Silicon grafted monodisperse poly(ethylene glycol) (PEG) silanes with various PEG chain lengths and mixtures of these were systematically analyzed with static time-of-flight secondary ion mass spectrometry (TOF-SIMS). The mass spectra show differences in the various relative signal intensities, an observation that was used to elucidate important aspects of the grafting process. The relationship between PEG-silane fragment ion abundances and Si(+) ion abundances were used to (i) qualitatively describe layer thicknesses of grafted mixtures of PEG-silanes on silicon, (ii) construct a calibration curve from which PEG chain length (or molecular mass) can be determined and (iii) quantitatively determine surface mixture compositions of grafted monodisperse PEG-silanes of different chain lengths (3, 7 and 11 PEG units). The results suggest that discrimination does take place in the adsorption process. The PEG-silane with the shorter PEG chain is discriminated for mixtures containing PEG3-silane, whereas the PEG-silane with the longer PEG chain is discriminated in PEG7/PEG11-silane mixtures. The origin of this difference in adsorption behavior is not well understood. Aspects of the grafting process and the TOF-SIMS analyses are discussed.  相似文献

The synthesis of branched poly(ethylene glycol) (PEG) derivatives, namely star PEG and dendronized PEG, can be challenging and their purity can be difficult to ascertain using traditional techniques, such as NMR and GPC. Herein, the detailed characterization of these branched PEGs using MALDI-ToF MS was investigated in order to confirm their structural purity. In this light, mass spectrometry offers a number of advantages for polymer characterization, including the ability to get detailed structural data, such as end group masses, from microgram-scale samples. In addition, the ability to rapidly acquire data from crude reaction aliquots makes MALDI-ToF MS ideal for monitoring end group transformations.  相似文献

A study of the fast-atom bombardment tandem mass spectrometry behavior of a number of ethylene glycol polymers (PEGs) has been carried out. Both linear (hydroxyl, amino, and/or alkyl end groups) and cyclic (crown ether) polymers were studied. One of the materials is a block copolymer of ethylene and propylene oxides. Collisional activation was carried out in the collision octapole of a BEoQ hybrid mass spectrometer at a translational energy of 50 eV, with collision gas air. For the linear polymers, the most intense product ions are lithiated, linear polyglycol oligomers. These ions are formed via internal hydrogen transfer reactions that are facilitated (charge-induced) by lithium. This series of product ions allows for the observation of consecutive losses of monomer units from the chain end; this is useful to determine the sequence of monomers in a copolymer. The most abundant product ions from cyclic PEGs are lithiated radical cations. An especially interesting finding in this work is the preferential loss of two internal ethylene oxide (EO) units (dioxane, 88 u) from some [M + Li]+ precursors. Factors that influence this loss include (a) the sequence length of EO repeat units in the oligomer and (b) the identity of the end group(s) on the oligomer. It is proposed that this elimination of dioxane involves a six-membered ring intermediate; this decomposition reaction is believed to be a lithium-mediated (charge-induced) rearrangement. A method of characterizing polyglycol esters, an important class of industrial polymer, has been developed using electrospray ionization ion trap mass spectrometry (ESI ITMS). The fragmentation behavior of polyglycol esters is found to be different from that of polyglycols whose functional end groups are linked to the polymer chain via ether bonds (i.e., polyglycol ethers). The fragmentation pattern of an oligomer ion generated by low-energy collision-induced dissociation is strongly dependent on the type of cation used for ionization. It is shown that structural information on the polymer chain and end groups is best obtained by examining the fragment ion spectra of oligomers ionized by ammonium, alkali, and transition metal ions. The application of this method is demonstrated in the analysis of two surfactants based on fatty acid methyl ester ethoxylates.  相似文献

The analysis of poly(ethylene glycol) (PEG)-containing particles by online single particle aerosol mass spectrometers equipped with laser desorption/ionization (LDI) is reported. We demonstrate that PEG-containing particles are useful in the development of aerosol mass spectrometers because of their ease of preparation, low cost, and inherently recognizable mass spectra. Solutions containing millimolar quantities of PEGs were nebulized and, after drying, the resultant micrometer-sized PEG-containing particles were sampled. LDI (266 nm) of particles containing NaCl and PEG molecules of average molecular weight<500 Da generated mass spectra reminiscent of mass spectra of PEG collected by other mass spectrometer platforms including the characteristic distribution of positive ions (Na+ adducts) separated by the 44 m/z units of the ethylene oxide units separating each degree of polymerization. PEGs of average molecular weight>500 Da were detected from particles that also contained the tripeptide tyrosine-tyrosine-tyrosine or 2,5-dihydroxybenzoic acid, which were added to nebulized solutions to act as matrices to assist LDI using pulsed 266 nm and 355 nm lasers, respectively. Experiments were performed on two aerosol mass spectrometers, one reflectron and one linear, that each utilize two time-of-flight mass analyzers to detect positive and negative ions created from a single particle. PEG-containing particles are currently being employed in the optimization of our bioaerosol mass spectrometers for the application of measurements of complex biological samples, including human effluents, and we recommend that the same strategies will be of great utility to the development of any online aerosol LDI mass spectrometer platform.  相似文献

