Abstract

A Chemiluminescence Enzyme‐Linked Immuno‐Sorbent Assay (CL‐ELISA) for determination and quantification of the fungicide thiram in honeybees was developed in an indirect competitive format. The assay was optimized by determining: the optimal coating conjugate concentration and anti‐thiram antiserum dilution, the effect of the incubation time on the competitive step, the tolerance to organic solvents. The IC50 and the limit of detection (LOD) values were 60 ng mL?1 and 9 ng mL?1, respectively, similar to those of colorimetric ELISA with a calibration range of 9–15,000 ng mL?1. Cross reactivity of some related compounds such as some dithiocarbamates, a thiocarbamate, the ethylenethiourea and the tetramethylthiourea were tested. The assay was then applied to honeybees sample extracts obtained by using the liquid‐liquid extraction or the graphitized carbon‐based solid phase extraction.

The calibration curves in honeybee extracts from liquid‐liquid procedure gave an IC50 of 141 ng mL?1 and a LOD of 17 ng mL?1. In case of extracts obtained by SPE these values were 139 ng mL?1 and 15 ng mL?1, respectively. The average recovery value from honeybee extracts spiked with 75 ng mL?1 of thiram was 72% for SPE, higher than for liquid‐liquid extraction (60%). On the opposite, when the honeybees were directly spiked with 2 and 10 ppm the average recovery was higher for liquid‐liquid extraction (54%), than for SPE (31%). Finally, the assay was applied to honeybee samples collected during monitoring activities in Italy and Russia.  相似文献

A simple, sensitive and rapid ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) method was developed and validated for the quantification of mitragynine in rat plasma using amitriptyline hydrochloride as an internal standard. Sample preparation involved a one-step liquid?Cliquid extraction using methyl t-butyl ether. Mitragynine was separated on an Acquity UPLC? BEH HILIC column using isocratic elution with a mobile phase of 10 mM ammonium formate buffer containing 0.1% formic acid:acetonitrile (15:85, v/v). At a flow rate of 0.2 mL min?1, the retention time of mitragynine was found to be 1.3 min. Ionization was performed in the positive ion electrospray mode. The selected mass-to-charge (m/z) ratio transition of mitragynine ion [M + H]+ used in the selected ion recording (SIR) was 399.1. The calibration curve was found to be linear over a concentration range of 1?C5,000 ng mL?1 (r = 0.999) with a lower limit of quantification (LLOQ) of 1 ng mL?1. Intra- and inter-day assay variations were found to be less than 15%. The extraction recoveries ranged from 85?C93% at the three concentrations (2, 400 and 4,000 ng mL?1) in rat plasma. This method was successfully used to quantify mitragynine in rat plasma following intravenous administration of the compound.  相似文献

A selective, sensitive, and accurate method has been developed and validated for the quantification of tangeretin in rat plasma. The application of LC-electrospray-ion trap mass spectrometry in full scan and multiple reactions monitoring modes were investigated. Following solid phase extraction using a hydrophilic–lipophilic balance cartridge, the analytes were separated on a C18 column using an isocratic mobile phase composed of acetonitrile/water (50:50, v/v) containing 0.3% formic acid. In full scan mode, the LOQ was 2 ng mL?1. The standard calibration curve was linear (R 2 = 0.9999) over the concentration range 2–200 ng mL?1. The precision over the concentration range was within 15% (RSD) and the accuracy was ranged from 86 to 115%. In multiple reaction monitoring mode, the LOQ was 1 ng mL?1 and the standard calibration curve was linear (R 2 = 0.9976) over the concentration range 1–100 ng mL?1 with a precision of 12% and accuracy rangeing from 91 to 113%.  相似文献

A combined homogeneous assay and colorimetric determination method using gold nanoparticles was developed for rapid determination of lead(II) in contaminated natural waters. The presence of lead(II) in the colloidal gold suspension causes a change in the absorbance of the suspension. An increase in the absorption property at 595 nm is accompanied by a change in the size of the gold nanoparticles. High concentrations of lead cause aggregation of the gold colloids. Colloidal gold nanoparticles were synthesized using tannic acid as the reducing agent; this reagent allowed selective determination of lead in 10 µL of water, with a detection limit of 310 ng mL?1 with an analysis time of 5 min. The coefficient of variation for lead(II) within the working range of the assay (520 ng mL?1–13 µg mL?1) varied from 1.3% to 9.2%. The limit of detection using this method with a sample volume of 50 µL was 60 ng mL?1. The coefficient of variation for lead over the working range of the determined concentrations (80 ng mL?1–25 µg mL?1) varied from 0.2% to 9.3%, while the values for the inter-day assay (n = 8) were less than 10%. The method was employed for the analysis of river, lake, marsh, and spring water; the recovery of lead was determined to be 72.5%–130% for 10 µL of water and 93.6%–114.7% for 50 µL.  相似文献

