A simple, economical and very effective method is demonstrated for simultaneous determination of 2,4-dichlorophenol, 2,4,6-trichlorophenol, 2,3,4,6-tetrachlorophenol and pentachlorophenol, in aqueous samples, by using purge-assisted headspace solid-phase microextraction (PA/HS-SPME) coupled to gas chromatography-mass spectrometry (GC-MS). In the new method, purging the sample enhances the removal of the trace chlorophenols without derivatization from the matrices to the headspace. Extraction parameters including extraction temperature, purge gas flow rate and extraction time were systematically investigated. Under optimal conditions, the relative standard deviations (RSDs) were 4-11% at 50 pg/mL and 5-14% at 5 pg/mL, respectively. The recoveries were in the range of 83-114%. Detection limits were determined at the fg level. These results indicate that PA/HS-SPME provides a significant contribution to highly efficient extraction of semi-volatile CPs, especially for pentachlorophenol, which has the smallest Henry's constant and large octanol-water partitioning coefficient. In addition, the proposed method was successfully applied to the analysis of chlorophenols in landfill leachate. New perspectives are opened for headspace extraction of relatively low vapor pressure compounds in complex matrices.  相似文献

In order to determine steroid compounds in GC/MS an analytical method using pressurized fluid extraction (PFE) was developed. While extracting in-house reference material (coastal sediment) typical recovery in PFE ranged from 80 to 120% (+/-2.5-14.5) and the average extraction yield in PFE in comparison to conventional soxhlet extraction was 115%. In particular, the PFE showed higher extraction efficiency for C29 and dien sterols. Optimizing parameters such as temperature and pressure is critical in achieving this efficiency. Sterols in the sediment were derivatized with silyl reagent BSTFA in acetone for the final determination. A short column florisil cleanup offered the best separation of the GC/MS sensitive derivatives from co-contaminants. Thirty-three coastal sediment samples were analyzed using PFE and Soxhlet extraction methods. The results on extraction efficiency, silyl derivatization kinetics and purification efficiency demonstrated that PFE is far superior in extracting sterols from sediment samples. It is simple, fast, efficient and amenable for automation.  相似文献

A single-drop microextraction (SDME) method and gas chromatography with mass spectrometry detection have been developed for the determination of acidic herbicides in water. The analytes were extracted from a 3 mL sample solution using 4 microL of hexyl acetate. After extraction, derivatization was carried out inside a glass microvial (1.1mm i.d.) using pentafluorobenzyl bromide (PFBBr). Triethylamine (TEA) was used as the reaction catalyst. The influence of derivatization reagent volume, catalyst amount, derivatization time and temperature on the yield of the in-microvial derivatization was investigated. Derivatization reaction was performed using 0.3 microL of PFBBr and 0.4 microL of TEA (10%, v/v in toluene) at 100 degrees C during 5 min. Also, the effects of different experimental SDME parameters such as selection of organic solvent, sample pH, addition of salt, extraction time and temperature of extraction were studied. Analytical parameters such as enrichment factor, precision, linearity and detection limits were also determined. The enrichment factors were between 83 and 157. The limits of detection (LOD) were in the range 1.2-7 ng/L (S/N=3). The relative standard deviations obtained were below 10.1% (n=5).  相似文献

A novel sorbent, dihydroxylated polymethylmethacrylate (DHPMM), coated on hollow-fiber membrane, is used for the polymer-coated hollow-fiber microextraction of trace amounts of natural and synthetic estrogens, such as diethylstilbestrol, estrone, 17beta-estradiol and 17alpha-ethynylestradiol, in aqueous samples. In this procedure, estrogens were extracted using the functionalized polar DHPMM polymer with derivatization using N-methyl-N-(trimethylsilyl)trifluoroacetamide followed by gas chromatography-mass spectrometric analysis. The detection limits for estrogens in aqueous sample were between 0.03 and 0.8 ng l(-1) and the calibration curves were linear over the concentration range 0.05-10 microgl(-1) and had correlation coefficients of >0.994. The relative standard deviations (RSDs) were <15% (n = 3). This simple, accurate, sensitive and selective analytical method is applicable to the determination of trace amounts of estrogens in reservoir and potable water samples.  相似文献

