An efficient and simple approach for the solid-phase synthesis of 2,4,6-triaminopyrimidines and 2-alkyl-4,6-diaminopyrimidines is described. Primary amines were immobilized on 2-(4-formyl-3-methoxyphenoxy)ethyl polystyrene resin via reductive amination. Attachment of two different 4,6-dichloropyrimidines led to the corresponding 4-chloro-6-aminopyrimidine intermediates. Aromatic nucleophilic substitution with various aliphatic amines or the corresponding lithium amides afforded the desired aminopyrimidines in high yield and excellent purity after acidic cleavage from the resin. The products were characterized by LC-MS, 1H and 13C NMR spectroscopy. Deuterium exchange experiments revealed that the investigated aminopyrimidines have a general tendency toward C-5 protonation.  相似文献

The preparation of functionalized 4,5,6,7-tetrahydroisoindole via a traceless solid-phase sulfone linker strategy is described. Thermolytic extrusion of SO(2) from polymer-bound 3-(phenylsulfonyl)-3-sulfolene (7) generated polymer-bound 2-(phenylsulfonyl)-1,3-butadiene (9) in situ which underwent Diels--Alder cycloaddition with various dienophiles to furnish vinyl sulfone resins 10-14. To complete a traceless linker cleavage strategy, (p-tolysulfonyl)methyl isocyanide or ethyl isocyanoacetate was employed to react with the vinyl sulfone moiety to liberate functionalized 4,5,6,7-tetrahydroisoindole products from the resin. Using this chemistry, nine tetrahydroisoindole derivatives (6, 15-22) were prepared in 32-41% overall yields from polystyrene/divinylbenzene sulfinate 1.  相似文献

Nucleosides of pyrrolo[2,3-d]pyridazin-4(5H)-ones were prepared by the single-phase sodium salt glycosylation of appropriately functionalized pyrrole precursors. The glycosylation of the sodium salt of ethyl 4,5-dichloro-2-formyl-1H-pyrrole-3-carboxylate ( 4 ), or its azomethino derivative 7 , with 1-bromo-2,3,5-tri-O-benzoyl-D-ribofuranose in acetonitrile afforded the corresponding substituted pyrrole nucleosides ethyl 4,5-dichloro-2-formyl-1-(2,3,5-tri-O-benzoyl-β-D-ribofuranosyl)-1H-pyrrole-3-carboxylate ( 5 ) and ethyl 4,5-dichloro-2-phenylazomethino-1-(2,3,5-tri-O-benzoyl-β-D-ribofuranosyl)-1H-pyrrole-3-carboxylate ( 8 ), respectively. The latter, upon treatment with hydrazine, afforded the annulated product 2,3-dichloro-1-β-D-ribofuranosyl-1H-pyrrolo[2,3-d]pyridazin-4(5H)-one ( 6 ), in good yield. The unsubstituted analog 1-β-D-ribofuranosyl-1H-pyrrolo[2,3-d]pyridazin-4(5H)-one ( 9 ), was obtained upon catalytic dehalogenation of 6 . This report represents the first example of the synthesis of nucleosides of pyrrolopyridazines.  相似文献

A series of mononuclear oxovanadium(v) complexes of tridentate Schiff bases HL(1)-HL(4) and H(2)L(5)-H(2)L(8) derived from 6-phenylsalicylaldehyde and 6-(2-hydroxyphenyl)salicylaldehyde and four different amines was synthesized. The systematically selected ligands contain aliphatic or aromatic nitrogen, or alkoxy- and phenoxy-oxygen as third donor atom. The complexes were characterised by spectroscopic methods in solution and the solid state. Single-crystal X-ray analyses were performed with VO(2)L(1)(), VO(2)L(3)x1/2EtOH (), VO(2)L(4)(), VO(OiPr)L(7)xiPrOH, VO(OiPr)L(8) and H(2)L(8). For all compounds the vanadium(v) cores contain distorted tetragonal pyramidal geometry around the dioxo- and oxovanadium site at which the N(2)O- and NO(2)-donor ligands bind equatorially. Complexes and display intramolecular hydrogen bonding of the pendant hydroxyphenyl group to a coordinated oxygen trans to a nitrogen atom and therefore serve as suitable models for the native site of vanadium dependent haloperoxidases. Variable-temperature (1)H NMR spectra revealed significant hydrogen bond interaction in acetonitrile solution. In situ prepared catalysts are active for hydrogen peroxide mediated oxygenation of ethyl phenyl sulfide and showed complete conversion of the substrate to ethyl phenyl sulfoxide, together with small amounts of the corresponding sulfone, as detected by GC/MS after 10 min. The complex of H(2)L(7) turned out to be most efficient while HL(1) and HL(2) were completely inactive. Catalysis is supported by the pendant OH group in the complex of HL(3), the catalyst is twice as active as the complex of HL(4).  相似文献

