A series of well-defined diblock copolymers, poly(ethylene oxide)-block-poly(4-vinylpyridine) (PEO-b-P4VP) used as physical coating of capillaries, were synthesized by atom transfer radical polymerization (ATRP). EOF measurement results showed that all synthesized PEO-b-P4VP diblock copolymer-coated capillaries in this report could suppress EOF effectively compared to the bare fused-silica capillaries, and efficient separations of basic proteins were achieved. The effects of the molecular weight of P4VP block in PEO-b-P4VP and buffer pH on the separation of basic proteins for CE were investigated in detail. Moreover, the relationships between morphologies of PEO-b-P4VP diblock copolymers in buffer, which were studied by transmission electron microscopy, and the separation efficiencies of basic protein with PEO-b-P4VP diblock copolymers coatings were discussed.  相似文献

A series of well‐defined triblock copolymers, poly(N, N‐dimethylacrylamide)‐block‐poly(ethylene oxide)‐block‐poly(N, N‐dimethylacrylamide) (PDMA‐b‐PEO‐b‐PDMA) synthesized by atom transfer radical polymerization, were used as physical coatings for protein separation. A comparative study of EOF showed that the triblock copolymer presented good capillary coating ability and EOF efficient suppression. The effects of the Mr of PDMA block in PDMA‐b‐PEO‐b‐PDMA triblock copolymer and buffer pH on the separation of basic protein for CE were investigated. Moreover, the influence of the copolymer structure on separation of basic protein was studied by comparing the performance of PDMA‐b‐PEO‐b‐PDMA triblock copolymer with PEO‐b‐PDMA diblock copolymer. Furthermore, the triblock copolymer coating showed higher separation efficiency and better migration time repeatability than fused‐silica capillary when used in protein mixture separation and milk powder samples separation, respectively. The results demonstrated that the triblock copolymer coatings would have a wide application in the field of protein separation.  相似文献

A novel method for the preparation of covalently linked capillary coatings of poly(N‐vinyl aminobutyric acid) (PVAA) obtained from hydrolyzed polyvinylpyrrolidone was demonstrated using photosensitive diazoresin (DR) as a coupling agent. A layer‐by‐layer self‐assembled film of DR and PVAA based on ionic bonding was first fabricated on the inner wall of capillary, then ionic bonding was converted into covalent bonding after treatment with UV light through a unique photochemical reaction of DR. The covalently bonded coatings suppressed protein adsorption on the inner surface of the capillary, and thus a baseline separation of lysozyme, cytochrome c, BSA, amyloglucosidase, and myoglobin was achieved using CE. Compared with bare capillary or noncovalently bonded DR/PVAA coatings, the covalently linked DR/PVAA capillary coatings not only improved the CE separation performance for proteins, but also exhibited good stability and repeatability. Due to the replacement of the highly toxic and moisture‐sensitive silane coupling agent by DR in the covalent coating preparation, this method may provide a green and easy way to make covalently coated capillaries for CE.  相似文献

The resolving ability of 2-O-(2-hydroxybutyl)-beta-CD (HB-beta-CD) with different degrees of substitution (DS = 2.9 and 4.0) as a chiral selector in CZE is reported in this work. Fourteen chiral drugs belonging to different classes of compounds of pharmaceutical interest such as beta-agonists, antifungal agents, ageneric agents, etc., were resolved. The effects of the DS of HB-beta-CD on separations were also investigated. The chiral resolution (R(s)) was strongly influenced by the concentrations of the CD derivative, the BGE, and the pH of the BGE. Under the conditions of 50 mmol/L Tris-phosphate buffer at pH 2.5 containing 5 mmol/L HB-beta-CD, all 14 analytes were separated. The very low concentration necessary to obtain separation was particularly impressive. The DS had a significant effect on the resolution of the chiral drugs and the ionic strength of the separation media; hence, the use of a well-characterized CD derivative is crucial.  相似文献