Two GC-MS methods, based on the application of N,O-bis(trimethylsilyl)trifluoroacetamide-derivatization-GC-MS (selected-ion monitoring) and GC-MS-MS without derivatization, respectively, were optimised and applied to the determination of a group of five selected endocrine disrupting compounds (EDCs) in wastewaters. Both methods included solid-phase extraction with Oasis HLB cartridges allowing an enrichment factor for wastewater samples of 100-fold. The investigated EDCs were estrone, 17beta-estradiol, 17alpha-ethynylestradiol, 4-tert-octylphenol and bisphenol A. Results obtained from the validation studies yielded comparable results in both cases. Recoveries in spiked wastewaters at 50 ng/l were higher than 90% for all the compounds, except for 4-tert-octylphenol (75%). Repeatability and reproducibility were adequate, varying from 1.6 to 14%, except for estrone which reproducibility was 28% when the derivatization-GC-MS method was applied. Limits of detection calculated ranged from 2.5 to 27.5 ng/l with differences between both methods from 1.1 (estrone) to 10.4 (bisphenol A) times. Both methods were successfully applied to the analysis of the target compounds in sewage treatment plant influents and effluents. Traces of bisphenol A, 4-tert-octylphenol, estrone and 17beta-estradiol were detected at concentration levels ranging from 13.3 to 1105.2 ng/l.  相似文献

Large volume injection-programmable temperature vaporization-gas chromatography-mass spectrometry (LVI-PTV-GC-MS) was optimized for the determination of estrone (E1), 17β-estradiol (E2), 17α-ethynyl estradiol (EE2), mestranol (MeEE2) and estriol (E3) for their determination in environmental samples (estuarine water, wastewater, fish bile and fish homogenate) after derivatization with 25 μL (BSTFA+1% TMCS) and 125 μL of pyridine. Experimental designs such as Plackett-Burman (PBD) and central composite designs (CCDs) were used to optimize the LVI-PTV variables (cryo-focusing temperature, vent time, vent flow, vent pressure, injection volume, purge flow to split vent, splitless time and injection speed). Optimized conditions were as follows: 45 μL of n-hexane extract are injected at 60°C and 6 μL/s with a vent flow and a vent pressure of 50 mL/min and 7.7 psi, respectively, during 5 min; then the split valve is closed for 1.5 min and afterwards the injector is cleaned at 100 mL/min before the next injection. The method was applied to the determination of estrogenic compounds in environmental samples such as estuarine water, wastewater, and fish homogenate and bile. Limits of detection (0.04-0.15 ng/L for water samples, 0.04-0.67 ng/g for fish bile and 0.1-7.5 ng for fish homogenate) obtained were approx. ten times lower than those obtained by means of a common split/splitless inlet.  相似文献

A rapid and sensitive method based on transient ITP and field enhancement in CE with electrochemical detection at copper disk electrode was developed for the simultaneous separation and determination of three estrogens: estrone, 17β‐estradiol, and estriol. The effects of several important factors that influence the separation and detection were investigated. Under the optimum conditions, the estrogens could be separated in 0.06 mol/L sodium hydroxide solution within 14 min. With transient ITP by addition of 0.5% NaCl, a good linear response was obtained for three estrogens from 0.2 to 10 μmol/L, with correlation coefficients higher than 0.9993. The detection limits were 8.9 × 10?8, 6.7 × 10?8, and 1.1 × 10?7 mol/L (S/N = 3) for estriol, 17β‐estradiol, and estrone, respectively. This method was successfully employed to analyze different water samples from waterworks, tap water, fishpond, and river samples with recoveries in the range of 90.8–108.9%, and RSDs < 4.69%. The satisfied results demonstrated that this method was of convenient preparation, high sensitivity, and good repeatability, which could be applied to the rapid determination of environmental water samples.  相似文献

