Adsorption behaviors of functional poly(amido amine) dendrimers with an azacrown core and long alkyl chain spacers were investigated on gold and self-assembled monolayer (SAM) by means of time course attenuated total reflection-surface enhanced infrared absorption and surface plasmon resonance spectroscopies. While 1.5th and 2.5th generation (G1.5 and G2.5) ester-terminated dendrimers were slightly adsorbed on all substrates examined, the adsorption of G2 amine-terminated dendrimer increased in the order dodecanethiol SAM相似文献

The present work describes, for the first time, in situ electrochemical preparation of dendrimer-encapsulated Cu nanoparticles using a self-assembled monolayer of fourth-generation amine-terminated polyamidoamine (PAMAM) dendrimer as the template. Atomic force microscopy (AFM) and X-ray photoelectron spectroscopy (XPS) studies of the modified surface confirmed the presence of Cu nanoparticles entrapped in dendrimer film. Au electrode modified with a monolayer of the dendrimer enables preconcentration and subsequent voltammetric detection of Cu2+ at picomolar concentrations. Further, Cu nanoparticles in the dendrimer monolayer could be electrochemically derivatised to Cu hexacyanoferrate, which exhibits specific crystal planes, unlike the random distribution of crystal planes in bulk-formed Cu hexacyanoferrate, which is another catalytically active material for sensor applications. In this study, electrochemical immunosensors were developed for the detection of prostate specific antigen (PSA) using ferrocene (Fc) and polyamidoamine dendrimer (PAMAM) constructs. The biosensor fabrication was designed by modifying the screen‐printed gold electrode (Au) with ferrocene cored dendrimers (FcPAMAM) synthesized in three different generations. The self‐assembled monolayer principle was followed, to obtain sensitive, selective and disposable electrodes. Therefore, the Au electrodes were modified with cysteamine (Cys) to obtain a functional surface for FcPAMAM dendrimers to bind. Dendrimer generations were attached to this surface using a cross‐linker (glutaraldehyde) so that a suitable surface was obtained for binding of biological components. The Monoclonal PSA antibody (anti‐PSA) was immobilized on the Au electrode surface which coated with dendrimer, and (Au/Cys/FcPAMAM/anti‐PSA) biosensing electrode was obtained. The PSA detection performances of electrochemical impedance spectroscopy (EIS) and Amperometry based immunosensors exhibited very low detection limits; 0.001 ng mL?1 and 0.1 pg mL?1, respectively. In addition, EIS and Amperometry based biosensors using Au/Cys/FcPAMAM/anti‐PSA sensing electrode were represented excellent linear ranges of 0.01 ng mL?1 to 100 ng mL?1 and 0.001 ng mL?1 to 100 ng mL?1. We report here a multistep route for the immobilization of DNA and proteins on chemically modified gold substrates using fourth-generation NH(2)-terminated poly(amidoamine) dendrimers supported by an underlying amino undecanethiol (AUT) self-assembled monolayer (SAM). Bioactive ultrathin organic films were prepared via layer-by-layer self-assembly methods and characterized by fluorescence microscopy, variable angle spectroscopic ellipsometry, atomic force microscopy (AFM), X-ray photoelectron spectroscopy (XPS), and attenuated total internal reflection Fourier transform infrared spectroscopy (ATR-FTIR). The thickness of the AUT SAM base layer on the gold substrates was determined to be 1.3 nm from ellipsometry. Fluorescence microscopy and AFM measurements, in combination with analyses of the XPS/ATR-FTIR spectra, confirmed the presence of the dendrimer/biopolymer molecules on the multilayer sensor surfaces. Model proteins, including streptavidin and rabbit immunoglobulin proteins, were covalently attached to the dendrimer layer using linear cross-linking reagents. Through surface plasmon resonance measurements, we found that sensor surfaces containing a dendrimer layer displayed an increased protein immobilization capacity, compared to AUT SAM sensor surfaces without dendrimer molecules. Other SPR studies also revealed that the dendrimer-based surfaces are useful for the sensitive and specific detection of DNA-DNA interactions. Significantly, the multicomponent films displayed a high level of stability during repeated regeneration and hybridization cycles, and the kinetics of the DNA-DNA hybridization process did not appear to be influenced by surface mass transport limiting effects.  相似文献