In this paper, we described a simple and rapid method, capillary electrophoresis with electrochemiluminescence (CE–ECL) detection using tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+), to simultaneously detect pethidine and methadone. Analytes were injected to separation capillary of 67.5 cm length (25 μm i.d., 360 μm o.d.) by electrokinetic injection for 10 s at 10 kV. Under the optimized conditions: ECL detection at 1.20 V, 30 mM sodium phosphate (pH 6.0) as running buffer, separation voltage at 14.0 kV, 5 mM Ru(bpy)32+ with 50 mM sodium phosphate (pH 6.5) in the detection cell, the linear range from 2.0 × 10− 6 to 2.0 × 10− 5 M for pethidine and 5.0 × 10− 6 to 2.0 × 10− 4 M for methadone and detection limits of 0.5 μM for both of them were achieved (S/N = 3). Relative standard derivations of the ECL intensity were 2.09% and 6.59% for pethidine and methadone, respectively.  相似文献

Surface plasmon resonance (SPR) was used to screen the interaction between a variety of affinity ligands and hemagglutinin (HA) from human influenza virus, with the aim of identifying low affinity ligands useful for the development of a rapid bioanalytical sensor. Three sialic acid-based structures and four lectins were evaluated as sensor ligands. The sialic acid-based ligands included a natural sialic acid-containing glycoprotein, human alpha1-acid glycoprotein (alpha1-AGP), and two synthetic 6'-sialyllactose-conjugates, with varying degree of substitution. The interaction of HA with the four lectin-based ligands, concanavalin A (Con A), wheat germ agglutinin (WGA), Maackia amurensis lectin (MAL), and Sambucus nigra agglutinin (SNA), showed a wide variation of affinity strengths. Affinity and kinetics data were estimated. Strong affinities were observed for Con A, WGA, alpha1-AGP, and a 6'-sialyllactose-conjugate with a high substitution degree, and low affinities were observed for MAL and a 6'-sialyllactose-conjugate with low substitution. The main objective, to identify a low affinity ligand which could be used for on-line monitoring and product quantification, was met by a 6'-sialyllactose-ovalbumin conjugate that had 0.6 mol ligand per mol carrier protein. The apparent affinity of this ligand was estimated to be 1.5+/-0.03 microM (K(D)) on the SPR surface. Vaccine process samples containing HA were analyzed in the range 10-100 microg HA mL(-1) and correlated with single-radial immunodiffusion. The coefficient of variation on the same chip was between 0.010 and 0.091.  相似文献

Various thioacridine derivatives are potential chemotherapeutics against various diseases which are intensively synthesized, characterized, and investigated by many research groups. Efficient, fast, and reliable separation and quantification methods for their analysis are still to be developed. MEKC and capillary LC (CLC) were applied for the separation and quantification of five highly hydrophobic, weakly basic, and structurally similar 9-(alkylthio)acridines. Since the common anionic and cationic surfactants failed to separate the strongly hydrophobic thioacridines by MEKC, sodium cholate was used in an alkaline BGE and successfully employed for their fast separation. In CLC, the weakly basic nature of the thioacridines necessitated use of LiChrosorb RP-select B sorbent as the stationary phase, which combined with a very simple mobile phase methanol/water yielded an efficient chromatographic separation system. Both, the MEKC and CLC optimized separation methods were then applied to quantify the thioacridines within a concentration range of 1.0 x 10(-5)-1.0 x 10(-3) mol/L and the obtained experimental results were critically compared. In practical terms, the MEKC analytical method can quantify the analytes much faster but with a lower reliability while the CLC method performs slower analysis with a higher repeatability of the experimental results.  相似文献

