Blood substitutes based on hemoglobin or hemoglobin-based oxygen carriers (HBOCs) are oxygen-carrying therapeutic agents developed for use in operations and emergencies in place of donated blood. Increased oxygen-carrying capacity through the use of blood substitutes could help elite athletes to lengthen endurance capacity and improve their performance. As blood substitutes become more readily available, it is essential that a qualitative detection method for their abuse in sport is available. Ideally, such a method would be simple and inexpensive. This study investigates methods that could be used as screening procedures to easily detect HBOCs in plasma and develops tests that can unequivocally confirm their presence. The investigation into the screening method indicates that the direct visual screening of plasma discoloration is the most appropriate with detection limits of less than 1% HBOC in plasma. Two methods are shown to confirm the presence of exogenous hemoglobin in plasma samples, size-exclusion chromatography with photodiode array detection and high-performance liquid chromatography analysis of enzymatic digests with detection by electrospray mass spectrometry. This work emphasizes the need for cooperation between drug developers and sports testing laboratories to ensure that methods for the detection of putative doping agents are available prior to product release. 相似文献

Hemoglobin-based oxygen carriers (HBOCs) are blood substitutes, synthesized by polymerizing hemoglobin, which are being developed and investigated as alternatives to blood for medical purposes. However, due to their ability to increase the oxygen carrying capacity when taken by healthy individuals, HBOCs have been used as a doping agent among endurance athletes and are included in the World Anti-Doping Agency's Prohibited List. To maintain the fairness of competitions and continue the battle against doping, it is essential to be able to detect HBOCs if present in an athlete's blood. To achieve this goal, it is necessary to differentiate HBOCs from the native hemoglobin and to do so in a cost and time effective manner. We have developed a rapid capillary zone electrophoresis (CZE), UV absorbance, method capable of detecting HBOCs, in whole blood samples, at levels below those considered necessary to provide a performance enhancement. Our approach to the analysis for HBOCs utilizes the whole blood sample, not just the plasma, and does not require the use of immunoprecipitants to ensure accurate analysis. By lysing the red blood cells and using centrifugal filtration, followed by our CZE separation, we are able to effectively distinguish between native hemoglobin and HBOCs. Through this method, we have been able to reliably detect concentrations of HBOCs at the equivalent of 5.5 g/L, the equivalent to a 3.5% increase in blood hemoglobin concentration for an athlete. 相似文献

Hemoglobin-based oxygen carriers (HBOCs) are blood substitutes based on hemoglobin of either bovine or human origin and they can potentially be misused in elite sports to improve endurance performance. Recently, three methods have been proposed in doping control analysis to allow HBOCs screening and identification by application of electrophoresis, size-exclusion chromatography coupled with HPLC and LC coupled with tandem mass spectrometry (LC/MSMS). In view of the Athens 2004 Olympic Games, modifications were introduced in order to increase the specificity of these methods. The sample preparation protocols of the electrophoretic and SEC-HPLC methods were modified with the introduction of sequential ultra filtration steps to remove all heme containing material below 100 kDa, thus leaving only HBOCs material for analysis. Furthermore, a modification of the LC/MSMS methodology was introduced to allow full scan MS-MS spectra of peptide segments arising from the tryptic digestion of bovine HBOCs. These relatively simple methodological modifications have major impact, as far as time and cost effectiveness is concerned in doping control procedures, because they provide a useful tool in order to identify which suspect samples from the initial visual screening are due to hemolysis and exclude them from further analysis. 相似文献

Various blood substitutes have been designed by researchers through continual efforts. With recent progress in nanotechnology, new types of artificial red blood cells with cellular structure are available. This article aims to describe some artificial red blood cells which encapsulate or conjugate hemoglobin molecules through various approaches, especially the nanoscale self-assembly technique, to mitigate the adverse effects of free hemoglobin molecules. The polarographic diffusion current of the O2 reduction was utilized to estimate the nature of O2 supply from O2 binding oxygen carriers. Perfluorocarbon (PFC) emulsion, hemoglobin (Hb) and erythrocyte were used as either artificial or natural oxygen carriers. The changes of O2 limiting current originated by the addition of an oxygen carrier into the electrolyte solution open or closed to the atmosphere under some restricted conditions were analyzed.It is concluded that the main rate-determining factor for O2 transport by PFC emulsion. Hb solution or erythrocyte suspension, may be attributed to the O2 diffusion inside the PFC particle, to the diffusion of O2 binding Hb in Hb solution, or to the diffusion of O2 binding Hb inside the erythrocyte, respectively. 相似文献

