A simple, accurate and reproducible capillary electrophoresis method with UV detection has been developed for the simultaneous determination of four iridoid glycosides, 6-O-methyl-catalpol, aucubin, harpagide, and harpagoside, in Scrophularia ningpoensis (Xuan-shen). The running buffer was 100 mM borate (pH 9.3) containing 20% methanol. Applied voltage was 20 kV and temperature was 25 °C. Diphylloside A was used as an internal standard (IS) and detection was at 200 nm. The effects on separation of buffer pH, buffer concentration, and organic modifiers were investigated. The extracts of S. ningpoensis were well separated within 45 min.

In this paper, the pressurized capillary electrochromatography (pCEC) with UV detection was utilized for the separation and determination of three structurally related estrogens, such as diethylstilbestrol (DES), hexestrol (HEX) and dienestrol (DE), which were difficult to be separated by capillary electrophoresis (CE) and HPLC due to their similarity in the structure and charge-to-mass ratios. Experiments were carried out in a commercially available pCEC instrument using a capillary column packed with 3 microm octadecyl silica (ODS). Surfactant sodium dodecyl sulfate (SDS) was introduced in the mobile phase to enhance the speed of analysis. The effective factors on the retention time and separation resolution, such as the applied voltage, supplementary pressure, the pH and the concentration of the buffer solution, the concentration of SDS, and the content of acetonitrile in the mobile phase, were evaluated. Based on the investigation, 31% (v/v) acetonitrile and 69% (v/v) of 10 mmol/L phosphate buffer (pH 6.5) containing 1.0 mmol/L SDS at an applied voltage of -12 kV and a supplementary pressure of 1000 psi were found to be the optimal conditions for pCEC to separate the three estrogens. The method also had been applied to the analysis of fish muscle samples spiked with estrogens.  相似文献

A novel and efficient stability-indicating, reverse phase ultra-performance liquid chromatographic (UPLC®) analytical method was developed and validated for the determination of hexoprenaline in an injectable dosage form. The development of the method was performed using analytical quality by design (AQbD) principles, which are aligned with the future requirements from the regulatory agencies using AQbD principles. The method was developed by assessing the impact of ion pairing, the chromatographic column, pH and gradient elution. The development was achieved with a Waters Acquity HSS T3 (50 × 2.1 mm i.d., 1.8 µm) column at ambient temperature, using sodium dihydrogen phosphate 5 mM + octane-1-sulphonic acid sodium salt 10 mM buffer pH 3.0 (Solution A) and acetonitrile (Solution B) as mobile phases in gradient elution (t = 0 min, 5% B; t = 1 min, 5% B; t = 5 min, 50% B; t = 7 min, 5% B; t = 10 min, 5% B) at a flow rate of 0.5 mL/min and UV detection of 280 nm. The linearity was proven for hexoprenaline over a concentration range of 3.50–6.50 µg/mL (R2 = 0.9998). Forced degradation studies were performed by subjecting the samples to hydrolytic (acid and base), oxidative, and thermal stress conditions. Standard solution stability was also performed. The proposed validated method was successfully used for the quantitative analysis of bulk, stability and injectable dosage form samples of the desired drug product. Using the AQbD principles, it is possible to generate methodologies with enhanced knowledge, which can eventually lead to a reduced regulatory risk, high quality data and lower operational costs.  相似文献

以十二烷基硫酸钠(SDS)胶束为准固定相,考察了头孢哌酮、头孢哌酮S-异构体、头孢哌酮杂质A及其他未知杂质在胶束电动毛细管色谱(MECC)分离模式下的分离行为。研究了运行缓冲液的pH值、磷酸盐浓度、SDS浓度、甲醇体积分数、分离电压、分离温度等因素对头孢哌酮、S-异构体、头孢哌酮杂质A及其他杂质的迁移时间、分离度以及可分离出的杂质个数的影响。结果发现,这些因素对头孢哌酮与诸杂质间的分离及检测有显著的影响,尤以pH值为最。它不仅影响它们的迁移时间和分离效率,还直接影响头孢哌酮及其杂质峰的检测。优化后的分离条件:运行缓冲液为70 mmol/L磷酸盐-100 mmol/L SDS (pH 6.5),分离电压为15 kV,分离温度为25 ℃。在此条件下,用非涂渍石英毛细管51.0 cm×75 μm(有效长度42.5 cm),压力进样5 kPa×5 s,在254 nm波长下进行检测,可分离出28个杂质,诸杂质彼此间及与头孢哌酮间可得到有效分离。并将该方法成功地用于测定注射用头孢哌酮钠的含量和有关物质,结果令人满意。  相似文献

A rapid and simple method has been developed and validated for the determination of m-nisoldipine and its five related substances using micellar electrokinetic capillary chromatography (MEKC). The key factors, including pH, buffer concentration and buffer additive, sodium dodecyl sulfate (SDS) concentration, applied voltage and temperature were systematically investigated in an uncoated fused silica capillary. The total analysis time was less than 9 min. The forced degradation products of m-nisoldipine, which were induced by hydrogen peroxide, acidic and basic conditions, heat and light, were also tested.