A simple, rapid and sensitive liquid chromatography/electrospray tandem mass spectrometry quantitative detection method, using amantadine as internal standard, was developed for the simultaneous analysis of paracetamol, pseudoephedrine and chlorpheniramine concentrations. Analytes were extracted from plasma samples by liquid–liquid extraction with n-hexane–dichloromethane–2-propanol (2:1:0.1, v/v), separated on a C18 reversed-phase column with 0.1% formic acid–methanol (40:60, v/v) and detected by electrospray ionization mass spectrometry in positive multiple reaction monitoring mode. Calibration curves for plasma were linear over the concentration range 10–10,000 ng mL?1 of paracetamol, 2–2,000 ng mL?1 of pseudoephedrine and 0.2–200 ng mL?1 of chlorpheniramine. The method has a lower limit of quantitation of 10 ng mL?1 for paracetamol, 2.0 ng mL?1 for pseudoephedrine and 0.2 ng mL?1 for chlorpheniramine. Recoveries, precision and accuracy results indicate that the method was reliable within the analytical range, and the use of the internal standard was very effective for reproducibility by LC-MS-MS. This method is feasible for the evaluation of pharmacokinetic profiles of a novel multicomponent sustained release formulation containing 325 mg of paracetamol, 30 mg of pseudoephedrine hydrochloride and 2 mg of chlorpheniramine maleate. It is the first time the pharmacokinetic evaluation of a novel sustained-action formulation containing paracetamol, pseudoephedrine and chlorpheniramine has been elucidated in vivo using LC-MS-MS.  相似文献

A sensitive and selective analytical method for the quantification of pregabalin, sildenafil and the active desmethyl metabolite of sildenafil (UK-103320) has been developed. The method can simultaneously quantify the three analytes within the expected in vivo concentration ranges using 50 ??L of rat plasma. It utilises solid-phase extraction followed by high performance liquid chromatography coupled with tandem mass spectrometry. Quantitation in rat plasma demonstrated good accuracy and precision over the following dynamic ranges for each analyte: pregabalin (70?C10,000 ng mL?1), sildenafil (1?C2,000 ng mL?1) and UK-103320 (1?C2,000 ng mL?1). For each analyte, the following lower limits of quantitation were obtained: 70 ng mL?1 for pregabalin and 1 ng mL?1 for sildenafil and UK-103320, respectively. The method was successfully used to analyse plasma samples from rats when pregabalin and sildenafil were administered in combination.  相似文献

A simple, sensitive and specific liquid chromatographic method with UV detection (228 nm) was developed for the simultaneous estimation of rosiglitazone and glimepride in human plasma. Rosiglitazone and glimepride were extracted from plasma using liquid–liquid extraction. Separation was achieved with an RP C18 Column using a mixture of phosphate buffer (50 mM) with octane sulfonic acid (10 mM), methanol and acetonitrile as a mobile phase (55:10:35, v/v). pH was adjusted to 7.0. Amlodipine was used as an internal standard (IS). LOD of the method was found to be 20 ng mL?1 for both drugs. Results were linear over the studied range 40.994–2007.556 ng mL?1 for rosiglitazone (r ≥ 0.99) and 41.066–2094.84 ng mL?1 for glimepride( r ≥ 0.99). The method was found to be simple, selective, precise and reproducible for the estimation of both drugs from spiked human plasma.  相似文献

