在发现18种氨基酸特别是色氨酸在TOYOPEARLHW-40S树脂所填色谱柱上表现出非理想排阻行为的基础上,以色氨酸在HW柱上的保留时间为指标,探讨不同的流动相改良剂对HW凝胶与色氨酸之间相互作用的影响,从而阐明这种反常行为是疏水、离子交换以及氢键三者综合作用的结果,而疏水作用是最主要的影响因素。实验结果说明流动相组成对HW凝胶色谱行为有明显的影响作用,这不但有助于确立HW凝胶理想的排阻色谱条件,而且为利用其所具有的吸附性能分离分析肽类和氨基酸等小分子物质奠定了基础。  相似文献

利用CIEMAT/NIST方法在液体闪烁计数装置上测量放射性核素55Fe,高质量样品是得到好的测量结果的关键。影响样品源质量的因素有闪烁液的选择,混合物的兼容性、均匀性、稳定性、放射性活度以及液闪计数瓶。在Ultima–GoldAB闪烁液中进行混合物配方试验,优化液体闪烁计数性能,完善满足测量需要的样品源制备方法和影响因素的解决办法,得到55Fe探测效率为0.48的理想结果。  相似文献

Gradient-corrected (GGA) and hybrid variants of density functional theory are used to compute geometries and 55Mn chemical shifts of MnO4 , Mn(CO)6 +, Mn2(CO)10, Mn(CO)5 X [X=H, Cl, C(O)Me], Mn(CO)5 , Mn(NO)3(CO), and Mn(C5H5)L x [L x =(CO)3, C6H6, C7H8]. For this set of compounds, substituent effects on δ(55Mn) are significantly underestimated with the pure GGA functional BPW91 and are well described with hybrid functionals such as mPW1PW91 and, in particular, B3LYP. The computed data provide evidence for solvent and counterion effects on δ(55Mn) of MnO4 and Mn(CO)6 +, respectively. The latter, in the presence of Cl, may be described as highly fluxional Mn(CO)5C(O)Cl. Electric field gradients computed with the B3LYP functional can be used for a qualitative rationalization of observed trends in 55Mn NMR line widths. The effect of magnetic nanoparticles(mNPs) on the devitrification crystallization of typical vitrification solution Vs55 was systematically explored by differential scanning calorimetry(DSC) and cryomicroscope system. The results show tliat,(i) the mNPs coated by both carboxylic acid(CA) and polyethylene glycol(PEG) had little effect on the glass transition temperature(rg) of Vs55, but had significant effect on the devitrification transition temperature(7a) and devitrification enthalpy(Hrd),(ii) in the range of the devitrification area(-85- 0℃), the MNPs coated by CA can significantly accelerate the devitrification of Vs55 as the isothermal temperatures and the cooling rates increased, and the ice growth rate was 0.37 p.m/s at the isothermal temperature of-85 ℃, and was about 2.19 gm/s at -75℃. Also, the ice growth rates rose from 1.72 pm/s to 3.54 pm/s when the cooling rates were increased from 2 ℃/min to 100 ℃/min(at the isothennal temperature of -75℃),(iii) magnetic nanoparticles coated by both PEG and CA could promote the devitrification of Vs55, for instance, without any crystal growth inside Vs55 at the isothermal temperature of-80℃, but 1.04 and 2.31 gm/s for adding magnetic nanoparticles coated by CA and PEG, respectively. Compared with the samples coated by CA, PEG promoted the devitrification of Vs55 in a much more positive way, and the ice growth rates were 0.62 and 6.25 μm/s at the isothermal temperatures of -85 and -75 ℃, respectively. Tliese results indicate that the surface coating of MNPs could significantly affect tlie recrystallization of Vs55, and further work should be conducted in the future research.  相似文献

Zusammenfassung Eisen-55 wurde in Neuherberg bei München seit März 1970 in monatlich gesammelten Luftstaubproben und seit Januar 1971 in Monatsniederschlagsproben bestimmt. Nach dem Veraschen der Proben wurde Eisen durch Extraktion' mit Di-iso-propyläther abgetrennt. Zur weiteren Entfernung störender Radionuklide des Fallouts wurde eine Ionenaustauschertrennung angeschlossen. Nach der elektrolytischen Abscheidung des Eisens auf Kupferplättchen wurden die Röntgenspektren mit einem Proportionalzähler sowie einem Si(Li)-Detektor aufgenommen.Die mittlere spezifische Luftaktivität betrug in Neuherberg 1,0 fCi/m3 im Jahr 1970 (ab März), 1,2 fCi/m3 im Jahr 1971 und 0,36 fCi/m3 im Jahr 1972. Die dem Boden durch den Niederschlag zugeführte Aktivität wurde zu 697 pCi/m2 · Jahr für 1971 und 178 pCi/m2 · Jahr für 1972 bestimmt.Vortrag anläßlich der Tagung Spurenanalyse, 2. bis 5. An electrochemiluminescence (ECL) sensor with good long-term stability and fast response time has been developed. The sensor was based on the immobilization of tris(2,2′-bipyridyl)ruthenium(II) (Ru(bpy)32+) into the Eastman-AQ55D–silica composite thin films on a glassy carbon electrode. The ECL and electrochemistry of Ru(bpy)32+ immobilized in the composite thin films have been investigated, and the modified electrode was used for the ECL detection of oxalate, tripropylamine (TPA) and chlorpromazine (CPZ) in a flow injection analysis system and showed high sensitivity. Because of the strong electrostatic interaction and low hydrophobicity of Eastman-AQ55D, the sensor showed no loss of response over 2 months of dry storage. In use, the electrode showed only a 5% decrease in response over 100 potential cycles. The detection limit was 1 μmol l−1 for oxalate and 0.1 μmol l−1 for both TPA and CPZ (S/N=3), respectively. The linear range extended from 50 μmol l−1 to 5 mmol l−1 for oxalate, from 20 μmol l−1 to 1 mmol l−1 for TPA, and from 1 μmol l−1 to 200 μmol l−1 for CPZ.  相似文献