Hemoglobin vesicles (HbV) or liposome-encapsulated Hbs are artificial oxygen carriers that have been developed for use as transfusion alternatives. The extremely high concentration of the HbV suspension (solutes, ca. 16 g/dL; volume fraction, ca. 40 vol %) gives it an oxygen-carrying capacity that is comparable to that of blood. The HbV suspension does not possess a colloid osmotic pressure. Therefore, HbV must be suspended in or co-injected with an aqueous solution of a plasma substitute (water-soluble polymer), which might interact with HbV. This article describes our study of the rheological properties of HbV suspended in a series of plasma substitute solutions of various molecular weights: recombinant human serum albumin (rHSA), dextran (DEX), modified fluid gelatin (MFG), and hydroxylethyl starch (HES). The HbV suspended in rHSA was nearly Newtonian. Other polymers-HES, DEX, and MFG-induced HbV flocculation, possibly by depletion interaction, and rendered the suspensions as non-Newtonian with a shear-thinning profile (10(-4)-10(3) s(-1)). These HbV suspensions showed a high storage modulus (G') because of the presence of flocculated HbV. However, HbV suspended in rHSA exhibited a very low G'. The viscosities of HbV suspended in DEX, MFG, and high-molecular-weight HES solutions responded quickly to rapid step changes in shear rates of 0.1-100 s(-1) and a return to 0.1 s(-1), indicating that flocculation is both rapid and reversible. Microscopically, the flow pattern of the flocculated HbV that perfused through microchannels (4.5 microm deep, 7 microm wide, 20 cmH2O applied pressure) showed no plugging. Furthermore, the time required for passage was simply proportional to the viscosity. Collectively, the HbV suspension viscosity was influenced by the presence of plasma substitutes. The HbV suspension provides a unique opportunity to manipulate rheological properties for various clinical applications in addition to its use as a transfusion alternative. 相似文献

A simple method for the isolation and purification of hemoglobin components from starch gel by electrophoresis is described. An inverted bottle with a cut off bottom is used. Pieces of gel containing the hemoglobin are embedded in potato starch, wetted with buffer, and placed in the neck of the inverted bottle, the mouth of which has been closed with filter paper and tape. Connection with the negative tank buffer is achieved by inserting a layer of starch gel between the potato starch and the negative tank buffer. By electrophoresis, the hemoglobin migrates and accumulates in a dialysing tube tied around the mouth of the bottle and hanging into the positive tank buffer. 相似文献

Reversible laser induced deoxygenation in the lyophilized phase of hemoglobin is demonstrated by means of resonant Raman scattering, luminescence, and optical transmission. Specific Raman modes, which are both sensitive to the spin states of Fe(II) in the hemes and resonant in the visible, are monitored as a function of time to evaluate the effect of the illuminating laser. These modes act as in-situ markers of the oxygen content of the protein. The reversible photoinduced deoxygenation can be observed through both the Raman spin-markers and the optical transmission experiments. In the former, reversible changes in the intensities of specific Raman modes are observed, while in the latter, the oscillator strength of the two main absorptions of oxyhemoglobin in the visible are seen to vary accordingly. The luminescence in lyophilized hemoglobin is found to have at least two different contributions, (i) a resonant component with the Raman modes and; (ii) a nonresonant contribution, which increases at high input laser powers and eventually masks the Raman signals. The nonresonant contribution is the luminescence of the photoproduct achieved by thermal denaturation of the protein and remains standing as a permanent nonreversible damage in the illuminated spot. Semiempirical electronic calculations of the wavefunction and total energy of the iron porphyrin reveal the underlying physical origin of the laser induced deoxygenation process in the hemes and are also presented. 相似文献

Chemical looping combustion (CLC) is a novel combustion technology with the capability for segregation of exhaust products (i.e., carbon dioxide/H2O or N2/O2). The combustion is performed in two interconnected reactors with a solid oxygen carrier circulating between them, transferring oxygen from the air to the fuel. The feasibility of a successful CLC system depends on the selection of an appropriate oxygen carrier. Cu-based oxygen carriers are good oxygen carriers due to high reactivity. However, it faces low melting point, agglomeration problems in fluidized bed. In this study, a circular reduction?Coxidation reaction simulated to the cyclic operation of the Cu-based oxygen carrier was conducted on the thermogravimetric analyzer (TG). The thermal behaviors of the potential Cu-based oxygen carrier were investigated by using an X-ray diffraction (XRD), scanning electron microscope (SEM), and surface analyzer. Multiple TG results show that the weight loss was 3.4%, indicating that the loading CuO amount was 17%. Moreover, the weight loss and weight gain was equal during 73 redox cycles, suggesting the good thermal stability of the oxygen carrier. The conversion rate of reduction and oxidation for each redox cycle remained constant even after 73 redox cycles. XRD results show the new phase formation of CuAl2O4 during redox cycles, which promotes the thermal stabilization of the oxygen carrier. The surface area of the oxygen carrier decreased from 105 to 13?m2?g?1 after 73 redox cycles and the particle size distribution shifted from 5?C15?nm to 15?C30?nm, suggesting that the micorpores were blocked or collapsed. However, the reactivity of the oxygen carrier didn??t decrease. SEM results show that CuO was evenly distributed on the surface of Al2O3 after 73 redox cycles. Overall, these results suggested that the Cu-based oxygen carrier was ready for fluidized bed tests. 相似文献