建立了基于毛细管电泳技术的新疆紫草指纹图谱的质量控制方法.优化后的电泳条件:分离柱为50μm×40 cm未涂层毛细管柱,运行缓冲液为pH 8.0、100 mmol/L硼酸盐缓冲液(含25 mmol/L SDS及20%(V/V)无水乙醇),进样量0.5 psi×5 s,分离电压25 kV,检测波长214 nm.应用此条件,35 min内可实现新疆紫草有效成分的高效分离.在方法学验证的基础上,建立了新疆紫草指纹图谱.以新疆紫草对照药材指纹图谱为对照图谱,通过特征指纹峰、SFˊ相似度评价、聚类分析对不同购买地的新疆紫草进行质量评价和鉴别.此研究结果与其它中药鉴定方法对照结果一致.本方法准确、可靠、用时短,且具有良好的重现性,为新疆紫草的质量控制与评价提供了一种新的快速有效的鉴别方法.  相似文献

A CE method was developed and validated for the assessment of the chiral purity of the drug tenofovir applying a quality by design approach. Following selection of a quaternary ammonium β-CD as chiral selector, a fractional factorial resolution V+ design was employed for identification of the critical process parameters, while a central composite design served for method optimization. The final method used a 40/50.2 cm, 50 μm id fused-silica capillary, a BGE composed of a 100 mM sodium phosphate buffer, pH 6.4, containing 45 mg/mL quaternary ammonium β-CD, an applied voltage of 18 kV, and a capillary temperature of 22°C. Robustness was assessed by a Plackett–Burman design. The method was validated according to guideline Q2(R1) of the International Council for Harmonization of Technical Requirements for Pharmaceuticals for Human Use and enabled the determination of the (S)-enantiomer of tenofovir at the 0.1% level.  相似文献

The operating parameters that affect the performance of the online preconcentration technique “analyte focusing by micelle collapse‐MEKC (AFMC‐MEKC)” were examined using a multivariate approach involving experimental design to determine the sunscreen agents in cosmetics. Compared to the single‐variable approach, the advantage of the multivariate approach was that many factors could be investigated simultaneously to obtain the best separation condition. A fractional factorial design was used to identify the fewest significant factors in the central composite design (cCD). The cCD was adopted for evaluating the location of the minimum or maximum response in this study. The influences of the experimental variables on the response were investigated by applying a chromatographic exponential function. The optimized condition and the relationship between the experimental variables were acquired using the JMP software. The ANOVA analysis indicated that the Tris pH value, SDS concentration, and ethanol percentage influenced the separation quality and significantly contributed to the model. The optimized condition of the running buffer was 10 mM Tris buffer (pH 9.5) containing 60 mM SDS, 7 mM γ‐CD, and 20% v/v ethanol. The sample was prepared in 100 mM Tris buffer (pH 9.0) containing 7.5 mM SDS and 20% v/v ethanol. The SDS concentration in the sample matrix was slightly greater than the CMC value that makes the micelle be easily collapsed and the analytes be accumulated in the capillary. In addition, sunscreen agents in cosmetics after 1000‐fold dilution were successfully determined by AFMC‐MEKC.  相似文献

A simple, accurate, precise, rapid and sensitive stability-indicating capillary electrophoresis (CE) method was optimized and validated for the simultaneous determination of norfloxacin and its inactive decarboxylated degradant in pharmaceuticals. The univariant method was used to optimize electrophoretic factors including injection time, separation voltage and column temperature. Electrolyte concentration and pH were optimized using the factorial design and response surface methods. The optimum conditions obtained were: 10 mmol l− 1 phosphate at pH 2.5, hydrodynamic injection time of 8 s at pressure 0.5 p.s.i., separation voltage 25 kV and column temperature 25 °C. The separation was carried out into a fused-silica capillary column (31.2 cm length × 50 μm i.d.) with detection at 301 and 285 nm for the intact drug and the degradant, respectively using a diode array detector. For both analytes, the method enjoys wide dynamic range (1-50 μg ml− 1) with good detectability (limits of detection 0.11 μg ml− 1). In addition, acceptable accuracy (recovery > 95%); and good repeatability and intermediate precision (RSD < 3.5%) were obtained.  相似文献

