In present study, a multiple columns and detectors liquid chromatography system for analysis of global components in traditional Chinese medicines was developed. The liquid chromatography system was consist of three columns, including size exclusion chromatography column, hydrophilic interaction chromatography column, and reversed phase chromato‐graphy column, and three detectors, such as diode array detector, evaporative light scattering detector, and mass spectrometry detector, based on column switching technique. The developed multiple columns and detectors liquid chromatography system was successfully applied to the analysis of global components, including macromolecular (polysaccharides), high (nucleosides and sugars)‐, and low (triterpenes)‐polarity small molecular compounds in Ganoderma, a well‐known Chinese medicinal mushroom. As a result, one macromolecular chromatographic peak was found in two Ganoderma species, 19 components were identified in Ganoderma lucidum (two sugars, three nucleosides, and 14 triterpenes), and four components (two sugars and two nucleosides) were identified in Ganoderma sinense. The developed multiple columns and detectors liquid chromatography system was helpful to understand comprehensive chemical characters in TCMs.  相似文献

We prepared hybrid particle-monolithic polymethacrylate columns for micro-HPLC by in situ polymerization in fused silica capillaries pre-packed with 3–5 μm C18 and aminopropyl silica bonded particles, using polymerization mixtures based on laurylmethacrylate–ethylene dimethacrylate (co)polymers for the reversed-phase (RP) mode and [2-(methacryloyloxy)ethyl]-dimethyl-(3-sulfopropyl) zwitterionic (co)polymers for the hydrophilic interaction (HILIC) mode. The hybrid particle-monolithic columns showed reduced porosity and hold-up volumes, approximately 2–2.5 times lower in comparison to the pure monolithic columns prepared in the whole volume of empty capillaries. The elution volumes of sample compounds are also generally lower in comparison to packed or pure monolithic columns. The efficiency and permeability of the hybrid columns are intermediate in between the properties of the reference pure monolithic and particle-packed columns. The chemistries of the embedded solid particles and of the interparticle monolithic moiety in the hybrid capillary columns contribute to the retention to various degrees, affecting the selectivity of separation. Some hybrid columns provided improved separations of proteins in comparison to the reference particle-packed columns in the reversed-phase mode. Zwitterionic hybrid particle-monolithic columns show dual mode retention HILIC/RP behaviour depending on the composition of the mobile phase and allow separations of polar compounds such as phenolic acids in the HILIC mode at lower concentrations of acetonitrile and, often in shorter analysis time in comparison to particle-packed and full-volume monolithic columns.  相似文献

Using a specially designed column system, we have systematically investigated the effect of mobile phase velocity on column efficiency. The performance of small bore columns operated at different linear velocities of mobile phase was examined for three different types of injection system. The theoretical advantages and drawbacks of using a multiple-, parallel column approach in on-line multidimensional liquid chromatography systems were investigated. Much time or peak capacity can be gained with the use of multiple parallel columns at the second-dimension while the aggregate time of separation increases only by the increment of the gradient time of the second-dimension. Multidimensional chromatographic systems are now used to perform many tasks ranging from routine, fast analyses to specialized, arduous separations. In this work, we focus on the advantages of a multiple, parallel columns approach to on-line multidimensional liquid chromatography systems. Calculations of the achievable peak capacities were made as functions of the number of columns operated in parallel. Increasing the number of second-dimension columns from one to two or three causes the largest increase in peak capacity with only a slight increase of aggregate time. We also present some practical aspects to consider when attempting multidimensional separations with multiple columns operated in parallel.  相似文献

A fast method using high-performance liquid chromatography based on two monolithic columns has been developed for the simultaneous determination of isoflavones extracted from soybeans and derived foods. The 12 main isoflavones were resolved in 10 min in two coupled monolithic columns working at 35 °C using a elution gradient of acidified water (0.1% acetic acid) and methanol (0.1% acetic acid) at a flow rate of 5 mL min−1. Retention time and relative area standard deviations were below 1% for all isoflavones. The method developed was successfully applied to several soy food samples and spiked samples. Total isoflavone concentration in sampled soy foods ranged from 34.28 mg L−1 to 4.29 mg g−1.  相似文献

Monolithic capillary columns were prepared via electron beam triggered free radical polymerization within the confines of 0.2 and 0.1mm I.D. capillary columns using ethyl methacrylate and trimethylolpropane triacrylate as monomers as well as 2-propanol, 1-dodecanol and toluene as porogenic system. The influence of column diameter on reproducibility and separation performance was investigated. For evaluation, a protein standard consisting of five proteins in the range of 5800-66,000 g mol(-1) was used. Reproducibility was checked by determining the relative standard deviations in retention times, peak widths at half height, asymmetry and resolution. Excellent run-to-run reproducibility was found for both 0.2 and 0.1mm I.D. columns; batch-to-batch reproducibility was good for both column types. In order to enhance the non-polar character of the monolithic columns, lauryl methacrylate-based capillary columns were prepared. These were successfully used for the separation of proteins and a cytochrome c digest.  相似文献

