In this study photoinduced cation generation, based on the photochemical properties of malachite green (MG), was used for the surface design and in vitro photochemical control of cell adhesion and proliferation. The MG-derivatized surface was prepared by coating a photoreactive polymer as a substrate onto a poly(ethylene terephthalate) (PET) sheet. The photoreactive polymer was radical copolymer of styrene with the MG-derivatized monomer diphenyl(4-vinylphenyl)methane leucohydroxide (degree of substitution of MG unit: 12.4 mol%). Water contact angle measurements and X-ray photoelectron spectroscopy revealed high hydrophobicity and homogeneous distribution of the MG groups on the outermost surface of the coated film, respectively. When the coated film was exposed to ultraviolet light (UV) irradiation at wavelengths of 290-410 nm, a time-dependent color change of the film was observed from pale yellow, before irradiation, to green. These results indicated generation of cations on the film surface by photochemical cation generation of the MG groups, which was quantitatively characterized by force versus distance curves measurements in atomic force microscopic (AFM) observation using a carboxylated AFM tip. The seeding and culture of endothelial cells showed a marked reduction in adhesion on the nonirradiated coated film surface, whereas the UV-irradiated surface promoted cell adhesion and proliferation except for incubation in serum-free medium, which was similar to commercial tissue culture PET sheet. These observations may be due to adsorption of cell adhesive proteins, typified by fibronectin, in serum-containing medium onto the cationized photoreactive copolymer surface by electrostatic interactions.  相似文献

Complexes between sodium (sulfamate‐carboxylate)isoprene/ethylene oxide double hydrophilic block copolymers and lysozyme, a globular protein, were formed in aqueous solutions, at pH 7, because of electrostatic interactions between the anionic groups of the polyelectrolyte block of the copolymers and the cationic groups of lysozyme. The structure of the complexes was investigated as a function of the anionic/cationic charge ratio of the two components in solution and ionic strength by static, dynamic, and electrophoretic light scattering, atomic force microscopy, and fluorescence spectroscopy. The mass and size of the micellar‐like complexes depend on the mixing ratio and the molecular characteristics (molecular weight, composition, and architecture) of the copolymer used. Complexation persists at 0.15M NaCl, the value for physiological saline, as a result of additional hydrophobic interactions between the copolymers and the enzyme. Fluorescence spectroscopy measurements indicate that the secondary structure of lysozyme does not change substantially after complex formation. © 2006 Wiley Periodicals, Inc. Water‐soluble crosslinked hollow nanoparticles were prepared using pH‐responsive anionic polymer micelles as templates. The template micelles were formed from pH‐responsive diblock copolymers (PAMPS‐PAaH) composed of the poly(sodium 2‐(acrylamido)‐2‐methylpropanesulfonate) and poly(6‐(acrylamido)hexanoic acid) blocks in an aqueous acidic solution. The PAMPS and PAaH blocks form a hydrophilic anionic shell and hydrophobic core of the core‐shell polymer micelle, respectively. A cationic diblock copolymer (PEG‐P(APTAC/CEA)) with the poly(ethylene glycol) block and random copolymer block composed of poly((3‐acrylamidopropyl)trimethylammonium chloride) containing a small amount of the 2‐(cinnamoyl)ethylacrylate photo‐crosslinkable unit can be adsorbed to the anionic shell of the template micelle due to electrostatic interaction, which form a core‐shell‐corona three‐layered micelle. The shell of the core‐shell‐corona micelle is formed from a polyion complex with anionic PAMPS and cationic P(APTAC/CEA) chains. The P(APTAC/CEA) chains in the shell of the core‐shell‐corona micelle can be photo‐crosslinked with UV irradiation. The template micelle can be dissociated using NaOH, because the PAaH blocks are ionized. Furthermore, electrostatic interactions between PAMPS and PAPTAC in the shell are screened by adding excess NaCl in water. The template micelles can be completely removed by dialysis against water containing NaOH and NaCl to prepare the crosslinked hollow nanoparticles. We have prepared a UV-responsive polymeric superamphiphile, formed by a malachite green derivative and the double hydrophilic block copolymer methoxy-poly(ethylene glycol)(114)-block-poly(l-lysine hydrochloride)(200) (PEG-b-PLKC) on the basis of electrostatic interactions. The malachite green derivative undergoes photo-ionization upon UV irradiation, which makes it more hydrophilic, resulting in changes in the self-assembly behavior of the polymeric superamphiphile. For this reason, the polymeric superamphiphile originally self-assembles to form sheetlike aggregates, which disassemble after UV irradiation because of the increased solubility of the malachite green derivative. By use of Nile red as a probe, the polarity of the polymeric superamphiphile solution is confirmed to be increased after UV irradiation by fluorescence spectra, which also explains the disassembly of the polymeric superamphiphile.  相似文献