A novel method based on pyrolysis-capillary gas chromatography-mass spectrometry (CGC-MS) was developed for the quantitative analysis of polyvinylchloride (PVC) in solid environmental samples like sludge and dust. The samples are extracted and the extract is fractionated by solid-phase extraction (SPE). Possibly interfering biological and frequently occuring synthetic polymers are removed by this clean-up. The final extract is analyzed by pyrolysis-CGC-MS. Selective detection of PVC is performed by using specific markers in the pyrogram. Quantitation is done on naphthalene. Good linearity was obtained in a range from 0.5 to 100 microg applied to the pyrolyser. The limit of quantitation (LOQ) in sludge and dust samples is 10 mg/kg dry mass. A correlation between PVC and phthalates was made for sewage sludge samples.  相似文献

An inexpensive and effective sample preparation procedure for the determination of three ethylphenolic off-flavours (4-ethylphenol, 4-ethylguaiacol and 4-ethylcathecol) in wine samples is presented. Analytes were in situ acetylated and concentrated using a disposable silicone sorbent (DSS) exposed to the diluted sample. After that, the analytes were recovered with ethyl acetate and determined by gas chromatography with mass spectrometry. The influence of different parameters (volume of acetic anhydride, basic catalyst, ionic strength, sorbent format, sampling mode and extraction time) on the efficiency of derivatization and extraction steps is discussed. Under optimized conditions, 2 mL of wine were diluted with 15 mL of an aqueous solution of potassium bicarbonate (5%, m/v) in a 22 mL vessel, containing 2 g of sodium chloride. The volume of acetic anhydride and the extraction time were set at 90 μL and 2 h, and the extraction was carried out at room temperature (20±2°C). Analytes were concentrated using a silicone disc (5 mm diameter × 0.5 mm thickness) and further desorbed with 0.2 mL of ethyl acetate. The achieved limits of quantification (LOQs), defined as the concentration of each compound providing a signal 10 times higher than the baseline noise, stayed between 5 and 15 ng mL(-1). The method provided a linear response range of up to 5000 ng mL(-1) and relative recoveries from 91% to 116%. The 4-ethylphenol off-flavour was detected in most red wine samples at concentrations of up to 2700 ng mL(-1).  相似文献

An analytical procedure has been developed for determination of eight selected natural and synthetic hormonal steroids in surface water and in effluent samples. Several methodological points have been investigated and are discussed; they include the choice of the solid-phase extraction sorbent, the influence of flow rate on recovery, the breakthrough volume for a given sorbent (Env+ and Oasis HLB), sample clean up, and sample storage. As regards the latter point, it was found that when no preservative was added to effluent from a sewage-treatment plant, severe loss of steroids occurred—85% of progesterone and about 30% of both estrone and estradiol were found to be degraded in 24 h. The procedure developed was applied to samples from the Seine river estuary. Sex steroids were not detected in surface water; estrone was the most commonly detected steroid in sewage-treatment plant effluent, with levels ranging from 1.8 to 8.3 ng L–1. Synthetic estrogens (ethynylestradiol and mestranol) and progestagens (levonorgestrel and norethindrone) were never detected, whatever the sampling season. Overall, for 162 out of 168 measurements levels were below the detection limits of the developed procedure.  相似文献

Butylated hydroxytoluene (BHT) is an antioxidant utilized as additive in foods and packaging plastic. Its presence in drinking water is possible if it is used as an antioxidant in the packaging plastic because it may migrate into the package's contents. A method for the determination of BHT in water by means of solid-phase microextraction and gas chromatography-mass spectrometry has been developed and evaluated with respect to the time of fiber exposure, limits of detection and quantitation, linearity and precision. Finally, the method was applied to evaluate the presence of this substance in samples of mineral and mineralized bottled drinking water, and it appeared to be present in seven out of a total of fifteen commercial brands.  相似文献