建立了快速、灵敏、准确的超高效液相色谱方法,用来分析4种商品人参(人参、红参、人参叶、人参须)中12种人参皂苷的含量,并用化学计量学方法评价了商品人参的质量。采用ACQUITY UPLCTM BEH C18色谱柱(50 mm×2.1 mm, 1.7 μm),以乙腈-水为流动相进行梯度洗脱。对所建立的测定12种人参皂苷的UPLC方法进行了线性方程、准确度、重复性、回收率等方法学考察。采用聚类分析和主成分分析的化学计量学方法对4种商品人参进行分析,评价了其质量。结果表明聚类分析和主成分分析2种化学计量学方法非常适合大样本、多成分的中药材质量分析。  相似文献

Porous ceramic bed supports for fused silica packed capillary columns utilized in liquid chromatography were prepared by polymerizing solutions containing potassium silicate in-situ within a column to create a mechanically stable, rugged, and easily constructed termination. The effect of the bed support length on efficiency, and comparisons to glass wool bed supports, were considered in terms of column efficiencies and hydrodynamic variables. Results obtained indicate better performance for the ceramic bed support.  相似文献

Summary Two methods to realize a porous retentive silica layer on the inner wall of 10–25 μm fused silica capillaries for OTLC, etching and precipitation of silica from solution, have been investigated. Etching of the fused silica capillaries with 1M KOH, creates an activated surface, but the capacity of the silica layer is too small to serve as retentive layer in OTLC. Better prospects are offered by the precipitation of silica from a solution of polyethoxysiloxane, dynamically coated on the inner wall of the fused silica capillary. It appears to be possible to deposite a porous silica layer up to 0.8 μm thick (in a 25 μm capillary) by this method, which seems to be suitable for liquid-solid an dynamically generated liquid-liquid chromatography in open tubular columns. The performance of these columns are demonstrated by means of efficient separations of test mixtures using on-column fluorescence detection.  相似文献

The efficiency of miniaturized liquid chromatography columns with inner diameters between 200 and 300 μm has been investigated using a dedicated micro‐liquid chromatography system. Fully porous, core–shell and monolithic commercially available stationary phases were compared applying van Deemter and kinetic plot analysis. The sub‐2 μm fully porous as well as the 2.7 μm core–shell particle packed columns showed superior efficiency and similar values for the minimum reduced plate heights (2.56–2.69) before correction for extra‐column contribution compared to normal‐bore columns. Moreover, the influence of extra‐column contribution was investigated to demonstrate the difference between apparent and intrinsic efficiency by replacing the column by a zero dead volume union to determine the band spreading caused by the system. It was demonstrated that 72% of the intrinsic efficiency could be reached. The results of the kinetic plot analysis indicate the superior performance of the sub‐2 μm fully porous particle packed column for ultra‐fast liquid chromatography.  相似文献

建立了固相萃取-超高效液相色谱测定人参中5种原人参二醇型人参皂苷的方法。人参药材经粉碎后通过水饱和正丁醇溶液进行超声提取,经过亲水作用固相萃取柱净化后,在ACQUITY UPLC BEH Shield RP18色谱柱(100 mm×2.1 mm, 1.7μm)上分离,利用乙腈/水作为流动相进行梯度洗脱,采用光电二极管阵列检测器检测。结果表明,5种原人参二醇型人参皂苷在5~500μg/mL范围内具有很好的线性关系,相关系数均大于0.999。方法精密度的RSD值在0.95%~2.62%(n=6)之间,22 h内样品稳定性的RSD值在0.90%~2.15%(n=8)之间,日内和日间重复性的RSD值分别为5.35%~6.47%(n=6)和5.56%~6.34%(n=8)。方法的加标回收率在87.16%~101.92%之间,相对标准偏差在1.54%~4.01%(n=6)之间。所建立的方法采用亲水作用色谱模式的固相萃取材料,药材的提取液可直接作为固相萃取的上样溶液进行人参皂苷的富集和净化,并且超高效液相色谱大大缩短了分析时间。该方法简单快速、通量高、重现性好,适用于人参中5种原人参二醇型人参皂苷的...  相似文献

建立了人参和黄芪中毒杀芬代表性单体残留量的检测方法。选择7种指示性毒杀芬单体为检测品种,样品用环己烷-丙酮(9:1,v/v)加速溶剂萃取法提取,经弗罗里硅土固相萃取柱净化后,以气相色谱-电子轰击离子源串联质谱(GC-EI-MS/MS)在多反应监测(MRM)模式下进行测定。在人参和黄芪两种中药材样品基质,0.005、0.01和0.1 mg/kg 3个加标水平条件下,7种毒杀芬单体的回收率为72.4%~105%,相对标准偏差为0.96%~10.4%;方法的检出限(S/N=3)为0.2~1.7 μg/kg,均符合农药残留检测要求。该方法提取效果好,检测灵敏度高,适用于人参和黄芪中7种毒杀芬残留量的检测。  相似文献

