The copper(I)-catalyzed carboxylation reaction of aryl- and alkenylboronic esters proceeded smoothly under CO(2) to give the corresponding carboxylic acid in good yield. This reaction showed wide generality with higher functional group tolerance compared to the corresponding Rh(I)-catalyzed reaction.  相似文献

An enzymatic reaction has been monitored by on-line direct infusion electrospray ionization (tandem) mass spectrometry. Using this fast and sensitive technique, a key and transient intermediate of Mycobacterium tuberculosis indole-3-glycerol phosphate synthase (IGPS)-catalyzed reaction has been trapped. The reaction catalyzed by indole-3-glycerol phosphate synthase is part of the tryptophan biosynthetic pathway, and is not present in mammals, including humans. This peculiarity renders this enzyme a potential target for the development of biospecific agents with potential anti-TB activity. The present results indicate the presence of two intermediates in the mechanism of this enzymatic reaction.  相似文献

The CuI-catalyzed domino reaction between 1-(2-bromophenyl)methanamines and amidines using K3PO4 as the base, pivalic acid as the additive, and aerial oxygen as the oxidant gives access to substituted quinazolines in a single step with yields in the range between 43 and 90%. It is assumed that the reaction proceeds as a Cu(I)-catalyzed intermolecular N-arylation followed by an intramolecular nucleophilic substitution and a Cu(II)-catalyzed oxidation. The amidines can be replaced with imidates and the reaction can also be run with 1-(2-iodophenyl)methanamine.  相似文献

We describe the synthesis of cyclam metal complexes derivatized with amino acids or a tripeptide using a copper(I)-catalyzed Huisgen "click" reaction. The linker triazole formed during the synthesis plays an active coordinating role in the complexes. The reaction conditions do not racemize the amino acid stereocenters. However, a methylene group adjacent to the triazole is susceptible to H/D exchange under ambient conditions, an observation which has potentially important implications for structures involving stereocenters adjacent to triazoles in click-derived structures. The successful incorporation of several amino acids is described, including reactive tryptophan and cysteine side chains. All complexes are formed rapidly upon introduction of the relevant metal salt, including synthetically convenient cases where trifluoroacetate salts of cyclam derivatives are used directly in the metalation. None of the metal complexes displayed any cytotoxicity to mammalian cells, suggesting that the attachment of such complexes to amino acids and peptides does not induce toxicity, further supporting their potential suitability for labeling/imaging studies. One Cu(II)-cyclam-triazole-cysteine disulfide complex displayed moderate activity against MCF-10A breast nontumorigenic epithelial cells.  相似文献

Several 2-imido substituted furans were found to undergo a rapid intramolecular [4 + 2]-cycloaddition to deliver oxabicyclo adducts in good to excellent yields. By using a Rh(I)-catalyzed ring opening of the resulting oxabicyclic adduct, it was possible to prepare several highly functionalized tetrahydro-1H-indol-2(3H)-one derivatives which were then used to prepare several erythrina alkaloids. By taking advantage of the Rh(I)-catalyzed reaction, it was possible to convert tert-butyl 3-oxo-5-carbomethoxy-10-oxa-2-azatricyclo[5.2.1.0(1,5)]dec-8-ene-2-carboxylate into the ring opened boronate by reaction with phenylboronic acid. Treatment of the boronate with pinacol/acetic acid afforded the corresponding diol which was used in a successful synthesis of racemic 3-demethoxyerythratidinone. During the course of these studies, several novel rearrangement reactions were encountered while attempting to induce an acid-initiated Pictet Spengler cyclization of a key lactam intermediate. The IMDAF/Rh(I)-catalyzed ring opening cascade sequence was also applied to the total synthesis of (+/-)-erysotramidine as well as the lycorine type alkaloid (+/-)-epi-zephyranthine.  相似文献

An efficient Bi(OTf)3-catalyzed synthesis of 3-alkenyl-2-pyrrolidine-2H-indazoles from triazenylaryl allylic alcohols via the intramolecular direct amination process is reported. Compared with the dodecyl benzenesulfonic acid (DBSA)-catalyzed method, the new method is more efficient and gives greater yields and functionality tolerance. Additionally, the 3-alkenyl-2-pyrrolidine-2H-indazoles can be transformed to a series of new products under different reaction conditions.  相似文献

