The stereochemistry at C-3 of the known compounds atomaric acid 2a, 5′a-desmethyl-5′-acetylatomaric acid 4a, and stypoquinonic acid 5a is revised to 2, 4, and 5 on the basis of a careful study of 2D NOESY experiments and also from comparison of their 1H and 13C chemical shifts with those of the related metabolites 6 and 7 isolated from Stypopodium zonale. Compound 7 is a novel unusually functionalized 1-keto-5′a-desmethyl atomaric acid derivative whose structure and stereochemistry were determined by spectroscopic means.  相似文献

Two novel briaranoidal derivatives, designated as briaexcavatins A (1) and B (2), along with two known metabolites, excavatolides B and C (3 and 4), were isolated from the octocoral Briareum excavatum. Both compounds 1 and 2 possess unprecedented 5,6-epoxy moiety in the 10-membered ring. Diterpenoid 1 possesses a novel pentacyclic skeleton with a ε-lactone. The structures of diterpenoids 1 and 2 were elucidated by interpretation of spectral data and the absolute stereochemistry was established by application of modified Mosher’s method on 3.  相似文献

Three dimeric γ-lactones (1-3), one dihydronaphthalene-2,6-dione (4), one hexahydroindenofuran (5), one cyclopentanone (6), and one lasiodiplodin (7) were isolated from the endophytic fungi Botryosphaeria rhodina PSU-M35 and PSU-M114 along with twelve known metabolites. The structures and the proposed stereochemistry of the new metabolites were established by spectral data analysis. The isolated compounds were submitted for evaluation of the antibacterial activity against Staphylococcus aureus, both standard and methicillin-resistant strains.  相似文献

Two novel 3,4-seco-25,26,27-trinorlanostane triterpeniod compounds, fornicatins A and B (1 and 2) have been isolated from the fruiting body of G. fornicatum. The structural elucidation of 1 and 2 were accomplished by extensive NMR analysis. The relative stereochemistry of 2 was established by single crystal X-ray crystallography, which also confirmed the novel carbon skeleton of the new triterpenoid. An ether linkage of C-4 with C-7 in 1 is unprecedented in natural triterpenoids. Both compounds were tested for their inhibitory effects on rabbit platelet aggregation induced by PAF, ADP, or AA.  相似文献

Three new brominated diterpenes (1, 2, 8), along with six previously reported metabolites (3-7, 9), were isolated from the organic extract of Sphaerococcus coronopifolius, collected in Palaiokastritsa bay at the west coasts of Corfu Island. The structures of the new natural products, as well as their relative stereochemistry, were established by means of spectral data analyses, including 2D experiments. The absolute stereochemistry of 2 and 4 as well as the structure revision of the previously reported metabolite 3 were established by X-ray crystallographic analyses. The cytotoxicity of the isolated metabolites was evaluated against the NSCLC-N6-L16 and A549 human lung cancer cell lines.  相似文献

Four new natural products (1-4), along with three previously reported metabolites (5-7), all belonging to the pregnane class of steroids were isolated from the organic extract of the gorgonian Eunicella cavolini. The structures and relative configurations of the isolated compounds were determined through NMR spectroscopic techniques and chemical interconversions, while their absolute stereochemistry was determined using the modified Mosher's method. Activity evaluation of 1-7 towards MCF-7 human breast cancer cells showed partial growth inhibitory effects for all metabolites bearing an 11α-acetoxy moiety.  相似文献

Three new cuparene sesquiterpenes 1-3 were isolated from the organic extract of the red alga Laurencia microcladia, collected at Chios island in the North Aegean Sea, Greece. The structures and the relative stereochemistry of the compounds are proposed on the basis of their spectral data. Metabolite 2 shows an unprecedented (for the cuparene class of sesquiterpenes) migration of the C-1 methyl group. All metabolites 1-3 were found to exhibit significant cytotoxic activity against two lung cancer cell lines.  相似文献

Three new sesquiterpenes (1-3), along with five known (5-9), were isolated from the organic extract of the red alga Laurencia obtusa, collected from the coastal rocks of Serifos in the Aegean Sea. A new dimeric sesquiterpene of the cyclolaurane-type (4) along with four previously reported (7, 10-12) metabolites, were isolated from the extract of Laurencia microcladia, collected at Chios island in the North Aegean Sea. The structures and the relative stereochemistry of the compounds are proposed on the basis of their spectral data. Five new unsaturated 2-amino-3-alcohols, crucigasterins A-E (2-6), were isolated together with known related compound 7 from the Mediterranean ascidian Pseudodistoma crucigaster and characterised as diacetyl derivatives (2a-6a) by spectroscopic methods. The threo-relative configuration of the amino alcohol portion was inferred by NOE analysis of the oxazolidinone derivative of crucigasterin A (2) as well as by 13C NMR comparison with synthetic threo and erythro model compounds. The co-occurring metabolites were assumed to have the same relative configuration as 2 by comparison of the diagnostic carbon value of C-1. The absolute stereochemistry of compound 7 that had not been previously reported was determined by applying the modified Mosher’s method on the corresponding N-acetyl derivative. The same absolute configuration was suggested for the other co-occurring crucigasterins by biogenetic considerations. Antibacterial and antifungal activities of selected crucigasterins were also evaluated.  相似文献

New degraded and rearranged diterpenoids, 5-8, have been isolated from the Antarctic sponge Dendrilla membranosa. The structure and relative stereochemistry of these compounds were determined by spectroscopic data. The absolute stereochemistry of 5 was determined by spectroscopic data using a chiral reagent. Conformational studies allowed us to assign also the absolute stereochemistry of 6-8, as well as those previously isolated spongian-derived metabolites with known relative stereochemistries.  相似文献

