An efficient domino approach for the synthesis of novel pyrrolo[2,3,4-kl]acridin-1-one derivatives has been established. This reaction represents the first facile conversion of an isatin to a pyrrolo[2,3,4-kl]acridin-1-one via a C-N bond cleavage reaction without the need for a multistep reaction process.  相似文献

A series of molecules of therapeutic interest, possessing the new skeleton of 1H-benzo[c]pyrido[2,3,4-kl]acridine with acyl or aminoacyl and methoxy or aminoalkoxy substituents on the aromatic homocycles were synthesized by means of a Friedl?nder-type reaction. The requisite 5-aminodihydroquinoline-4-ones 1, whose preparation is described, were reacted with the appropriate alpha-tetralones 2 using an acidic catalyst (PPTS) under azeotropic conditions. Optimized reaction time and yield depend on temperature, which must not be below 90 degrees C.  相似文献

Pyrido[4,3,2-kl]acridines represent a new class of heterocycles, isomers of marine alkaloids. The 7H-pyrido[4,3,2-kl]acridine reacts as an electron rich heterocycle, and in particular via electrophilic substitution such as H/D exchange and the Vilsmeier-Haack reaction. The reaction is fully regioselective and gives the corresponding 6-substituted derivatives. The pyrido[4,3,2-kl]acridin-4-one reacts with amines and thiol, via 1,4-Michael addition to give the 6-amino or 6-thio analogues in a very efficient way. Molecular calculations account for the observed regioselectivity.  相似文献

通过氮杂Wittig反应合成了一系列结构新颖的未见文献报道的取代四氢苯并[4',5']噻吩并[3',2':5,6]吡啶并[4,3-d]嘧啶-4(3H)-酮化合物6, 产物的结构通过核磁共振氢谱、质谱、红外光谱、元素分析的确证. 为进一步了解此类化合物的结构, 对化合物6c进行了X射线单晶衍射. 初步杀菌活性测定结果表明, 该类化合物具有较好的杀菌活性和选择性. 如6f在50 mg/L浓度时, 对黄瓜灰霉菌的抑制率为98%.  相似文献

利用串联Aza-Wittig法合成了两类共计16个未见文献报道的噻吩并吡啶并嘧啶类化合物, 通过IR, 1H NMR, EI-MS, 元素分析等方法对所合成的化合物进行了结构表征, 化合物II-1还通过了X单晶衍射的进一步确证. 并初步测定了所合成化合物的杀虫、杀菌和除草活性. 结果表明: 所测化合物的杀虫活性较差, 但大部分化合物具有一定的除草活性和良好的杀菌活性.  相似文献

Starting from commercially available educts, a straightforward synthetic route to new heterocyclic building blocks is exemplified with the one- or two-step synthesis of tri-, tetra-, or pentacyclic ring systems. Representatives of the following novel ring systems are prepared from 3-methyl-1-phenyl-2-pyrazolin-5-one and the corresponding o-halo-arenecarbonyl chloride using calcium hydroxide in refluxing 1,4-dioxane: pyrimidino[4',5':5,6]pyrano[2,3-c]pyrazol-4(1H)-one, thieno[3',2':5,6]pyrano[2,3c]pyrazol- 4-(1H)-one, thieno[3',4':5,6]pyrano[2,3-c]pyrazol-4(1H)-one, thieno[3',2':4',5']thieno[2',3':5,6]-pyrano[2,3-c]pyrazol-4(1H)-one, [1,3]dioxolo[5',6'][1]benzothieno[2',3':5,6]pyrano-[2,3-c]- pyrazol-4(1H)-one, pyridazino[4',3':5,6]pyrano[2,3-c]pyrazol-4(1H)-one and pyrazolo-[4',3':5',6']pyrido[3',4':5,6]pyrano[2,3-c]pyrazol-4(1H)-one. While the latter two ring systems are directly obtained due to a spontaneous intramolecular substitution reaction, in the other reactions uncyclised 4-aroylpyrazol-5-ols are produced, which are cyclised into the target heterocycles in a subsequent synthetic step (i.e. treatment with NaH in DMF). Detailed NMR spectroscopic investigations ((1)H-, (13)C-, (15)N-) with the obtained compounds were undertaken to unambiguously prove the new structures.  相似文献