Several electrospray mass spectrometry (ESI-MS) techniques have been described during the past years to enable the characterization work of large poly(ethylene glycol)s (PEGs) and PEGylated proteins. The proton transfer reaction ESI-MS method utilizes amines to charge reduce the electrospray envelope of PEGs, hence PEG molecules are aminated instead of protonated. This method simplifies the mass spectrum of large PEGs, and enables the interpretation of the charge state of the observable envelopes (R ≥ 3,000 (FWHM) measured at the (M + 6H)6+ ion from 40 K PEG compound 7,324.19). Hence, deconvolution of the MS data can be performed and relative molecular masses of the individual chain lengths of the PEGs can be calculated. However, as the poly-dispersity of PEGs may vary from batch to batch and from sample to sample, it was of interest to examine if the method could distinguish between these kinds of different material. Therefore, sample materials of each intermediate obtained at five synthetic steps during synthesis of a 40 kDa PEG molecule were collected. These four intermediates, starting material and the target molecule were examined by 1H-NMR spectroscopy and ESI-MS using a proton stripping base. The study revealed that the charge-stripping ESI-MS method is able to differentiate between even small changes in the structure of the polymeric molecules only when the analysis is assisted by 1H-NMR spectroscopy. A proper characterization of polymer molecules requires besides relative molecular mass, also poly-dispersity and end-group characterization. No end-group information is obtained based on MS data. Examination of the PEG polymers by 1H-NMR spectroscopy provides the needed information. In addition, the 1H-NMR spectra clearly distinguishes the examined polymers.  相似文献

Pyrene end-labeled double hydrophilic diblock copolymers, poly(N-isopropylacrylamide)-b-poly(oligo(ethylene glycol) methyl ether methacrylate) (Py-PNIPAM-b-POEGMA), were synthesized via consecutive reversible addition-fragmentation chain transfer polymerization using a pyrene-containing dithioester as the chain transfer agent. These diblock copolymers molecularly dissolve in pure methanol and water, but form well-defined and nearly monodisperse PNIPAM-core micelles in an appropriate mixture of them due to the cononsolvency of PNIPAM block. 1H NMR, laser light scattering, fluorescence spectroscopy, and transmission electron microscopy were employed to characterize the cononsolvency-induced PNIPAM-core micelles. When the volume fraction of water, phi water, in the methanol/water mixture is in the range of 0.5-0.8, the sizes of micelles are in the range of 20-30 nm in radius for Py-PNIPAM50-b- POEGMA18. At phi water = 0.5, the formed micelles possess the highest overall micelle density and the largest molar mass. The effects of varying the block lengths of Py-PNIPAM-b-POEGMA diblock copolymers on the structural parameters of PNIPAM-core micelles have also been explored. Although we can observe the immediate appearance of bluish tinge upon mixing the diblock copolymer solution in methanol with equal volume of water (phi water = 0.5), which is characteristic of the formation of micellar aggregates, the whole micellization process apparently takes a relatively long time to complete, as revealed by monitoring the time dependence of fluorescence emission spectra. The excimer/monomer fluorescence intensity ratios, IE/IM, continuously decrease with time and then reach a plateau value after approximately 20 min. The decrease of IE/IM after the initial formation of pseudo-equilibrium micelles should be ascribed to the structural rearrangement and further packing of PNIPAM segments within the micelle core, restricting the mobility of pyrene end groups and decreasing the probability of contact between them. Compared to the conventional cosolvent approach employed for the micellization of block copolymers in selective solvents, the reported cononsolvency-induced unimer-micelle-unimer transition of Py-PNIPAM-b-POEGMA in methanol/water mixtures has been unprecedented.  相似文献

The enzyme-mediated enantioselective hydrolysis of poly(ethylene glycol) (PEG)-supported carbonates is disclosed. The water-soluble carbonates were prepared by immobilization of a racemic secondary alcohol (4-benzyloxy-2-butanol) onto low-molecular weight (av MW 550 and 750) monomethoxy PEG through a carbonate linker. For the screening of the hydrolytic enzymes, the substrate was enantioselectively hydrolyzed by commercially available lipase from porcine pancreas (PPL; Type II, Sigma) to afford the optically active compounds. In this system, the separation of the remaining (S)-substrate and the resulting (R)-alcohol was achieved by an extraction process without a laborious column chromatography. The (S)-carbonate was easily hydrolyzed with K2CO3 to afford the corresponding (S)-alcohol. Other MPEG-supported substrates were also hydrolyzed to afford the corresponding optically active alcohols.  相似文献

