This investigation was carried out with the aim of determining the effect of paclobutrazol (PBZ) (0 and 2 mg l?1) and polyethylene glycol (PEG) (0, 2, 4 and 6 %?w/v of PEG 6000) treatments on antioxidant system of Stevia rebaudiana Bertoni under in vitro condition. Analysis of data showed that PEG treatment significantly increased hydrogen peroxide (H2O2) and phenolic contents, while PBZ treatment limited the effect of PEG on them. Our data revealed that PEG treatment significantly increased total antioxidant capacity, catalase (CAT), ascorbate peroxidase (APX), polyphenol oxidase (PPO) and peroxidase (POD) activity, while it inversely decreased glutathione reductase (GR) activity. The superoxide dismutase (SOD) activity was not affected by PEG treatment. PBZ treatment induced significantly higher levels of CAT and GR activity and lower levels of SOD activity in PEG-treated plants. PBZ in combination with PEG resulted in no significant difference on APX activity with PEG treatment alone. PBZ treatment prevented the effect of PEG on the PPO activity. PEG (with or without PBZ) treatment increased the ascorbate pool, whereas total glutathione level was not affected by PEG. Our finding indicated that PBZ reduced the negative effect of PEG treatment by quenching H2O2 accumulation and increasing the CAT activity. Collectively, the antioxidant capacity of S. rebaudiana in PEG treatment condition was associated with active enzymatic and non-enzymatic defence systems which partly could be improved by the PBZ treatment. In addition, a higher accumulation of phenolic compounds leads to a more potent reactive oxygen species scavenging activity in S. rebaudiana.  相似文献

The dietary properties of minikiwi make them, along with other fruits and vegetables, suitable as the basis for many slimming and pro-health diets. Prolonging the availability of minikiwi can be provided by different storage technologies. This experiment focused on evaluating the effect of various O2 and CO2 concentrations, i.e., low-oxygen atmosphere (DCA, 0.4% CO2:0.4% O2; ULO, 1.5% CO2:1.5% O2) or high-CO2 (CA, 5% CO2:1.5% O2) storage, in order to provide the consumer with fruits with comparable high nutritional values. Evaluation gave the basic characteristics of the fruits that characterize their health-promoting properties, i.e., total polyphenols (TPC), phenolic acids and flavonols, antioxidant activity (AA), monosaccharides, and acid content. The atmosphere with a higher CO2 content of 5% (CA) effectively influenced the high value of ascorbic acid even after 12 weeks of storage. DCA technology contributed to a significant inhibition of phenol loss but not as effectively as CA technology. In contrast, glucose and fructose contents were found to be significantly higher after storage in ULO or DCA, while sucrose content was more stable in fruit stored in CA or DCA. CA technology conditions stabilized the citric acid content of minikiwi, while DCA technology was less effective in inhibiting acid loss. The nutritional value of the fruit after storage in CA or DCA was not significantly reduced, which will allow the supply of fresh minikiwi fruit to be extended and provide a valuable component of the human diet.  相似文献

Redox homeostasis is one of the main reasons for reactive oxygen species (ROS) tolerance in hypoxic tumors, limiting ROS-mediated tumor therapy. Proposed herein is a redox dyshomeostasis (RDH) strategy based on a nanoplatform, FeCysPW@ZIF-82@CAT Dz, to disrupt redox homeostasis, and its application to improve ROS-mediated hypoxic tumor therapy. Once endocytosed by tumor cells, the catalase DNAzyme (CAT Dz) loaded zeolitic imidazole framework-82 (ZIF-82@CAT Dz) shell can be degraded into Zn2+ as cofactors for CAT Dz mediated CAT silencing and electrophilic ligands for glutathione (GSH) depletion under hypoxia, both of which lead to intracellular RDH and H2O2 accumulation. These “disordered” cells show reduced resistance to ROS and are effectively killed by ferrous cysteine-phosphotungstate (FeCysPW) induced chemodynamic therapy (CDT). In vitro and in vivo data demonstrate that the pH/hypoxia/H2O2 triple stimuli responsive nanocomposite can efficiently kill hypoxic tumors. Overall, the RDH strategy provides a new way of thinking about ROS-mediated treatment of hypoxic tumors.  相似文献