A bioassay-guided purification of the methanolic extract of Withania somnifera fruits yielded novel withanamides A-I (1-9) and withanolides (10-13). Among the withanolides, compound 10 is novel. The structures of these compounds were determined by using FABMS, HRFABMS, 1D and 2D NMR spectral and chemical methods. The withanamides possess novel chemical structures and consisted of serotonin, glucose and long-chain hydroxyl fatty acid moieties. The stereochemistry of the hydroxyl group in the long-chain fatty acid moiety in compound 1 was determined by the modified Mosher's ester method. Compounds 1-13 were tested for their ability to inhibit lipid peroxidation in a model system using large unilamellar vesicles. Withanamides 1-5 and 9 inhibited lipid peroxidation by 98, 93, 79, 94, 81 and 86%, respectively, at 1 μg/mL. However, compounds 6-8 inhibited the lipid peroxidation by 85, 82 and 90%, respectively, at 0.5 μg/mL. Withanolides 10-13 were also tested and only compound 12 inhibited the lipid peroxidation by 82% at 10 μg/mL. To evaluate the structure activity relationships of withanamides A-I, compounds 14-16 were purchased and their lipid peroxidation activity determined as in the case of compounds 1-9. Commercial antioxidants, butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and tert-butylhydroquinone (TBHQ), were also tested in this assay at 1 μg/mL and showed 80, 81 and 85% of inhibition, respectively. Our results suggest that the potent antioxidant activity exhibited by novel withanamides is probably due to the hydroxylated long-chain acyl group. This is the first report of withanamides, unique serotonin conjugates, from W. somnifera fruits.  相似文献

Two novel cyclopropyl diketones, hamavellone A (1) and B (2), and two new 14-membered nonaketide macrolactones, hamigeromycin A (3) and B (4), together with six known compounds, 89-250904-F1 (radicicol analogue A, 5), pseurotin A (6), emodin (7), ω-hydroxyemodin (8), and emodin bianthrones (9 and 10) were isolated from the soil fungus Hamigera avellanea BCC 17816. The structures of the new compounds were defined by analysis of NMR and MS data. The absolute stereochemistry of 3 was addressed by chemical correlation to 5. Hamavellone B (2) exhibited antimalarial activity with an IC50 of 5.2 μg/mL, whereas it also showed comparable cytotoxicity.  相似文献

An investigation was conducted to identify the structures and bioactive properties of five compounds isolated from the Halichondrida sponges Amorphinopsis foetida and Axinyssa aplysinoides. All compounds possessed the spiro-axane sesquiterpene core and all were substituted at C-2 with nitrogen containing functionality. The stereochemistry of one known compound has been revised to (2R,5R,10S)-2-formamido-6-axene (3). It exhibited mild selective solid tumor and mild antibacterial activity and was found from Axinyssa. A second known substance whose stereochemistry has also been revised, (2R,5R,10S)-2-isothiocyanato-6-axene (4) plus its undescribed diastereomer (5) were isolated from Amorphinopsis. Both sponges were the source of two new N-phenethyl-2-formamido-6-axene diastereomeric compounds 6 and 7. No solid tumor or antibacterial activity was found for 4-7.  相似文献

Chemical investigations on the acetone extract of the soft coral Lobophytum durum have afforded five new cembranoids with a trans-fused α-methylene-γ-lactone, durumolides A-E (1, 6, and 8-10), and five previously characterized cembrane-based diterpenoids (2-5 and 7). The structures of the isolated metabolites were elucidated through extensive spectroscopic analyses, while the relative stereochemistry of 4 was confirmed by X-ray diffraction analyses. Moreover, the absolute configurations of 3-5 and 8 were established by application of modified Mosher's method. The anti-inflammatory effects, antibacterial activities, and inhibition assay of HCMV (Human cytomegalovirus) endonuclease activity of these isolated metabolites 1-10 were evaluated in vitro.  相似文献

Chemical investigation on the gorgonian coral Briareum sp. has led to the isolation of six oxygenated briaran diterpenes 1-6, including three new compounds briarenolides A-C (1-3). The structures of 1-3 were determined on the basis of extensive spectroscopic analysis and by comparison of their spectral data with those of related metabolites. Among these metabolites, 1 and 2 are rarely found 9-ketobriaranes. Also, 1 is the first briarane derivative possessing a 20-hydroxy group.  相似文献

Three novel dimeric sesquiterpenoids, named parviflorenes G-I (1-3), have been isolated from Curcuma parviflora (Zingiberaceae), and their structures were elucidated by means of spectroscopic studies. Absolute stereochemistry of parviflorene G (1) as well as previously isolated related compounds, parviflorenes A (4), B (5), D (6), and F (7), was revealed by CD spectral data and chemical means. Parviflorenes G (1) and I (3) were cytotoxic against HeLa cells, while parviflorenes A (4) and F (7) were cytotoxic against all tested tumor cell lines in the human cancer cell line panel assay.  相似文献

Reinvestigation of the secondary metabolites from the marine mangrove fungus Aigialus parvus BCC 5311 led to the isolation of six new nonaketide metabolites, aigialomycins F (4) and G (5a/5b), 7′,8′-dihydroaigialospirol (7), 4′-deoxy-7′,8′-dihydroaigialospirol (8), and rearranged macrolides 9 and 10, along with six previously described compounds, hypothemycin (1), aigialomycins A (2) and B (3), aigialospirol (6), 4-O-demethylhypothemycin (11), and aigialone (12). Four new alkaloids, including two new meleagrin analogs, meleagrins B (2) and C (3), and two new diketopiperazines, roquefortines F (5) and G (6), together with six new diterpenes, conidiogenones B-G (7-12), were isolated from a deep ocean sediment derived fungus Penicillium sp. The structures and stereochemistry of the new compounds were elucidated by spectroscopic methods.
