Biocompatible and biodegradable ABC and ABCBA triblock and pentablock copolymers composed of poly(ε‐caprolactone) (PCL), poly(L ‐lactide) (PLA), and poly(ethylene glycol) (PEO) with controlled molecular weights and low polydispersities were synthesized by a click conjugation between alkyne‐terminated PCL‐b‐PLA and azide‐terminated PEO. Their molecular structures, physicochemical and self‐assembly properties were thoroughly characterized by means of FT‐IR, 1H‐NMR, gel permeation chromatography, differential scanning calorimetry, wide‐angle X‐ray diffraction, dynamic light scattering, and transmission electron microscopy. These copolymers formed microphase‐separated crystalline materials in solid state, where the crystallization of PCL block was greatly restricted by both PEO and PLA blocks. These copolymers self‐assembled into starlike and flowerlike micelles with a spherical morphology, and the micelles were stable over 27 days in aqueous solution at 37 °C. The doxorubicin (DOX) drug‐loaded nanoparticles showed a bigger size with a similar spherical morphology compared to blank nanoparticles, demonstrating a biphasic drug‐release profile in buffer solution and at 37 °C. Moreover, the DOX‐loaded nanoparticles fabricated from the pentablock copolymer sustained a longer drug‐release period (25 days) at pH 7.4 than those of the triblock copolymer. The blank nanoparticles showed good cell viability, whereas the DOX‐loaded nanoparticles killed fewer cells than free DOX, suggesting a controlled drug‐release effect. © 2010 Wiley Periodicals, Inc. A series of multibranched pentablock copolymer (mBr5BlC), poly(L ‐lactide)‐b‐HBP‐b‐poly(ethylene glycol)‐b‐HBP‐b‐poly(L ‐lactide) (HBP = hyperbranched polyglycidol), has been synthesized by ring‐opening multibranching polymerization of glycidol using bifunctional poly(ethylene glycol) [PEG; molecular weight (MW) = 1000] as an initiator, followed by ring‐opening polymerization (ROP) of L ‐lactide in the presence of stannous octoate. The ROP of different amounts of L ‐lactide on HBP‐b‐PEG‐b‐HBP [MW = 2550; polydispersity index (PDI) = 1.08] yielded a series of the targeted mBr5BlCs of the MW range of 4360–15,300 with narrow PDI. All the mBr5BlCs have been well demonstrated to be in possession of good biocompatibility as biomaterials for various applications in biological medicine areas. The mBr5BlCs with tunable MW exhibited promising controllability in self‐assembly into spherical micellar structures with an average diameter range of 59–140 nm, which have no acute and intrinsic cytotoxicity against normal cells and provide a convenient strategy for drug loading. The anticancer drug doxorubicin was demonstrated to have a good affinity with the copolymer system, and its controlled release was performed in various pHs. © 2012 Wiley Periodicals, Inc. The crystal unit‐cell structures and the isothermal crystallization kinetics of poly(L ‐lactide) in biodegradable poly(L ‐lactide)‐block‐methoxy poly(ethylene glycol) (PLLA‐b‐MePEG) diblock copolymers have been analyzed by wide‐angle X‐ray diffraction and differential scanning calorimetry. In particular, the effects due to the presence of MePEG that is chemically connected to PLLA as well as the PLLA crystallization temperature TC are examined. Though we observe no variation of both the PLLA and MePEG crystal unit‐cell structures with the block ratio between PLLA and MePEG and TC, the isothermal crystallization kinetics of PLLA is greatly influenced by the presence of MePEG that is connected to it. In particular, the equilibrium melting temperature of PLLA, T, significantly decreases in the diblock copolymers. When the TC is high so that the crystallization is controlled by nucleation, because of the decreasing T and thereafter the nucleation density with decreasing PLLA molecular weight, the crystallinity of PLLA also decreases with a decrease in the PLLA molecular weight. While, for the lower crystallization temperature regime controlled by the growth mechanism, the crystallizability of PLLA in copolymers is greater than that of pure PLLA. This suggests that the activation energy for the PLLA segment diffusing to the crystallization site decreases in the diblocks. © 2006 Wiley Periodicals, Inc. A model polyethylene‐poly(L ‐lactide) diblock copolymer (PE‐b‐PLLA) was synthesized using hydroxyl‐terminated PE (PE‐OH) as a macroinitiator for the ring‐opening polymerization of L ‐lactide. Binary blends, which contained poly(L ‐lactide) (PLLA) and very low‐density