用邻氨基噻吩腈与乙酰乙酸甲酯为原料,利用串联Aza-Wittig法合成了9个未见文献报道的噻吩并吡啶并嘧啶类化合物,通过IR,1H NMR,EI-MS,元素分析等方法对所合成的化合物进行了结构表征,并初步测定了所合成化合物的杀菌活性.生测结果表明:此类化合物对棉花枯萎(Fusarium oxysporium)、水稻纹枯(Rhizoctonia solani)、小麦赤霉(Gibberella zeae)、苹果轮纹菌(Dothiorella gregaria)、棉花炭疽菌(Colletotrichum gossypii)等5种菌均有一定的抑制活性.其中对苹果轮纹菌抑制活性最好,在浓度为5.0×10-5g/mL时抑制率为31.80%~82.84%.  相似文献

Heating 1,1-bis(5,5-dimethyl-1,3-cyclohexanedion-2-yl)acetone with ammonia gave a new heterocyclic compound, namely, 1,4,4,8,8-pentamethyl-2,3,4,5,7,8,9,10-octahydropyrrolo[4,3,2-m,n]acridin-10-one. Analogously, 3,6-dimethyl-9-benzoyl-1,2,3,4,5,6,7,8-octahydroxanthene-1,8-dione gave 4,8-dimethyl-1-phenyl-2,3,4,-5,7,8,9,10-octahydropyrrolo[4,3,2-m,n]acridin-10-one. Lithiation of 1-vinylbenzotriazole 9 with n-BuLi (2 equiv) generates dianion 10, which upon subsequent reaction with 1,2- and 1,4-diketones affords 14 and 13, representatives of the 5,6-dihydro-4H-[1,2,3]triazolo[4,5,1-ij]quinoline 1 and 5,6,7,8-tetrahydro-4H-[1,2,3]triazolo[4,5,1-kl][1]benzazocine 2 ring systems, respectively. Reactions of dianion 10 with isocyanates give 15a,b, which contain the 4,5,6,7-tetrahydro[1,2,3]triazolo[4,5,1-jk][1,4]benzodiazepine 3 ring system.  相似文献

本文以环己酮为原料,通过氮杂Wittig反应合成了一系列结构新颖的取代四氢苯并噻吩并吡啶并嘧啶衍生物,并采用MTT法考察所合成目标化合物对CNE2、KB、MGC-803、MCF-7和PC3这5种肿瘤细胞的抑制活性。初步的生物活性结果表明,目标化合物对5种肿瘤细胞均有抑制活性,尤其是对胃癌MGC-803细胞展现出了更强的抑制活性。其中3-(4-氟苯基)-2-((4-氟苯基)氨基)-5-甲基-8,9,10,11-四氢苯并[4',5']噻吩并[3',2':5,6]吡啶并[4,3-d]嘧啶-4(3H)-酮[化合物8c,IC_(50)=(0. 9±0. 25)μmol·L~(-1)]对MGC-803的活性最强,是5-氟尿嘧啶[IC_(50)=(18. 4±1. 43)μmol·L~(-1)]的20倍;同时,目标化合物对正常的胃黏膜上皮细胞GES-1没有毒性。四氢苯并[4',5']噻吩并[3',2':5,6]吡啶并[4,3-d]嘧啶类化合物具有良好的抗肿瘤活性,值得进一步深入研究。  相似文献

