Surprisingly similar reactions in the gas phase and in solution [Eq. (a)]: The ion 1 obtained by electrospray ionization behaves in the gas phase analogously to the corresponding complex [RuCl2(=CHPh)(PCy3)2] in solution. Measured relative rates for ring-opening metathesis are interpreted under the assumption of an intramolecular π complex, which leads to an estimation of the equilibrium constant for π complexation.  相似文献

In connection with efforts to build constrained peptidomimetics for the endocrine hormone TRH, a general strategy for the construction of bicyclic lactam peptide building blocks has been developed. This strategy used an anodic amide oxidation to selectively functionalize proline and then an olefin metathesis to build the desired lactam constraint. The route described provides a single approach for synthesizing both fully functionalized TRH analogs having seven- and eight-membered ring lactam constraints, as well as six- and seven-membered ring lactam analogs without the sidechain on the central amino acid.  相似文献

Bridging between (i)‐ and (i+3)‐positions in a β3‐peptide with a tether of appropriate length is expected to prevent the corresponding 314‐helix from unfolding (Fig. 1). The β3‐peptide H‐β3hVal‐β3hLys‐β3hSer(All)‐β3hPhe‐β3hGlu‐β3hSer(All)‐β3hTyr‐β3hIle‐OH ( 1 ; with allylated βhSer residues in 3‐ and 6‐position), and three tethered β‐peptides 2 – 4 (related to 1 through ring‐closing metathesis) have been synthesized (solid‐phase coupling, Fmoc strategy, on chlorotrityl resin; Scheme). A comparative CD analysis of the tethered β‐peptide 4 and its non‐tethered analogue 1 suggests that helical propensity is significantly enhanced (threefold CD intensity) by a (CH2)4 linker between the β3hSer side chains (Fig. 2). This conclusion is based on the premise that the intensity of the negative Cotton effect near 215 nm in the CD spectra of β3‐peptides represents a measure of ‘helical content’. An NMR analysis in CD3OH of the two β3‐octapeptide derivatives without (i.e., 1 ) and with tether (i.e., 4 ; Tables 1–6, and Figs. 4 and 5) provided structures of a degree of precision (by including the complete set of side chain–side chain and side chain–backbone NOEs) which is unrivaled in β‐peptide NMR‐solution‐structure determination. Comparison of the two structures (Fig. 5) reveals small differences in side‐chain arrangements (separate bundles of the ten lowest‐energy structures of 1 and 4 , Fig. 5, A and B ) with little deviation between the two backbones (superposition of all structures of 1 and 4 , Fig. 5, C ). Thus, the incorporation of a CH2? O? (CH2)4? O? CH2 linker between the backbone of the β3‐amino acids in 3‐ and 6‐position (as in 4 ) does accurately constrain the peptide into a 314‐helix. The NMR analysis, however, does not suggest an increase in the population of a 314‐helical backbone conformation by this linkage. Possible reasons for the discrepancy between the conclusion from the CD spectra and from the NMR analysis are discussed.  相似文献

A large-scale synthesis of known Ru olefin metathesis catalyst VII featuring an unsymmetrical N-heterocyclic carbene (NHC) ligand with one 2,5-diisopropylphenyl (DIPP) and one thiophenylmethylene N-substituent is reported. The optimised procedure does not require column chromatography in any step and allows for preparation of up to 0.5 kg batches of the catalyst from simple precursors. The application profile of the obtained catalyst was studied in environmentally friendly dimethyl carbonate (DMC). Although VII exhibited low efficiency in cross-metathesis (CM) with electron-deficient partners, good to excellent results were noted for substrates featuring easy to isomerise C−C double bonds. This includes polyfunctional substrates of medicinal chemistry interest, such as analogues of psychoactive 5F-PB-22 and NM-2201 and two PDE5 inhibitors—Sildenafil and Vardenafil. Finally, a larger scale ring-closing metathesis (RCM) of a Vardenafil derivative was conducted in DMC, allowing for straightforward isolation of the expected product (23 g) in high yield and with low Ru contamination level (7.7 ppm).  相似文献

Mono- and tris-3,5- and 2,6-pyridinediyl-containing macroheterocycles were synthesized by metathesis of olefin-substituted pyridines in the coordination sphere of mono- and tris-platinum complexes, respectively. Tris(2,6-pyridinediyl)-containing macroheterocycles were hydrogenated over palladium catalyst. The hydrogenated macrocycle was used as ligand for the triplatinum template. A diimine ligand having two [2.2]paracyclophanyl substituents at the N atoms (L1) was prepared from the reaction of amino[2.2]paracyclophane with acenaphtenequinone. The ligand reacts with NiBr2(dme) (dme: 1,2-dimethoxyethane) to form the dibromonickel complex with (R,R) and (S,S) configuration, NiBr2(L1). The structure of the complex was confirmed by X-ray crystallography. NiBr2(L1) catalyzes oligomerization of ethylene in the presence of methylaluminoxane (MAO) co-catalyst at 10–50 °C to form a mixture of 1- and 2-butenes after 3 h. The reactions for 6 h and 8 h at 25 °C causes further increase of 2-butene formed via isomerization of 1-butene and formation of hexenes. Reaction of 1-hexene catalyzed by NiBr2(L1)–MAO produces 2-hexene via isomerization and C12 and C18 hydrocarbons via oligomerization. Consumption of 1-hexene of the reaction obeys first-order kinetics. The kinetic parameters were obtained to be ΔG = 93.6 kJ mol−1, ΔH = 63.0 kJ mol−1, and ΔS = −112 J mol−1deg−1. NiBr2(L1) catalyzes co-dimerization of ethylene and 1-hexene to form C8 hydrocarbons with higher rate and selectivity than the tetramerization of ethylene.  相似文献

Phosphorodithioate-type short oligonucleotides were efficiently synthesized using bis(2,6-dimethylphenyl) phosphorochloridate as a coupling agent on a solid support by application of the H-phosphonothioate method, where oxidation was facilitated using elemental sulfur following completion of H-phosphonothioate oligomer assembly, as with the standard H-phosphonate method.  相似文献
