A new series of mesogenic compounds having a cholesteryl moiety has been synthesised by condensing p-amino benzoate of cholesterol and methoxy substituted 4-n-alkoxy cinnamoyl chlorides and their liquid crystalline properties has been studied. All the members of the series are enantiotropic and exhibit chiral nematic (N*) mesophase. The plot of transition temperatures versus number of carbon atoms in the alkoxy chain exhibits odd–even effect and falling tendency for N*–isotropic transition temperatures. The compounds exhibit oily streak textures that on slight disturbance change to the plane textures and show iridescent colours. High anisotropy, linearity and intermolecular hydrogen bonding confer rich mesomorphic properties on the system. Intermolecular hydrogen bonding arising from amide linkage can lead to supramolecular motifs.  相似文献

Various polymeric phosphonium salts and the corresponding low-molecular-weight model compounds were prepared and their antibacterial activities against Staphylococcus aureus and Escherichia coli were explored by the viable cell counting method in sterile distilled water. Antibacterial activity of the polymers was found to be higher than that of the corresponding model compounds, particularly against S. aureus. Furthermore, the polymeric phosphonium salt exhibited a higher activity by 2 orders of magnitude than the polymeric quaternary ammonium salt with the same structure except the cationic part. Compounds with the longest alkyl chain (octyl) studied were found to exhibit particularly high activity, and this finding may be ascribed to the contribution of the increased hydrophobicity of the compounds to the cidal activity. © 1993 John Wiley & Sons, Inc.  相似文献

Fabrication of an amperometric cholesterol biosensor by co-immobilization of cholesterol esterase (ChEt) and cholesterol oxidase (ChOx) onto conducting polypyrrole (PPY) films using electrochemical entrapment technique is described. Electrochemical polymerization was carried out using a two-electrode cell configuration at 0.8 V. Characterization of resulting amperometric biosensor for the estimation of cholesterol has been experimentally determined in terms of linear response range, optimum pH, applied potential, temperature, and shelf-life. These PPY/ChEt/ChOx electrodes can be used for cholesterol ester estimation from 1 to 8 mM and have shelf-life of about 4 weeks at 4 °C during which about 15 estimations of cholesterol ester could be made. The sensitivity of PPY/ChEt/ChOx electrode has been found to be 0.15 μA/mM and the apparent Km value for this electrode is 9.8 mM. Conductivity of the polymer films found to be about 3×10−3 S/cm.  相似文献

It is shown that cerium dioxide hydrosols can be stabilized by polycationic compounds of zirconium, and the optimum conditions of the stabilization are determined. Stable CeO2 hydrosols with concentrations of higher than 20 wt % are prepared; the sizes and electrophoretic mobility of their particles, as well as sol stability in the presence of some electrolytes, are investigated. The nature of the aggregation stability of the stabilized cerium dioxide sols is discussed.  相似文献

For the purpose of increasing the in vivo stability of polycation gene carriers, we prepared a kind of p H-sensitive poly(ethylene glycol)-poly(γ-benzyl-L-glutamate-co-glutamic acid)(PEG-PGA(65), 65 denotes the molar ratio of glutamic acid in poly(γ-benzyl-L-glutamate-co-glutamic acid)). PEG-PGA(65) showed low cytotoxicity and could shield the positive charge of DNA/PEI(1:1) polyplexes efficiently. The transfection was enhanced due to the partially charge shielding in He La cell line at pH of 7.4. There was almost no transfection efficiency when the surface charge of the ternary particles turned to negative at p H of 7.4. However, the transfection efficiency recovered a lot by culturing at p H of 6.0 at the beginning of transfection. Confocal microscopic observation and flow cytometry results showed DNA/PEI polyplexes should be efficiently released and endocytosized at p H 6.0, because of the p H triggered deshielding action of PEG-PGA(65). Due to the good biocompatibility and suitable p H triggered shielding/deshielding property, PEG-PGA(65) could be a potential shielding system for polycationic gene carriers used in vivo.  相似文献

