Abstract— Kinetic studies with the mustard seedling ( Sinapis alba L.) support the hypothesis that the so-called 'high energy reaction' of photomorphogenesis can be understood solely on the basis of phytochrome. Light-induced anthocyanin synthesis (a typical 'positive' photoresponse(1) and light dependent inhibition of hypocotyl lengthening (a typical 'negative' photoresponse(1)) have been investigated. In order to explain the experimental data we have to assume that there are two different types of phytochrome 730 which differ greatly as far as their resistance to irreversible destruction is concerned. The existence of these two different types of phytochrome 730 has already been postulated on the basis of spectrophotometric measurements in vivo .(2)  相似文献

Abstract— Lysozyme undergoes conformational changes when exposed to visible light in the presence of methylene blue. Ultracentrifugation, gel filtration and end group analysis showed that no peptide bond was cleaved. About 30 per cent of the tryptophan residues were, however, modified. The enzymic activity decreased by 50 per cent. The sedimentation coefficient and helical content decreased; the extinction in the u.v. region between 185 to 210 nm increased. The photooxidized enzyme was more susceptible to trypsinolysis than the native enzyme.  相似文献

The photoreaction between Pτ and the first detectable intermediate, lumi-R, of 124-kdalton oat phytochrome has been investigated at low temperatures. The temperature dependence of the quantum yields of the photoreactions, Pτ to lumi-R and lumi-R to Pτ, has been determined. From measurements over a temperature range from 119 to 155 K, an activation barrier of 3.6 ± 0.5 kJ mol 1 is found for the photoreaction of Pτ with 661-nm actinic light. A higher value (5.7 ± 0.7 kJ mol -1) is found for the photoreaction of lumi-R to Pτ. with 698-nm actinic light. Increased quantum yields are found in deuterated buffer solutions at low temperatures. The activation energies for deuterated phytochrome (3.2 ± 0.7 kJ mol–1 for Pτ with 661-nm irradiation and 6.2 ± 1.2 kJ mol-1 for lumi-R at 698-nm irradiation) are identical within the limits of error with those of protonated phytochrome. The lack of a deuterium effect for the activation energies favors the Z,E-isomerization rather than proton transfer or tautomerization for the chromophore photochemistry during Pτ⇄lumi-R conversion.  相似文献

蛋白质在折叠过程中其构象要发生明显变化 .采用精确计数法 ,计算了在蛋白质折叠的不同阶段其尺寸大小及其分布情况 .发现在折叠的初期 ,分子的尺寸比较大 ,其分布也比较宽 .在折叠的后期 ,分子的尺寸比较小 ,其分布比较窄 .不同的氨基酸序列 ,其分子尺寸的分布也不同 .对于可折叠的氨基酸序列 ,在平均尺寸大小附近出现的几率特别大 .同时还计算了比值SN DN.这里SN 为可设计序列数目 (Thenumberofdesigningsequences) ,DN 为可设计构象数目 (Thenumberofdesignableconformations) ,并有关系- 1 6 8 4 +0 32 5 5N≤SN DN ≤- 0 86 6 4 +0 312 5N  (N ≥ 13)通过这些研究以提高对折叠过程的认识  相似文献

The molecular mode of signal transduction triggered by phytochrome is unknown. One characteristic structural/topographic feature of the physiologically active form (Pfr) of phytochrome is that its tetrapyrrole chromophore becomes preferentially exposed in the Pfr form (compared to the Pr form). Phytochrome in its Pfr form appears to affect phosphorylation of cellular proteins. The literature on the phytochrome-mediated protein phosphorylation has been reviewed in an attempt to search for the role of the chromophore topography of phytochrome in the signal transduction process. In order to initiate a dephosphorylation-phosphorylation cascade as a possible step for the signal transduction, it may interact with a cellular protein kinase to inhibit its activity. This hypothesis has been reviewed with results from phosphorylation inhibition assays by the Pfr form of phytochrome and in light of the inhibition of protein kinase activity by tetrapyrroles in general.  相似文献

The photoreceptor phytochrome is widely distributed in the plant kingdom from angiosperms to ferns, mosses and algae. The epitope for the monoclonal antibody Z-3B1 which exhibits wide-ranging cross-reactivity with phytochromes from higher and lower plants was mapped by the combination of several methods: by Western blot with proteolytic fragments of known localization, by sequence comparison of phytochromes from various plants, and by production of overlapping fusion proteins. The only sequence which is common to all positively-reacting fusion proteins is the sequence A-830 to R-859. This sequence must contain the Z-3B1 epitope. The best candidate is suggested to be the T-cell antigenic sequence K-Y-V/I-E-A/C-L-L-T (= K-848 to T-855). The significance of the highly conserved epitope in all phytochromes is discussed.  相似文献