Hemoglobin was isolated from chicken blood and purified by biochemical methods.This hemoglobin was allowed to interact with copperglutathione (CuGSH) and the interaction pattern was monitored by spectrophotometric and spectrofluorimetric techniques. The interaction ratio was found to be 1:0.7 in both the cases. The amount of oxygen carried by hemoglobin is studied with a polyethyleneimine modified pyrolytic graphite electrode. The medicinal herb, salidroside, is shown to enhance the ability of hemoglobin to carry oxygen in vitro. When salidroside is added to the test solution, the reduction current in the presence of oxygen increases according to the amount of the herb added. No such increase is seen in the absence of oxygen. Furthermore, the percent increase is larger for lower concentrations of oxygen in the solution. UV–Vis spectrometry indicates that the salidroside does not greatly change the conformation of the hemoglobin. 相似文献

用浸渍法以橄榄石为载体负载CuO制备了Cu-橄榄石载氧体,并在固定床上开展了载氧体存在下煤焦的水蒸气气化实验。结果表明,提高气化温度和水碳物质的量比可以提高煤焦气化的碳转化率和合成气产率;增加载氧体CuO负载量和载氧体/煤焦质量比能提高碳转化率和水转化率,但会消耗产气中的合成气,导致合成气产率降低。高温再生后载氧体依然能保持活性,表明橄榄石载体抑制了Cu/CuO的烧结。载氧体经950℃再生八次后用于煤焦气化,实现碳转化率42.3%,水转化率57.3%,合成气产率2.12L/(g·h)。 相似文献

Summary Reduced copper has been used generally for removing the oxygen in the nitrogen gas in the microdetermination of oxygen, but instead of reduced copper the authors have used about 20 g of Raney-nickel in the dry state prepared through the development of Raney-alloy, and have found that the oxygen is removed completely from the nitrogen gas at room temperature; Raney-nickel has also a much greater capacity than the reduced copper.
Zusammenfassung Zur Reinigung des für die Mikrobestimmung des Sauerstoffs verwendeten Stickstoffs wurde üblicherweise reduziertes Kupfer verwendet. An dessen Stelle verwenden die Verfasser etwa 20 g trockenes Raney-Nickel und konnten feststellen, daß der Stickstoff damit schon bei Zimmertemperatur vollständig von Sauerstoff befreit wird. Außerdem hat Raney-Nickel eine viel höhere Kapazität als reduziertes Kupfer.

采用共沉淀法制备了Ce-M-O氧载体(M=Fe、Mn、Cu),并进行了XRD表征。研究了Ce-M-O中晶格氧部分氧化甲烷制合成气的反应。考察了再生时间、再生温度对氧载体部分氧化甲烷性能的影响。研究结果表明, Ce-Fe-O固溶体中的晶格氧适于部分氧化甲烷制合成气。在新鲜的Ce Fe O氧载体上存在少量的强氧化物种,导致开始阶段大部分甲烷被完全氧化,然后该氧载体能均匀地释放出具有高选择性的体相晶格氧将甲烷氧化为CO和H2。通过对氧载体再生条件的控制,可以有效提高目标产物的选择性,当再生温度为850℃,再生时间为7min时, 获得了最大的CO(96.68%)和H2(97.56%)选择性,同时H2与CO摩尔比达到2.02。在无气相氧存在下,用Ce-Fe-O中晶格氧实现甲烷部分氧化制合成气的方法是可行的。 相似文献

Photoinduced oxygen dynamics in hemoglobin is monitored by surface enhanced resonant Raman spectroscopy (SERRS) using silver colloids. The spectra are characterized by massive transient amplifications which we assign to a charge-transfer mechanism during the opening of the heme-pockets in the release of oxygen. This transient lifts the selection rule specificity of normal Raman active modes and allows for the full density of states of the hemes to be observed. The amplification of specific forbidden modes is controlled by the orientation alignment of individual molecules in proximity with the colloid surface. We propose that the technique can be used to monitor oxygen release in single proteins. 相似文献