Substituted pyrido,[3',2':4,5]thieno[3,2-b]pyndines were obtained by the reaction of 3-amino-2-benzoylthieno [2,3-b]pyridines with malononitrile and the reaction of 3-cyanopyridine-2(IH)-thiones with 2-aryl-3-bromo-I,I-dicyanopropene. 2-Amino-4-(4-bromophenyl)-7, 9-dimethyl-3-cyanopyrido [3',2':4,5]thieno[3, 2-b]-pyridine was used for the synthesis of a derivative of pyrido[3",2":4', 5']thieno[2',3':5,6]pyrido[2,3-d]-pyrimidine. The structure of these compounds was confirmed by spectral data and x-ray diffraction structural analysis.Deceased.  相似文献

The synthesis of 3-allylthio-5H-[1,2,4]triazino[5,6-b]indole has been carried out by the reaction of 3-mercapto-5H-[1,2,4]triazino[5,6-b]indole with allyl bromide in the NaOH–H2O–DMSO system and in a one-pot synthesis from isatin-β-thiosemicarbazide. Halocyclization of allylthio-5H-[1,2,4]-triazino[5,6-b]indole synthesized the 3-halomethyl-3,10-dihydro-2H-[1,3]thiazolo[3',2':2,3][1,2,4]-triazino[5,6-b]indolium halides.  相似文献

5,6-Dihydropyrimido[5′,4′: 5,6]pyrido[1,2-a]benzimidazole and pyrimido[4′,5′: 4,5]pyrimido[1,6-a]-benzimidazole derivatives were synthesized starting from 3-[4-hydroxy-6-methyl(hydroxy)-2-phenylpyrimidin-5-yl]propanoic and 4-hydroxy-2-phenylpyrimidine-5-carboxylic acids. 应用串联氮杂Wittig反应设计合成了一系列新的2-烷氨(芳氧)基-3,8-二苯基-5-芳基-6-甲基-5,8-二氢-4H-吡唑并[4',3':5,6]吡喃并[2,3-d]嘧啶-4(3H)-酮衍生物。通过氢核磁共振谱仪(1H NMR)、傅里叶变换红外光谱仪(FTIR)和元素分析等方法对所合成的化合物进行了结构表征,用X射线单晶衍射确证了化合物2-二正丙基氨基-3,8-二苯基-5-(4-甲基苯基)-6-甲基-5,8-二氢-4H-吡唑并[4',3':5,6]吡喃并[2,3-d]嘧啶-4(3H)-酮(Ⅲi)晶体结构。室内的杀菌和杀虫活性测试结果表明:目标化合物具有一定的杀菌活性,其中部分化合物对黄瓜灰霉菌(Botrytis cinereapers)在50 mg/L剂量下抑制率达到90%以上,显示了优异的杀菌活性;对水稻褐飞虱无明显的杀虫活性,但发现对粘虫具有较高的杀灭效果,大多数抑制率达到90%以上。  相似文献

The reaction of chromone-3-carbonitrile with thiobarbituric acid afforded 2-thioxochromeno[3′,2′:5,6]pyrido[2,3-d]pyrimidine-4,6(1H,3H)-dione, which utilized as a building block for construction of a novel heteroannulated compounds containing chromenopyridothiazolopyrimidine moiety. Reactions of the starting compound with a variety of bielectrophilic reagents namely; chloroacetonitrile, bromomalononitrile, 3-chloropentanedione, ethyl 2-chloro-3-oxobutanoate, phenacyl bromide, chloroacetic acid, dibromoethane, and oxalyl chloride afforded the first known chromenopyridothiazolopyrimidines. Cyclization of the starting compound with 2,3-dichloroquinoxaline gave the linear hepta-annulated chromenopyridopyrimidothiazoloquinoxaline. In addition, chromenopyridopyrimido thiazolopyrimidines were efficiently synthesized. The antimicrobial activity was evaluated for the prepared compounds and some of them seemed notable activity against the tested microorganisms. Analytical and spectral data confirmed the structures of the new products.  相似文献