polyethylene (LDPE), and ternary blends, which contained PLLA, LDPE, and PE‐b‐PLLA, were prepared by solution blending followed by precipitation and compression molding. Particle size analysis and scanning electron microscopy results showed that the particle size and distribution of the LDPE dispersed in the PLLA matrix was sharply decreased upon the addition of PE‐b‐PLLA. The tensile and Izod impact testing results on the ternary blends showed significantly improved toughness as compared to the PLLA homopolymer or the corresponding PLLA/LDPE binary blends. © 2001 John Wiley & Sons, Inc. Because poly(L ‐lactic acid) (PLLA) is a biodegradable polyester with low immunogenicity and good biocompatibility, it is used as a biomaterial. However, hydrophobic PLLA does not have any reactive groups. Thus, its application is limited. To increase the hydrophilicity of PLLA and accelerate its degradation rate, functionalized pendant groups and blocks were introduced through copolymerization with citric acid and poly(ethylene glycol) (PEG), respectively. This article describes the synthesis and characterization of poly(L ‐lactic‐co‐citric acid) (PLCA)‐PLLA and PLCA‐PEG multiblock copolymers. The results indicated that the hydrolysis rate was enhanced, and the hydrophilicity was improved because of the incorporation of carboxyl groups in PLCA‐PLLA. The joining of the PEG block led to improved hydrophilicity of PLCA, and the degradation rate of PLCA‐PEG accelerated as compared with that of PLCA‐PLLA. © 2003 Wiley Periodicals, Inc. Amphiphilic triblock copolymers, poly(ethyl cyanoacrylate)‐b‐poly(ethylene glycol)‐b‐poly(ethyl cyanoacrylate) (PECA‐b‐PEG‐b‐PECA), were synthesized via oxyanion‐initiated polymerization with sodium alcoholate‐terminated PEG as macroinitiator. PECA‐b‐PEG‐b‐PECA were characterized by gel permeation chromatography system, 1H NMR and FTIR. The results indicate that the copolymerization is well controlled with narrow molecular weight distribution. The dexamethasone (DXM)‐loaded PECA‐b‐PEG‐b‐PECA nanoparticles (NPs) were prepared by nanoprecipitation technique and then characterized by Laser Particle Size Analyzer, 1H NMR and transmission electron microscopy. The drug‐loaded PECA‐b‐PEG‐b‐PECA NPs are of spherical shape with average size of less than 100 nm. The drug‐loaded amount (DLA) and encapsulation efficiency of DLNPs were investigated by HPLC. The results show that DXM can be effectively incorporated into PECA‐b‐PEG‐b‐PECA NPs, which provides an optional delivery system for DXM and other hydrophobic drugs. © 2008 Wiley Periodicals, Inc. Random and block copolymerizations of L ‐ or D ‐lactide with ε‐caprolactone (CL) were performed with a novel anionic initiator, (C5Me5)2SmMe(THF), and they resulted in partial epimerization, generating D ,L ‐ or meso‐lactide polymers with enhanced biodegradability. A blend of PLLA‐r‐PCL [82/18; PLLA = poly(L ‐LA) and PCL = poly(ε‐caprolactone)] and PDLA‐r‐PCL [79/21; PDLA = poly(D ‐LA)] prepared by the solution‐casting method generated a stereocomplex, the melting temperature of which was about 40 °C higher than that of the nonblended copolymers. A blend of PLLA‐b‐PCL (85/15) and PDLA‐b‐PCL (82/18) showed a lower elongation at break and a remarkably higher tensile modulus than stereocomplexes of PLLA‐r‐PCL/PDLA‐r‐PCL and PLLA/PDLA. The biodegradability of a blend of PLLA‐r‐PCL (65/35) and PDLA‐r‐PCL (66/34) with proteinase K was higher than that of PLLA‐b‐PCL (47/53) and PDLA‐b‐PCL (45/55), the degradability of which was higher than that of a PLLA/PDLA blend. A blend film of PLLA‐r‐PDLLA (69/31)/PDLA‐r‐PDLLA (68/32) exhibited higher degradability than a film of PLLA/PDLLA [PDLLA = poly(D ,L ‐LA)]. A stereocomplex of PLLA‐r‐PCL‐r‐PDMO [80/18/2; PDMO = poly(L ‐3,D ,L ‐6‐dimethyl‐2,5‐morpholinedion)] with PDLA‐r‐PCL‐r‐PDMO (81/17/2) showed higher degradability than PLLA‐r‐PDMO (98/2)/PDLA‐r‐PDMO (98/2) and PLLA‐r‐PCL (82/18)/PDLA‐r‐PCL (79/21) blends. The tensile modulus of a blend of PLLA‐r‐PCL‐r‐PDMO and PDLA‐r‐PCL‐r‐PDMO was much higher than that of a blend of PLLA‐r‐PDMO and PDLA‐r‐PDMO. © 2004 Wiley Periodicals, Inc. Novel amphiphilic diblock copolymers from a combination of hydrophobic‐functional poly(lactides) (PLAs) with hydrophilic‐functional PLAs or poly(malic acid), respectively, toward fully biodegradable materials for medical applications, such as micellar drug delivery systems, are reported for the first time. The presented PLA‐based polymeric micelles are characterized by their small size