A rapid and specific high-performance liquid chromatographic method coupled with electrospray ionization mass spectrometric detection has been developed and validated for identification and quantification of wogonin and oroxylin A in rat plasma. Wogonin, oroxylin A, and diazepam (internal standard) were extracted from plasma samples by liquid–liquid extraction with ethyl acetate. Chromatographic separation was achieved on a C18 column with acetonitrile–0.6% aqueous formic acid 35:65 (v/v) as mobile phase at a flow rate of 0.2 mL min?1. Detection was performed with a single-quadrupole mass spectrometer in selected-ion-monitoring (SIM) mode. Linearity was good within the concentration range 14.4–360 ng mL?1 for wogonin and 10.8–271 ng mL?1 for oroxylin A; the correlation coefficients (r 2) were 0.9999. The intra-day and inter-day precision, as RSD, was below 12.4%, and accuracy ranged from 81.1 to 111.9%. The lower limit of quantification was 14.4 ng mL?1 for wogonin and 10.8 ng mL?1 for oroxylin A. This method was successfully used in the first pharmacokinetic study of wogonin and oroxylin A in rat plasma after oral administration of the active fraction from Xiao-xu-ming decoction.  相似文献

A sensitive, precise and simple LC method for the simultaneous quantification of aliskiren, valsartan and sitagliptin in rat plasma has been developed and validated. The chromatographic separation was achieved on a C18 column (250 mm × 4.6 mm, 5 μm) maintained at room temperature, using isocratic elution with acetonitrile/20 mM ammonium acetate buffer (35:65, v/v), pH adjusted to 4.85 with glacial acetic acid, and detected using a fluorescence detector. Liquid–liquid extraction of the aliskiren, valsartan and sitagliptin from the rat plasma with t-butyl methyl ether resulted in their high recoveries. LC calibration curves based on the extracts from the rat plasma were linear in the range of 25–2,000 ng mL?1 for aliskiren and sitagliptin and 50–4,000 ng mL?1 for valsartan. The limits of quantification were 25 ng mL?1 for aliskiren and sitagliptin and 50 ng mL?1 for valsartan. The precision and accuracy of the method were well within the generally accepted criteria for biomedical analysis. The described method was successfully applied to study the pharmacokinetics of aliskiren, valsartan and sitagliptin following oral administration, individually as well as in combination in Sprague–Dawley rats. The results of the study implied the occurrence of pharmacokinetic interaction upon the co-administration of these three drugs.  相似文献

A sensitive high performance liquid chromatographic (HPLC) method has been developed and validated for the simultaneous determination of four local anaesthetics: lidocaine, proparacaine, bupivacaine and oxybuprocaine. A full factorial design was used. The chromatographic separation was achieved using a Bondesil C8 (4.6 × 2.5 mm i.d., particle size 5 μm) analytical column. An optimised mobile phase consisted of acetonitrile and sodium dihydrogen phosphate (pH = 3.0, 20 mM) (30:70, v/v) at a flow rate of 1.2 mL min?1. Local anaesthetics detection was performed by UV-Vis detector at 220 nm. The retention times for lidocaine, proparacaine, bupivacaine and oxybuprocaine were 5.74, 9.28, 16.84 and 26.26 min, respectively. HPLC-UV-Vis method was linear in the range of 50–5,000 ng mL?1 for lidocaine and proparacaine and 100–5,000 ng mL?1 for bupivacaine and oxybuprocaine. The limit of detection (LOD) was 25 ng mL?1 for lidocaine, proparacaine and 30 ng mL?1 for bupivacaine and oxybuprocaine. The limit of quantification (LOQ) was found to be 50 ng mL?1 for lidocaine, proparacaine and 100 ng mL?1 for bupivacaine, oxybuprocaine. In intra-day and inter-day precision and accuracy analysis, the relative standard deviation was found to be less than 8%.  相似文献

