The double [3 + 2] photocycloaddition reaction involving arenyl-dienes has been used to assemble seven separate [5.5.5.5] fenestrane structures that include ether and aza variants. The primary photolysis step was a meta photocycloaddition reaction, while a secondary photocycloaddition step formed the fenestrane structure. Investigations involving the insertion of an additional methylene group into the basic arenyl-diene skeleton failed to afford the desired [5.5.5.6] fenestrane structure. The presence of an oxime moiety in the aromatic photosubstrate allowed the primary photolysis step to take place; however, an attempted secondary photocycloaddition reaction involving the oxime did not provide the intended polyheterocyclic fenestrane. An alternative strategy to form various "criss-cross" double meta photocycloadducts was investigated and led to the discovery of a Paterno-Büchi cycloaddition reaction between acetone and an angular meta photocycloadduct. Other novel thermally and photochemically mediated skeletal rearrangement reactions were also recorded.  相似文献

We have studied the base-promoted heterocyclization of alkyl N-(cis(trans)-3,trans(cis)-4-dibromocyclohex-1-yl)carbamates and N-(cis(trans)-3,trans(cis)-4-dibromocyclohex-1-yl)-2,2,2-trifluoroacetamides, investigating the effect of the nitrogen protecting group and the relative configuration of the leaving group at C3 and C4 on the outcome of this reaction. We have observed that the sodium hydride-promoted heterocyclization of alkyl N-(cis-3,trans-4-dibromocyclohex-1-yl)carbamates (10, 12, 14, 16, 18) is a convenient method for the synthesis of 7-azabicyclo[2.2.1]heptane derivatives. For instance, the reaction of tert-butyl N-(cis-3,trans-4-dibromocyclohex-1-yl)carbamate (10) with sodium hydride in DMF at room temperature provides 2-bromo-7-[(tert-butoxy)carbonyl]-7-azabicyclo[2.2.1]heptane (2) (52% yield), whose t-BuOK-promoted hydrogen bromide elimination affords 7-[(tert-butoxy)carbonyl]-7-azabicyclo[2.2.1]hept-2-ene (31) in 78% yield, an intermediate in the total synthesis of epibatidine (1). However, the NaH/DMF-mediated heterocyclization of alkyl N-(trans-3,cis-4-dibromocyclohex-1-yl)carbamates (11, 13) is a more structure dependent reaction, where the nucleophilic attack of the oxygen atom of the protecting group controls the outcome of the reaction, giving rise to benzooxazolone and 2-oxa-4-azabicyclo[3.3.1]non-3-ene derivatives, respectively, from low to moderate yields, in complex reaction mixtures. Conversely, the NaH/DMF heterocyclizations of N-(cis-3,trans-4-dibromocyclohex-1-yl)-2,2,2-trifluoroacetamide (40) or N-(trans-3,cis-4-dibromocyclohex-1-yl)-2,2,2-trifluoroacetamide (42) are very clean reactions giving 7-azabicyclo[2.2.1]heptane or 2-oxa-4-azabicyclo[3.3.1]non-3-ene derivatives, respectively, in good yields. Finally, a mechanistic investigation, based on DFT calculations, has been carried out to rationalize the formation of the different adducts.  相似文献

Two heterotrinuclear oligomeric complexes [trans-RuCl(C[triple bond, length as m-dash]Cpy-4)(dppm)(2)](2)[MCl(2)] (M = Pd ; M = Pt ) are prepared from the metalloligand trans-[RuCl(C[triple bond, length as m-dash]Cpy-4)(dppm)(2)] (dppm = Ph(2)PCH(2)PPh(2), ). The resultant linear alignment of the metals [Ru-M-Ru] is imposed by a combinative use of trans-directed spacers and planar metals with trans-juxtaposed donor sites. Ligand exchange of with [Pd(CH(3)CN)(4)][PF(6)](2) gives trans-[Ru(CH(3)CN)(C[triple bond, length as m-dash]Cpy-4)(dppm)(2)][PF(6)] (). All complexes are characterized by single-crystal X-ray crystallography and solution spectroscopy. Acid-base titration on suggested protonation of the pendant pyridyl. Complexes and also undergo protonation at the C[triple bond, length as m-dash]C moiety under acid conditions. The inter-conversion of alkynyl and vinylidene functionality is described. The dual acid and base characters of makes it a potential metalloligand towards basic and acidic fragments in multinuclear heterometallic assemblies.  相似文献

