Each drug has pharmacokinetics that must be defined for the substance to be used in humans and animals. Currently, one of the basic analytical tools for pharmacokinetics studies is high-performance liquid chromatography coupled with mass spectrometry. For this analytical method to be fully reliable, it must be properly validated. Therefore, the aims of this study were to develop and validate a novel analytical method for 4-acetamidobenzoic acid, a component of the antiviral and immunostimulatory drug Inosine Pranobex, and to apply the method in the first pharmacokinetics study of 4-acetamidobenzoic acid in pigs after oral administration. Inosine Pranobex was administered under farm conditions to pigs via drinking water 2 h after morning feeding at doses of 20, 40, and 80 mg/kg. For sample preparation, we used liquid–liquid extraction with only one step—protein precipitation with 1 mL of acetonitrile. As an internal standard, we used deuterium labeled 4-acetamidobenzoic acid. The results indicate that the described method is replicable, linear (r2 ≥ 0.99), precise (2.11% to 13.81%), accurate (89% to 98.57%), selective, and sensitive (limit of quantitation = 10 ng/mL). As sample preparation requires only one step, the method is simple, effective, cheap, and rapid. The results of the pilot pharmacokinetics study indicate that the compound is quickly eliminated (elimination half-life from 0.85 to 1.42 h) and rapidly absorbed (absorption half-life from 0.36 to 2.57 h), and that its absorption increases exponentially as the dose is increased.  相似文献

Sirolimus is a hydrophobic macrolide compound that has been used for long-term immunosuppressive therapy, prevention of restenosis, and treatment of lymphangioleiomyomatosis. In this study, a simple and sensitive liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed and validated for the simultaneous determination of sirolimus in both porcine whole blood and lung tissue. Blood and lung tissue homogenates were deproteinized with acetonitrile and injected into the LC-MS/MS system for analysis using the positive electrospray ionization mode. The drug was separated on a C18 reversed phase column with a gradient mobile phase (ammonium formate buffer (5 mM) with 0.1% formic acid and acetonitrile) at 0.2 mL/min. The selected reaction monitoring transitions of m/z 931.5 → 864.4 and m/z 809.5 → 756.5 were applied for sirolimus and ascomycin (the internal standard, IS), respectively. The method was selective and linear over a concentration range of 0.5–50 ng/mL. The method was validated for sensitivity, accuracy, precision, extraction recovery, matrix effect, and stability in porcine whole blood and lung tissue homogenates, and all values were within acceptable ranges. The method was applied to a pharmacokinetic study to quantitate sirolimus levels in porcine blood and its distribution in lung tissue following the application of stents in the porcine coronary arteries. It enabled the quantification of sirolimus concentration until 2 and 14 days in blood and in lung tissue, respectively. This method would be appropriate for both routine porcine pharmacokinetic and bio-distribution studies of sirolimus formulations.  相似文献

A rapid and sensitive liquid chromatography-tandem mass spectrometry(LC-MS/MS) method for the de- termination of cefotetan in human plasma was developed and validated. After the protein precipitation of sample with acetonitrile, the analyte and internal standard(IS), tramadol, were separated on a Zorbax XDB C8 column using ace- tonitrile/1%(volume fraction) formic acid(volume ratio 35:65, pH=2.5) as mobile phase at a flow rate of 1.0 mL/min with a 1 : 1 split. The detection was performed by electrospray ionization with positive ion mode, followed by multiple reaction monitoring of the transitions for cefotetan at m/z 576.3→460.2(quantifier) and m/z 576.3→432.2(qualifier) and for IS at m/z 264.1→58.1. Cefotetan and IS were eluted at 1.86 and 1.87 rain, respectively. The assay was linear over the concentration range of 0.1-100 gg/mL for 20 μL of human plasma only with intra- and inter-day preci- sions(expressed as the relative standard deviation) of less than 6.62% and accuracies(as relative error) of +1.31%. The method was applied to the pharmacokinetic study of a l-h intravenous infusion of 1.0 g of cefotetan disodium for human volunteers(n=6).  相似文献