Functionalized‐multiwall carbon nanotubes decorated with redox active copper nanoparticles have been fabricated for sensitive enzyme‐less H2O2 detection. The new nanocomposite was characterized by Transmission electron microscopy, energy dispersive X‐ray analysis and cyclic voltammetry. The response of the modified electrode to H2O2 was examined using amperometry at ?0.45 V vs. Ag/AgCl in a buffer solution at pH 10.0. The developed sensor displayed linear concentration ranges of 0.5–10.0 and 10.0–10000.0 µmol L?1 with a detection limit of 0.3 µmol L?1. The proposed sensor displayed good selectivity for H2O2 detection in the presence of common interferences such as ascorbic acid.  相似文献

The juice extracted from pomegranate fruits containing cyanin (flavylium) is utilized as the light-harvesting analog in a dye sensitized solid-state photovoltaic cell. Strong chelation of flavylium with TiO2 changes it to quinonoidal form. A higher incident photon to current conversion efficiency is observed in solid-state TiO2|pomegranate piment|CuI solar cell compare to that of the cells (TiO2|dye|CuI type) sensitized with other natural pigments.  相似文献

The generation of singlet oxygen (1O2) during photodynamic therapy is limited by the precise cooperation of light, photosensitizer, and oxygen, and the therapeutic efficiency is restricted by the elevated glutathione (GSH) levels in cancer cells. Herein, we report that an ultrathin two‐dimensional metal–organic framework of Cu‐TCPP nanosheets (TCPP=tetrakis(4‐carboxyphenyl)porphyrin) can selectively generate 1O2 in a tumor microenvironment. This process is based on the peroxidation of the TCPP ligand by acidic H2O2 followed by reduction to peroxyl radicals under the action of the peroxidase‐like nanosheets and Cu2+, and their spontaneous recombination reaction by the Russell mechanism. In addition, the nanosheets can also deplete GSH. Consequently, the Cu‐TCPP nanosheets can selectively destroy tumor cells with high efficiency, constituting an attractive way to overcome current limitations of photodynamic therapy.  相似文献

To understand the effect of enhanced UV-B radiation and low-energy N+ ion beam radiation on the response of photosynthesis, antioxidant enzymes, and lipid peroxidation in rice seedlings, Oryza sativa was exposed to three different doses of low-energy N+ ion beam and enhanced UV-B alone and in combination. Enhanced UV-B caused a marked decline in some photosynthetic parameters (net photosynthetic rate, transpiration rate, and stomatal conductance) and photosynthetic pigments, whereas it induced an increase in hydrogen peroxide (H2O2) accumulation, the rate of superoxide radical production, and the content of malondialdehyde (MDA). Enhanced UV-B also induced an increase in the activity of antioxidant enzymes (superoxide dismutase [SOD], peroxidase (POD), and catalase [CAT]) and some nonenzymatic antioxidants such as proline. Under the combined treatment of enhanced UV-B and low-energy N+ ion beam at the dose of 3.0?×?1017 N+ cm?2, the activity of antioxidant compounds (SOD, POD, CAT, proline, and glutathione), photosynthetic pigments, and some photosynthetic parameters (net photosynthetic rate, transpiration rate, and stomatal conductance) increased significantly; however, the MDA content, H2O2 accumulation, and rate of superoxide radical production showed a remarkable decrease compared with the enhanced UV-B treatment alone. These results implied that the appropriate dose of low-energy N+ ion beam treatment may alleviate the damage caused by the enhanced UV-B radiation on rice.  相似文献