Ultraresin 1 was prepared from highly branched polyethylene imine (Mn=10,000) via reductive cross-linking with terephthaldialdehyde. Following quaternization with methyl iodide, the polycationic Ultraresin 2, with iodide as a counterion, was obtained. These novel resins combine low swelling with high mechanical stability. By anion exchange polycationic Ultraresins carrying borohydride (3) and periodate (22) were generated and were investigated as very high loading polymer reagents. Ultra-borohydride resin 3 had a reducing activity of up to 12 mmol/g depending on the substrate. It proved successful in diverse reductions including those of aldehydes, ketones, and nitroolefines. The resin was employed in the reductive amination of aldehydes with an excess of amines, which were removed by the use of a scavenger resin. Periodate resin 22 was obtained with an active loading of up to 5.4 mmol/g and was employed in oxidations of sulfides, diols, hydroquinones, and hydrazines.  相似文献

The development of a cholesterol biosensor by co-immobilization of cholesterol esterase (ChEt) and cholesterol oxidase (ChOX) on oxygen electrode is described. The electrode consists of gold cathode and Ag/AgCl anode. The enzymes were immobilized by cross-linking with glutaraldehyde and Bovine Serum Albumin (BSA). The immobilized enzymatic membrane was attached to the tip of the electrode by a push cap system. The optimum pH and temperature of the sensor was determined, these are 6 and 25 degrees C respectively. The developed sensor was calibrated from 1-75 mg/dl of cholesterol palmiate and found linear in the range of 2-50 mg/dL. The calibration curve was drawn with V(i) (ppm/min)(initial velocity) vs different concentrations of cholesterol palmiate (mg/dL). The application of the sensor to determine the total cholesterol in different real food samples such as egg, meat was investigated. The immobilized enzymatic layer can be reused over 30 times and the stability of the enzymatic layer was studied up to 9 weeks.  相似文献

The synthesis of four lipids containing the hemithioindigo chromophore as part of the fatty acid is described. Heck reaction of bromophenyl thioacetate esters with acrylonitrile, followed by reduction, ester hydrolysis, and Friedel--Craft acylation--cyclization gave a substituted thioindoxyl that condensed with an alkoxy benzaldehyde to produce the hemithioindigo. "Solventless" nitrile hydrolysis followed by mixed anhydride coupling of the acid with glycerophosphocholine produced lipids bearing two hemithioindigo chromophores. The photochemistry of various hemithioindigo derivatives was studied to confirm the expected photoisomerization in both homogeneous organic solution, and in vesicle bilayer membranes. Characteristic changes in the UV--visible spectra are consistent with fully reversible Z--E photoisomerization. Chromatographic separation of the Z and E isomers of a compound containing a single hemithioindigo chromophore confirmed the spectroscopic analysis and provided a quantitative analysis of the compositions of Z--E isomer mixtures.  相似文献

Cholesterol amperometric biosensors constructed with enzymes entrapped in electropolymerized layers of polypyrrole and poly-naphthalene derivative polymers are compared. The biosensors are based on entrapment of cholesterol oxidase and/or cholesterol esterase in monolayer or multilayer films electrochemically synthesised from pyrrole, 1,8-diaminonaphthalene (1,8-DAN), and 1,5-diaminonaphthalene (1,5-DAN) monomers. Seven configurations were assayed and compared, and different analytical properties were obtained depending on the kind of polymer and the arrangement of the layers. The selectivity properties were evaluated for the different monolayer and bilayer configurations proposed as a function of the film permeation factor. All the steps involved in the preparation of the biosensors and determination of cholesterol were carried out in a flow system. Sensitivity and selectivity depend greatly on hydrophobicity, permeability, compactness, thickness, and the kind of the polymer used. In some cases a protective outer layer of non-conducting poly(o-phenylenediamine) polymer improves the analytical characteristics of the biosensor. A comparative study was made of the analytical performance of each of the configurations developed. The biosensors were also applied to the flow-injection determination of cholesterol in a synthetic serum.  相似文献

