Lactosucrose is a kind of trisaccharide that functions as a significant prebiotic in the maintenance of gastrointestinal homeostasis for human. In this study, a levansucrase from Brenneria goodwinii was further used for the lactosucrose production. The recombinant levansucrase showed efficiency in the lactosucrose production by transfructosylation from sucrose and lactose, and no other oligosaccharide or polysaccharide was detected in the reaction mixture. The transfructosylation product by this recombinant enzyme was structurally determined to be lactosucrose by FT-IR and NMR. The production condition was optimized as pH at 6.0, temperature at 35 °C, 5 U mL?1 enzyme, 180 g L?1 sucrose, and 180 g L?1 lactose. Under the optimal condition, the enzyme could approximately produce 100 g L?1 lactosucrose when the reaction reached equilibrium. The recombinant levansucrase could effectively and exclusively catalyze the formation of lactosucrose, which might expand the enzymatic choice for further preparation of lactosucrose.  相似文献

Fructooligosaccharides (FOS) and levan attract much attention due to a wide range of applications in food technology and pharmaceutical and cosmetic industry. Bacillus licheniformis ANT 179, isolated from Antarctica soil, produced levansucrase and levan in a medium containing sucrose as carbon substrate. In this study, characterization of levansucrase and production of short-chain FOS and levan were investigated. Temperature and pH optimum of the enzyme were found to be 60 °C and pH 6.0, respectively. The optimization of fermentation conditions for levan production using sugarcane juice by response surface methodology (RSM) was carried out. Central composite rotatable design was used to study the main and the interactive effects of medium components: sugarcane juice and casein peptone concentration on levan production by the bacterium. The optimized medium with sugarcane juice at 20 % (v/v) and casein peptone at 2 % (w/v) was found to be optimal at an initial pH of 7.0 and incubation temperature of 35 °C for 48 h. Under these conditions, the maximum levan concentration was 50.25 g/L on wet weight basis and 16.35 g/L on dry weight basis. The produced inulin type FOS (kestose and neokestose) and levan were characterized by Fourier transform infrared spectroscopy (FT-IR) and nuclear magnetic resonance (NMR) analysis. The study revealed that the levansucrase could form FOS from sucrose. The locally available low-cost substrate such as sugarcane juice in the form of a renewable substrate is proposed to be suitable even for scale-up production of enzyme and FOS for industrial applications. The levan and FOS synthesized by the bacterium are suitable for food applications and biomedical uses as the bacterium has GRAS status and devoid of endotoxin as compared to other Gram-negative bacteria.  相似文献

Yeasts are good candidates to utilize the hydrolysates of lignocellulose, the most abundant bioresource, for bioproducts. This study aimed to evaluate the efficiencies of single-cell protein (SCP) and xylitol production by a novel yeast strain, Candida intermedia FL023, from lignocellulosic hydrolysates and xylose. This strain efficiently assimilated hexose, pentose, and cellubiose for cell mass production with the crude protein content of 484.2 g kg?1 dry cell mass. SCP was produced by strain FL023 using corncob hydrolysate and urea as the carbon and nitrogen sources with the dry cell mass productivity 0.86 g L?1 h?1 and the yield of 0.40 g g?1 sugar. SCP was also produced using NaOH-pretreated Miscanthus sinensis straw and corn steep liquor as the carbon and nitrogen sources through simultaneous saccharification and fermentation with the dry cell productivity of 0.23 g L?1 h?1 and yield of 0.17 g g?1 straw. C. intermedia FL023 was tolerant to 0.5 g L?1 furfural, acetic acid, and syringaldehyde in xylitol fermentation and produced 45.7 g L?1 xylitol from xylose with the productivity of 0.38 g L?1 h?1 and the yield of 0.57 g g?1 xylose. This study provides feasible methods for feed and food additive production from the abundant lignocellulosic bioresources.  相似文献