Stationary phase selectivities for halogenated compounds in reversed-phase HPLC were compared using C18 monolithic silica capillary columns modified with poly(octadecyl methacrylate) (ODM) and octadecyl moieties (ODS). The preferential retention of halogenated benzenes on ODM was observed in methanol/water and acetonitrile/water mobile phases. In selectivity comparison of selected analytes on ODM and ODS, greater selectivities for halogenated compounds were obtained with respect to alkylbenzenes on an ODM column, while similar selectivities were observed with a homologous series of alkylbenzenes on ODM and ODS columns. These data can be explained by greater dispersive interactions by more densely packed octadecyl groups on the ODM polymer coated column together with the contribution of carbonyl groups in ODM side chains. For the positional isomeric separation of dihalogenated benzenes (ortho-, meta-, para-), the ODM column also provided better separation of these isomers for the adjacently eluted isomers that cannot be completely separated on an ODS column in the same mobile phase. These results imply that the ODM column can be used as a better alternative to the ODS column for the separation of other halogenated compounds.  相似文献

A systematic optimization of the HPLC separation of a mixture containing 11 pollutant phenols (PPs) using a Hypersil ODS (250 mm x 4.6 mm, 5 microm) column and UV-DAD detection has been carried out. The binary mobile phases used were obtained by mixing 50 mM phosphate (pH = 3.0) and methanol, ACN, or THF as organic modifiers. After selecting ACN as an organic modifier, the effects of pH and temperature on PPs separation were studied. A mobile phase of 50 mM acetate (pH = 5.0)-ACN (60:40 v/v) at 50 degrees C allowed the separation of 11 phenols but not to baseline in 17 min. To improve the performance of this separation, the following RP columns were tested: Luna C18 (2), Purospher C18, Synergi C12, Synergi Fusion C18, Gemini C18, Luna Cyano, Lichrospher C8, and Envirosep-PP (polymeric). In all the cases, the performance (analysis time, retention, selectivity, resolution, asymmetry factors, and efficiency) was evaluated. A further reoptimization of the mobile phase was carried out for all the columns by studying the ACN content and pH, with the aim of improving the above-mentioned separations and selecting the most suitable one for PPs analysis.  相似文献

As a traditional medicinal plant, Juglans mandshurica has been used for the treatment of cancer. Different organs of this plant showed anti‐tumor activity in clinic and laboratory. Comparative identification of constituents in different plant organs is essential for investigation of the relationship between chemical constituents and pharmacological activities. For this aim, the roots, branches, and leaves of J. mandshurica were extracted with 50% v/v methanol and then subjected to ultra‐high performance liquid chromatography with quadrupole time‐of‐flight mass spectrometry analysis conducted under low and high energy. As a result, we have to date identified 111 compounds consisting of 56 tannins, 29 flavonoids, 13 organic acids, 8 naphthalene derivatives, and 5 anthracenes. Five compounds, namely, diquercetin trihydroxy‐truxinoyl‐glucoside, two quercetin kaempferol dihydroxy‐truxinoyl‐glucosides, syringoyl‐tri‐galloyl‐O‐glucose, and dihydroxy‐naphthalene syringoyl‐glucoside, were tentatively identified as new compounds. Of the compounds identified, 76 were found in the root extract, 67 in the branch extract, and 37 in the leaf extract. Only six compounds including four organic acids and two tannins were found in all three extracts. We developed a rapid and sensitive ultra high performance liquid chromatography with quadrupole time‐of‐flight mass spectrometry approach to identify multiple constituents of complex extracts without separation and ion selection. The results presented provide useful information on further research of the bioactive compounds of J. mandshurica .  相似文献

A novel monolithic capillary column (530 microm i.d.) was prepared for capillary liquid chromatography (CLC) by in situ copolymerization of octyl methacrylate (MAOE) and ethylene dimethacrylate (EDMA) in the presence of a porogen solvent containing 1-propanol, 1,4-butanediol, and water with azobisisobutyronitrile as the initiator. The influences of the contents of the porogen solvent, EDMA and the various concentration ratios of 1-propanol to 1,4-butanediol in the polymerization mixture on the morphology, porosity, globule size, stability and column efficiency were investigated. The morphology and pore size distribution of monolithic capillary columns were characterized by SEM and mercury intrusion porosimetry, respectively. Chromatographic evaluations of the columns were performed under CLC mode. The results showed that good permeability and stability can be obtained under optimal experimental conditions. The separation results of some acid, neutral and basic analytes demonstrated the hydrophobicity and low affinity to basic analytes of the new column. Three metal ions, i.e. Mg(II), Zn(II) and Cd(II) were also separated under ion-pair mode on the new monolithic capillary column and the results were acceptable.  相似文献

The tuning of selectivity by changing the flow rate has been investigated in HPLC: two columns with different retention characteristics were coupled in series via a T-piece and the relative retention of components chromatographed on the system were changed by varying the individual flow rates in the coupled columns. The flow rate alteration was performed by adding a second flow after the first column. The flow rate ratio necessary for optimum resolution can be easily calculated on the basis of the capacity factors measured on the individual columns. The performance of this method for adjusting selectivity has been demonstrated by using different column combinations to separate several mixtures containing chlorophenols, nitroaromatic compounds, and aromatic hydrocarbons.  相似文献