Protein-resistant films derived from the fifth-generation poly(amidoamine) dendrimers (PAMAM G5) functionalized with oligo(ethylene glycol) (OEG) derivatives consisting of various ethylene glycol units (EG(n), n = 3, 4, and 6) were prepared on the self-assembled monolayers (SAMs) of 11-mercaptoundecanoic acid (MUA) on gold substrates. The resulting films were characterized by ellipsometry, contact angle goniometry, and X-ray photoelectron spectroscopy (XPS). About 35% of the peripheral amines of the dendrimers were reacted with N-hydroxysuccinimide-terminated EG(n) derivatives (NHS-EG(n)). The dendrimer films showed improved stability over octadecanethiolate SAMs on gold in hot solvents, attributed to the formation of multiple amide bonds per PAMAM unit with underlying NHS-activated MUA monolayer. The EG(n)-attached PAMAM surfaces with n = 3 reduced the adsorption of fibrinogen to approximately 20% monolayer, whereas 2-3% for n = 4 or 6. The dendrimer films with various densities of EG(n) molecules on PAMAM surfaces were prepared by immersion of the NHS-terminated MUA-functionalized gold substrates in ethanolic solutions containing PAMAM and NHS-EG(n) of various mole ratios. The density (r) of the EG(n) molecules on the PAMAM surfaces is consistent with the mole ratio (r') of NHS-EG(n)/free amine of PAMAM in solutions. The resistance to protein adsorption of the resulting surfaces is correlated with the surface density and the length of the EG chains. At their respective r, the EG(n)-modified dendrimer films resisted approximately 95% adsorption of fibrinogen on gold surfaces. Finally, the specific binding of avidin to the approximately 5% and approximately 40% biotinylated EG3 dendrimers (surface density of biotin with respect to the total number of terminal amino groups on PAMAM G5) gave rise to about 50% and 100% surface coverage by avidin, respectively.  相似文献

A newly designed 1.5th generation poly(amido amine) dendrimer with an azacrown core, hexylene spacers, and octyl terminals was spread on gold nanoparticle (Au-NP) suspension. The surface pressure-area isothermal curves indicated that the molecular area of dendrimer on Au-NP suspension was significantly smaller than that on water, indicating the formation of dendrimer/Au-NP composites. The dendrimer Langmuir films on the Au-NP suspension were transferred to copper grids at various surface pressures and observed by transmission electron microscopy. The transferred films consisted of a fractal-like network of nanoparticles at low surface pressure and of a defect-rich monolayer of nanoparticles at high surface pressure. From these results, it was suggested that the dendrimers bind Au-NPs, and dendrimer/Au-NP composites formed networks or monolayers at the interface. From the intensity decrease of the Au plasmon band of Au-NP suspension after the formation of composite, it was estimated that some (approximately 14) dendrimer molecules bind to one Au-NP. Furthermore, neutron reflectivity at the air/suspension interface and X-ray reflectivity of the film transferred on a silicon substrate revealed that the dendrimer molecules are localized on the upper-half surface of Au-NP. Metal affinity of azacrown, flexibility of hexylene spacer, and amphiphilicity of dendrimer with octyl terminals played important roles for the formation of dendrimer/Au-NP hybrid films. The present investigation proposed a new method to fabricate the self-assembled functional polymer/nanoparticle hybrid film.  相似文献

A novel one-step electrochemical method for the preparation of capping-free cadmium sulfide nanoparticles is described. With gold as the working electrode, capping-free CdS nanoparticles are synthesized very conveniently at 70°C in the ethylene glycol (EG) solution of elementary sulfur, cadmium salt, and supporting electrolyte at −0.1 V. By carefully selecting the reductive potential, elementary sulfur is reduced while the reduction of Cd2+ is blocked by the formation of a sulfur monolayer on the gold electrode surface. The produced S2− reacts with cadmium cations in the solution to produce CdS. In this method, magnetic stirring can effectively prevent the deposition of CdS on the electrode surface. XRD analysis indicates that the product is pure cubic-phase CdS. The size and morphology of the particles are studied by TEM. As shown by quartz-crystal microbalance measurements, in the potential range from 0.0 to 0.55 V (NHE), sulfide ions adsorbed on the gold electrode surface accelerate the electrode reaction of anodic dissolution of gold in acidic thiocarbamide solutions. The microbalance determination of kinetic parameters at a constant electrode surface coverage with sulfide ions includes a special procedure developed for the determination of the gold dissolution rate. The conditions (the potential range and the potential scan rate) of independence of the dissolution rate from the diffusion limitations associated with the ligand delivery is determined. Under these conditions, the polarization curve is shown to be linear on semilogarithmic coordinates and correspond to the Tafel equation. In this potential range, the transfer coefficient α and the reaction order with respect to the ligand p are determined at a constant electrode surface coverage θ with adsorbed sulfide ions. It is shown that with the transition from the surface coverage with sulfide ions θ = 0.1 to θ = 0.8, the transfer coefficient α changes from 0.25 to 0.55, the exchange current (i 0) changes from 10?5 to 5 × 10?5 A/cm2, and the effective reaction order p with respect to the ligand changes from 0.2 to 1.3. The mentioned changes are associated not only with the acceleration of gold dissolution in the presence of chemisorbed sulfide ions but also with the changeover in the mechanism of this process. Quartz-crystal microbalance data on the gold dissolution rate qualitatively agree with the results of voltammetric measurements of a renewable gold electrode. A possible version of explanation of the catalytic effect of sulfide ion adsorption on the gold dissolution is put forward.  相似文献