Isochrysis is a genus of marine algae without cell wall and capable of accumulating lipids. In this study, the lipid production potential of Isochrysis was assessed by comparing 15 Isochrysis strains with respect to their growth rate, lipid production, and fatty acid profiles. Three best strains were selected (lipid productivity, 103.0~121.7 mg L?1 day?1) and their lipid-producing capacities were further examined under different controlled parameters, e.g., growth phase, medium nutrient, and light intensity in laboratory cultures. Furthermore, the three Isochrysis strains were monitored in outdoor panel photobioreactors with various initial cell densities and optical paths, and the strain CS177 demonstrated the superior potential for outdoor cultivation. A two-stage semi-continuous strategy for CS177 was subsequently developed, where high productivities of biomass (1.1 g L?1 day?1) and lipid (0.35 g L?1 day?1) were achieved. This is a comprehensive study to evaluate the lipid-producing capability of Isochrysis strains under both indoor and outdoor conditions. Results of the present work lay a solid foundation for the physiological and biochemical responses of Isochrysis to various conditions, shedding light on the future utilization of this cell wall-lacking marine alga for biofuel production.  相似文献

l-ascorbyl palmitate (ASP) is an oil-soluble derivative of ascorbic acid which is used extensively in food, cosmetics industry, and medical hygiene. Enzymatic synthesis of ascorbyl palmitate in tert-butyl alcohol was carried out using indigenously immobilized lipase preparation PyCal with ascorbic acid and palmitic acid as starting material. The developed batch process under optimized reaction conditions resulted in conversion of 90% with relatively shorter reaction time of 6 h. Continuous process in packed bed reactor gave conversion of 50% with space time yield of 15.46 g/L/h which was found to be higher than the reported literature on enzymatic synthesis of ascorbyl palmitate. The immobilized lipase used in the present work showed good reusability. Characterization of formed ascorbyl palmitate was carried out by FTIR, MS/MS, H1-NMR, and C13-NMR. The enzymatic process resulted in selective synthesis of 6-O-l-ascorbyl palmitate with purity of 98.6% and no side product formation. The use of underivatized starting materials, high space time yield of 15.46 g L?1 h?1, high recyclability of catalyst, and no by-product formation make the overall process highly efficient and clean in terms of energy consumption and waste generation, respectively. The optimized reaction parameters for ascorbyl palmitate synthesis in the present study can be used as a useful reference for industrial synthesis of fatty acid esters of ascorbic acid by enzymatic route.  相似文献

Glutathione (γ-glutamyl-L-cysteinylglycine, GSH) is a pharmaceutical compound often used in food additives and the cosmetics industry. GSH can be produced biologically from L-glutamic acid, L-cysteine, and glycine through an enzymatic process traditionally involving two sequential adenosine triphosphate (ATP)-dependent reactions catalyzed by γ-glutamylcysteine synthetase (γ-GCS or GSHI, EC 6.3.2.2) and GSH synthetase (GS or GSHII, EC 6.3.2.3). Here, we report the enzymatic production of GSH by recombinant cell-free bifunctional γ-glutamylcysteine synthetase/glutathione synthetase (γ-GCS-GS or GshF) coupled with in vitro acetate kinase-based ATP generation. GSH production by an acetate kinase-integrated Escherichia coli Rosetta(DE3) mutant expressing Streptococcus thermophilus GshF reached 18.3 ± 0.1 g l?1 (59.5 ± 0.3 mM) within 3 h, with a molar yield of 0.75 ± 0.00 mol mol?1 added cysteine and a productivity of 6.1 ± 0.0 g l?1 h?1. This is the highest GSH titer reported to date. This newly developed biocatalytic process offers a promising approach for meeting the industrial requirements for GSH production.  相似文献

With the problems related to chemical methods of pyruvic acid (PA) synthesis, a fast-growing interest has been observed in research aiming at reducing the production cost of PA by applying biotechnological methods. This study aimed to investigate the potential applicability of Yarrowia lipolytica Wratislavia 1.31 yeast strain for valorisation of pure and crude glycerol through the production of industrially desired PA. Conditions required for the effective PA biosynthesis, i.e., pH value, thiamine concentration, agitation, and substrate concentration, were examined in batch and fed-batch cultivation modes. The efficient production of PA occurred under the limitation of thiamine (1 µg L?1) and was stimulated by moderate pH (4.5) and agitation (800 rev min?1) of the culture. Under optimal conditions, Y. lipolytica Wratislavia 1.31 was able to produce 85.2 g L?1 of PA with volumetric productivity of 0.90 g L?1 h?1. The yield of PA biosynthesis reached a high level of 1.03 g g?1. Obtained results confirmed the aptitude of Y. lipolytica yeast to produce high amounts of PA from simple glycerol-containing media. Presented process was very promising and might be considered as an attractive alternative for currently used chemical methods of PA synthesis.  相似文献