Ozone (O3) is an oxidating tropospheric pollutant. When O3 interacts with biological substrates, reactive oxygen and nitrogen species (RONS) are formed. Severe oxidative damage exhausts the endogenous antioxidant system, which leads to the decreased activity of antioxidant enzymes such as catalase (CAT), glutathione peroxidase (GPx), and superoxide dismutase (SOD). Curcumin (CUR) is a natural polyphenol with well-documented antioxidant and anti-inflammatory properties. The aim of this work is to evaluate the effects of curcumin on CAT, GPx, and SOD activity and the inhibition of oxidative damage after the acute and chronic exposure to O3. Fifty male Wistar rats were divided into five experimental groups: the intact control, CUR-fed control, exposed-to-O3 control, CUR-fed (preventive), and CUR-fed (therapeutic) groups. These two last groups received a CUR-supplemented diet while exposed to O3. These experiments were performed during acute- and chronic-exposure phases. In the preventive and therapeutic groups, the activity of plasma CAT, GPx, and SOD was increased during both exposure phases, with slight differences; concomitantly, lipid peroxidation and protein carbonylation were inhibited. For this reason, we propose that CUR could be used to enhance the activity of the antioxidant system and to diminish the oxidative damage caused by exposure to O3.  相似文献

Several modern drugs, which are derived from traditional herbal medicine are used in contemporary pharmacotherapy. Currently, the study of drug–plant interactions in pain has increased in recent years, looking for greater efficacy of the drug and reduce side effects. The antinociception induced by intragastric co-administration of the combination of pomegranate peel extract (PoPEx) and acetylsalicylic acid (ASA) was assessed using the isobolographic analysis in formalin test (nociceptive and inflammatory pain). The effective dose that produced 30% of antinociception (ED30) was calculated for both drugs from the logarithmic dose–response curves, subsequently generating a curve with the combination on fixed proportions (1:1) of PoPEx and ASA. Through isobolographic analysis, this experimental ED30 was compared with the calculated theoretical additive ED30. The result was a synergistic interaction, the experimental ED30 was significantly smaller (p < 0.05) than the theoretical ED30. The antinociceptive mechanism of the PoPEx-ASA combination involves the l-Arginine/NO/cGMP pathway, antioxidant capacity, and high content of total phenols. These findings suggest that an interaction between PoPEx and ASA could be a novel treatment for inflammatory and nociceptive pain, also diminish the secondary reactions of ASA.  相似文献

Anticancer treatment is largely affected by the hypoxic tumor microenvironment (TME), which causes the resistance of the tumor to radiotherapy. Combining radiosensitizer compounds and O2 self-enriched moieties is an emerging strategy in hypoxic-tumor treatments. Herein, we engineered GdW10@PDA-CAT (K3Na4H2GdW10O36·2H2O, GdW10, polydopamine, PDA, catalase, CAT) composites as a radiosensitizer for the TME-manipulated enhancement of radiotherapy. In the composites, Gd (Z = 64) and W (Z = 74), as the high Z elements, make X-ray gather in tumor cells, thereby enhancing DNA damage induced by radiation. CAT can convert H2O2 to O2 and H2O to enhance the X-ray effect under hypoxic TME. CAT and PDA modification enhances the biocompatibility of the composites. Our results showed that GdW10@PDA-CAT composites increased the efficiency of radiotherapy in HT29 cells in culture. This polyoxometalates and O2 self-supplement composites provide a promising radiosensitizer for the radiotherapy field.  相似文献

