A highly sensitive quantification method of aldosterone by liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) was investigated in a positive mode using recently developed picolinyl derivatization. Aldosterone was smoothly and quantitatively converted to the ethyl ether-picolinyl derivative by treatment with HCl-ethanol followed by the esterification with picolinic acid in the presence of 2-methyl-6-nitrobenzoic anhydride and 4-dimethylaminopyridine. The positive ion-ESI mass spectrum of the ethyl ether-picolinyl derivative was characterized by an appearance of protonated molecule ([M+H](+)) as a base peak. The ethyl ether-picolinyl derivatization gave a successful result in a separation of aldosterone from corticosterone, dehydrocorticosterone and cortexolone, and also provided an approximately 10-fold higher ESI response in the positive-LC-ESI-MS/MS (selected reaction monitoring; SRM) when compared to that of underivatized molecule (negative mode). The limit of quantification of aldosterone by SRM using ethyl ether-picolinyl derivatization (m/z 494-->m/z 448) was 1 pg/0.2 ml serum with accuracy and precision of 92.6% and 5.6%, respectively.  相似文献

Recently, promising fasciocidal activities of artesunate and artemether were described in rats and sheep. Therefore, a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed to quantify artesunate, artemether and their metabolites dihydroartemisinin and dihydroartemisinin-glucuronide in sheep plasma. Protein precipitation with methanol was used for sample workup. Reversed-phase high-performance liquid chromatography (HPLC) was performed using an Atlantis C18 analytical column with a mobile phase gradient system of ammonium formate and acetonitrile. The analytes were detected by MS/MS using selected reaction monitoring (SRM) with electrospray ionisation in the positive mode (transition m/z 267.4 → 163.0). The analytical range for dihydroartemisinin, dihydroartemisinin-glucuronide and artesunate was 10-1000 ng/ml and for artemether 90-3000 ng/ml with a lower limit of quantification of 10 and 90 ng/ml, respectively. Inter- and intra-day accuracy and precision deviations were < 10%. Consistent relative recoveries (60-80%) were observed over the investigated calibration range for all analytes. All analytes were stable in the autosampler for at least 30 h (6 °C) and after three freeze and thaw cycles. The validation results demonstrated that the LC-MS/MS method is precise, accurate and selective and can be used for the determination of the artemisinins in sheep plasma. The method was applied successfully to determine the pharmacokinetic parameters of artesunate and its metabolites in plasma of intramuscularly treated sheep.  相似文献

A sensitive, specific, and reliable liquid chromatography tandem mass spectrometry (LC-MS/MS) method was developed for detection and identification of zeranol in chicken or rabbit liver. A homogenized liver sample was hydrolyzed with beta-glucuronidase/arylsulfatase, and the hydrolysate was extracted with ethyl ether. The supernatant was evaporated to dryness, and the residue was dissolved in chloroform and re-extracted with sodium hydroxide. After acidification, the extract was cleaned up on a C18 solid-phase extraction cartridge and analyzed by electrospray LC-MS/MS in the negative ion mode. The multiple reaction monitoring transition from both m/z 321 to 277 and m/z 321 to 303 was monitored for confirmation, and the product ion of 277 was used for quantitation. Separation was performed on a Waters XTettra C18 column (50 x 2.1 mm, 3.5 microm) combined with a safeguard column (Symmetry C18, 20 x 3.9 mm, 5 microm), using a gradient elution with acetonitrile and 20 mM ammonium acetate. Calibration curves were prepared and good linearity was achieved over the concentration ranges tested. For all liver samples fortified at 3 different levels of 1, 5, and 50 microg/kg, the overall recoveries and relative standard deviations were in the range of 61-90 and 8-13%, respectively. The limit of quantitation based on the assay validation was 1 microg/kg. The method had been used on a routine basis for detection and identification of zeranol in liver samples.  相似文献