With unique 3‐D architecture, the application of core‐based hyperbranched polyethyleneimine (CHPEI), as a capillary coating in capillary electrophoresis, is demonstrated by manipulation of the electroosmotic mobility (EOF). CHPEI coatings (CHPEI5, Mw ≈? 5000 and CHPEI25, Mw ≈? 25 000) were physically adsorbed onto the inner surface of bare fused‐silica capillary (BFS) via electrostatic interaction of the oppositely charged molecules by rinsing the capillaries with different CHPEI aqueous solutions. The EOF values of the coated capillaries were measured over the pH range of 4.0–9.0. At higher pH (pH >6) the coated capillary surface possesses excess negative charges, which causes the reversal of the EOF. The magnitudes of the EOF obtained from the coated capillaries were three‐fold lower than that of BFS capillary. Desirable reproducibility of the EOF with % RSD (n = 5) ? 2 was obtained. Effect of ionic strength, stability of the coating (% RSD = 0.3) and the dependence of the EOF on pH (% RSD = 0.5) were also investigated. The CHPEI‐coated capillaries were successfully utilized to separate phenolic compounds, B vitamins, as well as basic drugs and related compounds with reasonable analysis time (<20 min) and acceptable migration‐time repeatability (<0.7% RSD for intra‐capillary and <2% RSD for inter‐capillary).  相似文献

A novel in-capillary reduction and capillary electrophoretic (CE)-chemiluminescence (CL) method was developed for the sensitive and selective determination of chromium(III) and chromium(VI). The proposed method was based on the in-capillary reduction of Cr(VI) with acidic H2O2 to form Cr(III) using the zone-passing technique and chemiluminescence detection of Cr(III). The sample [Cr3+ and CrO42−], hydrochloric acid, and H2O2 (reductant) solution segments were injected for specified periods of time in this order from the anodic end of a capillary, followed by application of an appropriate running voltage between both ends. As both chromium species have opposite charges, Cr3+ migrates to the cathode while CrO42− ion, moving oppositely to the anode, reacts with acidic H2O2, resulted in formation of Cr3+. Based on the migration time difference of both Cr3+ ions, they were separated by zone electrophoresis. Running buffer was composed of 0.02 mol l−1 HAc-NaAc (pH 4.7) with 1×10−3 mol l−1 EDTA. Parameters affecting CE-CL separation and detection, such as reductant concentration, mixing mode of the analytes with CL reagent, CL reaction reagent pH and concentration, stability of luminol-hydrogen peroxide mixed solution were optimized. The limits of detection for chromium(III) and chromium(VI) (3σ) were 6×10−13 mol l−1 (mass concentration 12 zmol) and 8×10−12 mol l−1 (160 zmol), respectively. This method offered potential advantages of simplicity, sensitivity, selectivity and applicability to the determination of Cr(III) and Cr(VI) in environmental water.  相似文献

The aim of this work was to characterize the fulvic acids (FA) extracted from peat by use of capillary electrophoresis (CE). Two FA extracts have been obtained using two different extractants (0.1 M NaOH and 0.1 M NaOH plus 0.1 M Na4P2O7, Na+PP). The CE runs wre performed using a coated silica capillary, and several buffer solutions at different PH values. The results obtained show that the Fa extracts may be well characterized by CE even if the isolelectric point of the fulvic compounds is very low. The best fractionation has been obtained using a buffer borate at PH 8.9 as funning buffer. The charcterization of the two extracts also showed some important differences between the NaOH extract and the Na+PP extract.  相似文献

Tris(2,2′-bipyridyl)ruthenium(II) electrochemiluminescence detection in a capillary electrophoresis separation system was used for the determination of diphenhydramine. In this study, platinum disk electrode (300 μm in diameter) was used as a working electrode and the influence of applied potential and buffer conditions were investigated. Under optimal conditions: 1.2 V applied potential, pH 8.50, 15 kV separation voltage and 10 mmol l−1 running buffer, the calibration curve of diphenhydramine was linear over the range of 4×10−8 to 1×10−5 mol l−1. This technique gave satisfactory precision, and relative standard deviations of migration times and chemiluminescence peak intensities were less than 1 and 6%, respectively. The technique was applied to animal studies for determination of diphenhydramine extracted from rabbit plasma and urine samples, and the extraction efficiency were between 92 and 98.5%.  相似文献