This study aimed to evaluate the properties of xanthan gum produced by Xanthomonas campestris pv. campestris 1866 and 1867 from lignocellulosic agroindustrial wastes. XG was produced using an orbital shaker in a culture medium containing coconut shell (CS), cocoa husks (CH), or sucrose (S) minimally supplemented with urea and potassium. The XG production results varied between the CS, CH, and S means, and it was higher with the CH in strains 1866 (4.48 g L?1) and 1867 (3.89 g L?1). However, there was more apparent viscosity in the S gum (181.88 mPas) and the CS gum (112.06 mPas) for both 1866 and 1867, respectively. The ability of XGCS and XGCH to emulsify different vegetable oils was similar to the ability of XGS. All gums exhibited good thermal stability and marked groups in the elucidation of compounds and particles with rough surfaces.  相似文献

Recombinant Escherichia coli cells harboring nitrilase from Alcaligenes faecalis were immobilized using tris(hydroxymethyl)phosphine (THP) as the coupling agent. The optimal pH and temperature of the THP-immobilized cells were determined at pH 8.0 and 55 °C. The half-lives of THP-immobilized cells measured at 35, 40, and 50 °C were 1800, 965, and 163 h, respectively. The concentration of R-mandelic acid (R-MA) reached 358 mM after merely 1-h conversion by the immobilized cells with 500 mM R,S-mandelonitrile (R,S-MN), affording the highest productivity of 1307 g L?1 day?1 and the space-time productivity of 143.2 mmol L?1 h?1 g?1. The immobilized cells with granular shape were successfully recycled for 60 batches using 100 mM R,S-MN as substrate at 40 °C with 64% of relative activity, suggesting that the immobilized E. coli cells obtained in this study are promising for the production of R-MA.  相似文献

In this study, a method for the efficient production of dehydroepiandrosterone (DHEA) from phytosterols in a vegetable oil/aqueous two-phase system by Mycobacterium sp. was developed. After the 3-hydroxyl group of phytosterols was protected, they could be converted into DHEA with high yield and productivity by Mycobacterium sp. NRRL B-3683. In a shake flask biotransformation, 15.05 g l?1 of DHEA and a DHEA yield of 85.39% (mol mol?1) were attained after 7 days with an initial substrate concentration of 25 g l?1. When biotransformation was carried out in a 30-l stirred bioreactor with 25 g l?1 substrate, the DHEA concentration and yield was 16.33 g l?1 and 92.65% (mol mol?1) after 7 days, respectively. The results of this study suggest that inexpensive phytosterols could be utilized for the efficient production of DHEA.  相似文献

Pulcherrimin, a potential biocontrol agent produced by microorganisms, has the promising applications in the agricultural, medical, and food areas, and the low yield of pulcherrimin has hindered its applications. In this study, the red pigment produced by Bacillus licheniformis DW2 was identified as pulcherrimin through the spectrometry analysis and genetic manipulation, and the component of the medium used for pulcherrimin production was optimized. Based on our results, the addition of 1.0 g L?1 Tween 80 could improve the yield of pulcherrimin, and glucose and (NH4)2SO4 were served as the optimal carbon and nitrogen sources for pulcherrimin synthesis, respectively. Furthermore, an orthogonal array design was applied for optimization of the medium. Under optimized condition, the maximum yield of pulcherrimin was 331.17 mg L?1, 5.30-fold higher than that of the initial condition, which was the maximum yield reported for pulcherrimin production. Collectively, this study provided a promising strain and a feasible approach to achieve the high-level production of antimicrobial pulcherrimin.  相似文献

Enantiopure 2-hydroxy acids (2-HAs) are important intermediates for the synthesis of pharmaceuticals and fine chemicals. Deracemization of racemic 2-HAs into the corresponding single enantiomers represents an economical and highly efficient approach for synthesizing chiral 2-HAs in industry. In this work, a novel ketoacid reductase from Leuconostoc lactis (LlKAR) with higher activity and substrate tolerance towards aromatic α-ketoacids was discovered by genome mining, and then its enzymatic properties were characterized. Accordingly, an engineered Escherichia coli (HADH-LlKAR-GDH) co-expressing 2-hydroxyacid dehydrogenase, LlKAR, and glucose dehydrogenase was constructed for efficient deracemization of racemic 2-HAs. Most of the racemic 2-HAs were deracemized to their (R)-isomers at high yields and enantiomeric purity. In the case of racemic 2-chloromandelic acid, as much as 300 mM of substrate was completely transformed into the optically pure (R)-2-chloromandelic acid (>?99% enantiomeric excess) with a high productivity of 83.8 g L?1 day?1 without addition of exogenous cofactor, which make this novel whole-cell biocatalyst more promising and competitive in practical application.  相似文献