ARV-110, a novel proteolysis-targeting chimera (PROTAC), has been reported to show satisfactory safety and tolerability for prostate cancer therapy in phase I clinical trials. However, there is a lack of bioanalytical assays for ARV-110 determination in biological samples. In this study, we developed and validated an LC-MS/MS method for the quantitation of ARV-110 in rat and mouse plasma and applied it to pharmacokinetic studies. ARV-110 and pomalidomide (internal standard) were extracted from the plasma samples using the protein precipitation method. Sample separation was performed using a C18 column and a mobile phase of 0.1% formic acid in distilled water–0.1% formic acid in acetonitrile (30:70, v/v). Multiple reaction monitoring was used to quantify ARV-110 and pomalidomide with ion transitions at m/z 813.4 → 452.2 and 273.8 → 201.0, respectively. The developed method showed good linearity in the concentration range of 2–3000 ng/mL with acceptable accuracy, precision, matrix effect, process efficiency, and recovery. ARV-110 was stable in rat and mouse plasma under long-term storage, three freeze-thaw cycles, and in an autosampler, but unstable at room temperature and 37 °C. Furthermore, the elimination of ARV-110 via phase 1 metabolism in rat, mouse, and human hepatic microsomes was shown to be unlikely. Application of the developed method to pharmacokinetic studies revealed that the oral bioavailability of ARV-110 in rats and mice was moderate (23.83% and 37.89%, respectively). These pharmacokinetic findings are beneficial for future preclinical and clinical studies of ARV-110 and/or other PROTACs.  相似文献

以氘代尼古丁、氘代可的宁、3-OH-氘代可的宁、氘代尼古丁葡萄糖醛酸复合物、氘代可的宁葡萄糖醛酸复合物、3-OH-氘代可的宁葡萄糖醛酸复合物作内标, 在LC-MS/MS的大气压化学电离(APCI)离子化模式下, 建立了快速检测吸烟者尿液中尼古丁及其9种代谢物的方法, 并对19个实际样品进行了测试和判别分析(DA). 实验前处理简单, 色谱运行时间仅为3.8 min. 结果表明, 尼古丁及其代谢物的精密度在0.5%~5.5%之间, 回收率在94%~109%之间, 线性相关系数均大于0.995. DA分析充分区别开了不同量的吸烟者, 进一步的相关性分析表明吸烟者24 h尿液中的尼古丁及其9种代谢物含量与卷烟抽吸量的分析因子之间均有较强正相关性.  相似文献

Houttuynia cordata Thunb. is a medicinal and edible plant that has been commonly used in traditional Chinese medicine since ancient times. This study used headspace solid-phase microextraction (HS-SPME) and direct injection, combined with gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS), to identify the volatile compounds in H. cordata. Extraction from different parts of the plant using different extraction techniques for the identification of volatile compounds were determined. A total of 93 volatile components were analyzed in the leaves, stems, rhizomes, and whole plant samples of H. cordata. The leaves contained more (Z)-3-hexenal, β-myrcene, (Z)-β-ocimene, and (4E,6E)-allo-ocimene; the stems contained more geranyl acetate and nerolidol; and rhizomes contained more α-pinene, β-pinene, limonene, 2-undecanone, and decanoyl acetaldehyde. Among them, the essential oil extracted by HS-SPME could produce more monoterpenes, while direct injection could obtain higher contents of aliphatic ketones, terpene esters, sesquiterpenes, and was more conducive to the extraction of 2-undecanone and decanoyl acetaldehyde.  相似文献