Cholesterol oxidase (ChOx) has been immobilized onto conducting poly[2-methoxy,5-(2′-ethyl-hexyloxy)-1,4-phenylene vinylene] (MEH-PPV)/stearic acid (SA) Langmuir-Blodgett film transferred onto octadecanethiol (ODT) modified gold plate. The ChOx/MEH-PPV/SA LB film bioelectrode exhibits has been characterized by FT-IR, contact angle, and atomic force microscopy. The response of the ChOx/MEH-PPV/SA LB film bioelectrode carried out using differential pulse voltammetry (DPV) studies reveal linearity from 1.29 to 12.91 mM of cholesterol concentration and response time as 30 s. This ChOx/MEH-PPV/SA bioelectrode exhibits values of correlation coefficient as 0.9939, standard deviation as 0.0029 μA and limit of detection as 1.66 mM. UV-visible spectrophotometer studies reveal that 5.2 × 10−3 U of ChOx are actively working per cm2 area of ChOx/MEH-PPV/SA LB film bioelectrode and this bioelectrode is thermally stable upto 55 °C with reusability of about 60 times.  相似文献

A rapid, simple and reproducible two-step method for constructing cholesterol biosensors by covalently bonding cholesterol oxidase (ChOx) to a 3,3′-dithiodipropionic acid di(N-succinimidyl ester) (DTSP)-modified gold electrode is described. Exhaustive characterizations of both the immobilization process and the morphological properties of the resulting ChOx monolayer were performed via a quartz crystal microbalance (QCM) and atomic force microscopy (AFM) operated under liquid conditions, respectively. In addition, scanning electrochemical microscopy (SECM) measurements were performed in order to check that the immobilized enzyme retains its catalytic activity. The replacement of the natural electron acceptor (O2) in the enzymatic reaction with an artificial mediator, hydroxymethylferrocene (HMF), was also studied. Finally, cholesterol was amperometrically determined by measuring the hydrogen peroxide produced during the enzymatic reaction at +0.5 V. The optimized cholesterol biosensor exhibited a sensitivity of 54 nA mM−1 and a detection limit of 22 μM.  相似文献

Novel glycolipids, which contain 2 and 15 oligomaltose units and a phosphatidylethanolamine, were synthesized and characterized by FTIR and (1)H-NMR spectroscopies. The well-defined linear structure of the glycolipids was assured by an end-point conjugation strategy using selective oxidation of the reducing end groups of maltose oligosaccharides, followed by aminolysis with distearoylphosphatidylethanolamine. The intermediate acids react selectively with amines to form amide linkages, catalyzed by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride and N-hydroxysuccinimide. Conformations of the glycolipids at the air-water interface were proposed based on the film balance measurements. The unique conformations of glycolipids at interfaces may offer advantages over traditional PEO-derived lipids in regard to their applications for sterically stabilizing liposomes. The glycolipids demonstrated the ability for sterically stabilizing liposome dispersions, as determined by turbidity measurements.  相似文献

Sensitization of lanthanide ions is important for lanthanide ion-based assays and sensing. To the best of our knowledge, there are very few reports of lanthanide ion sensitization after it is incorporated into the liposome surface. This paper describes the syntheses of several saturated and polymerizable metal-chelating lipids based on chelidamic acid. The lipids are synthesized either from (S)-ornithine or racemic 2,3-diaminopropanoic acid. These lipids as well as polymerized liposomes incorporating these lipids sensitize lanthanide ions. Liposomes from the lipid 18-Eu(3+) provided a probe that relies not only on the emission wavelengths of Eu(3+) but also on a reproducible lifetime that can be used for protein identification.  相似文献

The radiolysis of synthetic (based on diallyldimethylammonium) and natural (based on the biopolymer chitosan) polycations as antimutagens was studied by means of the ESR technique. It was shown that the introduction of gallic acid units into a polymer structure leads to stabilization of the macromolecule toward radiation and a reduction in radical formation during γ-irradiation, a change that correlates with a considerable increase in the antimutagenic activity of the modified polycations. Approaches to the targeted synthesis of effective animutagens and optimization of their composition were developed.  相似文献
