Summary Condensation of cyano acid hydrazide1 with cyclopentanone in refluxing ethanolic piperidine yields hydrazone2. With mixtures of aliphatic aldehydes and different active methylene reagents,2 reacts to 1,2,4-triazolo[1,5-a]pyridines (8a–f). Compound2 also reacts with arylidenes9a–g to give triazolopyridines10a–g. Reaction of2 with aromatic aldehydes affords compounds13a–c. Diazotation of2 with aryldiazonium chloride in ethanol at 0 °C leads to the azo adducts15a–d. The thieno-1,2,4-triazolopyridine16 is obtained by reaction of8a with elementary sulfur.16 undergoes cycloaddition with -nitrostyrene, maleic anhydride, N-arylmalemide, and acrylonitrile yielding the isoquinolines21–24. All new compounds have been characterized by their IR,1H NMR, and mass spectra.
Ein neuer Syntheseweg für 1,2,4-Triazolo[1,5-a]pyridine und 1,2,4-Triazolo[1,5-a]isochinoline
Zusammenfassung Kondensation des Cyanohydrazids1 mit Cyclopentanon in ethanolischem Piperidin bei Rückflußtemperatur ergibt das Hydrazon2. Mit Gemischen aus aliphatischen Aldehyden und verschiedenen Verbindungen mit aktiven Methylengruppen reagiert2 zu 1,2,4-Triazolo[1,5-a]pyridinen (8a–f). Verbindung2 reagiert außerdem mit den Arylidenen9a–g zu den Triazolopyridinen10a–g. Umsetzung von2 mit aromatischen Aldehyden führt zu den Verbindungen13a–c. Diazotierung von2 mit Aryldiazoniumchloriden in Ethanol bei 0°C ergibt die Azoaddukte15a–d. Das Thieno-1,2,4-triazolylpyridin16 erhält man durch Reaktion von8a mit elementarem Schwefel.16 geht it -Nitrostyrol, Maleinsäureanhydrid, N-Arylmaleimid und Acrylnitril eine Cycloaddition zu den Isochinolinen21–24 ein. Alle neuen Verbindungen wurden durch ihre IR-,1H-NMR- und Massenspektren charakterisiert.

A series of novel N-anilino-2-substituted-5-methyl-[1,2,4]triazolo[1,5-a]pyrimidine-7-amine derivatives was prepared and evaluated for their in vitro antiproliferative activity against two cancer cell lines,Bel-7402 and HT-1080.Most of the compounds inhibited the cell proliferation at a low concentration.Seven compounds,VI5,VI7,VI10,and VI12―VI15,possessed marked antiproliferative activity superior to that of cisplatin.Of these seven initial hits,compound VI10 was the most active.  相似文献

6-Functionalized 7-R-4,7-dihydro[1,2,4]azolo[1,5-a]pyrimidines (R?=?H or Me) were synthesized by Biginelli-like reaction of formaldehyde or acetaldehyde, aminoazoles, and corresponding β-dicarbonyl precursor. Alkylation of the obtained compounds proceeds smoothly at position 4, while oxidation leads to 7-R-[1,2,4]azolo[1,5-a]pyrimidines formation. 6-Ethoxycarbonyl derivatives could be reduced to the corresponding alcohols by LiAlH4 and hydrolyzed to the acids.  相似文献

The condensation of malonoaldehyde derivatives with either a 3‐amino‐[1,2,4]‐triazole or a 3,5‐diamino‐[1,2,4]‐triazole precursor was studied. In agreement with previous reports, two different bicycles, namely, bearing the regioisomeric [1,2,4]triazolo[1,5‐a]pyrimidine ( 1 ) or[1,2,4] triazolo [4,3‐a]pyrimidine ( 2 ) structural surrogates, could be obtained. We found that, depending on the triazole precursor, only one regioisomer resulted, either of the 1 or 2 series. We also observed that these two structural surrogates could be unambiguously differentiated by indirectly measuring their 15N chemical shifts by 1H? 15N HMBC experiments. The occasional conversion of [1,2,4]triazolo[4,3‐a]pyrimidines to the [1,2,4]triazolo[1,5‐a]pyrimidine counterparts could be unequivocally determined by 15N NMR data. In our efforts to identify novel potent anticancer agents, we synthesized a series of 2,7-disubstituted triazolo[1,5-a]pyrimidines (6–16). Their antiproliferative activity against Bel-7402, HT-1080 and WI-38 cell lines was tested by MTT assay in vitro. Four of the compounds (911 and 16) displayed promising antiproliferative activity superior to gefitinib, especially compound 9. A preliminary SAR study of these derivatives was performed.  相似文献