The major goal of this work was to develop a new solid-state electrochemiluminescence (ECL) detector suitable for capillary electrophoresis (CE). The detector was fabricated by coating a sol-gel derived zirconia (ZrO(2))-Nafion composite film on a graphite electrode, then the zirconia-Nafion modified electrode was immersed in tris(2,2'-bipyridyl)ruthenium(II) (Ru(bpy)(3) (2+)) solution to immobilize this active chemiluminescence reagent. The voltammetric and ECL behaviors of the detector were investigated and optimized in tripropylamine solution. The ratio of 53% for zirconia in the zirconia-Nafion composite provided the highest luminescence intensity of immobilized Ru(bpy)(3) (2+). The ECL can maintain its stability very well in the phosphate solution in the period of 5-90 h when the solid-state ECL detector was immersed in the solution all the time. The optimum distance of capillary outlet to the solid-state ECL detector has been found to be ca. 50-80 microm for a 75 microm capillary. The effects of ionic strength and pH of ECL solution on peak height were investigated. The CE with solid-state ECL detector system was successfully used to detect tripropylamine, lidocaine, and proline. The detection limits (S/N = 3) were 5 x 10(-9) mol.L(-1) for tripropylamine, 1 x 10(-8) mol.L(-1) for lidocaine and 5 x 10(-6) mol.L(-1) for proline, and the linear ranges were from 1.0 x 10(-8) to 1.0 x 10(-5) mol.L(-1) for tripropylamine, 5.0 x 10(-7) mol.L(-1) to 1.0 x 10(-5) mol.L(-1) for lidocaine and 1.0 x 10(-5) to 1.0 x 10(-3) mol.L(-1) for proline, respectively.  相似文献

In this paper, a poly(styrene-octadecene-divinylbenzene) (PS-OD-DVB) monolithic column was prepared in one step by introducing a C18 carbon chain as monomer. N,N-Dimethylformamide and decanol served as porogens to make a homogeneous polymerization mixture in a fused silica capillary (320 microm inner diameter). Its physical and chromatographic properties were compared with those of poly(styrene-divinylbenzene) (PS-DVB) monolithic column, which was also fabricated by in-situ polymerization in a fused silica capillary with the same inner diameter. Six standard proteins were used to evaluate the columns and their potential application for the separation of human hemoglobin was also discussed. It was shown that the PS-OD-DVB and PS-DVB monoliths appeared to have similar efficiency for rapid separation of six proteins within 3.5 min. The PS-OD-DVB monolith was found to have higher loading capacity and higher resolution for the separation of alpha and beta chains of hemoglobin because of the introduction of C18 carbon chains, and shows great potential for the separation of bio-macromolecules.  相似文献

Generation 2 to generation 5 poly(amidoamine) (PAMAM) dendrimers having different terminal functionalities were analyzed by capillary electrophoresis (CE). Polyacrylamide gel electrophoresis was also used to assess the composition of the individual generations for comparison with the CE results. Separation of PAMAMs can be accomplished by either using uncoated silica or silanized silica capillaries, although reproducibility is poor using the uncoated silica capillary. To improve run-to-run reproducibility, silanized capillary was used and various internal standards were also tested. Relative and normalized migration times of primary amine terminated PAMAM dendrimers were then determined using 2,3-diaminopyridine (2,3-DAP) as an internal standard. Using silanized capillaries and internal standards, the relative and normalized migration times are fully reproducible and comparable between runs. Apparent dimensionless electrophoretic mobilities were determined and the results were compared to theoretical calculations. It is concluded that for PAMAMs a complex separation mechanism has to be considered in CE, where the movement of the ions is due to the electric field, but the separation is rather the consequence of the adsorption/desorption equilibria on the capillary wall ("electrokinetic capillary chromatography"). The described method may be used for quality control and may serve as an effective technique to analyze polycationic PAMAM dendrimers and their derivatives with different surface modifications.  相似文献