The acetylido methyl iron(II) complexes, cis/trans-[Fe(dmpe)(2)(C[triple bond]CR)(CH(3))] (1) and trans-[Fe(depe)(2)(C[triple bond]CR)(CH(3))] (2) (dmpe = 1,2-dimethylphoshinoethane; depe = 1,2-diethylphosphinoethane), were synthesized by transmetalation from the corresponding alkyl halide complexes. Acetylido methyl iron(II) complexes were also formed by transmetalation from the chloride complexes, trans-[Fe(dmpe)(2)(C[triple bond]CR)(Cl)] or trans-[Fe(depe)(2)(C[triple bond]CR)(Cl)]. The structure of trans-[Fe(dmpe)(2)(C[triple bond]CC(6)H(5))(CH(3))] (1a) was determined by single-crystal X-ray diffraction. The methyl acetylido iron complexes, [Fe(dmpe)(2)(C[triple bond]CR)(CH(3))] (1), are thermally stable in the presence of acetylenes; however, under UV irradiation, methane is lost with the formation of a metal bisacetylide. Photochemical metathesis of cis- or trans-[Fe(dmpe)(2)(CH(3))(C[triple bond]CR)] (R = C(6)H(5) (1a), 4-C(6)H(4)OCH(3) (1b)) with terminal acetylenes was used to selectively synthesize unsymmetrically substituted iron(II) bisacetylide complexes of the type trans-[Fe(dmpe)(2)(C[triple bond]CR)(C[triple bond]CR')] [R = Ph, R' = Ph (6a), 4-CH(3)OC(6)H(4) (6b), (t)()Bu (6c), Si(CH(3))(3) (6d), (CH(2))(4)C[triple bond]CH (6e); R = 4-CH(3)OC(6)H(4), R' = 4-CH(3)OC(6)H(4), (6g), (t)()Bu (6h), (CH(2))(4)C[triple bond]CH (6i), adamantyl (6j)]. The structure of the unsymmetrical iron(II) bisacetylide complex trans-[Fe(dmpe)(2)(C[triple bond]CC(6)H(5))(C[triple bond]CC(6)H(4)OCH(3))] (6b) was determined by single-crystal X-ray diffraction. The photochemical metathesis of the bis-acetylene, 1,7-octadiyne, with trans-[Fe(dmpe)(2)(CH(3))(C[triple bond]CPh)] (1a), was utilized to synthesize the bridged binuclear species trans,trans-[(C(6)H(5)C[triple bond]C)Fe(dmpe)(2)(mu-C[triple bond]C(CH(2))(4)C[triple bond]C)Fe(dmpe)(2)(C[triple bond]CC(6)H(5))] (11). The trinuclear species trans,trans,trans-[(C(6)H(5)C[triple bond]C)Fe(dmpe)(2)(mu-C[triple bond]C(CH(2))(4)C[triple bond]C)Fe(dmpe)(2)(mu-C[triple bond]C(CH(2))(4)C[triple bond]C)Fe(dmpe)(2)(C[triple bond]CC(6)H(5))] (12) was synthesized by the photochemical reaction of Fe(dmpe)(2)(C[triple bond]CPh)(C[triple bond]C(CH(2))(4)C[triple bond]CH) (6e) with Fe(dmpe)(2)(CH(3))(2). Extended irradiation of the bisacetylide complexes with phenylacetylene resulted in insertion of the terminal alkyne into one of the metal acetylide bonds to give acetylide butenyne complexes. The structure of the acetylide butenyne complex, trans-[Fe(dmpe)(2)(C[triple bond]CC(6)H(4)OCH(3))(eta(1)-C(C(6)H(5))=CH(C[triple bond]CC(6)H(4)OCH(3)))] (9a) was determined by single-crystal X-ray diffraction.  相似文献

The [4 + 2] cycloaddition of trans,trans-2,4-hexadiene with C(60) proceeds via a concerted mechanism with retention of stereochemistry in the cycloadduct 1a. However, when cis,trans-2,4-hexadiene reacts with C(60), isomerization of the cis,trans to the thermodynamically more stable trans,trans isomer occurs. Subsequently, the cis,trans diene isomerized to the trans,trans isomer and cycloadds to C(60), to form adduct 1a. When the reaction is carried out at higher temperatures, the formation of cycloadduct 1b is also obtained. This result is consistent with a concerted cycloaddition of cis,trans-2,4-hexadiene with C(60), which is more reactive at elevated temperatures and leads to the formation of the Diels-Alder adduct 1b.  相似文献

