LC–DAD coupled with electrospray tandem mass spectrometry (LC–ESI–MS–MS) has been used to evaluate the quality of the traditional Chinese medicine Huang-Lian-Jie-Du-Tang (HLJDT). Twenty-five chromatographic peaks were obtained from a C18 analytical column by gradient elution with acetonitrile and formate buffer (containing 0.5% formic acid) at a flow rate of 1.0 mL min?1. The linearity, precision, and accuracy of the data obtained were acceptable. Thirteen components were identified by ESI–MS, and seven of these were quantitatively analyzed by LC–DAD. The method was used to analyze ten batches of HLJDT, and both chromatographic fingerprints and quantitative data were used to evaluate the quality of the HLJDT. It was concluded that this LC–DAD–ESI–MS method enables more fully validated and complete evaluation and monitoring of the quality of HLJDT.  相似文献

The purpose of this article was to develop a rapid and robust LC–MS–MS method for quantifying shikonin and deoxyshikonin simultaneously in rat plasma using emodin as internal standard. The LC system consisted of an Agilent ZORBAX SB-C18 (1.8 μm, 250 × 4.6 mm, 20 °C) column. Elution with an isocratic mobile phase consisted of methanol/10 mM ammonium acetate in water/acetonitrile containing 0.05% formic acid (45:10:45, v/v/v) at a flow rate of 0.8 mL min?1 yielded sharp, high-resolved peaks within 12 min. The lower limits of quantitation were 0.5 ng mL?1 for shikonin, and 8 ng mL?1 for deoxyshikonin. Correlation coefficient (r) values for the linear range of two analytes were greater than 0.99. Assay precision was <13% and accuracy was 87–99%. This newly developed method was used to the pharmacokinetic studies of the shikonin analogues in rats after intravenous administration (n = 4).  相似文献

For the first time, a validated high-performance liquid chromatographic method with diode-array detection has been developed for discrimination and quality assessment of three species of Baishouwu. LC with gradient elution was performed on extracts from 23 plant samples collected from different geographic locations. The fingerprint was established and 26 chromatographic peaks were selected as characteristic peaks. Similarities of chromatograms were greater than 0.9 for the same species but much lower than 0.6 for different species. Three bioactive constituents were simultaneously analyzed for further quantitative assessment of the quality of Baishouwu, and amounts of the three analytes in different species of Baishouwu samples were found to be significantly different. This LC–DAD method is thus a very reliable and useful method for assessment of the quality of Baishouwu.  相似文献

A rapid and sensitive liquid chromatography–electrospray ionization mass spectrometry method was developed for the determination of aesculin in rat plasma. The analyses were chromatographed on a Zorbax Extend-C18 analytical column (150 × 2.1 mm I.D., 5 µm) with 30:70 (v/v) methanol–0.1% formic acid as mobile phase. Detection was performed by triple-quadrupole tandem mass spectrometry in multi-reaction-monitoring mode with an electrospray ionization source. The method was validated for accuracy and precision, and linearity in the two matrices was good. The assay was linear in the range 12.5–1,800 ng mL?1. The lower limit of quantification of aesculin (LLOQ) was 12.5 ng mL?1. The recovery of aesculin and tinidazole (IS) were well above 85%. The within- and between-batch accuracy was 100–104% and 97–109%, respectively. There were no stability-related problems in the procedure for the analysis of aesculin. The method was successfully used in a preclinical study of the pharmacokinetics of aesculin in rats.  相似文献

To evaluate the quality of Flemingia philippinensis, a validated reversed-phase high-performance liquid chromatographic method with diode-array detection has been developed for chromatographic fingerprint analysis and for quantification of genistin, genistein, and 5,7,3′,4′-tetrahydroxy-6,8-diprenylisoflavone, the three main bioactive isoflavones. In the fingerprint analysis, 21 chromatographic peaks were selected as characteristic peaks; among these 14 flavonoids were identified on the basis of their retention times and UV spectra. In quantitative analysis, the recovery of the method was in the range 94.5–107.7%, and good linearity (r 2 > 0.9990) was observed for all the compounds over a relatively wide range of concentrations. The relative standard deviation of the method was less than 2.5% for intra-day and inter-day assays. The assay was successfully applied to analysis of the three active isoflavones in 19 samples. The results indicated the assay is reproducible and precise and could be used for convenient assessment of the quality of F. philippinensis.  相似文献