A novel and fully automated sequential injection analysis manifold coupled to a capillary electrophoresis apparatus with amperometric detection, is described. The sequential injection manifold was isolated from the high voltage by inserting an air plug into the circuit. Small buffer reservoirs were used to avoid the need to pump fresh buffer to the interface during the electrophoretic separation. No decoupling device was used to mitigate the interference from the high voltage electric field, instead the potential shift induced by the separation voltage, was accounted for. The new hydrodynamic injection method presented is based on the overpressure created in the circuit when a pinch valve is closed for a predetermined time. The injection method yields RSD values of peak height and area below 2.55 and 1.82%, respectively, at different durations of valve closure (n = 5). The capillary and working electrode alignment was achieved by adapting a commercial available capillary union. When the electrode was replaced, the alignment method proved to be very reliable, yielding RSD values of peak height and area lower than 2.64 and 2.08%, respectively (n = 8). Using this system with a gold microelectrode, dopamine, and epinephrine could be quantified within the concentration range of 1–500 μM and detected at a concentration of 0.3 μM. The methods here presented could be applied for the development of new capillary electrophoresis systems with amperometric detection and/or to the design of fully automated systems for online process monitoring purposes.  相似文献

An efficient micellar electrokinetic capillary chromatography method was developed to analyze three major active components including morroniside, loganin and gallic acid in Fructus corni and its six preparations for the first time. The factors that could affect the separation were studied, such as the pH of the buffer, concentrations of SDS, organic modifier and beta-CD, and the applied voltage. The optimum analysis conditions were 10 mmol/L NaH(2)PO(4)-5 mmol/L Na(2)B(4)O(7) (pH 6.8) buffer containing 140 mmol/L SDS, 1 mmol/L beta-CD, 5% (v/v) methanol and 12.5 kV applied voltage. The linearity between the peak-areas and the concentrations of the analytes were investigated, and they exhibit excellent linear behavior over the concentration ranges (correlation coefficients 0.9953-0.9995). In addition, the pK(a) of gallic acid was determined using capillary zone electrophoresis. The result was consistent with that reported by the literature.  相似文献

建立了测定红曲米中莫纳可林K含量的胶束电动毛细管色谱(MEKC)方法。考察了运行缓冲液的种类、pH及其浓度、有机添加剂、十二烷基硫酸钠(SDS)的浓度和分离电压等实验条件对电泳分离效果及检测灵敏度的影响。在优化的实验条件下,以20 mmol/L硼砂(pH 10.6,含10%(体积分数)乙醇和40 mmol/L SDS)作为缓冲溶液,莫纳可林K能在23 min内实现很好的基线分离,线性范围为5.00~100.00 mg/L,线性相关系数为0.9976,检出限(以信噪比(S/N)为3计)为0.13 mg/L,加标回收率为98.5%~99.5%。精密度和稳定性试验中,峰面积和迁移时间的相对标准偏差均小于3%,表明重复性良好。该方法简便、快速、灵敏,可用于红曲米中莫纳可林K含量的测定。  相似文献

建立了匹伐他汀钙对映体的毛细管区带电泳(CZE)拆分方法。分别考察了电泳电压,缓冲溶液种类、浓度及pH值,环糊精种类及浓度,添加剂种类及浓度等参数对实验结果的影响,从而确定了匹伐他汀钙对映体的最佳拆分条件: 电泳电压为18 kV;运行缓冲溶液为80 mmol/L的Tris-HCl缓冲体系,pH值为3.20,其中含有50 mmol/L HP-β-CD(羟丙基-β-环糊精)和5 mmol/L SDS(十二烷基磺酸钠);采用重力进样,进样高度17 cm,进样时间为2 s。在优化的实验条件下,匹伐他汀钙对映体得到了较好的分离,分离度可达2.17。实验结果表明该方法可用于匹伐他汀钙对映体的分离,具有快速、便捷、准确性好等优点。  相似文献

A practical chiral CE method, using sulfated‐β‐CD as chiral selector, was developed for the enantioseparation of glycopyrrolate containing two chiral centers. Several parameters affecting the separation were studied, including the nature and concentration of the chiral selectors, BGE pH, buffer type and concentration, separation voltage, and temperature. The separation was carried out in an uncoated fused‐silica capillary of (effective length 40 cm) × 50 μm id with a separation voltage of 20 kV using 30 mM sodium phosphate buffer (pH 7.0, adjusted with 1 M sodium hydroxide) containing 2.0% w/v sulfated‐β‐CD at 25°C. Finally, the method for determining the enantiomeric impurities of RS‐glycopyrrolate was proposed. The method was further validated with respect to its specificity, linearity range, accuracy and precision, LODs, and quantification in the expected range of occurrence for the isomeric impurities (0.1%).  相似文献