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) was used for the characterization of a partially transesterified poly(beta-hydroxyalkanoate) (PHA), a polymer produced by the bacterial strain Alcaligenes eutrophus with saponified vegetable oils as the sole carbon sources. The transesterification was carried out separately under acidic and basic conditions to obtain PHA oligomers weighing <10 kDa. The intact oligomers were detected in their cationized forms, [M + Na]+ and [M + K]+, by MALDI-TOFMS. A composition analysis, using the MALDI-TOF spectra, indicated that the oligomers obtained via acid catalysis contained a methyl 3-hydroxybutyrate end group, and those obtained by base catalysis had a methyl crotonate (olefinic) end group. In addition to hydroxybutyrate (HB), the oligomers were found to contain a small percentage of hydroxyvalerate, which was independently confirmed by gas chromatography/mass spectrometry. In comparison, analysis of a commercial PHA polymer, transesterified under identical conditions, showed only the presence of HB, i.e., a pure poly(HB) homopolymer.  相似文献

The selection of an appropriate internal standard (IS) for quantification by matrix-assisted laser desorption/ionization (MALDI) mass spectrometry is critical for the successful application of quantitative MALDI. Selection of the IS depends on the chemical similarity of the analyte and IS and the mass separation of the analyte and IS as a function of instrumental peak resolution. For the quantification of bovine insulin, a series of internal standards including horse heart cytochrome C, bovine insulin chain B, des-pentapeptide human insulin, and des-octapeptide porcine insulin was investigated. Des-pentapeptide human insulin was found to be the most appropriate internal standard (relative standard deviation of the standard curve slope = 2.99%, correlation coefficient = 0.988 in the range of 0.5-0.4 μmol/L). Two methods for measuring of the MALDI signal intensity were evaluated, direct peak integration following subtraction of a linear background and non-linear least squares curve fitting. The combination of electrospray ionization (ESI) with Fourier transform mass spectrometry (FTMS) is a powerful tool in characterizing synthetic polymers. ESI permits the generation of intact, multiply charged high mass ions, whereas FTMS provides high mass resolution and correspondingly improved mass accuracy. However, under “nonideal” empirically determined operating conditions, a mass discrimination effect occurs as a function of accumulation time that can result in significant differences for calculated average mass values (Mn, Mw) and polydispersities. A multidimensional tuning process to eliminate the deleterious effects of mass bias is demonstrated for several sodiated poly(ethylene glycol) samples containing oligomers with masses covering a 600–3350 Da mass regime. In addition, experiments are performed in order to elucidate the possible mechanism(s) that cause the mass discrimination effect. It is proposed that extended collisions (reactive and nonreactive) occur in the hexapole to alter the energy (and velocity) distributions of the ions before injection into the trap. By choosing higher skimmer potentials, ions over a narrower energy “window” are preselected in the hexapole and exhibit lower overall mass bias effects.  相似文献

Quantitativeness of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) was elucidated using an equimolar mixture of uniform poly(ethylene glycol) (PEG) oligomers with no molecular weight distributions. Uniform PEG oligomers with degrees of polymerization n = 6-40 were separated from commercial PEG samples by preparative super-critical fluid chromatography. MALDI-TOF mass spectra of an equimolar mixture of the uniform PEG oligomers were recorded by adding a mixture of 2,5-dihydroxybenzoic acid as a matrix reagent and four chlorinated salts, i.e. LiCl, NaCl, KCl and RbCl. Remarkable non-quantitative effects were observed in the MALDI-TOF mass spectra in both the lower and higher molecular mass regions. At higher molecular masses greater than about 10(3), PEG oligomers with larger molecular mass yielded lower spectral intensities irrespective of the species of adduct cations and higher laser powers induced larger decreases in mass spectral intensities with the increase in their molecular masses. On the other hand, in the lower molecular mass region, less than about 10(3), the observed non-quantitative effect greatly depends on the species of adduct cations, indicating that the stability of the PEG-cation complex affects the MALDI-TOF mass spectral intensities of uniform PEG oligomers.  相似文献

Poly(butylene terephthalate) (PBT) samples have been analyzed with size-exclusion chromatography (SEC) using a mixed solvent of 1,1,1,3,3,3-hexafluoroisopropanol (HFIP) and chloroform as the mobile phase. Several matrices and different sample deposition methods have been investigated to analyze PBT with matrix-assisted laser desorption/ionization (MALDI) time-of-flight (TOF) mass spectrometry (MS). Optimum results have been acquired by depositing PBT on top of a 2,4,6-trihydroxyacetophenone matrix. The found MALDI-TOF-MS method can be used to analyze the end group functionalities of PBT, as demonstrated with the samples at hand. By combining SEC (off-line) with MALDI-TOF-MS, absolute molecular masses of PBT can be measured, and these have been found to be considerably lower than those determined with SEC using polystyrene standards.  相似文献