将不同HW沸石含量(以W及L沸石的XRD特定峰强度Iw/IL值表示HW沸石在HL/HW复合沸石中的含量)的HL/HW复合沸石与REUSY沸石按相同比例复配后作为烃类催化裂化催化剂的活性组分,采用标准轻油微反方法对催化剂进行了活性评价(MAT),考察了不同HW含量的HL/HW复合沸石对催化剂活性、反应产物分布、汽油产品辛烷值及催化剂上积炭的影响.结果表明,当,Iw/IL值在0~0.12变化时,MAT指数和汽油收率逐渐上升,比积炭逐渐减少,气体收率先减后增;当Iw/IL值大于0.12时,MAT指数、气体收率和汽油收率均下降,比积炭总体呈增加趋势;当Iw/IL值为0.12时,评价的综合指标达到最佳,与不加HL及HW沸石的参比样品相比,汽油产品的芳烃含量和辛烷值分别提高了9.85%和3.01%,烯烃含量下降了1.30%.  相似文献

Cannabinoids act as pleiotropic compounds exerting, among others, a broad-spectrum of neuroprotective effects. These effects have been investigated in the last years in different preclinical models of neurodegeneration, with the cannabinoid type-1 (CB1) and type-2 (CB2) receptors concentrating an important part of this research. However, the issue has also been extended to additional targets that are also active for cannabinoids, such as the orphan G-protein receptor 55 (GPR55). In the present study, we investigated the neuroprotective potential of VCE-006.1, a chromenopyrazole derivative with biased orthosteric and positive allosteric modulator activity at GPR55, in murine models of two neurodegenerative diseases. First, we proved that VCE-006.1 alone could induce ERK1/2 activation and calcium mobilization, as well as increase cAMP response but only in the presence of lysophosphatidyl inositol. Next, we investigated this compound administered chronically in two neurotoxin-based models of Parkinson’s disease (PD), as well as in some cell-based models. VCE-006.1 was active in reversing the motor defects caused by 6-hydroxydopamine (6-OHDA) in the pole and the cylinder rearing tests, as well as the losses in tyrosine hydroxylase-containing neurons and the elevated glial reactivity detected in the substantia nigra. Similar cytoprotective effects were found in vitro in SH-SY5Y cells exposed to 6-OHDA. We also investigated VCE-006.1 in LPS-lesioned mice with similar beneficial effects, except against glial reactivity and associated inflammatory events, which remained unaltered, a fact confirmed in BV2 cells treated with LPS and VCE-006.1. We also analyzed GPR55 in these in vivo models with no changes in its gene expression, although GPR55 was down-regulated in BV2 cells treated with LPS, which may explain the lack of efficacy of VCE-006.1 in such an assay. Furthermore, we investigated VCE-006.1 in two genetic models of amyotrophic lateral sclerosis (ALS), mutant SOD1, or TDP-43 transgenic mice. Neither the neurological decline nor the deteriorated rotarod performance were prevented with this compound, and the same happened with the elevated microglial and astroglial reactivities, albeit modest spinal motor neuron preservation was achieved in both models. We also analyzed GPR55 in these in vivo models and found no changes in both TDP-43 transgenic and mSOD1 mice. Therefore, our findings support the view that targeting the GPR55 may afford neuroprotection in experimental PD, but not in ALS, thus stressing the specificities for the development of cannabinoid-based therapies in the different neurodegenerative disorders.  相似文献

The stereoselective synthesis of two broussonetine analogues 14·HCl and ent-14·HCl with differing stereochemistry of the polyhydroxylated pyrrolidine core and a simple C13 alkyl fragment has been achieved. For their construction, the known oxazolidinones 15 and 16 were chosen as appropriate advanced scaffolds. The common hydrophobic side chain was incorporated at an early stage of the synthesis through Grubbs’ cross metathesis chemistry. The required pyrrolidine skeleton was then formed by the cyclization of open chain intermediates 17 and 18. Four synthesized compounds were screened in vitro for antiproliferative/cytotoxic activity against six cancer cell lines by MTT assay. Compounds 14·HCl (HeLa and A-549) and ent-14·HCl (Caco-2 and Jurkat) showed comparable or higher potency than conventional anticancer agent cisplatin on at least two evaluated cancer cells, respectively.  相似文献

Using density functional calculations, we demonstrate a catalytic reaction path with activation barriers of less than 0.5 eV for CO oxidation on the neutral and unsupported icosahedral nanoclusters of Au(55), Ag(55), and Au(25)Ag(30). Both CO and O(2) adsorb more strongly on these clusters than on the corresponding bulk surfaces. The reaction path consists of an intermediate involving OOCO complex through which the coadsorption energy of CO and O(2) on these clusters is expected to play an important role in the reaction. Based on the studies for the Au and Ag nanoclusters, a model alloy nanocluster of Au(25)Ag(30) was designed to provide a larger coadsorption energy for CO and O(2) and was anticipated to be a better catalyst for CO oxidation from energetic analysis.  相似文献