An important building block, diethyl [(5-amino-4-cyano-3-methylsulfanyl-pyrazol-1-yl)–(4-fluorophenyl)methyl] phosphonate (3) was efficiently synthesized via the condensation of 1-hydrazino-1-(4-fluorophenyl)methyl phosphonate (1) with 2-[bis(methylthio)methylene]malononitrile (2).3 reacted with triethyl orthoformate to afford diethyl [(4-cyano-5-ethoxymethyleneamino-3-methylsulfanyl-pyrazol-1-yl)-(4-fluorophenyl)methyl] phosphonate (4), which reacted with various acyl hydrazines in refluxing 2-methoxyethanol to provide the target compounds (5) in good yields directly. The results of preliminary bioassay indicated that compounds 5 possess potent herbicidal activity against the roots of monocotyledonous (barnyard grass) and dicotyledonous (oil rape) plants, and could be further developed as potential herbicides.  相似文献

Hypercytokinemia, or cytokine storm, is one of the severe complications of viral and bacterial infections, involving the release of abnormal amounts of cytokines, resulting in a massive inflammatory response. Cytokine storm is associated with COVID-19 and sepsis high mortality rate by developing epithelial dysfunction and coagulopathy, leading to thromboembolism and multiple organ dysfunction syndrome. Anticoagulant therapy is an important tactic to prevent thrombosis in sepsis and COVID-19, but recent data show the incompatibility of modern direct oral anticoagulants and antiviral agents. It seems relevant to develop dual-action drugs with antiviral and anticoagulant properties. At the same time, it was shown that azolo[1,5-a]pyrimidines are heterocycles with a broad spectrum of antiviral activity. We have synthesized a new family of azolo[1,5-a]pyrimidines and their condensed polycyclic analogs by cyclocondensation reactions and direct CH-functionalization and studied their anticoagulant properties. Five compounds among 1,2,4-triazolo[1,5-a]pyrimidin-7-ones and 5-alkyl-1,3,4-thiadiazolo[3,2-a]purin-8-ones demonstrated higher anticoagulant activity than the reference drug, dabigatran etexilate. Antithrombin activity of most active compounds was confirmed using lipopolysaccharide (LPS)-treated blood to mimic the conditions of cytokine release syndrome. The studied compounds affected only the thrombin time value, reliably increasing it 6.5–15.2 times as compared to LPS-treated blood.  相似文献

A convenient and sustainable synthesis of pyrazolo[1,5-a]quinazolin-5(4H)-ones and [1,2,4]triazolo[1,5-a]quinazolin-5(4H)-one through copper-catalyzed cascade reactions of 2-bromobenzoates with 1H-pyrazol-5-amines or 1H-1,2,4-triazol-5-amine under ligand-free conditions in water is presented. It is notable that aqueous medium turned out to be crucial for the chemoselective formation of the title compounds. Compared with literature protocols, this new method showed advantages such as simple and sustainable procedure, commercially available starting materials, and convenient reuse of the reaction medium together with the copper catalyst.  相似文献

Twelve [1,2,4]-triazolo[1,5-a]pyrimidine derivatives had been designed and synthesized by using substituted hydrazine hydrate as starting materials.The synthesized compounds were confirmed by elemental analysis,Infrared spectra and 1~H NMR techniques.Preliminary bioassay indicated that some of them displayed good herbicidal activities.  相似文献

Ethyl 2-{2-[4-(2,3-dimethyl-5-oxo-1-phenyl-3-(pyrazolin-4-yl)]-2-cyano-1-(phenylamino)vinylthio}-acetate, 2-[4-(2,3-dimethyl-5-oxo-1-phenyl-(3-pyrazolin-4-yl))(1,3-thiazol-2-yl)]2-(4-oxo-3-phenyl-(1,3-thiazoilidin-2-ylidene))ethanenitrile, 2-[4-(2,3-dimethyl-5-oxo-1-phenyl(3-pyrazolin-4-yl))(1,3-thiazol-2-yl)]-2-(4-methyl-3-phenyl(1,3-thiazolin-2-ylidene))ethanenitrile, 2-(5-acetyl-4-methyl-3-phenyl(1,3-thiazolin-2-ylidene))-2-[4-(2,3-dimethyl-5-oxo-1-phenyl(3-pyrazolin-4-yl))(1,3-thiazol-2-yl)]ethanenitrile, and ethyl 2-(cyano(4-(2,3-dihydro-1,5-dimethyl-3-oxo-2-phenyl-1H-pyrazol-4-yl)thiazol-2-yl)methylene)-2,3-dihydro-4-methyl-3-phenylthiazole-5-carboxylate were synthesized by treatment of 2-(4-(2,3-dihydro-1,5-dimethyl-3-oxo-2-phenyl-1H-pyrazol-4-yl)thiazol-2-yl)-3-mercapto-3-(phenylamino)-acrylonitrile with appropriate halo ketones or halo esters. Also, 4-{2-[5,7-dimethyl-2-(phenylamino)(7a-hydropyrazolo[1,5-a]pyrimidin-3-yl](1,-thiazol-4-yl)}-2,3-dimethyl-1-phenyl-3-pyrazolin-5-one derivatives were synthesized via reaction of 4-{2-[5-amino-3-(phenylamino)pyrazolin-4-yl](1,3-thiazol-2-yl)}-2,3-dimethyl-1-phenyl-3-pyrazolin-5-one with β-diketone or β-keto ester. All synthesized compound were established by elemental analysis, spectral data, and alternative synthesis whenever possible.  相似文献