A multiresidue analysis method has been developed for the determination of pesticides in water by ultra-performance liquid chromatography (UPLC) combined with tandem mass spectrometry (MS/MS). The selected pesticides represent a broad range of polarity and volatility [benzoylcyclohexanedione (mesotrione and sulcotrione); chloroacetamide (acetochlor, alachlor, dimethenamide, and metolachlor); phenoxyacetic acid (2,4-D and MCPA); phenoxypropionic (dichloprop and mecoprop); phenylurea (chlortoluron, diuron, isoproturon, linuron, and metoxuron); sulfonylurea (foramsulfuron, iodosulfuron, and nicolsulfuron); triazine (atrazine, cyanazine, desethylatrazine (DEA), desisopropylatrazine (DIA), simazine, and terbutylazine)]. The analytes were extracted using solid-phase extraction (SPE). The separation was carried out on an acquity UPLC BEH C18 column (1.7 microm, 50 mm x 1 mm ID) using a gradient elution profile and mobile phase consisting of 0.1% formic acid in water and acetonitrile. The pesticides were detected with a tandem mass spectrometer after being ionised positively or negatively (depending on the molecule) using an electrospray ionisation (ESI) source. To achieve the suitable extraction conditions for sample preparation, several parameters affecting the efficiency of SPE such as the nature of the sorbent and the eluent, extractant volume and pH were studied. The best recovery was obtained by the extraction with an Oasis HLB cartridge and 3 mL of a solution of acetonitrile/dichloromethane (1:1, v/v) at pH 2. The average recoveries of the pesticides in different samples ranged from 82 to 109%. The weight least squares (WLS) linear regression was used to calculate the limits of detection and quantification (LOD and LOQ) because the dispersion was heteroskedastic. All the pesticides could be correctly quantified at a concentration level of 50 ng L(-1) and most of them could be detected at a concentration inferior or equal to 8 ng L(-1). Efficiency and robustness of this method were evaluated by the analysis of several samples of real natural water.  相似文献

A capillary liquid chromatography-electrospray tandem mass spectrometry (LC-ES-MS/MS) method was developed for the identification of metabolites of 4,4'-methylenedianiline (diaminodiphenyl methane, (DAPM)) derived from exposure of vascular smooth muscle cells (VSMC) to this compound. The use of precolumn concentration and column switching techniques prior to reversed-phase liquid chromatography coupled online to ES-MS enabled the separation and detection of low-level DAPM and its metabolites in the exposed cell samples. The employed LC-ES-MS method, and further LC-ES-MS/MS analysis enabled the structural assignments for two DAPM metabolites from vascular smooth muscle cells: N-acetyl methylenedianiline (N-acetyl-DAPM) and N,N'-diacetyl methylenedianiline (N,N'-diacetyl-DAPM).  相似文献

In this study, a new method for the determination of 12 nitroimidazoles and their hydroxymetabolites (metronidazole, hydroxymetronidazole, dimetridazole, ronidazole, hydroxydimetridazole, ipronidazole, hydroxyipronidazole, carnidazole, ornidazole, secnidazole, ternidazole, tinidazole) in beeswax has been developed and validated. The optimized sample preparation procedure included melting and dilution of beeswax in a mixture of n‐hexane and isopropanol followed by extraction with 2% acetic acid. The extracts were purified on strong cation exchange based solid‐phase extraction cartridges and evaporated in a vacuum system with vortex motion. The separation and detection of the nitroimidazoles in the beeswax extracts were achieved within 12 min by liquid chromatography tandem mass spectrometry using a pentafluorophenyl analytical column and applying a gradient elution with acetonitrile and 0.01% acetic acid as mobile phases. The method performance characteristics were evaluated at three concentration levels (1, 2, and 5 μg/kg) and the method was found to be suitable for determination of all tested nitroimidazoles. The limits of detection and quantification were 0.2–0.5 and 0.5–1 μg/kg, respectively. The recoveries varied from 71.2 to 104.9% while the relative standard deviations were less than 13.8% under the intermediate precision conditions.  相似文献

A method for the direct determination of paraquat, diquat, chlormequat and difenzoquat in water samples, using an on-line solid-phase extraction-liquid chromatography-tandem mass spectrometry system was developed. No sample preparation was required and the detection limits were below the European Union maximum residue levels. The chromatographic separation was performed using an XTera MS C8 column. The concentration of the ion pair reagent, the pH and the gradient elution were optimized to give high recoveries and good chromatographic resolution between quats. The detection was carried out using an ion trap as mass analyzer. Parameters such as the magnitude and duration of the resonant excitation voltage and the magnitude of the trapping RF voltage for full scan tandem mass spectrometry (MS-MS) experiments were studied to establish the optimal experimental conditions. Moreover, the accurate optimization of these parameters allowed MS-MS experiments of low mass ions, below m/z 200, providing unambiguous peak identification. Finally, the reproducibility of the proposed method was shown by good run-to-run and day-to-day precision values and its applicability to the determination of quats in drinking water was evaluated using spiked samples.  相似文献