The synthesis and spectroscopic properties of trans-[RuL4(C[triple bond]CAr)2] (L4 = two 1,2-bis(dimethylphosphino)ethane, (dmpe)2; 1,5,9,13-tetramethyl-1,5,9,13-tetraazacyclohexadecane, 16-TMC; 1,12-dimethyl-3,4:9,10-dibenzo-1,12-diaza-5,8-dioxacyclopentadecane, N2O2) are described. Investigations into the effects of varying the [RuL4] core, acetylide ligands, and acetylide chain length for the [(-)C[triple bond]C(C6H4C[triple bond]C)(n-1)Ph] and [(-)C[triple bond]C(C6H4)(n-1)Ph] (n = 1-3) series upon the electronic and electrochemical characteristics of trans-[RuL4(C[triple bond]CAr)2](0/+) are presented. DFT and TD-DFT calculations have been performed on trans-[Ru(L')4(C[triple bond]CAr)2](0/+) (L' = PH3 and NH3) to examine the metal-acetylide pi-interaction and the nature of the associated electronic transition(s). It was observed that (1) the relationship between the transition energy and 1/n for trans-[Ru(dmpe)2{C[triple bond]C(C6H4C[triple bond]C)(n-1)Ph}2] (n = 1-3) is linear, and (2) the sum of the d(pi)(Ru(II)) --> pi*(C[triple bond]CAr) MLCT energy for trans-[Ru(16-TMC or N2O2)(C[triple bond]CAr)2] and the pi(C[triple bond]CAr) --> d(pi)(Ru(III)) LMCT energy for trans-[Ru(16-TMC or N2O2)(C[triple bond]CAr)2]+ corresponds to the intraligand pi pi* absorption energy for trans-[Ru(16-TMC or N2O2)(C[triple bond]CAr)2]. The crystal structure of trans-[Ru(dmpe)2{C[triple bond]C(C6H4C[triple bond]C)2Ph}2] shows that the two edges of the molecule are separated by 41.7 A. The electrochemical and spectroscopic properties of these complexes can be systematically tuned by modifying L4 and Ar to give E(1/2) values for oxidation of trans-[RuL4(C[triple bond]CAr)2] that span over 870 mV and lambda(max) values of trans-[RuL4(C[triple bond]CAr)2] that range from 19,230 to 31,750 cm(-1). The overall experimental findings suggest that the pi-back-bonding interaction in trans-[RuL4(C[triple bond]CAr)2] is weak and the [RuL4] moiety in these molecules may be considered to be playing a "dopant" role in a linear rigid pi-conjugated rod.  相似文献

The thermal conversion of cis-bicyclo[4.2.0]oct-7-ene to cis,cis-1,3-cyclooctadiene might involve a direct disrotatory ring opening, or it might possibly take place by way of cis,trans-1,3-cyclooctadiene. This cis,trans-diene might possibly form the more stable cis,cis isomer through a [1,5] hydrogen shift or a trans-to-cis isomerization about the trans double bond. Deuterium kinetic isotope effect determinations for the isomerizations of 2,2,5,5-d(4)-bicyclo[4.2.0]oct-7-ene and 7,8-d(2)-bicyclo[4.2.0]oct-7-ene rule out these two alternatives because the observed effects are much smaller than would be anticipated for these mechanisms: k(H)/k(D)(d(4)) at 250 degrees C is 1.17 (1.04 per D), and k(H)/k(D)(d(2)) at 238 degrees C is 1.20 (1.10 per D). The direct disrotatory ring opening route remains the preferred mechanism.  相似文献