A microfluidic chip featuring laminar flow-based parallel gradient-generating networks was designed and fabricated. The microchip contains 5 gradient generators and 30 cell chambers where the resulting concentration gradients of drugs are delivered to stimulate on-chip cultured cells. The microfluidics exploits the advantage of lab-on-a-chip technology by integrating the generation of drug concentration gradients and a series of cell operations including seeding, culture, stimulation and staining into a chip. The microfluidic network was patterned on a glass wafer, which was further bonded to a PDMS film. A series of weir structures were fabricated on the cell culture reservoir to facilitate cell positioning and seeding. Cell injection and fluid delivery were controlled by a syringe pump. Steady parallel concentration gradients were generated by flowing two fluids in each network. Over time observation shows that the microchip was suitable for cell seeding and culture. The microchip described above was applied in studying the role of reduced glutathione (GSH) in mediating chemotherapy sensitivity of MCF-7 cells. MCF-7 cells were treated with concentration gradients of As2O3 and N-acetyl cysteine (NAC) for GSH modulation, followed by exposure to adriamycin. GSH levels were down-regulated upon As2O3 treatment and up-regulated upon NAC treatment. Suppression of intracellular GSH by treatment with As2O3 has been shown to increase sensitivity to adriamycin. Conversely, elevation of intracellular GSH by treatment with NAC leads to increased drug resistance. Four selected Amaranthus tricolor cultivars were grown under four irrigation regimes (25, 50, 80, and 100% field capacity) to evaluate the mechanisms of growth and physiological and biochemical responses against drought stress in randomized complete block design with three replications. Drought stress led to decrease in total biomass, specific leaf area, relative water content (RWC), photosynthetic pigments (chlorophyll a, chlorophyll b, chlorophyll ab), and soluble protein and increase in MDA, H2O2, EL, proline, total carotenoid, ascorbic acid, polyphenols, flavonoids, and antioxidant activity. However, responses of these parameters were differential in respect to cultivars and the degree of drought stresses. No significant difference was observed in control and LDS for most of the traits. The cultivars VA14 and VA16 were identified as more tolerant to drought and could be used for further evaluations in future breeding programs and new cultivar release programs. Positively significant correlations among MDA, H2O2, compatible solutes, and non-enzymatic antioxidant (proline, TPC, TFC, and TAC) suggested that compatible solutes and non-enzymatic antioxidant played vital role in detoxifying of ROS in A. tricolor cultivar. The increased content of ascorbic acid indicated the crucial role of the ASC–GSH cycle for scavenging ROS in A. tricolor.  相似文献

As an antioxidant, procyanidin B1(PB1) can improve the development of somatic cell nuclear transfer (SCNT) embryos; PB1 reduces the level of oxidative stress (OS) during the in vitro development of SCNT embryos by decreasing the level of reactive oxygen species (ROS) and increasing the level of glutathione (GSH) and mitochondrial membrane potential (MMP). Metabolite hydrogen peroxide (H2O2) produces OS. Catalase (CAT) can degrade hydrogen peroxide so that it produces less toxic water (H2O) and oxygen (O2) in order to reduce the harm caused by H2O2. Therefore, we tested the CAT level in the in vitro development of SCNT embryos; it was found that PB1 can increase the expression of CAT, indicating that PB1 can offset the harm caused by oxidative stress by increasing the level of CAT. Moreover, if H2O2 accumulates excessively, it produces radical-(HO-) through Fe2+/3+ and damage to DNA. The damage caused to the DNA is mainly repaired by the protein encoded by the DNA damage repair gene. Therefore, we tested the expression of the DNA damage repair gene, OGG1. It was found that PB1 can increase the expression of OGG1 and increase the expression of protein. Through the above test, we proved that PB1 can improve the repairability of DNA damage. DNA damage can lead to cell apoptosis; therefore, we also tested the level of apoptosis of blastocysts, and we found that PB1 reduced the level of apoptosis. In summary, our results show that PB1 reduces the accumulation of H2O2 by decreasing the level of OS during the in vitro development of SCNT embryos and improves the repairability of DNA damage to reduce cell apoptosis. Our results have important significance for the improvement of the development of SCNT embryos in vitro and provide important reference significance for diseases that can be treated using SCNT technology.  相似文献