A study has been undertaken to evaluate the usefulness of MALDI Q-TOF data for protein identification. The comparison of MS data of protein digests obtained on a conventional MALDI TOF instrument to the MS data from the MALDI Q-TOF reveal peptide patterns with similar intensity ratios. However, comparison of MS/MS Q-TOF data produced by nanoelectrospray versus MALDI reveals striking differences. Peptide fragment ions obtained from doubly charged precursors produced by nanoelectrospray are mainly y-type ions with some b-ions in the lower mass range. In contrast, peptide fragment ions produced from the singly charged ions originating from the MALDI source are a mixture of y-, b- and a-ions accompanied by ions resulting from neutral loss of ammonia or water. The ratio and intensity of these fragment ions is found to be strongly sequence dependent for MALDI generated ions. The singly charged peptides generated by MALDI show a preferential cleavage of the C-terminal bond of acidic residues aspartic and glutamic acid and the N-terminal bond of proline. This preferential cleavage can be explained by the mobile proton model and is present in peptides that contain both arginine and an acidic amino acid. The MALDI Q-TOF MS/MS data of 24 out of 26 proteolytic peptides produced by trypsin or Asp-N digestions were successfully used for protein identification via database searching, thus indicating the general usefulness of the data for protein identification. De novo sequencing using a mixture of 160/18O water during digestion has been explored and de novo sequences for a number of peptides have been obtained.  相似文献

Stable, pendant polyethylene oxide (PEO) layers were formed on medical-grade Pellethane? and Tygon? polyurethane surfaces, by adsorption and gamma-irradiation of PEO-polybutadiene-PEO triblock surfactants. Coated and uncoated polyurethanes were challenged individually or sequentially with nisin (a small polypeptide with antimicrobial activity) and/or fibrinogen, and then analyzed with time-of-flight secondary ion mass spectrometry (TOF-SIMS). Data reduction by robust principal components analysis (PCA) allowed detection of outliers, and distinguished adsorbed nisin and fibrinogen. Fibrinogen-contacted surfaces, with or without nisin, were very similar on uncoated polymer surfaces, consistent with nearly complete displacement or coverage of previously-adsorbed nisin by fibrinogen. In contrast, nisin-loaded PEO layers remained essentially unchanged upon challenge with fibrinogen, suggesting that the adsorbed nisin is stabilized within the pendant PEO layer, while the peptide-loaded PEO layer retains its ability to repel large proteins. Coatings of PEO loaded with therapeutic polypeptides on medical polymers have the potential to be used to produce anti-fouling and biofunctional surfaces for implantable or blood-contacting devices.  相似文献

The potential of matrix-assisted laser desorption ionization (MALDI) and MALDI-post-source decay (PSD) time-of-flight mass spectrometry for the characterization of peptides and proteins is discussed. Recent instrumental developments provide for levels of sensitivity and accuracy that make these techniques major analytical tools for proteome analysis. New software developments employing protein database searches have greatly enhanced the fields of application of MALDI-PSD. Peptides and proteins can be easily identified even if only a partial sequence information is determined. Derivatization procedures have been optimized for MALDI-PSD to increase the structural information and to obtain a complete peptide sequence even in critical cases. They are fast, simple and can be performed on target. MALDI-PSD is also a very powerful tool to characterize or elucidate post-translational or chemically induced modifications. In association with database searches, proteins issued from electrophoretic gels can be identified after specific enzymatic cleavages and peptide mapping.  相似文献

The surface of polyethersulfone (PES) membrane was modified by blending triblock copolymers of methoxyl poly(ethylene glycol)-polyurethane-methoxyl poly(ethylene glycol) (mPEG-PU-mPEG), which were synthesized through solution polymerization with mPEG Mns of 500 and 2000, respectively. The PES and PES/mPEG-PU-mPEG blended membranes were prepared through spin coating coupled with liquid-liquid phase separation. FTIR and (1)H NMR analysis confirmed that the triblock copolymers were successfully synthesized. The functional groups and morphologies of the membranes were studied by ATR-FTIR and SEM, respectively. It was found that the triblock copolymers were blended into PES membranes successfully, and the morphologies of the blended membranes were somewhat different from PES membrane. The water contact angles and platelet adhesion were decreased after blending mPEG-PU-mPEG into PES membranes. Meanwhile, the activated partial thromboplastin time (APTT) for the blended membranes increased. The anti-protein-fouling property and permeation property of the blended membranes improved obviously. SEM observation and 3-(4, 5-Dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assay proved the surfaces of the blended membranes promoted human hepatocytes adhesion and proliferation better than PES membrane.  相似文献