A five-step solid-phase synthesis of isoxazolinopyrrole-2-carboxylates (6) that employs a traceless sulfone linker strategy is reported. Resin-bound diene 4, obtained by acetylation and concomitant beta-elimination of acetate from resin-bound allylic alcohol 3, underwent regioselective 1,3-dipolar cycloadditons with nitrile oxides. Formation of the pyrrole products in a resin-releasing strategy was performed by pyrrole annulation with alkyl isocyanoacetates, which react with the vinyl sulfone moiety to generate the target isoxazolinopyrrole-2-carboxylates (6). Use of this chemistry afforded eight isoxazolinopyrrole-2-carboxylates in 6-24% overall yields from polystyrene/divinylbenzene sulfinate 1.  相似文献

3-Lithio-1-(trimethylsilyl)pyrrole ( 7 , Schem 2), obtained by halogen-metal interchange from the 3-bromo compound 2 , reacted with various electrophilic reagents to provide products, which on fluoride ion desilylation, gave 3-substituted pyrroles in good overall yields. One such pyrrole 13 (Schem 3), was converted into 2-formyl-3-octadeclpyrrole ( 14 ), reputed to be a metabolite of the marine sponge Oscarella lobularis. 3,4-Dibromo-1-(triisopropylsityl)pyrrole ( 5 ) was efficiently transformed, by a process involving two consecutive bromine-lithium exchange reactions (Scheme 4), into the antibiotic verrucarin E 17.  相似文献

The synthesis of the title compound 4 by cyclization of 1-(2-ethoxycarbonylthiobenzyl)pyrrole 9 , prepared by treating with ethyl chloroformate the 1-(2-mercaptobenzyl)pyrrole 7 previously obtained by debenzylation of 1-(2-benzylthiobenzyl)pyrrrole 6 , failed. On the other hand 4 was successfully synthesized by intramolecular cyclization of 1-(2-mercaptobenzyl)pyrrole-2-carboxylic acid 15 by DMAP -catalyzed DCC method. The pyrrole 6 and 1-(2-benzylthiobenzyl)pyrrole-2-carboxaldehyde 11 were useful as starting materials to obtain 1-(2-benzylthiobenzyl)pyrrole-2-carbonitrile 13 , which was hydrolyzed to corresponding amide 16 . Debenzylation of 16 afforded 1-(2-mercaptobenzyl)pyrrole-2-carboxyamide 17 , whose hydrolysis led to required acid 15 .  相似文献

Reactions of 4-hydroxy-3-methoxybenzaldehyde and 3-ethoxy-4-hydroxybenzaldehyde with methyl and ethyl chloroformates in the presence of pyridine gave the corresponding methyl and ethyl carbonates which were brought into condensation with biphenyl-4-amine, naphthalen-1-and-2-amines, and 3-and 4-aminobenzoic acids to obtain the corresponding Schiff bases. Alkyl 4-(9,9-dimethyl-11-oxo-7,8,9,10,11,12-hexahydrobenzo[a]acridin-12-yl)-2-alkoxyphenyl carbonates were selectively synthesized by cascade heterocyclization of naphthalen-2-amine, 5,5-dimethylcyclohexane-1,3-dione, and 4-formyl-2-methoxy(or ethoxy)phenyl methyl(or ethyl) carbonates. Three new compounds: 2R,3R-pterosin L 3-O-beta-D-glucopyranoside (1), beta-D-xylopyranosyl(1-->2)-7-O-benzoyl-beta-D-glucopyranoside (2) and 4-O-benzoyl-beta-D-xylo-pyranosyl(1-->2)-7-O-benzoyl-beta-D-glucopyranoside (3), together with nine known compounds, were isolated from the ethyl acetate extract of Pteris ensiformis. 5-[2-Hydroxyethylidene]-2(5H)-furanone (4), which had been synthesized, was isolated from natural sources for the first time. The structures of all isolated compounds were determined on the basis of mass and spectroscopic evidence. Compound 1 and pterosin B (5) show cytotoxicity against HL 60 cells (human leukemia) with the IC(50) values of 3.7 and 8.7 microg/mL, respectively.  相似文献