Two methodologies of C-C bond formation to achieve organometallic complexes with 7 or 9 conjugated carbon atoms are described. A C7 annelated trans-[Cl(dppe)2Ru=C=C=C-CH=C(CH2)-C[triple bond]C-Ru(dppe)2Cl][X] (X = PF6, OTf) complex is obtained from the diyne trans-[Cl(dppe)2Ru-(C[triple bond]C)2-R] (R = H, SiMe3) in the presence of [FeCp2][PF6] or HOTf, and C7 or C9 complexes trans-[Cl(dppe)2Ru-(C[triple bond]C)n-C(CH3)=C(R1)-C(R2)=C=C=Ru(dppe)2Cl][X] (n = 1, 2; R1 = Me, Ph, R2 = H, Me; X = BF4, OTf) are formed in the presence of a polyyne trans-[Cl(dppe)2Ru-(C[triple bond]C)n-R] (n = 2, 3; R = H, SiMe3) with a ruthenium allenylidene trans-[Cl(dppe)2Ru=C=C=C(CH2R1)R2][X]. These reactions proceed under mild conditions and involve cumulenic intermediates [M+]=(C=)nCHR (n = 3, 5), including a hexapentaenylidene. A combination of chemical, electrochemical, spectroscopic (UV-vis, IR, NIR, EPR), and theoretical (DFT) techniques is used to show the influence of the nature and conformation of the bridge on the properties of the complexes and to give a picture of the electron delocalization in the reduced and oxidized states. These studies demonstrate that the C7 bridging ligand spanning the metal centers by almost 12 angstroms is implicated in both redox processes and serves as a molecular wire to convey the unpaired electron with no tendency for spin localization on one of the halves of the molecules. The reactivity of the C7 complexes toward protonation and deprotonation led to original bis(acetylides), vinylidene-allenylidene, or carbyne-vinylidene species such as trans-[Cl(dppe)2Ru[triple bond]C-CH=C(CH3)-CH=C(CH3)-HC=C=Ru(dppe)2Cl][BF4]3.  相似文献

Conditions to prepare trans-[Ru2(dmba)4(C[triple chemical bond]CAr)2] from [Ru2(dmba)4(NO(3))2] (DMBA=N,N'-dimethylbenzamidinate) and HC[triple chemical bond]CAr were optimized; Et2NH was found to be the most effective among a number of weak bases in facilitating the product formation. Furthermore, a series of unsymmetric trans-[(ArC[triple chemical bond]C)Ru(2)(dmba)4(C[triple chemical bond]CAr')] compounds were prepared under optimized conditions, in which one or both of Ar and Ar' are donor (NMe2)-/acceptor (NO(2))-substituted phenyls. While the X-ray crystallographic studies revealed a minimal structural effect upon donor/acceptor substitution, voltammetric measurements indicated a significant influence of substituents on the energy level of frontier orbitals. In particular, placing a donor and an acceptor on the opposite ends of trans-[(ArC[triple chemical bond]C)Ru2(dmba)4(C[triple chemical bond]CAr')] moiety results in an energetic alignment of frontier orbitals that favors a directional electron flow, a necessary condition for unimolecular rectification.  相似文献

The reaction of trans-[RuCl(2)(PPh(3))(3)] (Ph = C(6)H(5)) with 2-thio-1,3-pyrimidine (HTPYM) and 6-thiopurines (TPs) produced mainly crystalline solids that consist of cis,cis,trans-[Ru(PPh(3))(2)(N,S-TPYM)(2)] (1) and cis,cis,trans-[Ru(PPh(3))(2)(N(7),S-TPs)(2)]X(2) (X = Cl(-), CF(3)SO(3)(-)). In the case of TPs, other coordination isomers have never been isolated and reported. Instead, the mother liquor obtained after filtration of 1 produced red single crystals of trans,cis,cis-[Ru(PPh(3))(2)(N,S-TPYM)(2)].2H(3)O(+).2Cl(-) (2.2H(3)O(+).2Cl(-)). Selected ruthenium(II)-thiobase complexes were studied for their structural, reactivity, spectroscopic, redox, and cytotoxic properties. Single crystals of 1 contain thiopyrimidinato anions chelated to the metal center via N and S. The Ru[bond]N bonds are significantly elongated for 1 [2.122(2) and 2.167(2) A] with respect to 2 [2.063(3) A] because of the trans influence from PPh(3). The coordination pseudo-octahedron for 2 is significantly elongated at the apical sites (PPh(3) ligands). Solutions of cis,cis,trans isomers in air are stable for weeks, whereas those of 2 turn green within 24 h, in agreement with the respective redox potentials. cis,cis,trans- and trans,cis,cis-[Ru(PH(3))(2)(N,S-TPYM)(2)], as optimized through the DFT methods at the Becke3LYP level are in good agreement with experimental geometrical parameters (1 and 2), with cis,cis,trans being more stable than trans,cis,cis by 3.88 kcal. The trend is confirmed by molecular modeling based on semiempirical (ZINDO/1) and molecular mechanics (MM) methods. Cytotoxic activity measurements for cis,cis,trans-[Ru(PPh(3))(N-THZ)(N(7),S -H(2)TP)(2)]Cl(2) (4) (THZ = thiazole, H(2)TP = 6-thiopurine) and cis,cis,trans-[Ru(PPh(3))(2)(N(7),S-HTPR)2]Cl(2) (5) (HTPR = 6-thiopurine riboside) against ovarian cancer cells A2780/S gave IC(50) values of 17 +/- 1 and 29 +/- 9 microM, respectively. Furthermore, the spectral analysis of HTPYM, TPs, and their Ru(II) complexes in solution shows that intense absorptions occur in the UVA/vis region of light, whereas standard nucleobases absorb in the UVB region.  相似文献