Haloacetaldehydes (HAs) are becoming the most widespread disinfection by-products (DBPs) found in drinking water, besides trihalomethanes and haloacetic acids, generated by the interaction of chemical disinfectants with organic matter naturally present in water. Because of their high potential toxicity, HAs have currently received a singular attention, especially trichloroacetaldehyde (chloral hydrate, CH), the most common and abundant compound found in treated water. The aims of this study are focused on the miniaturisation of EPA Method 551.1, including some innovations such as the use of ethyl acetate as the extracting solvent, the enhancement of HAs stability in aqueous solutions by adjusting the pH ~3.2 and the use of a large-volume sample injection (30 μL) coupled to programmed temperature vaporizer-gas chromatography-mass spectrometry to improve both sensitivity and selectivity. In optimised experimental conditions, the limits of detection for the 7 HAs studied ranged from 6 to 20 ng/L. Swimming pools have recently been recognized as an important source of exposure to DBPs and as a result, in this research for the first time, HAs have been determined in this type of water. Two HAs have been found in the analysed water: CH at concentrations between 1.2-38 and 53-340 μg/L and dichloroacetaldehyde between 0.07-4.0 and 1.8-23 μg/L in tap and swimming pool waters, respectively.  相似文献

The use of estrogens, gestagens and androgens (EGAs) in animal fattening is prohibited in the European Community. Based on the general detection capabilities of Belgian laboratories, National Minimum Required Performance Limits (National MRPLs) for a number of EGAs have been imposed by the inspection services. Selective hyphenated techniques, e.g. GC–MS and GC–MS2, with high detection capability are needed. β-Trenbolone, which is meant to be a “problem” molecule for GC–MS, can be detected at the 2 μg/kg level using GC–MS2. Based on the National MRPLs in different matrices, our laboratory has divided the EGAs into a class system. In this set-up, analysis of EGAs in kidney fat and meat is discussed.  相似文献

Analysis of exhaled air is of particular interest as an indicator of health as well as a tool for the diagnosis of diseases. It is also a very attractive procedure for the biological control of the exposition to hazardous solvents. This kind of analysis presents numerous advantages over other methods, the most important being that it is not an invasive procedure and, therefore, it is well accepted and can be applied to a wide range of compounds. Furthermore, the analysis is simplified since the matrix is less complex that in the case of blood or urine. In spite of these obvious advantages and the good results obtained, analysis of exhaled air is not in daily use, probably due to the fact that there are no normalized systems of sampling, thus making the interpretation of the results difficult. In this paper, a method for the determination of tetrachloroethylene in exhaled air using solid-phase microextraction is presented. This method, which can be applied to other volatile organic compounds, was developed with special emphasis of end-exhaled breath sampling. The sample is collected in a glass tube whose ends are closed once the exhalation is finished. The tube has an orifice sealed with a septum through which the fiber is inserted. Then, the fiber is desorbed in the injector of a gas chromatograph and the analysis is accomplished using mass spectrometry for the identification and quantification of the components. The proposed system avoids the need of complex sampling equipment and allows analysis of the alveolar fraction. Additionally, the system is economical and easy to handle, thus facilitating the development of normalized methods and its routine use in field studies.  相似文献

A simple and novel speciation method for the determination of volatile organic compounds of selenium (dimethylselenide (DMSe) and dimethyldiselenide (DMDSe) has been developed using a headspace hollow fiber protected liquid-phase microextraction (HS-HF-LPME) combined with capillary gas chromatography-mass spectrometry (GC-MS). The organic solvent impregnated in the pores and filled inside the porous hollow fiber membrane was used as an extraction interface in the HS-HF-LPME of the compounds. The effect of different variables on the extraction efficiency was studied simultaneously using an experimental design. The variables of interest in the HS-HF-LPME were sample volume, extraction time, temperature of sample solution, ionic strength, stirring rate and dwelling time. A Plackett-Burman design was performed for screening in order to determine the significant variables affecting the extraction efficiency. Then, the significant factors were optimized by a Box-Behnken design (BBD) and the response surface equations were derived. Under optimum conditions, preconcentration factors up to 1250 and 1170 were achieved for DMSe and DMDSe respectively. The detection limit and relative standard deviation (RSD) (n=5, c=50 μg L(-1)) for DMSe were 65 ng L(-1) and 4.8%, respectively. They were also obtained for DMDSe as 57 ng L(-1) and 3.9%, respectively. The developed technique was found to be applicable to spiked environmental and biological samples.  相似文献

