2-Halophenyl ketones 1a-e (1a, o-IC(6)H(4)COCH(3)) undergo carbocyclization with alkyl propiolates (2a, CH(3)(CH(2))(4)C[triple bond]CCO(2)CH(3); 2b, TMSC[triple bond]CCO(2)Et 2c, CH(3)C[triple bond]CCO(2)CH(3); 2d, CH(3)OCH(2)C[triple bond]CCO(2)CH(3); 2e, CH(3)(CH(2))(3)C[triple bond]CCO(2)CH(3); 2f, PhC[triple bond]CCO(2)CH(3); and 2g, (CH(3))(3)C[triple bond]CCO(2)CH(3)) in the presence of Ni(dppe)Br(2) and zinc powder in acetonitrile at 80 degrees C to afford the corresponding indenol derivatives 3a-m with remarkable regioselectivity in good to excellent yields. The nickel-catalyzed carbocyclization reaction was successfully extended to other simple disubstituted alkynes. Thus, the reaction of 2-halophenyl ketones 1a-e with disubstituted alkynes (2h, PhC[triple bond]CPh; 2i, CH(3)C(6)H(4)C[triple bond]CC(6)H(4)CH(3); 2j, CH(3)CH(2)C[triple bond]CCH(2)CH(3); 2k, PhC[triple bond]CCH(3); 2l, TMSC[triple bond]CCH(3); and 2m, PhC[triple bond]C(CH(2))(3)CH(3)) proceeded smoothly to afford the corresponding indenols 4a-t in good to excellent yields. For unsymmetrical alkynes 2k-m, the carbocyclization gave two regioisomers with regioselectivities ranging from 1:2 to 1:12 depending on the substituents on the alkyne and on the aromatic ring of halophenyl ketone. A possible mechanism for this nickel-catalyzed carbocyclization reaction is also proposed.  相似文献

Simple enolizable ketones such as acetophenones and benzalacetones were treated with malononitrile under Vilsmeier-Haack reaction conditions to afford 2-chloronicotinonitriles. The reaction proceeds via a one-pot chloromethyleneiminium salt mediated three-component reaction followed by sequential cyclization and aromatization under Vilsmeier-Haack reaction conditions.  相似文献

A series of aryltriethoxysilanes was synthesized from the reaction mixture of aryl bromide, Mg powder, 1,2-dibromoethane and tetraethyl orthosilicate in THF under ultrasound. This sonochemical Barbier-type reaction provides a simple and efficient method for preparation of aryltriethoxysilanes. The Hiyama cross-coupling reaction of phenyltriethoxysilane with bromobenzene was investigated under different palladium catalysts and Pd(PhCN)2Cl2 was found to be the best choice.  相似文献

A new route toward 4-acylamino- and 4-amino-substituted tetrahydroindazoles is disclosed. The title compounds are obtained in good to excellent yields in the Ritter reaction between 4-hydroxy-tetrahydroindazoles and various nitriles. The reactivity of the tetrahydroindazole-derived carbenium ion is both sufficiently high to react with trichloroacetonitrile and sufficiently selective to resist the azide functionality within its structure. The present approach adds a method to the toolbox of tetrahydroindazole chemistry and facilitates the structural modifications of the scaffold, which has found important applications in medicinal chemistry.  相似文献

A series of new bisquinoline systems, ethyl 2-((quinolin-8-yloxy)methyl)quinoline-3-carboxylate derivatives (3a–i), were synthesized by Williamson reaction of ethyl 2-(halogenmethyl)quinoline-3-carboxylates (4a–c) with 8-hydroxyquinolines (5a–c) in CH3CN as solvent and in the presence of K2CO3 as base. Characterization of these bisquinoline compounds was primarily achieved by use of spectral data and X-ray crystallography. This procedure enables easy access to bisquinoline compounds in a short reaction time and the products are obtained in high yields.  相似文献

Abstract

A new series of regioselective dispiropyrrolidine analogs via efficient one-pot four-component tandem reaction has been discovered. The reaction was carried out in the presence of four common reactants and magnesium silicate nanoparticles (NPs) in ethanol under microwave irradiation. The reaction proceeds rapidly and can be completed within 60–90?min. This methodology offers several significant advantages such as high yield, mild catalyst, and easy to perform. The synthesized compound was further screened for antibacterial and antiproliferative activities. The results showed that compound 5c and 5a were potent antibacterials against Gram-positive and Gram-negative bacteria. Further, 5a showed best antiproliferative activity against tested cell lines.  相似文献

Treatment of hindered borinic esters, R2 BOR′; with an equimolar quantity of α,α-dichloromethyl methyl ether and lithium triethylcarboxide yields the α-chloroboronic esters, R2 CHClB(OCH3)OR′, in excellent yields. Abstract  

New organic reactions allow chemical transformations which were previously unknown. Therefore, new reactions are important contributions to progress in the field of organic synthesis. Herein, we are reporting a simple, one-pot, efficient three-component synthesis of novel 3-chloro-4-[4-(2-oxo-2H-chromen-4-ylmethoxy)phenyl]-1-phenylazetidin-2-one derivatives using 4-(2-oxo-2H-chromen-4-ylmethoxy)benzaldehydes, anilines, and chloroacetyl chloride in the presence of triethyl amine as a catalyst under different conditions. Taking into account environmental and economic considerations, the protocol presented here has the merits of simple operation, convenient work-up, being environmentally benign, and providing good yields. The synthesized compounds were characterized by IR, 1H NMR, 13C NMR, MS, and elemental analysis.  相似文献

In the presence of a catalytic amount of Pd(PPh3)4, the reaction took place smoothly to give the corresponding 4-fluoroalkylated isoquinoline in high yield as a single isomer. A novel tandem one-pot method for the synthesis of polysubstituted pyridine derivatives has been developed via Knoevenagel-type reaction and subsequent 6π electrocyclization of the Blaise reaction intermediates and 3-formylchromones. An efficient synthesis of 4-arylcoumarins has been accomplished via Kostanecki reactions of 2-hydroxybenzophenones with acetic anhydride employing DBU at ambient temperature. Using the same strategy, several 2-acyloxybenzophenone derivatives were readily converted to 3,4-difunctionalized coumarins. This protocol offers a notable improvement in reaction conditions for coumarin synthesis and takes advantage of its synthetic capability, especially for highly functionalized 4-arylcoumarins with structural diversity.  相似文献

A simple and efficient procedure was developed for the synthesis of multisubstituted 2-aminothiophene derivatives. In the presence of catalytic amount of imidazole, ketones or aldehydes, dicyanomethane and elemental sulfur were converted into the corresponding 2-aminothiophene derivatives in moderate to high yields in DMF at 60 °C.  相似文献

The reaction of 1-adamantyl triflate (1) with carbon monoxide and adamantane catalyzed by triflic acid affords 3-hydroxy-4-homoadamantyl 1-adamantanecarboxylate (2) as a major product, which is easily converted to 3,4-homoadamantanediol (5) — a promising starting material for 3,4-bifunctional homoadamantane derivatives.  相似文献