below 100 nm, low critical micellar concentrations, good in vitro stabilities at room and body temperature, and efficient incorporation capability of hydrophobic compounds, particularly with regard to potential drug substances. A novel type of biodegradable/biocompatible amphiphilic hyperbranched copolymer (H40‐PLA‐b‐MPEG) was synthesized. Its micellar properties were studied by DLS, fluorescence spectroscopy and TEM. The drug release profile showed that the H40‐PLA‐b‐MPEG micelles provide an initial burst release, followed by a sustained release of the entrapped hydrophobic model drug over a period of 4 to 58 h. The copolymer degraded hydrolytically within 6 weeks under physiological conditions. The MTT assay showed no obvious cytotoxicity against a human endothelial cell line at a concentration range of 0–400 µg · mL−1. These results indicate that the H40‐PLA‐b‐MPEG micelles have great potential as hydrophobic drug delivery carriers.

The solubility nature of many medicines presents a challenge for successful delivery of these drugs to the body. Polymeric carriers are potentially viable as vessels for both the protection and transport of these medicinal substances. In an effort to generate polymeric materials for this desired application, A‐B‐A triblock copolymers have been synthesized with a central block composed of hydrophilic poly (ethylene glycol) (PEG) and flanking hydrophobic sequences composed of five valine units terminated with end groups of varying hydrophobicity. These copolymers were constructed by adding amino acids stepwise to the hydrophilic block using solution phase chemistry. The self‐assembly behavior of all polymers was investigated using fluorimetry with a pyrene probe. In general, copolymers with more hydrophobic end groups exhibited lower critical aggregation concentrations (CACs). Fmoc‐terminated copolymers displayed the lowest CAC of 0.032 mg/mL and demonstrated little cytotoxicity when exposed to SW620 colorectal cancer cells. Transmission electron micrographs show the presence of multiple compartments within these spherical assemblies, which may prove useful in encapsulating medicinal substances. © 2008 Wiley Periodicals, Inc. Biodegradable star‐shaped poly(ethylene glycol)‐block‐poly(lactide) copolymers were synthesized by ring‐opening polymerization of lactide, using star poly(ethylene glycol) as an initiator and potassium hexamethyldisilazide as a catalyst. Polymerizations were carried out in toluene at room temperature. Two series of three‐ and four‐armed PEG‐PLA copolymers were synthesized and characterized by gel permeation chromatography (GPC) as well as 1H and 13C NMR spectroscopy. The polymerization under the used conditions is very fast, yielding copolymers of controlled molecular weight and tailored molecular architecture. The chemical structure of the copolymers investigated by 1H and 13C NMR indicates the formation of block copolymers. The monomodal profile of molecular weight distribution by GPC provided further evidence of controlled and defined star‐shaped copolymers as well as the absence of cyclic oligomeric species. The effects of copolymer composition and lactide stereochemistry on the physical properties were investigated by GPC and differential scanning calorimetry. For the same PLA chain length, the materials obtained in the case of linear copolymers are more viscous, whereas in the case of star copolymer, solid materials are obtained with reduction in their Tg and Tm temperatures. © 2007 Wiley Periodicals, Inc. ABA triblock copolymers of L ‐lactide (LL) and ε‐caprolactone (CL), designated as PLL‐P(LL‐co‐CL)‐PLL, were synthesized via a two‐step ring‐opening polymerization in bulk using diethylene glycol and stannous octoate as the initiating system. In the first‐step reaction, an approximately 50:50 mol% P(LL‐co‐CL) random copolymer (prepolymer) was prepared as the middle (B) block. This was then chain extended in the second‐step reaction by terminal block polymerization with more L ‐lactide. The percentage yields of the triblock copolymers were in excess of 95%. The prepolymers and triblock copolymers were characterized using a combination of dilute‐solution viscometry, gel permeation chromatography (GPC), 1H‐ and 13C‐NMR, and differential scanning calorimetry (DSC). It was found that the molecular weight of the prepolymer was controlled primarily by the diethylene glycol concentration. All of the triblock copolymers had molecular weights higher than their respective prepolymers. 