A conjugation of bile acids with peptides via Cu(I)-catalyzed click chemistry has been described. Novel bile acid-peptide conjugates linked via a 1,2,3-triazole moiety based on cholic, deoxycholic and lithocholic acid derivatives were synthesized using Cu(I)-catalyzed 1,3-dipolar cycloaddition ("click" reaction). It was shown that up to three peptide fragments can be attached to a central steroid core, thus forming complex three-dimensional polyconjugate structures, which can find important applications in biochemistry, medicinal chemistry, and coordination chemistry.  相似文献

An efficient strategy for the synthesis of a variety of 3-substituted isocoumarins has been developed. The reaction proceeded from o-halobenzoic acids and 1,3-diketones via a copper(I)-catalyzed domino reaction in DMF under the action of K(3)PO(4) at 90-120 °C without a ligand to afford the corresponding 3-substituted isocoumarin derivatives in good to excellent yields. o-Halobenzoic acids could be o-iodobenzoic acid, o-bromobenzoic acid, and o-chlorobenzoic acid derivatives. 1,3-Diketones could be alkyl- and aryl-substituted 1,3-diketones.  相似文献

Kinetics of the hydroxylaminolysis of acetamide, glycinamide, glycylglycine and triglycine have been studied in the range of temperatures 37-60 degrees C as a function of pH and hydroxylamine concentration. Rate constants for specific acid, general-acid and general-base catalyzed pathways have been determined for all substrates (for glycine derivatives rate constants for different protonation forms were obtained). Testing different metal ions as possible reaction catalysts revealed a significant catalytic effect of Zn(II) on the hydroxylaminolysis of glycine substrates, but not acetamide. On the basis of the kinetic results, a mechanism of Zn(II) catalysis is proposed, which involves the coordination of the metal ion to the alpha-amino group of the substrate and the base-assisted nucleophilic attack of hydroxylamine on the bound substrate. The product analysis by proton NMR shows that the primary reaction product in the catalytic reaction is glycine hydroxamic acid, which undergoes further Zn(II)-catalyzed hydrolysis to glycine. Thus the final result of the Zn(II)-catalyzed treatment of peptides by hydroxylamine is hydrolytic cleavage.  相似文献

研究了在有机介质叔丁醇中魔芋葡甘聚糖(KGM)的分子量及酰基供体对固定化脂肪酶Novozym 435催化KGM乙酰化反应的影响.KGM的分子量对酶促其酰化反应的活性及产物取代度有显著影响.随着KGM分子量的增大,酶催化反应的速率逐渐下降,产物的取代度逐渐减小.KGM分子量对该反应的影响与不同分子量KGM的溶解度、体系粘度、空间位阻及颗粒形态等因素有关.以不同链长的脂肪酸乙烯酯为酰基供体时,随着酰基供体中脂肪酸碳链的增长,酶促KGM酰化反应速率逐渐下降,产物的取代度逐渐减小,且该酰化反应具有高度的区域选择性,反应均发生在C6-OH上.  相似文献

Abstract— The photochemical interactions between tryptophan and nucleic acid bases were studied. When aqueous solutions of tryptophan were irradiated (Λ > 260 nm) at neutral pH in the presence of each of the nucleic acid bases, pyrimidine bases but not purine bases were altered. Air was found to decrease the rate of reaction. Two classes of photoproducts were isolated by thin layer and ion-exchange chromatography and partially characterized. One was the dihydro-pyrimidine forms of the base (see Reeve and Hopkins, 1979) and the other consisted of tryptophan-pyrimidine photoadducts. Four tryptophan-uracil and two tryptophan-thymine adducts each with a 1:1 molecular stochiometry were found. Spectroscopic measurements and a positive reaction with Ehrlich's reagent indicate that the indole nucleus remained intact, but that the pyrimidine base was reduced at the 5, 6 double bond. The absence of a positive ninhydrin reaction and the effect of pH on the quantum yield of the photoadduct formation indicated that the ionized a-amino acid group of tryptophan was involved in photoadduct formation. Indole derivatives lacking an a-amino group were also found to form photoadducts with pyrimidine bases. The experimental results are consistent with a reaction mechanism involving tryptophan excitation to the first excited singlet state as the initial event. Radical scavenging experiments indicate that tryptophan free radicals, formed by electron dissociation from the excited state, react with the ground state pyrimidine.  相似文献