A rapid, sensitive and specific assay method has been developed to simultaneously determine human plasma concentrations of hydrocodone and its metabolites, norhydrocodone, hydromorphone, using high-performance liquid chromatography with an electrospray ionization (ESI) tandem mass spectrometry (HPLC?CMS?CMS). Hydrocodone, its metabolites, and internal standard, hydrocodone-d 3, norhydrocodone-d 3, hydromorphone-d 3, were separated from human plasma using solid-phase extraction (Empore MPC-SD Solid Phase Extraction Disk). The eluate was dried, reconstituted and injected into the LC?CMS?CMS system. Chromatographic separation was performed on a Kromasil 100-5SIL-Dimensions C18 column (100 × 2.1 mm, 5.0 ??m, Thermo Hypersil-Keystone, USA) using a gradient mobile phase with 20 mmol L?1 ammonium formate in water with 0.2% formic acid and 0.1% formic acid in acetonitrile. Detection and quantitation were performed by MS/MS using electrospray ionization and multiple reactions monitoring in the positive ion mode. The calibration curves were linear over the concentration ranges 0.05?C50 ng mL?1 for hydrocodone (r 2 = 0.9991) and norhydrocodone (r 2 = 0.9990), and 0.01?C10 ng mL?1 for hydromorphone (r 2 = 0.9990). The limit of quantification was 0.05 ng mL?1 for hydrocodone and norhydrocodone, and 0.01 ng mL?1 for hydromorphone. The extraction recovery was above 64.36, 68.51 and 71.78% for hydrocodone, norhydrocodone and hydromorphone. The accuracy was higher than 99.06, 97.70 and 100.07% for hydrocodone, norhydrocodone and hydromorphone. The intra- and inter-day precisions were <5.80, 5.90 and 3.02% for hydrocodone, norhydrocodone and hydromorphone. The method was accurate, sensitive and simple and was successfully applied to a pharmacokinetic study after a single oral administration of hydrocodone bitartrate at a dose of 5 mg in 12 healthy Chinese volunteers.  相似文献

A highly sensitive liquid chromatographic-atmospheric pressure chemical ionization-tandem mass spectrometric method is developed to quantitate phenacetin and its metabolite paracetamol in rabbit plasma. The analytes and internal standard oxazepam are extracted from plasma by liquid–liquid extraction using ethyl acetate, and separated on a Zorbax SB-C18 column (2.1 mm × 150 mm, 5 μm) using acetonitrile–0.1% formic acid in water (40:60 v/v) at a flow of 0.4 mL min?1. Detection is carried out by multiple reaction monitoring on a ion-trap LC-MS-MS system with an atmospheric pressure chemical ionization interface. The assay is linear over the range 4–1,600 ng mL?1 for phenacetin and 3–2,000 ng mL?1 for paracetamol, with a lower limit of quantitation of 4 ng mL?1 for phenacetin and 3 ng mL?1 for paracetamol. Intra- and inter-day precision are less than 7.1% and the accuracy are in the range 97.3–103.5%. The validated method is successfully used to analyze the drug in samples of rabbit plasma for pharmacokinetic study.  相似文献

A sensitive, simple, and accurate method for determination and pharmacokinetic study of ferulic acid and isoferulic acid in rat plasma was developed using a reversed-phase column liquid chromatographic (RP-LC) method with UV detection. Sample preparations were carried out by protein precipitation with the addition of methanol, followed by evaporation to dryness. The resultant residue was then reconstituted in mobile phase and injected into a Kromasil C18 column (250 × 4.6 mm i.d. with 5 μm particle size). The mobile phase was methanol-1% formic acid (33:67, v/v). The calibration plots were linear over the range 5.780–5780 ng·mL?1 for ferulic acid and 1.740–348.0 ng·mL?1 for isoferulic acid. Mean recoveries were 85.1% and 91.1%, respectively. The relative standard deviations (RSDs) of within-day and between-day precision were not above 15% for both of the analytes. The limits of quantification were 5.780 ng·mL?1 for ferulic acid and 1.740 ng·mL?1 for isoferulic acid. This RP-LC method was used successfully in pharmacokinetic studies of ferulic acid and isoferulic acid in rat plasma after intravenous injection of Guanxinning Lyophilizer.  相似文献

A simple, sensitive, and precise high performance liquid chromatographic method for the analysis of pantoprazole, rabeprazole, esomeprazole, domperidone and itopride, with ultraviolet detection at 210 nm, has been developed, validated, and used for the determination of compounds in commercial pharmaceutical products. The compounds were well separated on a Hypersil BDS C18 reversed-phase column by use of a mobile phase consisting of 0.05 M, 4.70 pH, potassium dihydrogen phosphate buffer - acetonitrile (720:280 v/v) at a flow rate of 1.0 mL min?1. The linearity ranges were 400–4,000 ng mL?1 for pantoprazole, 200–2,000 ng mL?1 for rabeprazole, 400–4,000 ng mL?1 for esomeprazole, 300–3,000 ng mL?1 for domperidone and 500–5,000 ng mL?1 for itopride. Limits of detection (LOD) obtained were: pantoprazole 147.51 ng mL?1, rabeprazole 65.65 ng mL?1, esomeprazole 131.27 ng mL?1, domperidone 98.33 ng mL?1 and itopride 162.35 ng mL?1. The study showed that reversed-phase liquid chromatography is sensitive and selective for the determination of pantoprazole, rabeprazole, esomeprazole, domperidone and itopride using single mobile phase.  相似文献