The synthesis and characterization of 10-(o-alkoxyphenyl)phenoxarsines 2-ROC6H4As(C6H4)2O (R = H, Me, and Pri, As(C6H4)2O = phenoxarsine) and their platinum(II) and palladium(II) complexes cis-[PtCl2{2-PriOC6H4As(C6H4)2O-kappaAs}2] (1), trans-[PdCl2{2-PriOC6H4As(C6H4)2O-kappaAs}2] (2), cis-[PtCl2{2-HOC6H4As(C6H4)2O-kappaAs}2] (3), cis-[PdCl2{2-HOC6H4As(C6H4)2O-kappaAs}2] (4), cis-[PtI2{2-MeOC6H4As(C6H4)2O-kappaAs}2] (5), and trans-[PdCl2{2-MeOC6H4As(C6H4)2O-kappaAs}2] (6) are reported. The chelate complex cis-[Pt{2-OC6H4As(C6H4)2O-kappaAs,O}2] (7) is also described. The molecular structures of 1-4 and 7 were determined. The short As...O intramolecular interaction found in complexes 1-4 in the solid state was also verified by calculations at the B3LYP/LANL2DZ level for complex 2 and for 10-(o-isopropoxyphenyl)phenoxarsine in the gas phase, and this suggests that the interaction is a characteristic of the ligand rather than a packing effect. Calculations at the B3LYP/LANL2DZ and Oniom(B3LYP/LANL2DZ:uff) levels for complexes 1-4 showed that the solvent plays a crucial role in the crystallization (through geometry constraints) of the kinetically stable cis isomers.  相似文献

The reaction of tetrapyrazine Pt(II) with adenine under basic conditions yielded two products both disubstituted by adenine and bound in all cases to the N-9 nitrogen of adenine. Crystals amenable to X-crystallographic analysis were obtained for one product which was consequently identified as trans-[Pt(II)(N9-adeH)2(pz)2](NO3)(4).H2O. The other product, though, was identified as the cis isomer based on extensive and comparative NMR structural studies whereby the two compounds were examined as both neutral and tetraprotonated species in solution. The major product of the reaction was unexpectedly the cis isomer, thus obtained in contrast to the trans effect, and the reason for this result is speculated on. Computational calculations using DFT at the B3LYP/TZVP-MARI-J level of theory provided the head-to-tail conformer as the overwhelmingly more stable species over the head-to-head conformer for both compounds whilst the trans product was found to be more stable than the cis. Thus the reaction does not follow a thermodynamic course and rather is kinetically controlled in concert with the speculated mode of reaction.  相似文献

Enantiomerically pure cis and trans isomers of 4-acetoxy-[eta3(1,2,3)-cyclohexenyl]palladium chloride dimers (cis-1 and trans-1) were prepared from enantiomerically pure trans-1-acetoxy-4-chloro-2-cyclohexene. X-ray analyses of these complexes show that in the trans complex (trans-1) the six-membered ring prefers a chair conformation, whereas in the cis complex (cis-1) the cyclohexenyl ring has a boat conformation. According to the X-ray structure of trans-1 the Pd-C3 bond is shorter than the other allylic terminal palladium-carbon bond (Pd-C1). On the other hand, in cis-1 the Pd-C3 and Pd-C1 bond lengths are identical within the experimental error. The calculated structures (B3PW91/LANL2DZ + P) of trans-1 and cis-1 also display differences in the allylpalladium bonding. The asymmetric allylpalladium bonding in trans-1 is explained on the basis of pi-sigma electronic interactions between the 4-acetoxy substituent and the allyl-metal moiety.  相似文献