A high-performance liquid chromatography coupled with photodiode array detection and electrospray ionization tandem mass spectrometry (HPLC-PAD–ESI-MS) method has been developed for the simultaneous identification and quantification of active compounds (rutin and quercetin) contained in Traditional Chinese Medicine (TCM) Euonymus alatus (Thunb.) Siebold (EAS). The herb samples from ten main origins and five medicinal portions (leaf, fruit, stem, pterygium and root) were investigated. The separation was performed on a Shim C18 column at 30 °C with an isocratic elution. Methanol (A) and water (0.5% methanoic acid, v/v) (B) were used as mobile phases. The recoveries of the two compounds were 100.184% and 100.417%, respectively, and all of them showed good linearity (r2 ? 0.9993) in relatively wide concentration ranges. The developed method was applied to identify and quantify the two major active compounds in the collected herb samples, and the results indicated that contents of the two compounds in EAS varied significantly from habitat to habitat. It was demonstrated that the proposed method was helpful for the quality evaluation of EAS.  相似文献

In this work, an analytical multiresidue method using liquid chromatography tandem-mass spectrometry (LC–MS–MS) with triple quadrupole in selected reaction monitoring (SRM) mode for the simultaneous determination of 54 pesticides in vegetables (pepper and tomato) and citrus fruits (orange and lemon) has been developed. The procedure involves initial single phase extraction of sample with acetonitrile by agitation, followed by liquid–liquid partition aided by “salting out” process using NaCl. The average recovery by the LC–MS–MS method obtained for these compounds varied from 65.5 to 114.5% with a relative standard deviation between 2.3 and 8.3%. The method presents good linearity over the range assayed 10–500 μg L?1 (except famoxadone 50–1,000 μg L?1) and the detection limits for the pesticides studied varied from 0.03 to 14.9 μg kg?1. The proposed method was used to determine pesticide levels in vegetables and citrus fruit samples from different experimental orchards and greenhouses from the Region of Murcia.  相似文献

A liquid chromatographic tandem mass spectrometric validated method was developed for the detection of chemicals attributing color, flavor, taste and medicinal properties to saffron (Crocus sativus L. stigma). Ultrasonic extractions of saffron stigmas were followed by LC procedure with Pinnacle II Cyano (5 μm 150 × 2.1 mm) column and acetonitrile: water (70:30, v/v) as mobile phase. Deprotonated ions formed by a turbo ion spray in negative MS mode were used to detect the analytes. MS–MS detection was by monitoring precursors (m/z) fragmentations; of 149 → 113 (safranal), 327 → 283 (crocetin), 329 → 167 (picrocrocin), 355 → 327 (dimethyl crocetin), 489 → 327 (crocin E), 535 → 489 (carotenes), 651 → 327 (crocin C), 813 → 652 (crocin B), 975 → 651 (crocin A) and 1,137 → 813 (crocin F). The method was validated for linearity, precision, repeatability and specificity.  相似文献

A rapid, selective and convenient liquid chromatography–mass spectrometric method for the simultaneous determination of paracetamol and caffeine in human plasma was developed and validated. Analytes and theophylline [internal standard (I.S.)] were extracted from plasma samples with diethyl ether-dichloromethane (3:2, v/v) and separated on a C18 column (150 × 4.6 mm ID, 5 μm particle size, 100 Å pore size). The mobile phase consisted of 0.2% formic acid–methanol (60:40, v/v). The assay was linear in the concentration range between 0.05 and 25 μg mL?1 for paracetamol and 10–5,000 ng mL?1 for caffeine, with the lower limit of quantification of 0.05 μg mL?1 and 10 ng mL?1, respectively. The intra- and inter-day precision for both drugs was less than 8.1%, and the accuracy was within ±6.5%. The single chromatographic analysis of plasma samples was achieved within 4.5 min. This validated method was successfully applied to study the pharmacokinetics of paracetamol and caffeine in human plasma.  相似文献

A new, rapid, and efficient method, multiple reaction monitoring liquid chromatography–tandem mass spectrometry, has been developed for analysis of nanafrocin in foodstuffs of animal origin. The researchers used a C18 stationary phase coupled with triple-quadrupole tandem mass spectrometry in negative-electrospray mode. The limits of detection (LOD) and quantification (LOQ) were 0.005 and 0.01 mg kg?1, respectively, in the matrixes. Detector response was found to be a linear function of concentration over the range 0.005–0.1 mg kg?1 in each matrix. Mean overall recovery (n = 10) of nanafrocin varied from 71 to 101%. The results show that identification and quantification of nanafrocin residues in foodstuffs of animal origin can be successfully achieved by use of the proposed LC–MS–MS method.  相似文献

In the present paper, study on the multiple absorbed bioactive components of Radix Astragali in rabbit was performed by alternative moving window factor analysis (AMWFA) and LC–DAD–MS. AMWFA was used to confirm the common components both in its abstract fingerprint and plasma fingerprint. This method could be utilized not only to determine the number of common components in different systems but also to identify their corresponding spectra. As can be seen from the results, the method determines the common components in different chromatographic fingerprints more efficient and reliable, especially for none or only weak selective information of some chromatographic peak clusters. With the aid of AMWFA both its abstract fingerprint and plasma fingerprint, one could find that 25 chemical components in RA were absorbed into the rabbit body after oral solution of RA administration.  相似文献

