保留的影响,并讨论了相关保留机理。在优化的色谱条件下,即流动相为0.05 mmol/L氢氧化四丁铵-0.038 mmol/L柠檬酸-35%乙腈(pH 5.5),流速1.0 mL/min,色谱柱温度40℃时,PF6-与其它常见阴离子(F-、Cl-、Br-、NO3-、SO24-、BF4-)达到基线分离且保留时间在15 min内。方法检出限(S/N=3)为0.25 mg/L,标准曲线的线性范围为0.5~100.0 mg/L,峰面积和保留时间的相对标准偏差(n=5)分别为0.17%和0.15%。该法用于1-丁基-3-甲基咪唑六氟磷酸盐和1-丙基-2,3-二甲基咪唑六氟磷酸盐两种离子液体中PF6-的测定,加标回收率分别为99%和104%。该方法简单、准确、可靠,实用性好。  相似文献

建立了以多糖衍生物为手性固定相的高效液相色谱-串联质谱(HPLC-MS/MS)直接拆分氰戊菊酯对映体的方法。在反相液相色谱条件下,考察了手性固定相的种类、流动相组成、柱温、流速对氰戊菊酯4个立体异构体分离的影响。同时,利用热力学方法对氰戊菊酯的立体异构体与固定相之间的色谱保留和分离的热力学机理进行了探讨。结果表明:采用Lux Cellulose-3(纤维素-三(4-甲基苯甲酸酯))手性色谱柱,在以流动相为乙腈-水(5 mmol/L甲酸铵)=(55:45,V:V)流速0.4 mL/min,柱温30℃的条件下,可在14 mins内实现氰戊菊酯4个立体异构体的基线分离。拓展了HPLC-MS/MS在菊酯类手性农药对映体分离及检测上的应用。  相似文献

Capillary columns were packed with 3 microns C18 stationary phase, interfaced with an ultraviolet-laser based thermo-optical absorbance detector, and evaluated for separation of a mixture of phenylthiohydantoin-amino acids. These columns demonstrated consistent performance with a relative standard deviation (RSD) for migration time of less than 1.5% and a separation efficiency of 216,000 plates/m for the electroosmotic flow marker, thiourea. The thermo-optical absorbance detector was based on a 248 nm krypton-fluoride excimer laser. Detection limits (3 sigma) ranged from 1.6 to 4.8 x 10(-7) M phenylthiohydantoin (PTH)-amino acid injected onto the column, which is a factor of three superior to those obtained in micellar electrokinetic chromatographic analysis of these compounds. A mixture of 17 PTH amino acids was injected onto the capillary; 13 components were nearly baseline resolved in 14 min.  相似文献

The performance of isocratic separations of 11 pollutant phenols (PP) using monolithic (Chromolith RP-18e) and conventional reversed-phase 5 microm (Luna and Purospher C18) and 4 microm (Synergi C12) particulate size columns, selected from high purity silica materials, has been compared. The separations have been optimized based on a previously optimized separation in which a reversed-phase C18 Luna column and acetonitrile as organic modifier were used, allowing the separation of all phenols tested in 23 min. The optimization process was carried out for each column by studying the effect of the mobile phase (acetonitrile as organic modifier, pH, flow-rate) on phenols separation. Under the optimized separation conditions, all phenols were separated in less than 23 min for all columns tested. Asymmetry factors were further evaluated and used to estimate column efficiency using the Dorsey-Foley equation. The efficiency and asymmetry factors were lower for Chromolith than for Purospher and Luna columns respectively. The Chromolith column was finally selected, due to its lower flow resistance, analysis time and good efficiency and asymmetry factors. The PPs separation was achieved in 3 min. The asymmetry factors were in the range 0.9-1.5 using 50mM acetate buffer (pH = 5.25)-ACN (64:36, v/v) as mobile phase, T=45 degrees C and 4.0 ml min(-1) flow-rate.  相似文献