A series of new water‐soluble cationic pyrene‐dendron derivatives, G1 , G2 , and G3, was successfully synthesized and characterized. These new dendrons were designed with the quaternized amino moieties at the periphery of the dendrons for DNA detection and functionalized with pyrene as a fluorescent probe. The electrostatic interactions between the plasmid DNA (pDNA) and cationic charged dendrons in an aqueous solution resulted in a change in the photophysical properties of pyrene, which could be shown in the UV‐vis and fluorescence spectra. Pyrene dendrons showed a high and rapid fluorescence response upon the addition of pDNA, which was strongly dependent on the size and hydrophobicity of the dendrons. © 2011 Wiley Periodicals, Inc. Here we report fluorescence turn‐on synthetic lipid rafts by self‐assembly of a cationic distyrylanthracene derivative on a negatively‐charged sheet in an aqueous solution. First, the negatively‐charged 2D membrane structure is formed by lateral associations of aromatic rods with carboxylate groups. Then, the synthetic rafts are floated on the surface of the negatively‐charged sheets through electrostatic interactions. The fluorescence of the synthetic rafts is turned on due to the aggregation of the positively‐charged AIE dye on the sheets, facilitating monitoring of the formation of rafts. Concanavalin A (Con A) protein can load hierarchically onto the synthetic rafts at neutral pH to provide discrete Con A aggregates with a uniform size of ≈12 nm. The uniform aggregates of Con A on the synthetic rafts can stimulate Jurkat cells with enhanced efficiency, as compared with random‐sized aggregates of Con A.  相似文献