The interaction of avidin with biotin on a functional Au surface containing fourth generation amine-terminated polyamidoamine (G4-NH2 PAMAM) dendrimers was investigated through the use of Fourier transform infrared reflection–adsorption spectroscopy (FT-IRRAS). The first step in the fabrication of the functional surfaces used was the construction of an aldehyde-terminated self-assembled monolayer (SAM) through the treatment of Au-coated glass slides with ethanol solutions of self-synthesized 2-hydroxypentamethylene sulfide (HPMS). The as-formed aldehyde-terminated monolayer was subsequently immersed in methanol solutions of G4-NH2 PAMAM dendrimer to obtain well-organized primary amine-terminated surfaces. Biotinylation of the amine-terminated layers thus obtained was accomplished by use of the N-succinimidyl ester of biotin. Each step of the synthetic process, as well as the performance of final surface for protein recognition was monitored by FT-IRRAS. In particular, the molecular recognition ability was examined and quantified by use of an alkyne dicobalt hexacarbonyl probe coupled with avidin. Non-specific adsorption of avidin was determined by exposure of the amine-terminated and/or biotinylated surfaces to solutions of biotin-saturated avidin. The results indicate that the biotinylated G4-NH2 PAMAM dendrimer layers formed according to this procedure have a high capacity for binding avidin with relatively high specificity. The performance of these layers (i.e. both binding capacity and specificity) improve substantially when 6-mercapto-1-hexanol (MH) is present as a co-adsorbent during the formation of the initial aldehyde-terminated layers. This effect can be attributed to the dilution of the initial aldehyde-terminated SAM, leading to a more favorable spatial arrangement of the subsequent biotinylated surfaces.  相似文献

Divergent growth of surface-initiated dendritic nanostructures on gold surfaces in a highly controlled, stepwise manner is demonstrated, using metal-organic coordination as the binding motif. The repeat unit for dendrimer growth was a branched, C3-symmetrical ligand building block bearing three bis-hydroxamate groups. The surface initiation sites for dendrimer growth were obtained by the formation of a mixed monolayer comprising isolated bis-hydroxamate disulfide anchor ligands and octanethiol (OT) at very low anchor/OT ratios. Following functionalization of the surface with spaced anchors, alternate immersion in solutions of Zr4+ ions and the branched ligand afforded surface-confined dendrimers of increasing generation, where the number of generations is conveniently controlled by the number of coordination binding sequences. The heights of different generation dendrimers are in excellent agreement with values predicted by molecular models, as well as with thicknesses of branched multilayers prepared by the same procedure on full anchor monolayers. At higher generation numbers, gradual dendrimer overlap and coalescence are observed, eventually resulting in a continuous overlayer and a transition from 3D to 1D growth. A mechanism for the development of dendritic coordination nanostructures on surfaces is discussed.  相似文献

New types of microfabricated surface‐enhanced Raman spectroscopy (SERS) active substrates produced by electron beam lithography and ion beam etching are introduced. In order to achieve large enhancement factors by using the lightning rod effect, we prepare arrays consisting of sharp‐edged nanostructures instead of the commonly used dots. Two experimental methods are used for fabrication: a one‐stage process, leading to gold nanostar arrays and a two‐stage process, leading to gold nanodiamond arrays. Our preparation process guarantees high reproducibility. The substrates contain a number of arrays for practical applications, each 200×200 μm2 in size. To test the SERS activity of these nanostar and nanodiamond arrays, a monolayer of the dye crystal violet is used. Enhancement factors are estimated to be at least 130 for the nanodiamond and 310 for the nanostar arrays.  相似文献