13C‐NMR analysis confirmed that the prepolymers contained at least some random character and that the triblock copolymers consisted of additional terminal PLL end (A) blocks. From their DSC curves, the triblock copolymers were seen to be semi‐crystalline in morphology. Their glass transition, solid‐state crystallization, and melting temperature ranges, together with their heats of melting, all increased as the PLL end (A) block length increased. New Y‐shaped (AB2‐type) amphiphilic copolymers of poly(ethylene glycol) (PEG) with poly(trimethylene carbonate) (PTMC), PEG‐b‐(PTMC)2, were successfully synthesized by the ring‐opening polymerization (ROP) of TMC with bishydroxy‐modified monomethoxy‐PEG (mPEG). First, a bishydroxy functional ROP initiator was synthesized by esterification of acryloyl bromide with mPEG, followed by Michael addition using excess diethanolamine. A series of Y‐shaped amphiphilic PEG‐(PTMC)2 block copolymers were obtained via ROP of TMC using this PEG with bishydroxyl end groups as macroinitiator and ZnEt2 as catalyst. The amphiphilic block copolymers with different compositions were characterized by gel permeation chromatography (GPC) and 1H NMR, and their molecular weight was measured by GPC. The results showed that the molecular weight of Y‐shaped copolymers increased with the increase of the molar ratio of TMC to mPEG‐(OH)2 initiator in feed while the PEG chain length was kept constant. The Y‐shaped copolymer mPEG‐(PTMC)2 could self‐assemble into micelles in aqueous medium and the critical micelle concentration values of the micelles decrease with increase in hydrophobic PTMC block length of mPEG‐(PTMC)2. Novel poly(l ‐lactide) (PLLA)/poly(d ‐lactide) (PDLA)/poly(tetrahydrofuran) (PTHF) multiblock copolymers with designed molecular structure were synthesized by a two‐stage procedure. Well‐defined PDLA‐PLLA‐PTHF‐PLLA‐PDLA pentablock copolymers were prepared by sequential ring opening polymerization of l ‐ and d ‐lactides starting from PTHF glycol, with the length of the (equimolar) PLLA and PDLA blocks being varied. Then, these dihydroxyl‐terminated pentamers were transformed into multiblock copolymers by melt chain‐extension with hexamethylene diisocyanate–being the first time that the coupling of pentablock units is reported. The successful formation of macromolecular chains with a multiblock and well‐defined architecture was demonstrated by 1H NMR spectroscopy. The thermal properties and structuring of the resulting materials were investigated by means of DSC and WAXD measurements and DMA analysis. Stereocomplexation was found to be promoted during solution and melt crystallization. This approach affords materials combining the high rigidity and strength (other than improved thermal resistance) of the hard stereocomplex crystallites with the flexibility imparted by the soft block, whereby their properties can be finely tailored through the composition of the basic pentablock units without limitations on the final molecular weight. PEG-PLA copolymers with dumbbell- and Y-shaped structures are prepared. They can self-assemble from nanoparticles to micro-sized fusiform micellar particles in aqueous solution. In particular the micelles formed by the (PLA)2-PEG-(PLA)2 particles show a better drug loading capacity and encapsulation efficiency than those formed by linear MPEG-PLA. In vitro studies show that the particles formed by Y-shaped copolymers show a particularly quick drug release. The copolymers have good biocompatibility with low cytotoxicity. These unique self-assembled systems thus have many possible biomedical applications, such as a sustained delivery of high-dosed water insoluble drugs, quick effective drugs for trauma, controlled delivery of the oral-administration drugs, and so forth.  相似文献

In this study, a novel type of amphiphilic block copolymers poly(lactic acid)‐block‐poly(ascorbyl acrylate) (PLA‐block‐PAAA) with biodegradable poly(lactic acid) as hydrophobic block and poly(ascorbyl acrylate) (PAAA) as hydrophilic block was successfully developed by a combination of ring‐opening polymerization and atom transfer radical polymerization, followed by hydrogenation under normal pressure. The chemical structures of the desired copolymers were characterized by 1H NMR and gel permeation chromatography. The thermal physical properties and crystallinity were investigated by thermogravimetric analysis, differential scanning calorimetry, and wide angle X‐ray diffraction, respectively. Their self‐assembly behavior was