A rapid and sensitive method for the quantitative determination of picroside II in rat plasma was developed and validated using liquid chromatographic separation with tandem mass spectrometric detection. The analytes of interest were extracted from rat plasma samples by ethyl acetate after acidification with 1.0% acetic acid solution. Chromatographic separation was achieved on a Hypersil GOLD column (50 × 2.1 mm I.D., 5 μm) using a mobile phase consisting of acetonitrile–0.1% formic acid solution (30:70, v/v) at a flow rate of 0.2 mL min?1. Detection was performed on a triple quadrupole tandem mass spectrometer by selected reaction monitoring (SRM) mode via electrospray ionization (ESI). The calibration curve was linear in the concentration range of 1.00–400 ng mL?1 in rat plasma, with a 1.00 ng mL?1 lower limit of quantification (LLOQ). Satisfactory results were achieved for intraday repeatability [relative standard deviation (RSD) = 6.4–12.4%] and inter-day precision (RSD = 6.8–14.7%). The accuracy in terms of relative error ranged from ?2.1 to 10.0%. The extraction recoveries of picroside II and icariin (internal standard) were 80.0 and 89.3%, respectively. The developed method was successfully employed to determine picroside II plasma concentrations after oral administration to Wistar rats.  相似文献

Based on the available rabbit monoclonal antibody (RabMAb), a rapid and sensitive lateral flow immunoassay (LFA) platform has been developed for quantitative detection of four sulfonamide residues(SRs) of sulfadiazine (SD), sulfathiazole (STZ), sulfapyridine (SP), and sulfamethoxazole (SMX).Within the designed LFA competitive format assay, which was based on antigen-antibody properties, the hapten conjugate N1-[4-(carboxymethyl)-2-thiazolyl] sulfanilamide linked to protein ovalbumin (TS-OVA) and goat anti-rabbit antibody were sprayed as capture and control reagents, respectively, and then the antibody was conjugated to colloidal gold particles as the detection reagent. With quantitative assessment aided by a colorimetric strip reader, the sensitivities of the established LFA method for SD, STZ, SP, and SMX were 0.91 ng mL?1, 0.10ng mL?1,0.12ng mL?1, and 2.13ng mL?1, and the half-maximum inhibition concentrations (IC50) were 5.19 ng mL?1, 1.25 ng mL?1, 0.66 ng mL?1, and 24.14 ng mL?1, respectively. The recoveries at three spiked levels (5, 20, 50 ng mL?1for SD, STZ, and SP; 20, 50, 100 ng mL?1 for SMX) were in the range of 78.02–135.10% and 76.40–137.16% for milk and swine urine, respectively. More importantly, the detection performance of the established platform was consistent with that of in-parallel LC-MS/MS analysis. In conclusion, the proposed LFA platform has showed the potential for fast, sensitive and relatively accurate quantification of four sulfonamide residues in practical uses.  相似文献

The IC50 and the limit of detection (LOD) values were 14.8 ng mL?1 and 0.11 ng mL?1, respectively, similar to those of colorimetric ELISA with a calibration range of 0.1–2600 ng mL?1. Cross reactivity of some related compounds such as three imidacloprid metabolites, 6-chloro nicotinic acid, 5-hydroxy-imidacloprid, and imidacloprid olefin, and one other chloronicotinoid insecticide, acetamiprid, were tested. The assay was then applied to honeybee extracts obtained by using the liquid-liquid extraction. The calibration curves in honeybee extracts from the liquid-liquid procedure gave an IC50 of 23.7 ng mL?1 and a LOD 1.6 ng mL?1. The average recovery value from honeybee extracts spiked with 100 and 1000 ng mL?1 of imidacloprid were 73% and 76%, respectively. Finally, the assay was applied to honeybee samples collected during monitoring activities in Italy; it was found that only five of the 27 samples were positives, with low concentrations of imidacloprid ranging between 1.2 and 15.4 ng g?1.  相似文献