Chlorination of [14]aneS(4)-ol (1,4,8,11-tetrathiatetradecan-6-ol) and cis/trans-[14]aneS(4)-diol (cis/trans-1,4,8,11-tetrathiatetradecane-6,13-diol) yields the corresponding dichloro-substituted macrocycles [14]aneS(4)-Cl (1,4,8,11-tetrathiatetradecane 6-chloride) and cis/trans-[14]aneS(4)-Cl(2) (cis/trans-1,4,8,11-tetrathiatetradecane 6,13-dichloride) in good yield. Thiomethylation of the chlorides produces the ring-contracted pendent thioether macrocycles [13]aneS(4)-CH(2)SCH(3) (1,4,7,10-tetrathiatridecane-5-(methylthio)methane) and cis/trans-anti-[12]aneS(4)-(CH(2)SCH(3))(2) (1,4,7,10-tetrathiadodecane-5,11-bis((methylthio)methane)). The mechanism of the ring contraction reaction is discussed in terms of the reactivity of the monochlorinated macrocycle toward ring contraction and the stereochemistry of the chlorinated intermediates and the thiomethylated products, which are based on the X-ray crystal structure analyses of trans-[14]aneS(4)-Cl(2) and trans-anti-[12]aneS(4)-(CH(2)SCH(3))(2).  相似文献

Reactions of or with acetone oxime , cyclic nitrone (-)O(+)N = CHCH(2)CH(2)C[upper bond 1 end]Me(2) or N,N-diethylhydroxylamine give access, in moderate to good yields, to the unsymmetric mixed ligand oxadiazoline and/or imine complexes trans-[PtCl(2)(oxadiazoline-a)(imine-a)] , trans-[PtCl(2)(oxadiazoline-a)(oxadiazoline-b)] [oxadiazoline-b = [upper bond 1 start]N[double bond, length as m-dash]C(CH(2)CO(2)Me)O[lower bond 1 start]NC[upper bond 1 end](H)CH(2)CH(2)C[lower bond 1 end]Me(2)], trans-[PtCl(2)(imine-a)(imine-b)] [imine-b = NH = C(CH(2)CO(2)Me)ONEt(2)] or trans-[PtCl(2)(imine-a)(oxadiazoline-b)] . The cis mono-imine mixed ligand complex cis-[PtCl(2)(imine-a)(NCCH(2)CO(2)Me)] is the major product from the reaction of cis-[PtCl(2)(NCCH(2)CO(2)Me)(2)] with the oxime , while the di-imine compound cis-[PtCl(2)(imine-a)(2)] is a minor product. Reaction of cis-[PtCl(2)(imine-a)(NCCH(2)CO(2)Me)] with N,N-diethylhydroxylamine or the cyclic nitrone affords, in good yields, the unsymmetric mixed ligand complexes cis-[PtCl(2)(imine-a)(imine-b)] or cis-[PtCl(2)(imine-a)(oxadiazoline-b)] , respectively. All these complexes were characterized by elemental analyses, IR and (1)H, (13)C and (195)Pt NMR spectroscopies, and FAB(+)-MS. The X-ray structural analysis of trans-[PtCl(2){NH=C(CH(2)CO(2)Me)ON=CMe(2)}(NCCH(2)CO(2)Me)] is also reported.  相似文献

