Abstract Porphyrins, which may be introduced into the eye as a result of abnormal porphyrin metabolism (uroporphyrin–Uro) or when used in the diagnosis or photodynamic therapy of certain tumors, including intraocular tumors (hematoporphyrin–Hp and'hematoporphyrin derivative'–Hpd and mesotetra( P -sulfonatophenyl)porphyrin–TPPS) are efficient photosensitizers in biological systems. We have been studying the potential phototoxic side effects of these drugs in the lens of the eye. Encapsulated in the human lens is a mixture of soluble protein crystallins. With little turnover of protein in the lens, any photosensitized modifications will accumulate and may result in an opacification of the lens. To evaluate the potential of different porphyrins to induce such damage, a series of porphyrins were photolyzed (transmission above 295 nm) in the presence of calf lens protein (2 mg m−1). Marked photopolymerization and histidine destruction were observed for the lens protein photolyzed in the presence of all of the drugs. We have found that the relative effectiveness of the following porphyrins to induce that damage is: Uro = TPPS Hpd = Hp. Both the singlet oxygen quencher, azide, and the free radical scavenger, penicillamine, decrease this photosensitized oxidative damage to lens protein. TPPS binds significantly to lens protein and this binding leads to conformational changes in that protein.  相似文献

After excitation at room temperature with a 15 ns dye laser pulse, phytochrome (60 kDalton) from etiolated oat seedlings was studied by optoacoustic spectroscopy, which records the heat emission caused by non-radiative deactivation processes of the photoexcited molecule. The action spectrum for the heat emitted by Pr deviates from the absorption spectrum around 610 and 695 nm, indicating the build-up of photoproduct(s) within 15 ns after excitation. It is proposed that the 695 nm product is identical with I700 and lumi-R known to be the first intermediate on the Pr× Pfr pathway, and that the photochemical back reaction of I700 to Pr occurs on the same time scale. About 90% of the absorbed light energy was lost by radiationless deactivation. Values for the rate constants of excited state deactivation of Pr and for the internal energy content difference of the primary photoproducts are calculated.  相似文献

通过检测缝线的力学性能、分子量、失重、大分子取向和结晶度变化研究了聚二氧杂环己酮可吸收缝合线(PDS)在体外的初期(4星期)降解过程。研究结果表明,本文工艺条件下研制的PDS缝线在体外降解过程中随着分子量和大分子取向程度下降,力学性能明显下降,4星期时强度基本消失。分子量明显下降而失重变化很小的结果,证明PDS缝线的酯水解在链中和链端等活性,水解的几率只与水分子的可及程度相关。WAXD和DSC测试结果表明,随着体外降解进行,缝线原晶区基本保持不变,无定形区产生新的有序区,并且含量不断增加和完善。采用结晶高聚物微原纤结构模型解释PDS缝线无定形区在水中的降解机理与研究结果相吻合。PDS缝线的体外降解机理与聚羟基乙酸(PGA)缝线基本相同  相似文献

The effect of applied thickness of sunscreen on the degree of protection against ultraviolet exposure has been investigated using an in vitro model (excised human epidermis) and phototesting in human subjects. The agreement between the protection factors obtained by each technique was excellent. It is shown that the influence of applied thickness on sunscreen protection factors is much less than would be predicted by Beer's law.  相似文献

Abstract— The absorption, fluorescence, and chemiluminescence (CL) characteristics of luminol have been studied in basic dimethylsulfoxide (DMSO) and various basic DMSO — water mixed solvents. It has been shown that the luminol dianion can be produced quantitatively in carefully deoxygenated ‘dry’ DMSO using potassium t-butyl alcoholate (BTO) as the base. A direct correlation has been found between the intensity of CL and the concentration of luminol dianion, Indicating that the dianion is the reactive species in the chemiluminescent reaction in DMSO. Increasing concentrations of water in the mixed solvents greatly reduced the CL intensity because of the decrease in luminol dianion concentration. Solvent effects on the fluorescence of 3-aminophthalic acid samples was noted and identified. Chemiluminescent spectra of luminol were run as a function of solvent composition, and compared with fluorescence spectra in the same media. The need for correcting these spectra for comparison was noted. Stopped flow kinetic studies were run in DMSO and DMSO-water mixtures. These data can be interpreted on the basis of two second-order steps and one first-order step in the reaction. The first-order step is probably last and probably arises from decomposition of a ‘peroxy’ intermediate. The rate constant for this step is k= 1·2±0·3×10-1 sec-1.  相似文献