The cycloaddition of azides to alkynes is one of the most important synthetic routes to 1H-[1,2,3]-triazoles. Here a novel regiospecific copper(I)-catalyzed 1,3-dipolar cycloaddition of terminal alkynes to azides on solid-phase is reported. Primary, secondary, and tertiary alkyl azides, aryl azides, and an azido sugar were used successfully in the copper(I)-catalyzed cycloaddition producing diversely 1,4-substituted [1,2,3]-triazoles in peptide backbones or side chains. The reaction conditions were fully compatible with solid-phase peptide synthesis on polar supports. The copper(I) catalysis is mild and efficient (>95% conversion and purity in most cases) and furthermore, the X-ray structure of 2-azido-2-methylpropanoic acid has been solved, to yield structural information on the 1,3-dipoles entering the reaction. Novel Fmoc-protected amino azides derived from Fmoc-amino alcohols were prepared by the Mitsunobu reaction.  相似文献

Singlet molecular oxygen, O(2)(a(1)Δ(g)), can influence many processes pertinent to the function of biological systems, including events that result in cell death. Many of these processes involve a reaction between singlet oxygen and a given amino acid in a protein. As a result, the behavior of that protein can change, either because of a structural alteration and/or a direct modification of an active site. Surprisingly, however, little is known about rate constants for reactions between singlet oxygen and amino acids when the latter are in a protein. In this report, we demonstrate using five separate proteins, each containing only a single tryptophan residue, that the rate constant for singlet oxygen reaction with tryptophan depends significantly on the position of this amino acid in the protein. Most importantly, the reaction rate constant depends not only on the accessibility of the tryptophan residue to oxygen, but also on factors that characterize the local molecular environment of the tryptophan in the protein. The fact that the local protein environment can either appreciably inhibit or accelerate the reaction of singlet oxygen with a given amino acid can have significant ramifications for singlet-oxygen-mediated events that perturb cell function.  相似文献

Density functional theory studies have been carried out to investigate the mechanism of the Pd(II)(bpy)- and Rh(I)(bpy)-catalyzed conjugate additions and their competitive Heck reactions involving α,β-unsaturated carbonyl compounds. The critical steps of the mechanism are insertion and termination. The insertion step favors 1,2-addition of the vinyl-coordinated species to generate a stable C-bound enolate intermediate, which then may isomerize to either an oxa-π-allyl species or an O-bound enolate. The termination step involves a competition between β-hydride elimination, leading to a Heck reaction product, and protonolysis reaction that gives a conjugate addition product. These two pathways are competitive in the Pd(II)-catalyzed reaction, while a preference for protonolysis has been found in the Rh(I)-catalyzed reaction. The calculations are in good agreement with the experimental observations. The potential energy surface and the rate-determining step of the β-hydride elimination are similar for both Pd(II)- and Rh(I)-catalyzed processes. The rate-determining steps of the Pd(II)- and Rh(I)-catalyzed protonolysis are different. Introduction of an N- or P-ligand significantly stabilizes the protonolysis transition state via the O-bound enolate or oxa-π-allyl complex intermediate, resulting in a reduced free energy of activation. However, the barrier of the β-hydride elimination is less sensitive to ligands. For the Rh(I)-catalyzed reaction, protonolysis is calculated to be more favorable than the β-hydride elimination for all investigated N and P ligands due to the significant ligand stabilization to the protonolysis transition state. For the Pd(II)-catalyzed reaction, the complex with monodentate pyridine ligands prefers the Heck-type product through β-hydride elimination, while the complex with bidentate N and P ligands favors the protonolysis. The theoretical finding suggests the possibility to control the selectivity between the conjugate addition and the Heck reaction by using proper ligands.  相似文献

[reaction: see text] The catalytic dihydroxylation of olefins represents a unique synthetic tool for the generation of two C,O-bonds with defined relative configuration. Whereas OsO(4) has been established as a very general dihydroxylation catalyst within the past 30 years, the less expensive and toxic isoelectronic RuO(4) has found only limited use for this type of oxygen-transfer reaction. High catalyst loading and undesired side reactions were severe drawbacks in RuO(4)-catalyzed oxidations of C,C-double bonds. Recently, we were able to improve the RuO(4)-catalyzed dihydroxylation by addition of Bronsted acids to the reaction mixture. This protocol proved to be of general applicability, however, certain limitations were observed. To address these problematic functional groups a new Lewis acid accelerated oxidation was developed. The use of only 10 mol % of CeCl(3) allowed a further decrease in the catalyst concentration down to 0.25 mol % while broadening the scope of the reaction. Silyl ethers and nitrogen containing functional groups are now tolerated in this optimized protocol. Furthermore, competing scission reactions are supressed in the presence of Lewis acid allowing longer reaction times and the successful oxidation of electron-deficient tetrasubstituted double bonds that cannot be oxidized using known dihydroxylation protocols.  相似文献
