The NH,NH-dihydrocycloadduct 1 (6-chloro-9,9-dimethyl-7,8-dihydro-9H-pyrazolo[4,3-d]tetrazolo[1,5-b]-pyridazine) obtained by the cycloaddition of 2-diazopropane to 6-chlorotetrazolo[1,5-b]pyridazine, is transformed with protected or unprotected carbohydrates 2, 5, 7,9, 11, 13 and 15 in the presence of methyl acrylate ( 3 ) into the corresponding C-nucleosides 4,6,8,10,12,14 and 16 . In this one-pot synthesis two new chiral centers are formed stereoselectively, dependent on the chirality at C-atom next to aldehydo group in the carbohydrate.  相似文献

Trifluoroacetic acid and benzenesulfonic acid in dichloromethane at ambient temperature have been found to be efficient co-catalysts for the epimerization of C-nucleosides with electron-withdrawing substituents. This approach provides a convenient route to the β anomer of the nucleosides with high yields in the range 54-78%. Synthesis of the 3′-phosphoramidite of 2,4-difluorophenyl C-nucleoside suitable for solid-phase DNA synthesis is described.  相似文献

An efficient synthesis of pyrimidine-containing butenolides is reported, starting from C-alkoxycarbonyl isoxazolidines. Two competitive reaction routes are operating: the pathway leading to homo-N,O-nucleosides, based on the reduction of an ester group at C3, and the reaction channel leading to butenolides promoted by the removal of the hydrogen atom at C3. The two reaction pathways can be easily controlled according to the adopted experimental conditions  相似文献

We report the design and synthesis of triazolyl donor/acceptor unnatural C-nucleosides via alkyne (sugar)—azide (aromatic) 1, 3-dipolar cyclo-addition reaction as a key step and studies on their photophysical properties. We have chosen β-1′-ethynyl-2′-deoxyribose as a precursor to synthesize triazolyl-C-nucleosides. Overcoming the difficulties, we obtain β-1′-ethynyl-2′-deoxyribose as a major product following a Co2(CO)8 catalyzed intramolecular Nicholas reaction. The 1,3-diaxial interaction is the driving force for the α to β-anomeric conversion while performing cobalt complexation followed by oxidation to afford β-1′- ethynyl-2′-deoxyribose as the major product. A Cu(I)-catalyzed click reaction between different aromatic donor/acceptor azides and β-1′- ethynyl-2′-deoxyribose generates the desired unnatural triazolyl donor-acceptor aromatic C-nucleosides (cTBDo/Ac) within 30 min. Single crystal X-ray structure shows the puckered conformation of sugar as C3′-exo. Studies on the photophysical properties suggests good fluorophoric as well as solvatochromic characteristics of these nucleosides. Two of the synthesised nucleosides, cTAnthBDo and cTPyBDo, are found to interact with BSA as the only tested protein with quenching of fluorescence signal. The designed bases, thus, might find applications in stabilizing a DNA and in the biophysical study thereof, if a pair of such donor acceptor C-nucleosides could be incorporated into a DNA sequence.  相似文献

A new class of homo-N,O-nucleosides has been designed, based on the 1,3-dipolar cycloaddition of C-substituted nitrones with allyl nucleobases. The N-methyl-C-ethoxycarbonyl nitrone 1, and the C-α-silyloxymethyl-N-methyl nitrone 7 have been exploited: the stereochemical features of the obtained nucleosides are dependent on the nature of the dipole. The results obtained with DFT calculations fully agree with the experimental results and successfully reproduce the experimentally observed reversal of endo/exo selectivity for nitrones 1 and 7.  相似文献

The sequential reactions of 2-C-methyl-d-arabinose with cyanamide and methyl propiolate afford an anhydronucleoside, which may be opened under acid conditions with inversion at C2′, to give 2′-C-methyl uridine; ring opening with sodium hydroxide gave 2′-C-methyl arabino-uridine with retention of configuration at C2′. This gives complete stereospecific control to yield only β-nucleosides.  相似文献