Since cultivations of Arthrospira platensis have a high water demand, it is necessary to develop treatment methods for reusing the exhausted medium that may prevent environmental problems and obtaining useful biomass. The exhausted Schlösser medium obtained from A. platensis batch cultivation in bench-scale mini-tanks was treated by varying concentrations of different coagulants, ferric chloride (6, 10, and 14 mg L?1) or ferric sulfate (15, 25, and 35 mg L?1) and powdered activated carbon (PAC, 30 and 50 mg L?1). Such treated effluent was restored with NaNO3 and reused in new cultivations of A. platensis performed in Erlenmeyer flasks. Reusing media through the cultivation of A. platensis showed satisfactory results, particularly in the medium treated with ferric chloride and PAC. The maximum cell concentration obtained in the flasks was 1093 mg L?1, which corresponded to the medium treated with ferric chloride (6 mg L?1) and PAC (30 mg L?1). This cellular growth was higher than in the medium treated with ferric sulfate and PAC, in which values of maximum cell concentration did not exceed 796 mg L?1. The cultures in the media after treatment did not modify the biomass composition. Thus, combined coagulation/adsorption processes, commonly used in water treatment processes, can be efficient and viable for treating exhausted medium of A. platensis, allowing the production of such biomass with the reduction of production cost and saving water.  相似文献

Bacillus licheniformis TISTR 1010 was used for glutamic acid-independent production of poly-γ-glutamic acid (γ-PGA). A fed-batch production strategy was developed involving feedings of glucose, citric acid, and ammonium chloride at specified stages of the fermentation. With the dissolved oxygen concentration controlled at ≥50% of air saturation and the pH controlled at ~7.4, the fed-batch operation at 37 °C afforded a peak γ-PGA concentration of 39.9 ± 0.3 g L?1 with a productivity of 0.926 ± 0.006 g L?1 h?1. The observed productivity was nearly threefold greater than previously reported for glutamic acid-independent production using the strain TISTR 1010. The molecular weight of γ-PGA was in the approximate range of 60 to 135 kDa.  相似文献

Numerous desulfurizing bacteria from the Rhodococcus genus harbor conserved dsz genes responsible for the degradation of sulfur compounds through 4S pathway. This study describes a newly identified desulfurizing bacterium, Rhodococcus sp. FUM94, which unlike previously identified strains encodes a truncated dsz operon. DNA sequencing revealed a frameshift mutation in the dszA gene, which led to an alteration of 66 amino acids and deletion of other C-terminal 66 amino acids. The resulting DszA polypeptide was shorter than DszA in Rhodococcus sp. IGTS8 reference strain. Despite the truncation, desulfurizing activity of the operon was observed and attributed to the removal of an overlap of dszA and dszB genes, and lack of active site in the altered region. Desulfurization experiments resulted in specific production rate of 6.3 mmol 2-hydroxy biphenyl (kgDCW)?1 h?1 at 2 g l?1 biocatalyst concentration and 68.8% biodesulfurization yield at 20 g l?1 biocatalyst concentration, both at 271 μM dibenzothiophene concentration which is comparable to similar wild-type biocatalysts.  相似文献

A new strain, Candida tropicalis UFMG BX 12-a, was found to produce higher yields of xylitol on poplar wood hemicellulose hydrolysate. The hemicellulose hydrolysate liquor was detoxified using a novel method we developed, involving vacuum evaporation and solvent separation of inhibitors which made the hydrolysate free of toxins while retaining high concentrations of fermentable sugars. The effect of the detoxification method on the fermentation was also reported and compared to well-known methods reported in literature. In this study, the new strain C. tropicalis UFMG BX 12-a was used on the detoxified hydrolysate to produce xylitol. It was also compared to Candida guilliermondii FTI 20037, which has been reported to be one of the best strains for fermentative production of xylitol. To further improve the efficiency of the fermentation process, these strains were immobilized in calcium alginate beads. The yield (0.92 g g?1) and productivity (0.88 g L?1 h?1) obtained by fermenting the wood hydrolysate detoxified by our new detoxification technique using an immobilized new Candida strain were found to be higher than the values reported in literature.  相似文献