Abstract— In vitro phosphorylation of some polypeptides was affected in extracts obtained from 5-and 6-day-old plants irradiated with 5 min of red light. The phosphorylation of 55 kDa polypeptide in both 5- and 6-day-old plants, a 60 kDa, and 76 kDa polypeptide in 6-day-old plants and 70 kDa, 67 kDa polypeptide in 5-day-old plants was stimulated by red light. This effect was reversible by far-red light. The extent of stimulation by red light and reversal by far-red light varied for different polypeptides. No differential effect of red and far-red light was seen on the phosphorylation of 94 and 40 kDa polypeptides. In fact, phosphorylation of 94 kDa polypeptide in 6-day-old plants decreased on red light irradiation. These results show that the phosphorylation or dephosphorylation of some proteins is affected by phytochrome and the effect of light is also dependent on the age of the plant.  相似文献

Abstract —We present physiological evidence using the threshold control of lipoxygenase synthesis† by Pfr in the mustard seedling (LOG response) that there is no dark reversion of phytochrome
which would be relevant for this response. Such Ptr which can be detected with the lipoxygenase response disappears exclusively through degradation with a half-life of 45 min at 25°C. De novo synthesis of P*r in the hypocotylar hook takes place at a constant rate (zero order rate constant) irrespective of the level of Pr or P*tot, i.e. there is no detectable feedback control of Pr synthesis during the period of experimentation. The data of the present paper are consistent with a quantitative phytochrome model (Scheme 1) which has been advanced and treated quantitatively in the companion paper (Oelze-Karow and Mohr, 1976).  相似文献

本工作在合成了几种不同结构吡唑啉衍生物的基础上,测定了它们在相同极性,不同粘度介质中的荧光量子产率并表明:和1-位氮原子相联苯基的旋转松弛对该类化合物的发光具有决定意义,通过对化合物在极性与非极性溶剂内和在冻结与流动条件下(低温与室温下)荧光强度的比较,证明了二苯基吡唑啉化合物的发光态具有扭曲构象。  相似文献

Abstract. A rapid procedure for the purification of phytochrome from rye by fast protein liquid chromatography (FPLC) is described. A pre-purification with Q-Sepharose fast flow and hydroxylapatite fast flow is necessary in order not to exceed the total capacity of the Mono-Q HR 5/5 column. The whole fractionation is finished within 24 h and yields an intact 124-kDa phytochrome of high purity.  相似文献

利用PPP-SCF-CI-DV量子化学程序拟合胆红素以及胆汁酸盐-胆红素水溶液的UV和CD谱, 借以研究胆红素及其与胆汁酸盐复合物的构象性质。发现胆红素与胆汁酸盐作用时, 两个双吡咯生色团的扭角由104°变成112°, 仍保持形成分子内氢键的状态, 其UV及CD谱两个吸收峰间隔的增宽, 主要是胆红素的一侧双吡咯生色团与胆汁酸盐呈疏水性结合的原故; 二羟基和三羟基胆汁酸盐所形成的二元复合物园二色谱的差别, 是胆红素的对映选择性结合造成的。  相似文献

用紫外分光光度法研究了不同价态金属离子对DNA大分子溶液构象的影响。研究结果显示: 金属离子与DNA的作用使DNA溶液的紫外吸收值下降, 即呈现减色效应, 同时减色效应的强弱与金属离子的价态有关。随着价态升高, 减色效应增强。由此说明, 金属离子与DNA的作用使DNA的构象趋于缩拢; 且随着金属离子价态升高, 缩拢程度增强, 甚至产生缩合。还用现代多聚电解质理论对以上现象进行了讨论, 结果是令人满意的。  相似文献

Abstract— To probe the nature of primary photoprocess and the mechanism of the phototransformation of undegraded 124 kDa oat phytochrome, solvent deuterium isotope effects on the fluorescence and phototransformation of phytochrome have been investigated. The fluorescence intensity and lifetime of phytochrome (Pr form) are greater in D20-buffer than in H2O-buffer, suggesting a possible involvement of proton transfer in the primary photoprocess of phytochrome. Although the photostationary equilibrium (Pr to Pfr ratio) was not altered by deuterium oxide, in contrast to degraded phytochrome, the rate constants of both transformations, Pr→ Pfr and Pfr→ Pr were enhanced by up to 24%. The Pr to Pfr phototransformation of degraded phytochrome, however, was retarded by about the same percentage in D2O. These opposite effects of D2O with degraded and undegraded phytochromes underscore the fact that the Pr form from the former reverts to the Pr form in the dark, apparently catalyzed by deuterated general and/or conjugate acidic group(s). With the degraded phytochrome the deuterium oxide enhancement of the rate of dark reversion was approximately 2-fold (Sarkar and Song, 1981). Both the fluorescence intensity and the rates of phototransformation of phytochrome were enhanced in D2O with successive photocyclings (Pr→ Pfr→ Pr→ Pfr→ Pr etc.) with alternating red and far-red irradiation. It has been proposed that successive photocycling of phytochrome in D2O results in proton-deuteron exchange in the partially exposed Ptr chromophore and/or its surrounding amino acid residues.  相似文献