The purpose of this study was to develop a simple and accurate analytical method for determination of norepinephrine, epinephrine, and dopamine in urine. The method involves liquid–liquid extraction then liquid chromatography–mass spectrometry (LC–MS). Alkyl chloroformate derivatives were prepared, as the N(O,S)-alkoxycarbonyl alkyl esters of the analytes, in the aqueous samples. The optimum derivatizing reagent for preparation of the N(O,S)-alkoxycarbonyl alkyl esters was chosen by comparing the efficiency of LC of the derivatized analytes after liquid–liquid extraction. The optimum conditions for liquid–liquid extraction from the aqueous matrix were pH 3.0, no salt, and diethyl ether as extraction solvent. Limits of detection (LOD) were 0.5 ng mL?1 for dopamine and epinephrine and 0.1 ng mL?1 for norepinephrine. Limits of quantification (LOQ) for urine samples were 1.0 ng mL?1 for all three compounds. The precision of intra- and inter-day assays was 1.65–581 and 7.17–9.73% (relative standard deviation, RSD), respectively. The range of inaccuracy for intra- and inter-day assays was ?6.47 to 11.9% and ?7.5 to 7.76% (bias) at concentrations of 5 and 50 ng mL?1, respectively.  相似文献

The aim of this research was to develop a sensitive liquid chromatographic–electrospray ionization–mass spectrometric (LC–MS) method for direct measurement of the concentration of Atorvastatin in human plasma. Plasma samples (1 mL) were extracted with 3 mL ethyl acetate, and by a simple reversed-phase chromatography. Pitavastatin was used as internal standard (IS). The LOQ was 0.25 ng mL?1 (RSD 4.24%). The assay was linear from 0.25–20 ng mL?1. And the correlation coefficient for the calibration regression line was 0.9996 or better. Intra-day and inter-day accuracy were better than 15%. The method has been successfully used for a pharmacokinetic study with human subjects. A two-period crossover designed bioequivalence research was also progressed in healthy Chinese volunteers. Among the pharmacokinetic data obtained, T max was 1.36 ± 0.68 h for reference formulation and 0.81 ± 0.54 h for test formulation. C max was 8.54 ± 5.06 ng mL?1 for reference formulation and 9.54 ± 3.68 ng mL?1 for test formulation. t 1/2 was 8.50 ± 2.74 h for reference formulation and 9.24 ± 3.17 h for test formulation. AUC 0?48h was 54.77 ± 21.82 h ng mL?1 for reference formulation and 55.66 ± 20.91 h ng mL?1 for test formulation. The method was successfully applied to the study of pharmacokinetics of Atorvastatin in healthy Chinese volunteers.  相似文献

A sensitive fluorescence liquid chromatographic analytical method was developed for the simultaneous determination of carnosine enantiomers in rat plasma. The method was applied to pharmacokinetic studies. Chiral separation of carnosine enantiomers was achieved by pre-column derivatization with o-phthaldialdehyde and the thiol N-acety-l-cysteine as derivating reagents. They were separated on an ODS column and detected by fluorescence detection (λex = 350 nm, λem = 450 nm). γ-Aminobutyric acid was used as internal standard. The method was linear up to 6,000 ng mL?1 for l-carnosine, 4,000 ng mL?1 for d-carnosine. Low limit of quantitation (LLOQ) was 40 ng mL?1 for each isomer. The relative standard deviations obtained for intra- and inter-day precision were lower than 12% and the recoveries were higher than 75% for both enantiomers. The method was applied to a stereoselective study on the pharmacokinetics of carnosine after oral administration with a single dose (carnosine, 75 mg kg?1 for each isomer) to a rat. The initial data indicated that l-carnosine had a larger value of the highest plasma concentration than d-carnosine (C max 5,344 vs. 1,914 ng mL?1), and that of l-carnosine had a lower value of AUC(0?∞) and t 1/2(h) (AUC(0?∞) 5,306 vs. 6,321 ng h mL?1, t 1/2 1.43 vs. 3.37 h). Our results indicated that the pharmacokinetic of l-carnosine and d-carnosine revealed enantioselective properties significantly.  相似文献