Reactions of 5-R-3-amino-1,2,4-triazoles with ethoxymethylideneacetylacetone and ethyl ethoxymethylideneacetoacetate proceeded regioselectively, giving 2-R-7-methyl[1,2,4]triazolo[2,3-a]-pyrimidines in good yields. The compounds obtained were characterized by elemental analysis, 1H NMR spectroscopy, and X-ray diffraction analysis (for ethyl 2-ethylthio-7-methyl[1,2,4]triazolo[2,3-a]pyrimidine-6-carboxylate). The frontier orbitals, the molecular electrostatic potential, and the geometries of the reagent molecules were calculated by the DFT method (B3LYP/6-31G**). A novel series of pyrazolo[1,5-a]pyrimidines 14a–j and pyrazolo[1,5-a]quinazolines 18a, b were synthesized via condensation of 5-amino-1H-pyrazoles 10a, b with 3-(dimethylamino)-1-aryl-prop-2-en-1-ones 11a–e and 2-((dimethylamino)methylene)-5,5-dimethylcyclohexane-1,3-dione (15), respectively, in glacial acetic acid. Finally, treatment of 10a, b with sodium nitrite (NaNO2) afforded pyrazolo[3,4-d]triazines 20a, b. Structures of compounds were confirmed by their spectral data. These compounds were screened for their in vitro cytotoxic activities against human cancer cell lines (HepG-2 and MCF-7) using 3-[4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. The results reveal that, the compounds 14b and 14h were the most potent in comparison with doxorubicin. The structure–activity relationship was discussed.  相似文献

The herbicidal activities of a series of 1,2,4-triacolo[l,5-a]pyrimidine-2-sulfonanilides containing various substituents on the benzene ring were quantitatively analyzed with physicochemical parameters by using Hansch-Fujita method. Variations in the activity were parabolically related to electronic parameters with the optimum pKa Value being about 6.93. The hydrophobic factor in addition to the electronic property seemed to have important effect on the activity.  相似文献

以2-苄硫基-5-甲基-7-羟基-1,2,4-三唑并[1,5-a]嘧啶为原料,经醚化、肼解、成盐、关环和席夫碱反应合成了20个新型的含1,2,4-三唑-5-硫酮席夫碱的1,2,4-三唑并[1,5-a]嘧啶类化合物,通过IR,1H NMR,MS和元素分析对所合成的化合物进行了结构表征.初步生物活性测试结果表明,部分化合物表现出一定的抑菌或较好的抗烟草花叶病毒(TMV)活性.在500μg/mL浓度下,化合物6b,6f和6p对TMV的抑制率分别为41%,43%和40%.  相似文献

A series of novel 6‐[(1,3,4‐thiadiazol‐2‐yl)sulfanyl]‐7‐phenylpyrazolo[1,5‐a]pyrimidines, 5‐phenyl‐6‐[(1,3,4‐thiadiazol‐2‐yl)sulfanyl]imidazo[1,2‐a]pyrimidines, and 2‐phenyl‐3‐[(1,3,4‐thiadiazol‐2‐yl)sulfanyl]pyrimido[1,2‐a]benzimidazoles have been synthesized in four steps starting with 2‐hydroxyacetophenone. The intermediate 3‐[(1,3,4‐thiadiazol‐2‐yl)sulfanyl]‐4H‐1‐benzopyran‐4‐ones reacted with pyrazol‐3‐amines, 5‐methylpyrazol‐3‐amine, and 1H‐imidazol‐2‐amine, 1H‐benzimidazol‐2‐amine via a cyclocondensation to give the title compounds in the presence of MeONa as base, respectively. The approach affords the target compounds in acceptable‐to‐good yields. The new compounds were characterized by their IR, NMR, and HR mass spectra.  相似文献

以5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-甲硫醚为起始原料, 设计合成了15个新型的5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-氧乙酰腙及10个(R)-5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-氧(α-甲基)乙酰腙类化合物, 通过元素分析、 MS和 1H NMR对所合成的化合物进行了结构表征. 初步生物活性测试结果表明, 部分化合物表现出不同程度的除草及杀菌活性. 目标化合物中引入手性中心有利于生物活性的提高.  相似文献