Melamine is attracting much attention because of its toxicity. In the work discussed in this paper, microwave-assisted extraction in combination with ion-exchange high-performance liquid chromatography with diode-array detection was used for analysis of melamine in pet food. Trichloroacetic acid–N,N-dimethylformamide 10:1 was the best extractant, because of the strong polarity of melamine. Separation was performed on a 250 mm × 4.6 mm i.d., 5-μm particle, cation-exchange column; isocratic elution with a mixture of ammonium formate solution (pH 3.0) and acetonitrile was complete within 10 min. UV absorbance DAD detection was performed at 240 nm. Response was a linear function of melamine concentrations from 0.1 to 50 μg mL?1, with a detection limit of 1.0 mg kg?1. Intra-day and inter-day precision, as RSD, was <3% and the recovery of the assay was in the range 95.4–106.8%. In analysis of spiked pet food, the new method yielded satisfactory results. Because of its simplicity and accuracy this straightforward method is particularly suitable for routine melamine analysis.  相似文献

Melarsoprol is the only currently available drug for treatment of the late stage of African trypanosomiasis (sleeping sickness). Unfortunately, the arsenic-containing drug causes serious side effects, for which the mechanisms have not been elucidated so far. This investigation describes the study of the melarsoprol biotransformation processes by electrochemical (EC) techniques. Based on EC, potential oxidation reactions of melarsoprol are examined. Moreover, the reactivity of melarsoprol, its metabolite melarsen oxide, and their oxidation products toward the tripeptide glutathione and the proteins hemoglobin and human serum albumin is evaluated. The combination of different analytical techniques allows the identification as well as the quantification of the biotransformation products. The hyphenation of liquid chromatography (LC) and electrospray ionization mass spectrometry (ESI–MS) is applied for identification and structure elucidation, which implies the determination of exact masses and fragmentation patterns. For the selective detection of arsenic containing metabolites, LC coupled to inductively coupled plasma mass spectrometry is utilized. Based on the obtained data, the oxidative biotransformation of melarsoprol can be predicted, revealing novel species which have been suspected, but not been identified up to now. The results of the protein studies prove that melarsen oxide, the active derivative of melarsoprol, strongly binds to human hemoglobin and forms different adducts via the free cysteinyl groups of the hemoglobin α- and β-chain.  相似文献

Liquid chromatography–ion trap mass spectrometry was employed to investigate the metabolism of linarin in rats. Identification and structural elucidation of the metabolites were performed by comparing the differences in molecular masses, retention times, and full scan MS n spectra between linarin and its metabolites. Six metabolites (acacetin, apigenin, acacetin glucuronide, apigenin glucuronide, acacetin sulfate, apigenin sulfate) were detected in rat urine after oral administration of linarin at the dose of 50 mg kg?1. Furthermore, a selective and sensitive liquid chromatography–triple quadruple mass spectrometry assay was developed and validated for the simultaneous determination of linarin and acacetin (the major metabolite of linarin) in rat urine. Chromatographic separation was carried out on a C18 column, and mass spectrometric detection was performed using a triple-quadrupole mass spectrometer coupled with an electrospray ionization source in the positive ion mode. Quantitation of linarin and acacetin was performed using selected reaction monitoring of precursor–product ion transitions at m/z 593 → 285 for linarin, 285 → 242 for acacetin, and 303 → 153 for hesperitin (internal standard), respectively. The assay exhibited good linearity (r > 0.9900) for both linarin and acacetin. The intra- and inter-day precisions were <13.4 % and the accuracy was between ?8.1 and 3.1 %. The method was successfully applied to the urinary excretion study of linarin in rats after oral administration of linarin.  相似文献

An efficient HPLC–DAD method was developed for simultaneous determination of eight adulterants in weight management supplements and herbs. The eight adulterants were phenolphthalein, sibutramine, nuciferine, and five anthraquinone compounds including aloe-emodin, rhein, emodin, chrysophanol, and physcion. The analytes were ultrasonically extracted with 70% (v/v) methanol aqueous solution followed by centrifugation. The supernatant was subjected to HPLC analysis. A Phenomenex Luna C18 column was applied for chromatographic separation. The mobile phase was consisted of methanol and aqueous solution of 0.05% (v/v) phosphoric acid–0.025% (m/v) sodium dodecyl sulfonate. The flow rate of mobile phase was 0.8?ml?min?1 with gradient elution. Clenbuterol and ibuprofen were used as internal standards. The retention times and the characteristic UV spectrograms were used for qualitative analysis. Quantifications were based on the internal standard curves. Good linearities (r?>?0.9996) for all analytes were obtained with the intra- and inter-day precision (n?=?6) ranging from 0.76 to 5.9% and 0.90 to 8.1%, respectively. The average recoveries from the spiked samples with different matrices varied from 73.4 to 114%. Validations were subsequently performed using LC–MS/MS. The proposed method successfully determined the target adulterants in eight commercial weight management supplements and five weight reducing herbs with satisfactory results.  相似文献