This paper proposes a new analytical procedure based on the headspace solid‐phase microextraction (HS‐SPME) technique and gas chromatography‐selected ion monitoring‐mass spectrometry (GC‐SIM‐MS) for the determination of 16 phenols extracted from leather samples. The optimized conditions for the HS‐SPME were obtained through two experimental designs – a two‐level fractional factorial design followed by a central composite design – using the commercial SPME fiber polyacrylate 85 μm (PA). The best extraction conditions were as follows: 200 μL of derivatizing agent (acetic anhydride), 20 mL of saturated aqueous NaCl solution and extraction time and temperature of 50 min and 75°C, respectively. All optimized conditions were obtained with fixed leather sample mass (250 mg), vial volume (40 mL) and phosphate buffer pH (12) and concentration (50 mmol/L). Detection limits ranging from 0.03 to 0.20 ng/g, and relative standard deviation (RSD) lower than 10.23% (n=6) for a concentration of 800 ng/g (chlorophenols) and 1325 ng/g (2‐phenylphenol) in the splitless mode were obtained. The recovery was studied at three concentration levels by adding different amounts of phenols to the leather sample and excellent recoveries ranging from 90.0 to 107.2% were obtained. The validated method was shown to be suitable for the quantification of phenols in leather samples, as it is simple, relatively fast and sensitive.  相似文献

建立了同时测定体外透皮接收液中丙胺卡因和利多卡因的气相色谱-质谱(GC-MS)分析方法。以罗哌卡因为内标,样品经氢氧化钠碱化、乙酸乙酯萃取后采用GC-MS分析,采用选择离子监测(SIM)模式进行定量分析。丙胺卡因和利多卡因两种药物均在0.016~50.0 mg/L范围内呈良好的线性关系;回收率为85.3%~109.7%,日内和日间测定的相对标准偏差均小于10%;检出限分别为3 μg/L和2 μg/L。该方法操作简便,选择性好,灵敏度高,适用于局部麻醉药物快速透皮吸收研究中丙胺卡因和利多卡因含量的分析。  相似文献

This paper reports an experimental design optimization of a recently proposed silylation procedure that avoids the introduction of false positives and false negatives in the simultaneous determination of steroid hormone estrone (E1) and 17-alpha-ethinylestradiol (EE2) by gas chromatography-mass spectrometry (GC/MS). The figures of merit for several calibration procedures were evaluated under optimum conditions in the silylation step. Internal standardization strategies were applied and global models were constructed by gathering signals recorded on three non-consecutive days. Three calibration models were examined: a univariate model with a sum of six monitorized ions and a three-way PARAFAC-based model (the analyte scores were standardized on the basis of the scores of the internal standard). The global PARAFAC-based calibration model showed the best performance with detection capabilities of 4.3 microg l(-1) and 7.0 microg l(-1) for E1 and EE2, respectively, when the probability of false positives was fixed at 1% and that of false negatives at 5%. Mean relative error in absolute terms for E1 and for EE2 was 11.1% and 8.5%, respectively, and trueness was likewise confirmed. The proposed optimized derivatization procedure using a three-way calibration function was also applied in the determination of E1, 17-beta-estradiol (E2) and EE2 in bovine urine samples: recovery values were 68.5%, 40.4% and 43.4%, respectively, and the detection capability was 18.4, 19.3 and 18.6 microg l(-1) when the probability of false positives was fixed at 1% and that of false negatives at 5%. Mean relative error in absolute terms for E1, E2 and EE2 was 7.4%, 9.4% and 8.6%, respectively, and trueness was likewise confirmed.  相似文献