Living cationic polymerization of 4‐methyl‐7‐(2‐vinyloxyethoxy)coumarin (CMVE) was achieved using SnCl4 in the presence of nBu4NBr as an added salt at 0 °C. The number‐average molecular weight of the resulting polymers increased in direct proportion to the monomer conversion while retaining relatively low polydispersity. Structural analysis revealed that the resulting polymers carried pendant coumarinyl moieties. These coumarinyl moieties were crosslinked by irradiation with UV light at λmax = 366 nm, and the crosslinked sites were then cleaved by irradiation with UV light at λmax = 254 nm. The crosslinking behaviors of the polymers were studied by UV and FTIR spectroscopic measurement. PolyCMVE was soluble in dichloromethane but was found to be insoluble upon UV light irradiation. We also synthesized amphiphilic block polymers bearing coumarinyl moieties by living cationic copolymerization with an amphiphilic vinyl ether. The resulting block polymers were soluble in an aqueous medium and also formed micelle‐like aggregates. Upon UV irradiation of aqueous solutions above the critical micelle concentration, an efficient crosslinking reaction occurred. Arylenevinylene‐based π‐conjugated polymers containing imidazolium cationic units in the main chain and their model compounds were synthesized and characterized in terms of optical and electrochemical properties. 9,9‐Bisoctylfluorene, 2,5‐bisdodecyloxybenzene, and 3‐dodecylthiophene were introduced as arylene units with different donor characteristics to evaluate the effect on the highest occupied molecular orbital‐lowest unoccupied molecular orbital (HOMO‐LUMO) gap energy. The UV–vis and fluorescence spectra of cationic polymers and model compounds with iodide counter anion exhibited a significant blue shift with respect to the parent neutral molecules. X‐ray single crystal analysis for model compounds revealed that the effective π‐conjugation length of cationic model compounds decreased compared to the neutral model compounds by means of twisted conformation directed by CH‐π interactions between N‐methyl groups of imidazolium and neighboring aryl units. The cyclic voltammetry measurement suggested the negative shift of LUMO levels by the conversion of imidazole to imidazolium, indicating the electron‐accepting characteristics of cationic imidazolium unit. © 2011 Wiley Periodicals, Inc. The well‐defined azoindazole‐containing homopolymer, poly(6‐{6‐[(4‐dimethylamino) phenylazo]‐indazole}‐hexyl methacrylate) (PDHMA), and amphiphilic diblock copolymer, poly({6‐[6‐(4‐dimethylamino)phenylazo]‐indazole}‐hexyl methacrylate)‐b‐poly(2‐(dimethylamino)ethylmethacrylate) (PDHMAmb‐PDMAEMAn), were successfully prepared via reversible addition‐fragmentation chain transfer polymerization technique. The homopolymer and amphiphilic diblock copolymer in CH2Cl2 exhibited intense fluorescence emission accompanied by trans–cis photoisomerization of azoindazole group under UV irradiation. The experiment results indicated that the intense fluorescence emission may be attributed to an inhibition of photoinduced electron transfer of the cis form of azoindazole. On the other hand, the intense fluorescence emission of amphiphilic diblock copolymers in water‐tetrahydrofuran mixture was observed, which increased with the volume ratio of water in the mixed solvent. The self‐aggregation behaviors of three amphiphilic diblock copolymers were examined by transmission electron microscopy, laser light scattering, and UV–vis spectra. The restriction of intramolecular rotation of the azoindazole groups in aggregates was considered as the main cause of aggregation‐induced fluorescence emission. © 2011 Wiley Periodicals, Inc. A dicarboxylate host (1) binds cationic monosaccharides such as D-glucosamine HCl (2), D-galactosamine-HCl (3), and D-mannosamine-HCl (4) with high affinity (K1 = 8.0 x 10(4)-2.0 x 10(5) M(-1)) in methanol. In circular dichroism (CD) spectroscopy a positive exciton-coupling band was observed near 290 nm; this indicates that the saccharides are recognized by multiple point interactions. Since the corresponding neutral monosaccharides are not significantly bound, one may conclude that complex formation is primarily due to the electrostatic interaction between NH3+ in the guest and one carboxylate in the host and secondarily due to hydrogen-bonding interactions of OH groups with the other carboxylate and/or nitrogen bases. Molar ratio plots and Job plots indicate that host 1 and cationic monosaccharide guests form CD-active, pseudo-cyclic 1:1 complexes at low guest concentration followed by the formation of CD-silent, acyclic 1:2 1-saccharide complexes at high guest concentration. The possible binding modes are discussed in detail on the basis of molecular mechanics calculations and chemical shift changes in 1H NMR spectra. The results of competition experiments with several cationic reference compounds bearing fewer OH groups than 2-4 are consistent with the proposed binding model. Thus, the present study is a rare example of saccharide recognition in a protic solvent, where in general, hydrogen-bonding interactions are rarely useful because of strong solvation energy. These are apparently the strongest saccharide complexes involving noncovalent interactions between host and guest. We believe that the findings are significant as a milestone toward development of new saccharide recognition systems ultimately useful in aqueous solution.  相似文献

Nonlinear optical probe, second harmonic generation (SHG), of the adsorption of the dye molecule malachite green (MG), in cationic form at pH < or = 5, on polystyrene microspheres in aqueous solution is used to study the effect of surface charge and composition on molecular adsorption. Three types of polystyrene microspheres with different surface composition are investigated: (1) a sulfate terminated, anionic surface, (2) a neutral surface without any functional group termination, and (3) an amine terminated, cationic surface. The cationic dye was found to adsorb at all three surfaces, regardless of surface charge. The adsorption free energies, DeltaG's, measured for the three surfaces are -12.67, -12.39, and -10.46 kcal/mol, respectively, with the trend as expected from the charge interactions. The adsorption density on the anionic surface, where attractive charge-charge interaction dominates, is determined by the surface negative charge density. The adsorption densities on the neutral and cationic surfaces are on the other hand higher, perhaps as a result of a balance between minimizing repulsive charge interaction and maximizing attractive molecule-substrate and intermolecular interactions. The relative strength of the SH intensity per molecule, in combination of a model calculation, reveals that the C(2) axis of the MG molecule is nearly perpendicular to the surface on the anionic surface and tilts away from the surface norm when the surface is neutral and further away when cationic. Changing the pH of the solution may alter the surface charge and subsequently affect the adsorption configuration and SH intensity.  相似文献