Coordination compounds of Cu(II), VO(II), Ni(II), and Mn(II) with the Schiff base obtained through the condensation of 4-amino-2,3-dimethyl-1-phenyl-3-pyrazolin-5-one with 3-formyl-6-methyl-chromone were synthesized. The characterization of the newly formed compounds was done by 1H NMR, UV–Vis, IR, ESR spectroscopy, elemental analysis and molar electric conductibility. The crystal structure of 1-phenyl-2,3-dimethyl-4-(N-3-formyl-6-methyl-chromone)-3-pyrazolin-5-one (HL) has been determined by X-ray diffraction studies, as well as the one of its copper(II) complex [CuL(OAc)]·CH3OH which contains an anionic ligand and an acetate in the coordination sphere of the metal. The single crystal X-ray structure for (HL) was analyzed for its various weak H-bonding and dimeric association.  相似文献

本文使用组合化学方法以聚苯乙烯亚磺酸钠树脂(1)为载体合成了2,5-取代-1,3,4-二唑化合物库。树脂1首先与溴代乙酸乙酯反应生成聚合物支载的乙酸酯(2)。肼解后所得的肼树脂进一步与取代的苄氯反应得双欧讲树脂(4)。树脂4与三氯氧磷回流得聚合物支载的2,5-取代-1,3,4-二唑(5)。用W-溴代乙酰苯酮烷基化后聚合物支载的产物用三乙胺解脱得2-芳基-5-芳甲酰亚乙烯基-1,3,4-二唑化合物库,产率15%-36%。  相似文献

Novel electrophilic trisubstituted ethylene monomers, methyl and methoxy ring- substituted 2-cyano-3-phenyl-2-propenamides, RPhCH=C(CN)CONH2, where R is 2,3-dimethyl, 2,4-dimethyl, 2,5-dimethyl, 2-(3-methoxyphenoxy), 2-(4-methoxyphenoxy), 3-(4-methoxyphenoxy), 4-(4-methylphenoxy), 2,3-methylenedioxy were prepared and copolymerized with styrene. The monomers were synthesized by potassium hydroxide catalyzed Knoevenagel condensation of ring-substituted benzaldehydes and cyanoacetamide, and characterized by CHN elemental analysis, IR, 1H- and 13C-NMR. Novel copolymers of the ethylenes and styrene were prepared at equimolar monomer feed composition by solution copolymerization in the presence of a radical initiator, ABCN at 70°C. The composition of the copolymers was calculated from nitrogen analysis, and the structures were analyzed by IR, 1H- and 13C-NMR, GPC, DSC, and TGA. High Tg of the copolymers in comparison with that of polystyrene indicates a substantial decrease in chain mobility of the copolymer due to the high dipolar character of the trisubstituted ethylene monomer unit. Decomposition of the copolymers in nitrogen occurred in two steps, first in the 200–500°C range with residue (5.8–33.8 wt%), which then decomposed in the 500–800°C range.  相似文献

Attachment of two different 4-chloro-2-methylthiopyrimidines, followed by sulfide oxidation, led to their corresponding sulfone intermediates. Aromatic nucleophilic substitution with various anilines or heteroaromatic amines in the presence of trimethyl aluminum afforded the desired 2,4-diaminopyrimidines after acidic cleavage from the resin. Three new unsymmetrical compartmental dinucleating ligands, 4-bromo-2-(4-methylpiperazin-1-ylmethyl)-6-[{2-(1-piperidyl)ethyl}aminomethyl]phenol (HL1), 4-bromo-2-(4-methylpiperazin-1-ylmethyl)-6-[{2-(morpholin-4-yl)ethyl}aminomethyl]phenol (HL2), and 4-bromo-2-(4-methylpiperazin-1-ylmethyl)-6-[{2-(thiomorpholin-4-yl)ethyl}aminomethyl]phenol (HL3), have been synthesized in order to model the active site of type 3 copper proteins. The dicopper(II) complexes of these ligands give first hints about the influence of a thioether group close to the metal site. The bromophenol-based ligands have one piperazine arm and one other bidentate arm in positions 2 and 6 of the phenolic ring, respectively. With each ligand a dinuclear copper(II) complex was prepared and structurally characterized. The copper ions were found to have square pyramidal environments and a mixture of endogenous phenoxo and exogenous acetate bridging. The influence of a heteroatom in one arm of the ligand on catecholase activity and speciation in solution was studied by UV/Vis spectroscopy, ESI-MS experiments and, DFT calculations.  相似文献