The contents of five pharmacologically active flavone and xanthone glycosides, namely, swertianolin, swertisin, isoorientin, mangiferin, and 7-O-[α-L-rhamnopyranosyl-(1 → 2)-β-D-xylopyranosyl]-1,8-dihydroxy-3-methoxyxanthone, extracted from Tibetan folk medicinal species Swertia mussotii and S. franchetiana were determined by capillary electrophoresis with diode-array detection. The separation of five components has been optimized with a capillary column with a total length of 48.5 cm and effective length of 40 cm (50 μm i.d). The influence of the running buffer, the sodium dodecyl sulfonate (SDS) concentration, organic modifier, etc. on the resolution was evaluated. The background electrolyte contained 30 mM borate buffer, 28 mM SDS, 1.0% (v/v) acetonitrile, and was adjusted to pH 9.0 with 0.1 M NaOH. A good baseline resolution was obtained for the separation of five components within 5 min with the working voltage of 24 kV and a column temperature of 25°C. The established method was rapid and reproducible for the separation and determination of five flavone and xanthone glycosides from the extracts of S. mussotii and franchetiana plant samples. Microchip capillary electrophoresis in mixed hydro-organic solvent combined with laser-induced fluorescence detection was developed for the separation and detection of physcion and rhein in rhubarb. In contrast to the conventional capillary electrophoresis method, ammonium acetate-dimethyl sulfoxide was used as the basic buffer system in this method. The effects of background buffer, buffer apparent pH*, buffer concentration, water ratio, sample preparation method, and separation voltage on separation and detection were investigated. Optimized separation and detection conditions were obtained: the buffer consisted of 20 mmol/L of ammonium acetate in hydro-organic solvent composed dimethyl sulfoxide, formamide, and water mixed at 60/20/20 (v/v/v) ratio. The separation voltage was 1.9 kV. Under these conditions, the physcion, rhein, and other components of rhubarb can be completely separated within 150 s. Under the methodological verification, good linearity (R ≥ 0.9995) for physcion and rhein, and low limits of detection (0.085 μg·mL−1 and 0.077 μg·mL−1, respectively), satisfactory peak area precisions, migration time precisions (1.74%–3.09%), and accuracy (recovery rate 97.8% and 101.4%) were achieved. It is shown that the proposed method is simple, efficient, fast, sensitive, simple instrument, consumes few samples, has low operating cost, and is linear.  相似文献

A capillary electrophoretic (CE) method with β-cyclodextrin (CD) as modifier has been developed to enable separation, for the first time, of bioactive pentacyclic triterpene acids from the fruits of Rubus chingii. The effects of conditions such as the concentration of the running buffer, amounts of added β-cyclodextrin and organic modifier, applied voltage, and column temperature were systematically investigated to optimize the separation conditions. Baseline separation was achieved for the seven triterpenes by use of background electrolyte consisting of 200 mmol L−1 disodium tetraborate, 15 mmol L−1 β-cyclodextrin, and 12.5% (v/v) methanol. Binding constants between β-cyclodextrin and the triterpenes in the capillary electrophoresis buffer were calculated from the order of migration to elucidate the separation mechanism. 为防止消毒剂与抗抑菌产品中违规添加核苷类抗病毒药物,建立了胶束电动毛细管色谱(MEKC)同时分离测定消毒剂与抗抑菌产品中3种核苷类抗病毒药物(更昔洛韦、阿昔洛韦和喷昔洛韦)的新方法。通过3因素7水平均匀试验设计和4因素4水平的正交试验设计优化了十二烷基硫酸钠(SDS)、磷酸二氢钠(NaH2PO4)及硼砂(Na2B4O7)在分离缓冲液中的浓度,最大限度地实现了3种核苷类抗病毒药物在短时间内的有效分离。以30.2 cm×50 μ m(有效长度为20 cm)未涂层熔融石英毛细管为分离柱,以25 mmol/L NaH2PO4+10 mmol/L Na2B4O7(pH 7.41)+140 mmol/L(SDS)为分离缓冲溶液,分离电压为10 kV;进样压力为0.003 Pa,进样时间为4 s;检测波长为250 nm。3种抗病毒药物在一定的浓度范围内线性关系良好,相关系数r均不小于0.9995。3种抗病毒药物的检出限均为2.0 mg/kg,定量限均为7.0 mg/kg。低、中、高浓度加标回收率为85.4%~104.1%,相对标准偏差(RSD)均小于8.0%。该方法简便,快速,适于检测消毒剂与抗抑菌产品中更昔洛韦、阿昔洛韦和喷昔洛韦。  相似文献