The interaction has been studied in aqueous solutions between a negatively charged conjugated polyelectrolyte poly{1,4-phenylene-[9,9-bis(4-phenoxybutylsulfonate)]fluorene-2,7-diyl} copolymer (PBS-PFP) and several cationic tetraalkylammonium surfactants with different structures (alkyl chain length, counterion, or double alkyl chain), with tetramethylammonium cations and with the anionic surfactant sodium dodecyl sulfate (SDS) by electronic absorption and emission spectroscopy and by conductivity measurements. The results are compared with those previously obtained on the interaction of the same polymer with the nonionic surfactant C12E5. The nature of the electrostatic or hydrophobic polymer-surfactant interactions leads to very different behavior. The polymer induces the aggregation with the cationic surfactants at concentrations well below the critical micelle concentration, while this is inhibited with the anionic SDS, as demonstrated from conductivity measurements. The interaction with cationic surfactants only shows a small dependence on alkyl chain length or counterion and is suggested to be dominated by electrostatic interactions. In contrast to previous studies with the nonionic C12E5, both the cationic and the anionic surfactants quench the PBS-PFP emission intensity, leading also to a decrease in the polymer emission lifetime. However, the interaction with these cationic surfactants leads to the appearance of a new emission band (approximately 525 nm), which may be due to energy hopping to defect sites due to the increase of PBS-PFP interchain interaction favored by charge neutralization of the anionic polymer by cationic surfactant and by hydrophobic interactions involving the surfactant alkyl chains, since the same green band is not observed by adding either tetramethylammonium hydroxide or chloride. This effect suggests that the cationic surfactants are changing the nature of PBS-PFP aggregates. The nature of the polymer and surfactant interactions can, thus, be used to control the spectroscopic and conductivity properties of the polymer, which may have implications in its applications.  相似文献

In this work, cationic and anionic microgels which are mainly formed from thermal responsive polymer, poly(N‐isopropylacrylamide), are prepared and mixed in water. These microgels interact with each other due to the electrostatic interaction, and aggregate voluntarily. By applying the microgel aggregating system, photo‐responsive aggregating system is constructed by using o‐nitrobenzaldehyde (NBA), which reacts and releases hydrogen triggered by photo stimuli. The microgel aggregates in an aqueous solution of NBA re‐disperse depending on the irradiation time of UV light. In addition, by masking the UV irradiated area, the resultant shapes of microgel aggregates are controlled. An amphiphilic diblock copolymer composed of a photoresponsive dialkoxycyanostilbene polymethacrylate and poly(ethylene oxide) (PDACS‐b‐PEO) was synthesized and its photophysical and aggregation properties were investigated. The amphiphilic nature of the polymer caused it to self‐assemble in water, and dynamic light scattering studies indicated formation of spherical aggregates with an average size of 160 nm. Atomic force microscopy images of dried films cast from solutions containing the polymer aggregates revealed supramolecular aggregates with a spherical morphology. Photoisomerization of the stilbene chromophore in PDACS‐b‐PEO on UV irradiation resulted in the destruction of the self‐assembled superstructures which could be attributed both to change in shape of the chromophore from the linear trans isomer to the bent cis isomer which would hinder self‐aggregation of the molecules and the higher dipole moment of the cis isomer leading to a reduction of the hydrophobic nature of the stilbene containing block of PDACS‐b‐PEO. It was observed that hydrophobic dyes such as curcumin could be encapsulated within the hydrophobic interior of the spherical micellar aggregates from which the encapsulated dye could be released on UV irradiation. © 2011 Wiley Periodicals, Inc. The underlying mechanism of UV light-induced dissociation and visible light-induced reformation of vesicles formed by an azobenzene diblock copolymer was investigated. These processes were studied in situ by monitoring changes in optical transmittance of the vesicular solution while being exposed to UV or visible light irradiation. The results indicate that the UV-induced dissociation of the vesicles results from their thermodynamic instability due to a shift of the hydrophilic/hydrophobic balance arising from the trans-cis isomerization, while their reaggregation takes place upon visible light irradiation that shifts the hydrophilic/hydrophobic balance in the opposite direction after the reverse cis-trans isomerization. The study suggests a specific design principle for obtaining UV light-dissociable and visible light-recoverable vesicles based on azobenzene block copolymers. On one hand, the structure of azobenzene moiety used in the hydrophobic block should have a small (near zero) dipole moment in the trans form and a significantly higher dipole moment in the cis form, which ensures a significant increase in polarity of the hydrophobic block under UV light irradiation. On the other hand, the hydrophilic block should be weakly hydrophilic. The conjunction of the two conditions can make the light-induced shift of the hydrophilic/hydrophobic balance important enough to lead to the reversible change in vesicular aggregation.  相似文献