Summary A sensitive and selective liquid chromatographic method coupled with electrospray ionization tandem mass spectrometry (LC–ESI–MS–MS) has been developed for quantification of felodipine in human and dog plasma. Compounds were separated on a 2.0 mm × 150 mm, 5.0 m particle, C8 column with 1 m m ammonium acetate–acetonitrile, 20:80, pH 6.0, as mobile phase at a flow rate of 200 L min–1. Nifedipine was used as internal standard. Plasma samples were extracted with diethyl ether, the centrifuged upper layer was evaporated, the residue was reconstituted with mobile phase, and the reconstituted samples were injected. The analytical column lasted for at least 1000 injections. By use of multiple reaction monitoring (MRM) mode in MS–MS felodipine and nifedipine were detected without severe interference from the human or dog plasma matrix. Felodipine produced a protonated precursor ion ([M + H]+) at m/z 384 and a corresponding product ion at m/z 338. And internal standard (nifedipine) produced a protonated precursor ion ([M + H]+) at m/z 347 and a corresponding product ion at m/z 315. Detection of felodipine in human and dog plasma was accurate and precise, with a limit of quantification of 0.05 ng mL–1. The method has been successfully applied to preliminary pharmacokinetic study of felodipine in human and dog plasma.  相似文献

A simple and specific analytical method for the quantitative determination of shikimic acid from the methanol extract of the fruits of Illicium species and from various plant samples was developed. The LC–UV separation was achieved by reversed-phase chromatography on a C18 column with potassium dihydrogen phosphate and methanol as the mobile phase. In the LC–MS method, the separation was achieved by a C12 column using water and acetonitrile, both containing 0.1% acetic acid as the mobile phase. The methods were successfully used to study the percentage compositions of shikimic acid present in nine species of Illicium and various other plant samples. The detector response was linear with concentrations of shikimic acid in the range from 1.0–500.0 μg mL?1 by LC–UV and 100–1000 ng mL?1 by LC–MS. Mass spectrometry coupled with electrospray ionization interface is described for the identification of shikimic acid in various plant samples. This method involved the use of the [M-H]? ions of shikimic acid at m/z 173.0455 (calculated mass) in the negative ion mode with extractive ion monitoring.  相似文献

High-performance liquid chromatography coupled with diode-array detection and electrospray ionization quadrupole-time of flight mass spectrometry (ESI–Q–TOF MS) has been used for global chemome analysis of Qishenyiqi dropping pills. Under gradient conditions, two sub-chemomes extracted from Qishenyiqi dropping pills were analyzed on the same C18 reversed-phase column by use of different binary mobile phases. In the chromatogram obtained from sub-chemome I, 19 compounds with strong ultraviolet absorption at 260 nm were detected; 27 compounds with weak ultraviolet absorption at 203 nm were detected in the chromatogram obtained from sub-chemome II. All the compounds were identified by ESI–Q–TOF MS. This study furnished much more important information about the pharmacological activity of Qishenyiqi dropping pills than previous studies and laid a good foundation for further exploration of chemomics.  相似文献

A liquid chromatography separation with electrospray ionisation and tandem mass spectrometry detection method was developed for the simultaneous quantification of ten commonly handled cytotoxic drugs in a hospital pharmacy. These cytotoxic drugs are cytarabine, gemcitabine, methotrexate, etoposide phosphate, cyclophosphamide, ifosfamide, irinotecan, doxorubicin, epirubicin and vincristine. The chromatographic separation was carried out by RPLC in less than 21 min, applying a gradient elution of water and acetonitrile in the presence of 0.1% formic acid. MS/MS was performed on a triple quadrupole in selected reaction monitoring mode. The analytical method was validated to determine the limit of quantification (LOQ) and quantitative performance: lowest LOQs were between 0.25 and 2 ng mL(-1) for the ten investigated cytotoxic drugs; trueness values (i.e. recovery) were between 85% and 110%, and relative standard deviations for both repeatability and intermediate precision were always inferior to 15%. The multi-compound method was successfully applied for the quality control of pharmaceutical formulations and for analyses of spiked samples on potentially contaminated surfaces.  相似文献