A sensitive, simple, and specific liquid chromatographic method coupled with electrospray ionization-mass spectrometry (MS) is presented for the determination of sertraline in plasma. With zaleplon as the internal standard, sertraline is extracted from the alkalized plasma with cyclohexane. The organic layer is evaporated and the residue is redissolved in the mobile phase of methanol-10 mmol/L ammonium acetate solution-acetonitrile (62:28:10, v/v/v). An aliquot of 20 microL is chromatographically analyzed on a Shimadzu ODS C18 column (5 microm, 150- x 4.6-mm i.d.) by means of selected-ion monitoring mode of MS. The calibration curve of sertraline in plasma exhibits a linear range from 0.5 to 25.0 ng/mL with a correlation coefficient of 0.9998. The limit of quantitation is 0.5 ng/mL. The intra- and interday variations (relative standard deviation) are less than 7.8% and 9.5% (n = 5), respectively. The application of this method is demonstrated for the analysis of sertraline plasma samples in a human pharmacokinetic study.  相似文献

The measurement of catecholamines in human body fluids is requested frequently for the differential diagnosis and monitoring of pheochromocytoma. The methods in most clinical laboratories focus on high‐performance liquid chromatography coupled with electrochemical detection, which suffers from high background noise, low sensitivity, and poor separation. We reported and developed a robust high‐throughput liquid chromatography tandem mass spectrometry method in routine clinical laboratories for the measurement of urinary catecholamines for diagnosis of pheochromocytoma. The method was validated for consistent linearity, good recovery (88–112%), excellent stability and low carryover. Intra‐ and inter‐assay precision values for catecholamines were all below 3.35 and 4.83% respectively. Dilution linearity was investigated with satisfactory linearly dependent coefficients (r > 0.9988). The reference intervals were obtained from 310 results derived from patients in which the diagnosis of pheochromocytoma was excluded. This method was successfully used in our laboratory. The clinical characteristics of patients have been explored with satisfactory sensitivity and specificity. Therefore, we have developed a reliable assay for the liquid chromatography tandem mass spectrometry measurement of catecholamines in a routine clinical laboratory. The assay requires a small volume of urine, and all analytes are measured simultaneously. The assay is rapid and reliable to be executed, offering the potential for routine clinical laboratories.  相似文献

A rapid method for detection and quantification of metabolites of specific olive oil phenolic compounds (hydroxytyrosol monoglucuronide, hydroxytyrosol monosulfate, tyrosol glucuronide, tyrosol sulfate and homovanillic acid sulfate) in low-density lipoprotein (LDL) fractions by solid-phase extraction (SPE) and high-performance liquid chromatography coupled to electrospray ionization tandem mass spectrometry (HPLC/ESI-MS/MS) is described. A 3 microm particle size fast C18 Luna column, 5 cm x 2.0 mm I.D., was used at a flow rate of 0.6 mL/min with a mobile phase consisting of 0.1% (v/v) formic acid (A) and acetonitrile (B). A linear gradient profile was used for separation at column temperature 40 degrees C. The proposed chromatographic procedure is rapid without loosing its separation efficiency and sensitivity. Validation proofs were carried out for the method described, showing a linear system (r>0.99) and a recovery of 81.9 and 101.3% for hydroxytyrosol and homovanillic acid, respectively. The results show that this method is effective and can be used in routine analysis.  相似文献

Tetrodotoxin (TTX) exhibits the therapeutic potential in blocking pain and in low doses can safely relieve severe pain. The urinary excretion profiles of TTX in humans have not been reported due to the extremely low lethal dose. In this study, a rapid and specific method based on protein precipitation coupled to liquid chromatography tandem mass spectrometry was developed to determine the level of TTX in human urine samples. 11-Deoxytetrodotoxin was used as an internal standard (IS). Multiple reaction monitoring mode was used for quantification using target fragment ions m/z 320.0 → 162.1 for TTX and m/z 304.0 → 176.0 for 11-deoxyTTX. The separation of analytes was achieved on a hydrophilic interaction liquid chromatography column (250 × 4.6 mm, 5.0 μm). The mobile phase consisted of 5 mM ammonium formate in water (pH = 4.50) and 5 mM ammonium formate in acetonitrile (pH = 4.50). The flow rate was set at 0.80 mL/min in a gradient condition. Calibration plots were linear throughout the range 0.986–98.6 ng/mL of TTX in human urine. The intra-assay accuracies and precisions were within the acceptable range. The method was successfully applied to a urinary excretion study after intravenous administration of TTX to healthy volunteers. The developed method will be helpful for future pharmacological studies of TTX.  相似文献