Flavonols (FLA) from Vaccinium macrocarpon (V. macrocarpon) were identified using high-performance liquid chromatography coupled with mass spectrometry detection. Nanoparticles were prepared using highly crosslinked keratin (KER) from human hair and silver and entrapped with flavonols [KER + FLA + AgNPs]. Nanocomposites were characterized using UV–Vis spectroscopy, transmission electron microscopy (TEM), X-ray diffraction, zeta potential, and dynamic light scattering, and release profiles. The interactions between the capping agent and the silver core have been investigated using FTIR spectroscopy·H2O2 is a source of Reactive Oxygen Species (ROS) and acts as an activator of oxidative stress affecting NS-1 cells, and the protective effect of the nanocomposites were evaluated against H2O2-induced pancreatic β-cell damage. LC-MS/MS and HPLC analyses revealed the presence of 12 flavonols in V. macrocarpon plant extract. The cell apoptosis and proliferation, were evaluated by Hoechst 33342 staining, flow cytometry and Cell Counting Kit-8 respectively. Preincubation of the NS-1 cells with 250 µg/mL of H2O2 induced oxidative stress conditions that show pancreatic β-cell dysfunction, including ROS, cell death, mitochondrial function, antioxidant enzymes, and lipid peroxidation. Nevertheless, pretreatment with FLA and [KER + FLA + AgNPs] prevented mitochondria disruption, maintained cellular ATP levels, inhibited LDH release, intracellular ROS production, decreased lipid peroxidation, increased expression of antioxidant enzymes (CAT, SOD, and GPx) and GSH levels. These results indicate that nanocomposites could protect rat INS-1 pancreatic β-cell from H2O2-induced oxidative damage, apoptosis and proliferation by reducing the production of intracellular reactive oxygen species.  相似文献

The novel hierarchical flower-like superstructure NiCo2O4/reduced graphene oxide (rGO) hybrids have been successfully synthesized with a facile one-step hydrothermal process for the determination of fungicide pyrimethanil (PMT). For comparison, various structures of NiCo2O4/rGO including hexagonal nanoplates and nanorods were also synthesized. Among them, three-dimensional (3D) flower-like NiCo2O4/rGO exhibited the highest electrocatalytic activity for the oxidation of PMT. With the synergistic effect of [OMIM]PF6 ionic liquid (IL), the electrochemical sensor film (NiCo2O4/rGO/IL) further facilitated interfacial electron transfer and enhanced electrocatalytic activity for the oxidation of PMT. Under the optimum conditions, the electrochemical sensor exhibited two linear ranges of 0.1–10.0 μmol/L and 20.0–140 μmol/L for PMT with a low detection concentration of 11.0 nmol/L. Besides, the interference, repeatability, reproducibility and stability measurements were also evaluated. The proposed method was successfully applied to the detection of PMT in water, seawater, fruits and vegetables with good recovery ranging from 93% to 105%, and possessed potential applications in the analysis of real samples.  相似文献

The present study is designed to investigate the neuroprotective effect of a kind of phlorotannins, diphlorethohydroxycarmalol (DPHC) isolated from Ishige okamurae against hydrogen peroxide (H2O2)-induced oxidative stress in murine hippocampal neuronal cells, HT22. H2O2 treatment induced neurotoxicity, whereas DPHC prevented cells from H2O2-induced damage then restoring cell viability was significantly increased. DPHC slightly reduced the expression of Bax induced by H2O2 but recovered the expression of Bcl-xL as well as caspase-9 and -3 mediated PARP cleavage by H2O2. Intracellular reactive oxygen species (ROS) and lipid peroxidation was overproduced as the result of the addition of H2O2; however, these ROS generations and lipid peroxidation were effectively inhibited by addition of DPHC in a dose-dependent manner. Moreover, DPHC suppressed the elevation of H2O2-induced Ca2+ release. These findings indicate that DPHC has neuroprotective effects against H2O2-induced damage in neuronal cells, and that an inhibitory effect on ROS production may contribute to the underlying mechanisms.  相似文献