A rapid, simple and sensitive LC‐MS/MS method for the quantification of vinflunine in plasma was developed and validated. The analysis involved a simple liquid–liquid extraction. After making alkaline with NaOH, plasma was extracted with methyl tert‐butyl ether and the organic extract was then evaporated and the residue was reconstituted in mobile phase. The reconstituted solution was injected into an HPLC system and was subjected to reverse‐phase HPLC on a 5 µm ODS‐3 column at a flow‐rate of 0.2 mL/min. The mobile phase consisted of ammonium acetate (0.02 mol/L, pH = 3.0) and acetonitrile (20:80). Vinflunine was detected in the single ion monitoring mode using target ions at m/z 817.4/160.1/142.3 for vinflunine and m/z 447.2/128.3/112.1 for gefitinib (internal standard). Standard curves were linear over the concentration range of 5–1000 ng/mL. The mean predicted concentrations of the quality control samples deviated by less than 2% from the corresponding nominal values; the intra‐assay and inter‐assay precisions of the assay were within 7% relative standard deviation. The extraction recovery of vinflunine was more than 80%. The validated assay was applied to a pharmacokinetic study of vinflunine in plasma following the administration of a single vinflunine injection (2 mg/kg). Isoginkgetin is a biflavonoid compound isolated from the leaf extracts of Ginkgo biloba. In this study, an liquid chromatography–tandem mass spectrometry (LC/MS/MS) with liquid–liquid extraction was developed and validated for the analysis of isoginkgetin in rat plasma. In the process of chromatographic separation, selected reaction monitoring transitions for isoginkgetin and IS were m/z 566.8 → 134.7 and m/z 430.8 → 269.3, respectively. The validation parameters including selectivity, linearity, LLOQ, accuracy, precision, matrix effect, stability and recovery were satisfactory. The intra‐ and inter‐batch precision (RSD) were <12.1% in plasma, while the accuracy (RE) was within ±14.3%. This method was employed in a pharmacokinetic study on rats after the intravenous administration of isoginkgetin.  相似文献

建立了LC-MS/MS法定性定量分析甲卡西酮。采用三重串联四极杆液质联用仪(LC/QQQ),AgilentZorbax Eclipse Plus C18色谱柱(100 mm×2.1 mm,1.8μm),流动相为0.1%甲酸-乙腈,梯度洗脱,流速为0.3mL/min。质谱应用ESI源、正离子模式、多反应监测(MRM)方式。在0.1~10 000 ng/mL质量浓度范围内线性关系良好,r2=0.999 8,日内与日间保留时间和峰面积的相对标准偏差不大于5.28%,检出限为0.04 ng/mL,回收率为95.6%~100.7%。该方法适用于甲卡西酮的定性、定量分析。  相似文献

This study was performed to evaluate and compare the pharmacokinetic parameters between two dosage formulations of hesperidin and naringenin: mixture and tablet. Our objective was to determine that the flavonoid tablet does not significantly modify the pharmacokinetic parameters compared with the mixture. For this study, we administered 161 mg/kg of either mixture (Mix-160) or tablet composed of hesperidin and by intragastric administration. Blood microsamples were collected from tail vein up to 24 h. Serum flavonoid extraction was performed by solid phase extraction and analyzed by LC-MS/MS of triple quadrupole (QqQ). Serum concentration vs. time plot showed that data fitted for a first-order model. The pharmacokinetic parameters were calculated by a noncompartmental model. The results showed that the absorption constant is higher than the elimination constant. The first concentration was found at five minutes, and minimal concentration at 24 h after administration, suggesting a enterohepatic recirculation phenomena and regulation of liver cytochromes’ activity. We did not find meaningful differences between the pharmacokinetic parameters of both samples. We concluded that tablet form did not interfere with the bioavailability of hesperidin and naringenin, and it could be a suitable candidate for developing a drug product.  相似文献

Cucurbitacin B (CuB), one of the most abundant forms of cucurbitacins, is a promising natural anticancer drug candidate. Although the anticancer activity of CuB has been well demonstrated, information regarding the pharmacokinetics is limited. A rapid, selective and sensitive UPLC‐MS/MS for CuB was developed and validated using hemslecin A (HeA) as internal standard (IS). Plasma samples were pre‐treated by liquid–liquid extraction with dichloromethane. Separation was achieved on a reversed‐phase C18 column (50 × 4.6 mm, 5 µm) at 35°C using isocratic elution with water–methanol (25:75, v/v) at a flow rate of 0.3 mL/min. The analytes were monitored by a triple quadrupole tandem mass spectrometer with positive electrospray ionization mode. The calibration curve was linear (r > 0.995) in a concentration range of 0.3–100 ng/mL with a limit of quantification of 0.3 ng/mL. Intra‐ and inter‐day accuracy and precision were validated by percentage relative error and relative standard deviation, respectively, which were both lower than the limit of 15%. A simple and sensitive liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the simultaneous analysis of three tropane alkaloids in blood and urine. After 1 mL of a blood or urine sample was extracted using a liquid–liquid extraction method with ethyl acetate at pH 8 and homatropine as the internal standard, the tropane alkaloids were separated. An Allure PFP propyl column (50 mm × 2.1 mm, 5 µm) separated the tropane alkaloids using an acetonitrile-buffer solution (20 mmol/L ammonium acetate and 0.1% formic acid, pH 4) (70:30) as the mobile phase at a flow-rate of 0.2 mL/min in isocratic mode, with the LC-MS/MS in the positive ionization mode. For each compound, detection was related to two daughter ions (scopolamine: m/z 304.4 → 138.1 and 155.9; atropine: m/z 290.3 → 124.0 and 93.1; anisodamine: m/z 306.3 → 140.1 and 91.1; and homatropine: m/z 276.3 → 124.3 and 142.1). The tropane alkaloids exhibited excellent linearity in the range of 0.05–100 ng/mL in blood and 0.2–100 ng/mL in urine, with a limit of detection range from 0.02 to 0.05 ng/mL for biological materials. The extraction recoveries of atropine, scopolamine, and anisodamine were more than 53% in the blood and urine; the interday and intraday RSDs were less than 10%; the within-day and between-day accuracy were between ?9.8% and +8.8%. The present method is simple and rapid, as shown by its application to a clinical case. This method is useful for routine analysis of tropane alkaloids in cases of suspected tropane alkaloid poisoning.  相似文献