monitored by fluorescence‐probe technique and turbidity change using UV–vis spectrometer, and the morphology and size of the nanocarriers via self‐assembly were detected by cryo‐transmission electron microscopy and dynamic light scattering. These polymeric micelles with PAAA shell extending into the aqueous solution have potential abilities to act as promising nanovehicles for targeting drug delivery. © 2011 Wiley Periodicals, Inc. Polymerization of L ‐lactide (LA) was performed in the presence of trifluoromethanesulfonic acid (CF3SO3H) via an activated monomer mechanism to synthesize various block copolymers composed of polyethyleneglycol (PEG) and poly(L ‐lactide) (PLLA). The PLLAs obtained had molecular weights close to theoretical values calculated from LA/PEG molar ratios and exhibited monomodal GPC curves. A 1H NMR spectroscopic study showed that the LA carbonyl carbon signal exhibited a change in chemical shift to lower field, caused by electron delocalization of the carbonyl carbon by CF3SO3H. We successfully prepared PEG and PLLA block copolymers using this activated monomer mechanism. We concluded that synthesis proceeded by LA ring‐opening polymerization caused by PEG in the presence of CF3SO3H to yield PEG and PLLA block copolymers. © 2009 Wiley Periodicals, Inc. Poly(ethylene oxide/polylactide/poly(ethylene oxide) (PEO/PL/PEO) triblock copolymers, in which each block is connected by an ester bond, were synthesized by a coupling reaction between PL and PEO. Hydroxyl‐terminated PLs with various molecular weights were synthesized and used as hard segments. Hydroxyl‐terminated PEOs were converted to the corresponding acid halides via their acid group and used as a soft segment. Triblock copolymers were identified by Fourier transform infrared spectroscopy, 1H NMR, and gel permeation chromatography. Differential scanning calorimetry (DSC) and X‐ray diffractometry of PEO/PL/PEO triblock copolymers suggested that PL and PEO blocks were phase‐separated and that the crystallization behavior of the PL block was markedly affected by the presence of the PEO block. PEO/PL/PEO triblock copolymers with PEO 0.75k had two exothermic peaks (by DSC), and both peaks were related to the crystallization of PL. According to thermogravimetric analysis, PEO/PL/PEO triblock copolymer showed a higher thermal stability than PL or PEO. © 2002 Wiley Periodicals, Inc. Novel, biodegradable poly(?‐caprolactone)‐block‐poly(trans‐4‐hydroxy‐N‐benzyloxycarbonyl‐L ‐proline)‐block‐poly(?‐caprolactone) triblock copolymers were synthesized by ring‐opening polymerization from dihydroxyl‐terminated macroinitiator poly(trans‐4‐hydroxy‐N‐benzyloxycarbonyl‐L ‐proline) (PHpr) and ?‐caprolactone (?‐CL) with stannous octoate as the catalyst. The molecular weights were characterized with gel permeation chromatography and matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry. With an increase in the contents of ?‐CL incorporated into the copolymers, a decrease in the glass‐transition temperature (Tg) was observed. The Tg values of copoly(4‐phenyl‐?‐caprolactone) and copoly(4‐methyl‐?‐caprolactone) were higher than Tg of copoly(?‐caprolactone). Their micellar characteristics in an aqueous phase were investigated with fluorescence spectroscopy, dynamic light scattering, and transmission electron microscopy. The block copolymers formed micelles in the aqueous phase with critical micelle concentrations in the range of 1.00–1.36 mg L?1. With higher molecular weights and hydrophobic components in the copolymers, a higher critical micelle concentration was observed. As the feed weight ratio of antitriptyline hydrochloride (AM) to the polymer increased, the drug loading increased. The micelles exhibited a spherical shape, and the average size was less than 250 nm. LPEIs, which are efficient DNA transfection agents, were found to be far less effective for the delivery of siRNAs. Here, two amphiphilic triblock copolymers LPEI(50) -b-PPG(36) -b-LPEI(50) (2) and LPEI(14) -b-PPG(68) -b-LPEI(14) (4) have been synthesized. The transfection assays showed that compound 2 was efficient for DNA transfection whilst it was almost inactive for siRNA delivery. In contrast, polymer 4 was inefficient for DNA transfection while it showed capabilities for siRNA delivery. Taken together, our results indicate that the properties required for DNA and siRNA delivery are different. Moreover, we show that introduction of a hydrophobic segment that allows self-assembly confers siRNA delivery capacities.  相似文献