Biodegradable polycaprolactone (PCL) nanosphere encapsulating superoxide dismutase (SOD) and catalase (CAT) were successfully synthesized using double emulsion (w/o/w) solvent evaporation technique. Characterization of the nanosphere using dynamic light scattering, field emission scanning electron microscope, and Fourier transform infrared spectroscopy revealed a spherical-shaped nanosphere in a size range of 812?±?64 nm with moderate protein encapsulation efficiency of 55.42?±?3.7 % and high in vitro protein release. Human skin HaCat cells were used for analyzing antioxidative properties of SOD- and CAT-encapsulated PCL nanospheres. Oxidative stress condition in HaCat cells was optimized with exposure to hydrogen peroxide (H2O2; 1 mM) as external stress factor and verified through reactive oxygen species (ROS) analysis using H2DCFDA dye. PCL nanosphere encapsulating SOD and CAT together indicated better antioxidative defense against H2O2-induced oxidative stress in human skin HaCat cells in comparison to PCL encapsulating either SOD or CAT alone as well as against direct supplement of SOD and CAT protein solution. Increase in HaCat cells SOD and CAT activities after treatment hints toward uptake of PCL nanosphere into the human skin HaCat cells. The result signifies the role of PCL-encapsulating SOD and CAT nanosphere in alleviating oxidative stress.  相似文献

Although endogenous H2O2 is overexpressed in tumor tissue, the amount of endogenous H2O2 is still insufficient for chemodynamic therapy (CDT). In addition, the abundant cellular glutathione (GSH) could also consume ?OH for reduced CDT. Thus, the elevation of H2O2 and the consumption of GSH in tumor tissue are essential for the increased ?OH yield and amplified CDT efficacy. In this paper, host-guest interactions based supramolecular complexes self-assemblies (SCSAs) were fabricated by incorporating cinnamaldehyde (CA) and PEG-modified cyclodextrin host units (mPEG-CD-CA) with ferrocene-(phenylboronic acid pinacol ester) conjugates (Fc-BE) on the basis of CD-induced host-guest interactions. After being internalized by cancer cells, CA can be released from SCSAs through the pH-responsive acetal linkage, elevating the H2O2 level by activating NADPH oxidase. Then, Fc can catalyze the H2O2 to higher cytotoxic hydroxyl radicals (?OH). Moreover, quinone methide (QM) can be produced through H2O2-induced aryl boronic ester rearrangement and further consume the antioxidant GSH. In vitro and in vivo experiments demonstrate that SCSAs can be provided as potential amplified CDT nanoagents.  相似文献

A carbon fiber microelectrode, surface of which ruthenium and glucose oxidase (GOx) were electrochemically codeposited, has been investigated. The Ru deposition onto the microelectrode increased current response to H2O2 oxidation, while decreased oxidation currents due to interfering substances, such as ascorbic acid, uric acid, p-acetamidophenol, l-cysteine and dopamine. The codeposition of Ru and GOx gave further suppression of the interfering signals with keeping the current response to H2O2. When amperometric glucose sensing was conducted by using the GOx and Ru modified microelectrode, an increase in GOx concentration in the deposition bath enlarged oxidation current of H2O2 generated from glucose oxidation by GOx. The presence of ascorbic acid in analyte gave no error in detection of glucose and errors caused by uric acid was +3% at the most for measuring 5 mM glucose, which is the normal physiological level in blood.  相似文献

Oxidative stress in aquatic organisms might suppress the immune system and propagate infectious diseases. This study aimed to investigate the protective effect of polyphenolic extracts from spent coffee grounds (SCG) against oxidative stress, induced by H2O2, in C. viridis brain cells, through an in vitro model. Hydrophilic extracts from SCG are rich in quinic, ferulic and caffeic acids and showed antioxidant capacity in DPPH, ORAC and FRAP assays. Furthermore, pretreatment of C. viridis brain cells with the polyphenolic extracts from SCG (230 and 460 µg/mL) for 24 h prior to 100 µM H2O2 exposure (1 h) significantly increased antioxidant enzymes activity (superoxide dismutase and catalase) and reduced lipid peroxidation (measured by MDA levels). These results suggest that polyphenols found in SCG extracts exert an antioxidative protective effect against oxidative stress in C. viridis brain cells by stimulating the activity of SOD and CAT.  相似文献