A rapid and sensitive analytical method based on liquid chromatography coupled to tandem mass spectrometry detection with positive ion electrospray ionization was developed for the determination of febuxostat in human plasma using d7‐febuxostat as the internal standard (IS). A simple protein precipitation was performed using acetonitrile. The analyte and IS were subjected to chromatographic analysis on a Capcell PAK C18 column (4.6 × 100 mm, 5 µm) using acetonitrile–5 mm ammonium acetate–formic acid (85:15:0.015, v/v/v) as the mobile phase at a flow rate of 0.6 mL/min. An Agilent 6460 electrospray tandem mass spectrometer was operated in the multiple reaction monitoring mode. The precursor‐to‐product ion transitions m/z 317 → m/z 261 (febuxsotat) and m/z 324 → m/z (261 + 262) (d7‐febuxostat, IS) were used for quantitation. The results were linear over the studied range (10.0–5000 ng/mL), and the total analysis time for each chromatograph was 3 min. The intra‐ and inter‐day precisions were less than 7.9 and 7.2%, respectively, and the accuracy was within ±4.2%. No evidence of analyte instability in human plasma was observed storage at ?20°C for 31 days. A system was developed for the separation of sulfophthalimide (SPI), sulfophthalamide (SPAM), sulfophthalamic acid (SPAA) and sulfophthalic acid (SPA) by ion-pair liquid chromatography and their detection by electrospray ionization tandem mass spectrometry (ESI-MS-MS). Except for SPAM, the 3- and 4-sulfo-isomers of the analytes were separated by HPLC using volatile tributylamine as ion-pairing agent. Initial fragmentations of the analytes in the negative mode involve losses of CO2 or HNCO or condensation via H2O or NH3 elimination. ortho-Effects of the sulfonate group were recognized in the fragmentation of the respective 3-sulfo-isomers and allowed us to assign the order of elution of the SPAA isomers. Quantitative analysis of these sulfonated aromatic compounds with MRM detection was elaborated and resulted in detection limits ranging from 1 pg for SPA to 13 pg for SPAA isomers and in limits of quantification of 2-10 microg/L for 5 microL volumes of injected tap water, municipal wastewater or industrial effluents up to salt contents of 0.5-1 g/L. The method was applied to study the isomer-specific chemical and microbial transformations of SPI, which was previously shown to be formed by white-rot fungi from sulfophthalocyanine textile dyes.  相似文献

We have developed a simple method for extracting and purifying (+)-abscisic acid (ABA) and eight ABA metabolites - phaseic acid (PA), dihydrophaseic acid (DPA), neophaseic acid (neoPA), ABA-glucose ester (ABAGE), 7′-hydroxy-ABA (7′-OH-ABA), 9′-hydroxy-ABA (9′-OH-ABA), ABAaldehyde, and ABAalcohol - before analysis by a novel technique for these substances, ultra-performance liquid chromatography-electrospray ionisation tandem mass spectrometry (UPLC-ESI-MS/MS). The procedure includes addition of deuterium-labelled standards, extraction with methanol-water-acetic acid (10:89:1, v/v), simple purification by Oasis® HLB cartridges, rapid chromatographic separation by UPLC, and sensitive, accurate quantification by MS/MS in multiple reaction monitoring modes. The detection limits of the technique ranged between 0.1 and 1 pmol for ABAGE and ABA acids in negative ion mode, and 0.01-0.50 pmol for ABAGE, ABAaldehyde, ABAalcohol and the methylated acids in positive ion mode. The fast liquid chromatographic separation and analysis of ABA and its eight measured derivatives by UPLC-ESI-MS/MS provide rapid, accurate and robust quantification of most of the substances, and the low detection limits allow small amounts of tissue (1-5 mg) to be used in quantitative analysis. To demonstrate the potential of the technique, we isolated ABA and its metabolites from control and water-stressed tobacco leaf tissues then analysed them by UPLC-ESI-MS/MS. Only ABA, PA, DPA, neoPA, and ABAGE were detected in the samples. PA was the most abundant analyte (ca. 1000 pmol/g f.w.) in both the control and water-stressed tissues, followed by ABAGE and DPA, which were both present at levels ca. 5-fold lower. ABA levels were at least 100-fold lower than PA concentrations, but they increased following the water stress treatment, while ABAGE, PA, and DPA levels decreased. Overall, the technique offers substantial improvements over previously described methods, enabling the detailed, direct study of diverse ABA metabolites in small amounts of plant tissue.  相似文献