1,3-Dimethyluracil ( 1 ), a versatile synthon for the synthesis of various heterocycles, reacted readily with 3-aminopyrazoles 2 in sodium ethoxide to give pyrazolo[1,5-a]pyrimidines 3 . Under similar conditions, 3-aminopyrazole C-nucleosides 4 and the synthon 1 gave a mixture of pyrazolo[1,5-a]pyrimidine C-nucleosides, which was separated on a silica gel column. Attempts to remove the protecting groups yielded pyranose derivative 10 . Another synthon 1,3-dimethyl-5-azauracil and 3-aminopyrazoles 12 gave pyrazolo[1,5-a]triazines 13 . In a similar reaction with 3-aminopyrazole C-nucleosides 4 gave the corresponding pyrazolo[1,5-a]-triazine C-nucleosides 14 and 15 .  相似文献

Methyl 2-deoxy-2-isothiocyanato-α-d-glucopyranoside 2, which exists in equilibrium with the corresponding 2,3-cyclic carbamate 1, reacts with d-glucosamine producing the pseudo-C and N-nucleoside of chiral imidazolidine-2-thione 3, in good yield and high stereoselectivity. Starting from 3, different pseudo-C- and N-nucleosides of imidazoline-2-thione 611, and of imidazole 12 and 13 are obtained. An efficient synthesis of [1,2,5]oxadiazolo[3,4-d]pyridazine 1,5,6-trioxides (1) from 3,4-bis(hydroxyimino)methyl)-1,2,5-oxadiazole 2-oxides using a mixture of concentrated nitric and trifluoroacetic acids has been developed. The scope of the unconventional reaction was established. The 4,7-dinitro[1,2,5]oxadiazolo[3,4-d]pyridazine 1,5,6-trioxide 1f represents a new high energy compound, unfortunately with low thermal stability. The parent [1,2,5]oxadiazolo[3,4-d]pyridazine 1,5,6-trioxide 1c was studied by single-crystal X-ray diffraction analysis which revealed a planar molecule with an unusually long intracyclic NN bond of 1.668(5)?Å and unexpected exo-cyclic bond angles at the nitroxyl nitrogen atoms. In the crystal, the molecules of 1c are bound to each other by strong π-π stacking and CH?O hydrogen bonding interactions into a three-dimensional framework that results in a high crystal density of 1.833?gcm?3.  相似文献

A one step synthesis of s-triazolo[4,3-b]pyridazine using 3-chloro-6-hydrazinopyridazine and an appropriate thioformimidate is described. Applying this procedure to 5-O-benzoyl-1-benzylthio-1-formimidate-D-ribofuranose, 5′ benzoyl-6-chloro-3β-D-ribofuranosyl-s-triazolo[4,3-b]pyridazine was obtained. Substitutions of chlorine by nucleophilic reagents afforded some derivatives of a new series of C-nucleo-sides. Structural determination including anomeric configuration assignment is discussed based mainly on 1H nmr spectroscopy.  相似文献

A new method for the synthesis of 3-halo-6-(N-tosyl-2-pyrrolyl)pyridazine 7 was developed. Suzuki cross-coupling reactions of 7 with arylboronic acids and in situ de-tosylation gave a variety of novel 3-aryl-6-(2-pyrrolyl)pyridazines. It found that protection of the pyrrolyl moiety was necessary for efficient coupling reaction.  相似文献

A versatile intermediate 2 for C-nucleoside synthesis was treated with thiosemicarbazide to obtain thiosemicarbazone 6 , which was then converted to 3-aminopyrazol-2N-thiocarboxamide derivatives 7 and 8 by the reaction of 6 and sodium ethoxide. 4-Thioxo-4H-pyrazolo[1,5-a]-1,3,5-triazine C-nucleosides 11 and 12 were obtained by the ring closure reaction of 7 and 8 with triethyl orthoformate. Brief treatment of 11 and 12 with 10% methanolic hydrogen chloride afforded C-nucleosides 4 and 13 , respectively, without anomerization. Identification of compounds 4 and 13 was made on the basis of 1H nmr and uv spectra, as well as chemical conversion to known compounds with established configurations. Model compounds were also synthesized in order to confirm the heterocyclic moieties.  相似文献