Abstract— We have measured a calcium and magnesium dependent change in cell shape when mammalian cell monolayers are being prepared for irradiation by replacing their growth medium with certain buffers. In some cases, flattened cells (umbonate) assumed a spherical configuration. In order to assume a centrally located target molecule, we used a DNA-dependent cellular function–pacity for herpes viral growth–as the parameter to measure ultraviolet (UV) sensitivity of cells irradiated while in either of the two shapes. Umbonate cells were more sensitive to UV than were spherical cells. Exposures to the cell that lowered the cellular capacity of umbonate cells to the 10% survival level only lowered spherical cells to the 50% level. Twenty-seven per cent additional UV exposure to spherical cells was required to get the same effect as with umbonate cells. Included in the text are photographs of both cell types, survival curves for cellular capacity, a measure of the absorbance of cell homogenates, and a calculation of the relative number of photons absorbed by each cell nucleus.  相似文献

Abstract— –The intracellular pigment of the ciliate protozoan Blepharisma in the presence of oxygen sensitizes the cells to bright visible light (2700 foot candles (fc)). Illumination of the cells with dim visible light (200 fc) changes the color of the pigment to blue-gray; such cells are no longer sensitive to bright visible light. The pigment which exists in granules can be extruded by cold treatment and is slowly regenerated. The suspension of red cells, the extruded pigment from them and an ethanol extract of the pigment all have very similar absorption spectra. Illumination of the red pigment in ethanol converts it to the blue form of the pigment but only if oxygen is present, indicating a photooxidation. The pigment can be oxidized in darkness to the blue form by ozonation. A suspension of blue cells, the extruded pigment from them and an ethanol extract from them, all have similar absorption spectra. The pigments in red and blue form are very similar spectrophotometrically and in solubility in three species of Blepharisma studies: B. americanum, B. intermedium and B. japonicum. The purified pigment has strong absorption in the far (200–300 nm) ultraviolet (u.v.) and may serve as a screen against damaging U.V. radiation, especially as Blepharisma shows poor photoreactivation.  相似文献

Abstract– Spectroscopic measurements of retinochrome and the related photoproduct, metaretino-chrome, were carried out to determine the conformation of the retinal and the protein. Absorption spectra with fourth derivatives indicate that the tryptophan residues are located in a hydrophobic core and that the environment around these residues does not change after light irradiation. Circular dichroism measurements indicate that retinochrome has a high helical content which is not altered by the conversion of retinochrome to metaretinochrome. Fourier transform infrared difference spectra demonstrate that retinochrome has the all-trans retinal and metaretinochrome has the 11-CM retinal. Retinochrome has an absorption due to amino acid residue(s) which changes in metaretinochrome. This work demonstrates that conformational changes are not induced during the conversion but the electrical charge(s) of the protein are changed by irradiation.  相似文献

用DSC和X ray衍射法对聚对二氧环己酮 乙交酯 (PDG)缝合线的初期体外降解过程进行了研究 ,并使用纤维摄影仪对纤维在降解过程中的表面形态变化进行了观察 .研究结果表明 ,在本工艺条件下得到的PDG缝合线在体外降解过程中分为三个阶段降解 ,即无定形区降解、原纤间非晶区降解和晶区水解 .在体外降解过程中 ,PDS缝合线以横向断裂为主 ,而PDG则通过表面积的大块脱落来完成降解过程  相似文献

用SDS-PAGE法研究了酸溶胶原中的α121112组份在中性水溶液以及加入盐(AuCl3等)后的水解能力。实验表明:在水溶液中,α1与α2的水解能力相近,β11与β12的水解能力也相差不多。但在加入与乳剂成熟条件一致的金属盐[AuCl3(0.167‰,NH4CNS(2‰)和Na2S203(1‰)]之后,由于金属离子(尤其是Au3+)的影响,α1的水解能力明显大于α211的水解能力明显大于β12.本文还讨论了影响胶原中不同组份水解能力的各种因素。  相似文献

Kinetics of the addition of p-toluenesulfinic acid to p-substituted benzylideneacetophenones has been investigated. The reaction was second order, first order each in the concentration of sulfinic acid and chalcone. Effects of the p-substituents on the reaction rate were small (rho = 0.4).