Copolymers containing oxime‐urethane groups were prepared by the copolymerization of methyl methacrylate and benzophenoneoximinocarbonylaminoethyl methacrylate (BCM), and their photochemical properties were examined from the UV and IR absorption spectral changes. The decomposed fraction of oxime‐urethane groups in the copolymer increased with irradiation time, but it decreased with the content of BCM units in the copolymer. Changes of the surface properties of the copolymer film on irradiation were studied by measurements of the contact angle and dyeing with an acid dye. The surface of the copolymer film changed to become more hydrophilic upon irradiation with 254 nm of UV light. After the irradiated copolymer films were treated with HCl or methanol, changes of the contact angle of water on irradiation were compared. The copolymer film was dyed by acid dyes after treatment of the irradiated film with HCl. The degree of dyeing increased with irradiation time and BCM units in the copolymer, but it was unaffected by the film thickness. Magnetite (Fe3O4) nanoparticles have been synthesized and complexed with carboxylate-functional block copolymers, and then aqueous dispersions of the complexes were investigated as functions of their chemical and morphological structures. The block copolymer dispersants had either poly(ethylene oxide), poly(ethylene oxide-co-propylene oxide), or poly(ethylene oxide-b-propylene oxide) outer blocks, and all of them had a polyurethane center block that contained pendent carboxylate groups. The complexes were formed through interactions of the carboxylates with the surfaces of the magnetite nanoparticles. The magnetite cores of the magnetite-copolymer complexes were near 10 nm in diameter, and the particles were superparamagnetic. Complexes with mass ratios of polymer to magnetite varying from 50:50 to 85:15 were studied. One of our objectives is to design complexes that form stable dispersions of discrete particles in water, yet that can be actuated (moved together) upon exposure to a uniform magnetic field. DLVO calculations that accounted for magnetic attractive interparticle forces, as well as van der Waals, steric, and electrostatic forces are presented. Compositions were identified wherein a shallow, attractive interparticle potential minimum appears once the magnetic term is applied. This suggests that it may be possible to tune the structures of superparamagnetic nanoparticle shells to allow discrete dispersions without a field, yet weak flocculation could be induced upon exposure to a field.  相似文献

The anionic porphyrin, meso-tetrakis(4-sulfonatophenyl)porphine, is found to tightly bind to an engineered 14-residue peptide, resulting in induced alpha-helix formation when mixed in aqueous solutions. The small porphyrin-peptide dissociation constant (2 muM) observed is related to the energetics of peptide helix formation coupled with electrostatic interactions between the anionic porphyrin and cationic residues in the coiled peptide. Analytical ultracentrifugation measurements indicate the porphyrin-peptide complexes dimerize, probably into a coiled coil, and weakly associate to form even higher order structures.  相似文献

Molecular clip 1 remains monomeric in water and engages in host–guest recognition processes with suitable guests. We report the Ka values for 32 1? guest complexes measured by 1H NMR, UV/Vis, and fluorescence titrations. The cavity of 1 is shaped by aromatic surfaces of negative electrostatic potential and therefore displays high affinity and selectivity for planar and cationic aromatic guests that distinguishes it from CB[n] receptors that prefer aliphatic over aromatic guests. Electrostatic effects play a dominant role in the recognition process whereby ion–dipole interactions may occur between ammonium ions and the C=O groups of 1 , between the SO3? groups of 1 and pendant cationic groups on the guest, and within the cavity of 1 by cation–π interactions. Host 1 displays a high affinity toward dicationic guests with large planar aromatic surfaces (e.g. naphthalene diimide NDI+ and perylene diimide PDI+) and cationic dyes derived from acridine (e.g. methylene blue and azure A). The critical importance of cation–π interactions was ascertained by a comparison of analogous neutral and cationic guests (e.g. methylene violet vs. methylene blue; quinoline vs. N‐methylquinolinium; acridine vs. N‐methylacridinium; neutral red vs. neutral red H+) the affinities of which differ by up to 380‐fold. We demonstrate that the high affinity of 1 toward methylene blue (Ka=3.92×107 m ?1; Kd=25 nm ) allows for the selective sequestration and destaining of U87 cells stained with methylene blue.  相似文献