The effect of the gold surface renewal on the polarization curve is studied at various gold dissolution stages in electrolytes containing 0.1 M thiourea, (1.4–4.0) 10?5 M sodium sulfide, and 0.5 M sulfuric acid, at 20°C. The behavior of the curves after the cutting-off of a surface layer of gold in the potential region where a current decay is observed (i.e. passivation of the process) and the dependence of the current in the maximum of a polarization curve on the thiourea concentration are explained by “deactivation” of catalytically active adsorbed sulfide ions.  相似文献

We report on a highly sensitive chemiluminescent (CL) biosensor for the sequenc-specific detection of DNA using a novel bio barcode DNA probe modified with gold nanoparticles that were covered with a dendrimer. The modified probe is composed of gold nanoparticles, a dendrimer, the CL reagent, and the DNA. The capture probe DNA was immobilized on magnetic beads covered with gold. It first hybridizes with the target DNA and then with one terminal end of the signal DNA on the barcoded DNA probe. CL was generated by adding H2O2 and Co(II) ions as the catalyst. The immobilization of dendrimer onto the gold nanoparticles can significantly enhance sensitivity and gives a detection limit of 6 fmol L-1 of target DNA.

This work reports a new electrochemical monitoring platform for sensitive detection of Cu2+ coupling click chemistry with nanogold‐functionalized PAMAM dendrimer (AuNP‐PAMAM). The system involved an alkyne‐modified carbon electrode and an azide‐functionalized AuNP‐PAMAM. Initially, the added Cu2+ was reduced to Cu+ by the ascorbate, and then the azide‐modified AuNP‐PAMAM was covalently conjugated to the electrode via Cu+‐catalyzed azide‐alkyne click reaction. The carried AuNPs accompanying PAMAM dendrimer could be directly monitored by stripping voltammetry after acidic pretreatment. By introduction of high‐loading PAMAM dendrimer with gold nanoparticles, as low as 2.8 pM Cu2+ (ppt) could be detected, which was 125‐fold lower than that of gold nanoparticle‐based labeling strategy. The method exhibited high specificity toward target Cu2+ against other potentially interfering ions, and was applicable for monitoring Cu2+ in drinking water with satisfactory results.  相似文献

Self-assembled monolayer of 8-mercaptoquinoline (MQ) on the surface of gold from MQ dilute ethanolic solutions is investigated by electrochemical methods. Some aqueous redox probes, such as ferrocene carboxylic acid and Fe(CN)6 4–/3– can sufficiently diffuse into the monolayer because significant diffusion-limited current peaks are observed when the redox reactions take place, showing that the monolayer is very loosely packed or dominated by defects. However, the study on the electron transfer of other aqueous probes, such as Cu2+ and Ru(NH3)6 3+/2+, confirm that the monolayer can block the electron transfer on the gold electrode surface rather effectively for its low ratio of pinhole defects. These studies show that the MQ monolayer on the electrode can provide an excellent barrier for penetration of some probes but cannot resist the penetration of other probes effectively. The unusual properties of the self-assembled monolayers are attributed to the entity of the very large heterocyclic moiety.  相似文献

Flameless atomic absorption spectrometry (flameless a.a.s.) was applied to study the state of mercury deposited on a gold plate electrode from very dilute mercury(II) solution by controlled-potential electrolysis. A stable monolayer is formed on the gold electrode by the electrolysis at a potential about 200 mV more positive than the reversible Nernst potential for the reduction of mercury(II) to mercury(0). After the monolayer formation, bulk mercury is deposited on the monolayer at the reversible potential and an adatom layer is also found. The difference of activation free energies between the evaporation of mercury from the monolayer and that from bulk mercury corresponds to the underpotential shift for the electrodeposition of mercury on the gold electrode.  相似文献