The paper deals with the exploitation of Ipomoea carnea as a feedstock for the production of bioethanol. Dilute acid pretreatment under optimum conditions (3 %H2SO4, 120 °C for 45 min) produced 17.68 g L?1 sugars along with 1.02 g L?1 phenolics and 1.13 g L?1 furans. A combination of overliming and activated charcoal adsorption facilitated the removal of 91.9 % furans and 94.7 % phenolics from acid hydrolysate. The pretreated biomass was further treated with a mixture of sodium sulphite and sodium chlorite and, a maximum lignin removal of 81.6 % was achieved. The enzymatic saccharification of delignified biomass resulted in 79.4 % saccharification with a corresponding sugar yield of 753.21 mg g?1. Equal volume of enzymatic hydrolysate and acid hydrolysate were mixed and used for fermentation with a hybrid yeast strain RPRT90. Fermentation of mixed detoxified hydrolysate at 30 °C for 28 h produced ethanol with a yield of 0.461 g g?1. A comparable ethanol yield (0.414 g g?1) was achieved using a mixture of enzymatic hydrolysate and undetoxified acid hydrolysate. Thus, I. carnea biomass has been demonstrated to be a potential feedstock for bioethanol production, and the use of hybrid yeast may pave the way to produce bioethanol from this biomass.  相似文献

Due to be able to migrate or leach from food packaging materials into the foods and/or beverages, development of a new, sensitive and selective analytical methods for low levels of antimony as a food contaminant is of great importance in terms of food safety. In this context, an ultrasonic-assisted cloud point extraction method was developed for the preconcentration and determination of antimony as Sb(III) using 4-(2-thiazolylazo)resorcinol (TAR) and 2-(2-thiazolylazo)-p-cresol (TAC) as chelating agents and sodium dodecyl sulfate as signal enhancing agent at pH 6.0 and mediated by nonionic surfactant, t-octylphenoxypolyethoxyethanol by flame atomic absorption spectrometry. Using the optimized conditions, the calibration curves obtained from Sb(III) with TAR and TAC were linear in the concentration ranges of 0.5–180 and 1–180 μg L?1 with detection limits of 0.13 and 0.28 μg L?1, respectively. The precision (as relative standard deviations, RSDs) was lower than 3.9 % (25 and 100 μg L?1, n: 6). The method accuracy was validated by the analysis of two standard reference materials. The results obtained were statistically in a good agreement with the certified values at 95 % confidence limit. The method has successfully been applied to the determination of Sb(III) and total Sb in selected beverages, milk and fruit-flavored milk products before and after pre-reduction of Sb(V) to Sb(III) with a mixture of KI/ascorbic acid in acidic media. The Sb(V) contents of samples were quantitatively calculated from analytical difference between total Sb and Sb(III) levels.  相似文献

This study presents an easy and cost-effective flow-based cloud point extraction (CPE) method for determining partial amounts of two organophosphorus pesticides (phosalone and ethion) in seawater by HPLC–UV–Vis. In continues CPE methodology, the effect of the different column packing type such as carbon nanotube, polyacrylonitrile nanofiber and fiberglass on pesticide extraction was investigated. The Triton X-100 was utilized as nonionic surfactant, and moreover, effect of different parameters such as pH, temperature, extraction time, surfactant concentration, type and volume of the eluent solution on the extraction efficiency was optimized. Under optimum conditions, the figures of merit of the method for phosalone and ethion were obtained as: the enrichment factor (172 and 166), line range (0.8–300 and 0.5–300 µg L?1, R 2 = 0.9973 and 0.9982), relative standard deviation in concentration of 200 µg L?1 (%RSD = %5.4 and %7.99, N = 5) and limit of detection (LOD = 0.24 and 0.14 µg L?1). The suggested method was successfully used for determination of phosalone and ethion in Chabahar Bay seawaters with satisfactory results.  相似文献

Temperature control is a very important factor on triterpene productivity in submerged liquid fermentation. Temperature effects from 23 to 32 °C on triterpene production by Ganoderma lucidum G0119 were investigated in 6-L stirred fermentor. Logistic and Luedeking-Piret equations were used to estimate the mycelial growth and triterpene production kinetics by regression analysis. On that basis, a temperature-shifting fermentation control strategy was established. From 0 to 61 h, culturing was performed at 32 °C to get high specific mycelial growth rate. Between 62 and 127 h, the temperature was decreased stepwise from 31 to 30 °C to maintain high triterpene productivity. After 128 h, temperature was maintained at 29 °C to minimize triterpene production inhibition and sustain high productivity. Elevated triterpene level (0.269 g L?1), yield (0.0101 g g?1), and productivity (0.00207 g (L h)?1) were achieved representing 27.32, 13.94, and 37.11 % higher than submerged liquid fermentation at constant temperature of 29 °C, respectively, feasible for the industrial scale.  相似文献