The synthesis, characterization, and cytotoxicity of eight new platinum(IV) complexes having the general formula cis,cis,trans-[Pt(NH(3))(2)Cl(2)(O(2)CNHR)(2)] are reported, where R = tert-butyl (4), cyclopentyl (5), cyclohexyl (6), phenyl (7), p-tolyl (8), p-anisole (9), 4-fluorophenyl (10), or 1-naphthyl (11). These compounds were synthesized by reacting organic isocyanates with the platinum(IV) complex cis,cis,trans-[Pt(NH(3))(2)Cl(2)(OH)(2)]. The electrochemistry of the compounds was investigated by cyclic voltammetry. The aryl carbamate complexes 7-11 exhibit reduction peak potentials near -720 mV vs Ag/AgCl, whereas the alkyl carbamate complexes display reduction peak potentials between -820 and -850 mV vs Ag/AgCl. The cyclic voltammograms of cis,cis,trans-[Pt(NH(3))(2)Cl(2)(O(2)CCH(3))(2)] (1), cis,cis,trans-[Pt(NH(3))(2)Cl(2)(O(2)CCF(3))(2)] (2), and cis-[Pt(NH(3))(2)Cl(4)] (3) were measured for comparison. Density functional theory studies were undertaken to investigate the electronic structures of 1-11 and to determine their adiabatic electron affinities. A linear correlation (R(2) = 0.887) between computed adiabatic electron affinities and measured reduction peak potentials was discovered. The biological activity of 4-11 and, for comparison, cisplatin was evaluated in human lung cancer A549 and normal MRC-5 cells by the MTT assay. The compounds exhibit comparable or slightly better activity than cisplatin against the A549 cells. In MRC-5 cells, all are equally or slightly less cytotoxic than cisplatin, except for 4 and 5, which are more toxic.  相似文献

Diethylaluminum trimethylsilylacctylide reacted stereospecifically with the mesylate of trans-2-(N- pyrrolidinyl)cyclohexanol (2) to give trans-N-[2-(trimethylsilylacetylcnyl)cyclohexyl]pyrrolidine (3). The trimethylsilyl group in 3 was displaced with a 3,4-dichlorobenzoyl group to give ynone 4, which then underwent 1,4-addition with lithium dimethylcuprate to give trans enone 6 and its cis isomer 5 in a ratio 5:1. Reduction of the enones with NaBHVCeCl3 resulted in cis and trans allylic alcohols 9 and 10. Both 9 and 10 were successfully deoxygenated with Znl2-NaCNBH3 reducing system to give trans-(E)-N-{2-[4-(3,4- dichlorophenyl)bul-2-en-2-yl]cyclohexyl}pyrrolidine (11) as the onty product. Olefin 11, a carbon-carbon double bond isostere of the prototypek-selective opioid analgesic U-50488, showed a 103 fold reduction in K-affinity (Kik = 1.6 × 104 nM vs 15 nM for U-50488).  相似文献

A single-crystal study of cis,trans-(L-N2S2)MoVOCl (1) doped into cis,trans-(N2S2)MoVIO2 (3) has enabled the g-tensor of 1 and its orientation with respect to the molecular structure to be determined. The EPR parameters (g1, 2.004; g2, 1.960; g3, 1.946; A1, 71.7 x 10(-4) cm(-1); A2, 11.7 x 10(-4) cm(-1); A3, 32.0 x 10(-4) cm(-1)) of cis,trans-(L-N2S2)MoVOCl [L-N2S2H2 = N,N'-dimethyl-N,N'-bis(mercaptophenyl)ethylenediamine] mimic those of the low-pH form of sulfite oxidase and the "very rapid" species of xanthine oxidase. The principal axis that corresponds to g1 is rotated approximately 10 degrees from the Mo[triple bond]O vector, while the principal axis that corresponds to g3 is located in the equatorial plane and approximately 38 degrees from the Mo-Cl vector. Independent theoretical calculations of the g-tensor of 1 were performed using two types of techniques: (1) the spectroscopically parametrized intermediate neglect of differential overlap technique (INDO/S) combined with single-excitation configuration interaction (CIS); (2) a scalar relativistic DFT (BP86 and B3LYP functionals) treatment using the zeroth order regular approximation to relativistic effects (ZORA) in combination with recently developed accurate multicenter mean field spin-orbit operators (RI-SOMF) and the estimation of solvent effects using dielectric continuum theory at the conductor-like screening model (COSMO) level. The excellent agreement between experiment and theory, as well as the high consistency between the INDO/S and BP86/ZORA results, provides a sound basis for analysis of the calculated orientation of the g-tensor for cis,trans-(L-N2S2)MoVO(SCH2Ph) (2), for which single-crystal EPR data are not available but which contains three equatorial sulfur donor atoms, as occurs in sulfite oxidase and xanthine oxidase. The implications of these results for the EPR spectra of the Mo(V) centers of enzymes are discussed.  相似文献