Electrogenerated chemiluminescence (ECL) for DNA hybridization detection is demonstrated based on DNA that was self-assembled onto a bare gold electrode and onto a gold nanoparticles modified gold electrode. A ruthenium complex served as an ECL tag. Gold nanoparticles were self-assembled on a gold electrode associated with a 1,6-hexanedithiol monolayer. The surface density of single stranded DNA (ssDNA) on the gold nanoparticle modified gold electrode was 4.8?×?1014 molecules per square centimeter which was 12-fold higher than that on the bare gold electrode. Hybridization was induced by exposure of the target ssDNA gold electrode to the solution of ECL probe consisting of complementary ssDNA tagged with ruthenium complex. The detection limit of target ssDNA on a gold nanoparticle modified gold electrode (6.7?×?10?12 mol L?1) is much lower than that on a bare gold electrode (1.2?×?10?10 mol L?1). The method has been applied to the detection of the DNA sequence related to cystic fibrosis. This work demonstrates that employment of gold nanoparticles self-assembled on a gold electrode is a promising strategy for the enhancement of the sensitivity of ECL detection of DNA.  相似文献

Introducing sodium sulfide (about 10?5 M) into acidic thiocarbamide solutions reduces the gold reduction overpotential. The reaction rate passes through a maximum at a potential of 0.1 V. The overpotential depends on the sulfide ion concentration and the time of electrode exposure to solution prior to the beginning of scanning. Transients of potential measured on a renewable gold electrode in thiocarbamide electrolytes containing catalytically active species served as the basis for calculations of the coefficient of trapping of sulfide ions by the growing gold deposit. The kinetics of gold electrodeposition at fixed surface coverages with adsorbed sulfide ions θ is studied. It is shown that at θ = const, the dependence of the reaction rate on the overpotential is described by the Tafel equation. It is shown that with an increase in θ, the effective values of exchange current and transfer coefficient increase from i 0 ≌ 10?5 A/cm2 and α ≌ 0.25 in pure solutions to α ≌ 0.5 and i 0 ≌ 10?4 A/cm2 at θ ≥ 0.3 and then remains virtually unchanged. The reaction order decreases in the absolute magnitude, remaining negative. Thus for θ ≌ 0, p k = ?logi/?logc = ?1, whereas for θ ≥ 0.3, p k = ?0.3. A possible explanation is proposed for the catalytic effect of the sulfide ion adsorption on the mechanism of the gold reduction from acidic thiocarbamide electrolytes.  相似文献

The electrochemical detection of alpha‐feto protein based on novel gold nanoparticles‐ poly(propylene imine) dendrimer platform is reported. The platform was prepared by co‐electrodeposition of gold nanoparticles and generation 3 poly (propylene imine) dendrimer on a glassy carbon electrode. Each modifying step was characterised by cyclic voltammetry and electrochemical impedance spectroscopy. The electrochemical measurements showed that the platform was stable, conducting and exhibited reversible electrochemistry. Results obtained from the electrochemical impedance spectroscopy interrogation in [Fe(CN)63−/4−] redox probe showed a marked reduction in charge transfer resistance (Rct) after each modification step. The immunosensor was prepared by immobilisation of a probe anti‐alpha feto protein (AFP) on the platform for 3 hrs at 35 °C followed by blocking the surface with bovine serum albumin to minimise non‐specific binding. The prepared immunosensor was used to detect AFP over a wide concentration range from 0.005 to 500 ng/mL and detection limits of 0.0022 and 0.00185 ng/mL were obtained for SWV and EIS measurements respectively. The immunosensor gave good stability over a period of fourteen days when stored at 4 °C.  相似文献

Platinum and palladium dendrimer-encapsulated nanoparticles (DENs) were prepared within commercially available, fourth-generation, amine-terminated, poly(amidoamine) dendrimers (G4-NH2). The synthesis is carried out by selectively encapsulating metal complexes within the dendrimer and then reducing the resulting composite. Intradendrimer complexation requires control over the solution pH to prevent attachment of the metal complexes to primary amine groups on the dendrimer periphery. That is, the surface primary amines of the dendrimer must be selectively protonated in the presence of the interior tertiary amines. The metal-ion encapsulation and reduction processes were characterized by UV-vis spectroscopy. Forty-atom Pt and Pd DENs were examined by high-resolution transmission electron microscopy, which showed that the mean particle sizes were 1.4 and 1.5 nm, respectively, and that both were nearly monodisperse (standard deviation = 0.3 nm). The free amine groups on the dendrimer surface were used to link Pd DENs to monolithic Au surfaces via an intermediate self-assembled monolayer adhesion layer.  相似文献