建立了烤鳗中苯胺灵、嘧菌酯、氨苯乙酯、氟酮唑草、丙蝇驱、唑螨酯6种农药残留的同时检测方法.烤鳗样品采用乙腈、中性氧化铝超声基体分散吸附、涡旋混合提取,Florisil固相萃取小柱净化、浓缩,以乙腈-乙酸铵缓冲液为流动相进行色谱分离,串联质谱检测.结果表明,6种化合物在0.5 ~1 000 μg/L范围内线性关系良好,方法的检出限为0.03 ~2.3 μg/kg,定量下限为0.09 ~7.7 μg/kg,平均加标回收率为75% ~104%,相对标准偏差(RSD)为1.6% ~15%,满足当前进出口残留控制要求.  相似文献

建立了血液中卡马西平的固相萃取/液相色谱-串联质谱(LC-MS/MS)定量检测方法。血液中的卡马西平用固相萃取柱(Bond Elut Certify)提取,采用Waters AtlantisTMdC18(150 mm×3.9 mm,5μm)色谱柱,电喷雾离子源,正离子检测,多反应监测方式进行定量分析,以SKF-525A为内标。结果表明,该方法对卡马西平的检出限为0.1μg/L,卡马西平的质量浓度在100~6 000μg/L范围内线性关系良好(r=0.997 6),日内、日间精密度RSD(n=6)不高于8.6%,血液中卡马西平的回收率为81%~90%。该方法具有良好的灵敏度、重现性、稳定性和专属性,可用于法庭与临床的毒物分析。  相似文献

A quick, sensitive, and reproducible analytical method for the determination of 77 multiclass pesticides and their metabolites in Capsicum and tomato by gas and liquid chromatography tandem mass spectrometry was standardized and validated. The limit of detection of 0.19 to 10.91 and limit of quantification of 0.63 to 36.34 µg·kg−1 for Capsicum and 0.10 to 9.55 µg·kg−1 (LOD) and 0.35 to 33.43 µg·kg−1 (LOQ) for tomato. The method involves extraction of sample with acetonitrile, purification by dispersive solid phase extraction using primary secondary amine and graphitized carbon black. The recoveries of all pesticides were in the range of 75 to 110% with a relative standard deviation of less than 20%. Similarly, the method precision was evaluated interms of repeatability (RSDr) and reproducibility (RSDwR) by spiking of mixed pesticides standards at 100 µg·kg−1 recorded anRSD of less than 20%. The matrix effect was acceptable and no significant variation was observed in both the matrices except for few pesticides. The estimated measurement uncertainty found acceptable for all the pesticides. This method found suitable for analysis of vegetable samples drawn from market and farm gates.  相似文献

在运用GC/MS技术的基础上采用HELP(直观推导式演进特征投影法)方法研究了九种唇形科植物的化学成分, 并以唇形科植物半枝莲为例详细介绍了HELP的解析过程. 应用总体积积分法测定各成分的相对百分含量. 鉴定出相对共有成分达70余种, 大多数为萜类化合物及其衍生物. 不同的唇形科植物的挥发油化学成分与特征成分有明显差异. 九种药材挥发性成分中均含有桉油精(Eucalyptol, 含量0.10%~1.01%)和芳樟醇(Linalool, 0.11%~3.05%). 利用GC/MS分析法结合化学计量学分辨方法鉴定挥发油化学成分, 比单独使用GC-MS法结果更准确、可靠.  相似文献