Currently, there is growing interest in the degradation pathways of organic contaminants such as pesticides. In the case of pesticides, the determination of metabolites in agricultural products and environment is necessary as some of them could present similar toxicity to or even higher toxicity than the parent compound. The development of analytical methodology for the identification and quantification of carbendazim fungicide and its metabolites in cucumber was studied. Cucumber (cucumis sativus) is a global food in terms of economic importance and nutritional quality. Careful optimisation of the liquid chromatography–mass spectrometry (LC-MS)/MS parameters was achieved in order to attain a fast separation with the best sensitivity. The detection was carried out on an Ion-Trap tandem mass spectrometer (MS/MS) by electrospray ionisation in positive ion mode (ESI+) with multiple reaction monitoring (MRM).  相似文献

A liquid chromatographic-tandem mass spectrometric multiresidue method for the simultaneous quantitative determination of the tetrahydroimidazole, levamisole and the benzimidazoles thiabendazole, oxfendazole, oxibendazole, albendazole, fenbendazole, febantel and triclabendazole in milk has been developed and validated. The anthelmintic residues were extracted with ethyl acetate. The liquid chromatographic separation was performed on a reversed-phase C18 column with gradient elution. The analytes were detected by tandem quadrupole mass spectrometry after positive electrospray ionisation by multiple reaction monitoring. The confirmatory method is very sensitive and each component can be detected at a residue level lower than 1 microgram/l. The method is validated according to the revised European Union requirements and all parameters were found conform the criteria. The evaluated parameters were linearity, specificity, stability, recovery, precision (repeatability and within-laboratory reproducibility) and analytical limits (detection limit, decision limit and detection capability). This analytical method is applied in the Belgian monitoring programme for classical anthelmintic veterinary drugs in raw farm cow's milk.  相似文献

Trace amounts of diuretics were determined in human urine by hollow fiber liquid-phase microextraction (LPME) combined with liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS) in this study. Chromatography was performed on a C(8) reversed-phase column. A 25 microL n-octanol was used to extract analytes in urine. Extraction was optimized using a pH 2 solution spiked with 0.15 g/mL NaCl for 40 min at 40 degrees C with 1010 rpm stirring. The limits of detection of diuretics in urine were 0.3-6.8 ng/mL, and linearity range was 1-1000 ng/mL. Recoveries of spiked 50 ng/mL diuretics were 97.7-102.5%. The intra-day precision and inter-day precision were 3-18% and 4-21%, respectively. The diuretics concentration profiles in patient urine were also determined. The results of this study reveal the adequacy of LPME-LC-MS/MS method for analyzing diuretics in urine and quantification limits exceed World Anti-Doping Agency requirements.  相似文献

In the present study we developed and validated a liquid chromatography/tandem mass spectrometry (LC‐MS/MS) assay for the determination of flucloxacillin in human plasma and microdialysis samples and cloxacillin in microdialysis samples, using oxacillin as the internal standard for the assay. The samples were separated on a UPLC BEH C18,1.7 µm column (2.1 × 50 mm) and analyzed by a tandem–quadrupole mass spectrometer in multiple reaction monitoring mode using an electronspray ionization interface. For flucloxacillin the method was demonstrated to be accurate and precise in the linearity range of 1–30 mg/L in plasma and 0.05–5.0 mg/L for microdialysate with a regression coefficient (r) of 0.9986 and 0.9989 in plasma and microdialysate respectively. For cloxacillin it was accurate and precise in the range of 0.1–5.0 mg/L for microdialysate with a regression coefficient of 0.9972. The method presents a high sensitivity for flucloxacillin (lower limit of quantification of 1 mg/L for plasma and 0.05 mg/L for microdialysis samples) combined with a low within‐ and between‐day variation (<5.0% for flucloxacillin and cloxacillin in microdialysis samples and <6.5% for plasma samples of flucloxacillin). The validation experiments for the microdialysis probes showed a relative recovery of 85.5% for flucloxacillin at a flow rate of 1.0 μL/min. The results justify the use of this assay for clinical studies for measuring free unbound tissue concentrations of flucloxacillin in patients with a Staphylococcus aureus bacteremia.