称取蜂蜜样品2.00g,加入同位素内标40ng,用pH7的磷酸二氢钾-磷酸氢二钠缓冲溶液溶解并稀释至10.0mL。充分混匀后,加入乙腈10.0mL和氯化钠2.0g,离心涡旋提取5min。取上层乙腈相保留待用,于下层水相中再加入乙腈10mL重复提取1次。合并两次提取液,加乙腈定容至20.0mL。分取此提取液1.0mL,加入于己预置N-丙基乙二胺(PSA)25mg、十八烷基硅烷(C18)10mg和MgSO430mg的离心管中,充分混匀后,高速离心,进行分散固相萃取(dSPE)净化。转移全部上清液,加水定容至2.0mL,此溶液供液相色谱-串联质谱法(LC-MS/MS)分析。色谱分离中采用XDB-C18色谱柱为固定相,用不同比例的φ0.15%甲酸溶液(A)和乙腈(B)的混合液作为流动相进行梯度淋洗。所得各洗脱液按工作条件进行MS/MS测定双甲脒及其代谢物单甲脒、2,4-二甲基苯基甲酰胺和2,4-二甲基苯胺的残留量。由于采用空白蜂蜜作基体加入混合标准溶液制作工作曲线,并加入同位素内标参与定量,有效地克服了基质效应。上述4种化合物工作曲线的线性范围为0~100μg·kg^-1,测定下限(10S/N)依次为0.10,0.20,5.0,2.0μg·kg^-1。在实际样品基体中加入3个浓度水平的标准溶液(5.0,10,20μg·kg^-1)进行回收试验,测得回收率均大于80%,测定值的相对标准偏差(n=6)均小于10%。所提出方法具有简便、快速的优点,其测定下限能满足目前国内外的规定要求。  相似文献

A rapid and sensitive liquid chromatography-tandem mass spectrometry method(LC-MS/MS)was developed and validated for the quantification of fexofenadine in human plasma,to conduct comparative bioavailability studies.Human plasma was extracted with a mixture of dichloromethane-diethyl ether(volume ratio 2∶3)in a basic environment and the extract was separated on a C18 column with a mobile phase consisting of acetonitrile-methanol-10 mmol/L ammonium acetate(volume ratio 45∶45∶10).The analytes were detected via electrospray ionization(ESI)tandem mass spectrometry in the multiple-reaction-monitoring(MRM)mode.The linearity was within a range of 1-1000 ng/mL.The intra-and inter-day precision were〈4.1% and〈4.8%,respectively,and the accuracy was in the range of 95.0%-105%.The method was applied to the quantification of fexofenadine human plasma from 20 healthy male Chinese volunteers,according to a single dose,randomized,two-way crossover design with a two-week washout period.The mean values of major pharmacokinetic parameters of ρmax,AUC0-48,AUC0-∞,tmax,and t1/2 were determined from the plasma concentration.The analysis of variance(ANOVA)did not show any significant difference between the two products of fexofenadine and 90% confidence intervals fell within the acceptable range for bioequivalence.  相似文献

以动植物油脂为实验材料,建立了测定食用油中天然辣椒素、二氢辣椒素和合成辣椒素含量的凝胶渗透色谱–高效液相色谱–串联质谱(GPC–HPLC–MS/MS)法。样品经凝胶渗透色谱净化后,采用液相色谱串联质谱法(HPLC–ESI–MS/MS)分析,多反应监测模式(MRM)下外标法定量。在0.1~5.0μg/L范围内线性良好,天然辣椒素、二氢辣椒素和合成辣椒素的相关系数分别为0.999 6,0.999 8,0.999 8,检出限为0.5μg/kg。在5μg/kg添加水平下,空白加标回收率为71.5%~82.5%,测定结果的相对标准偏差为3.0%~8.3%(n=6)。该方法样品处理过程简便快捷,测定结果准确,可满足实验室大量、快速分析的需求。  相似文献
