Blue organic light-emitting supramolecular microfibers: the self-assembly of a 1,2,4-triazolo[1,5-a]pyrimidine derivative

The design and synthesis of 5,7-dimethyl-1,2,4-triazolo[1,5-a]pyrimidine-2-thioaceto-(2-fluorobenzyl) hydrazone (TPTH), which self-assembled into supramolecular microfibers with blue organic light-emitting properties, is reported. This is the first occurrence of the molecular self-assembly of 1,2,4-triazolo[1,5-a]pyrimidine derivatives.

以5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-甲硫醚为起始原料, 设计合成了15个新型的5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-氧乙酰腙及10个(R)-5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-氧(α-甲基)乙酰腙类化合物, 通过元素分析、 MS和 1H NMR对所合成的化合物进行了结构表征. 初步生物活性测试结果表明, 部分化合物表现出不同程度的除草及杀菌活性. 目标化合物中引入手性中心有利于生物活性的提高.     

含肟醚的新型三唑并嘧啶衍生物的合成及除草活性研究

2-巯基-5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶(6)与氯乙醛或氯丙酮反应生成5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-硫丙酮(硫乙醛) (7). 中间体7与O-烷基(芳基)羟胺(5)反应得到目标化合物1-(5,7-二甲基-1,2,4三唑[1,5-a]嘧啶-2-硫)乙醛-O-烷基肟醚(9), 2-(5,7-二甲基-1,2,4三唑[1,5-a]嘧啶-2-硫)丙酮O-烷基肟醚(10). 初步生物活性测试结果表明部分化合物具有较好的除草活性.     

On Triazoles. XIII. The reaction of 5-benzalimino-1,2,4-triazoles with substituted acetyl chlorides

The reaction of Schiff bases prepared from 1- and 2-substituted-5-amino-1,2,4-triazoles with phenoxyacetyl chlorides in the presence of triethylamine and a mixture of phosphorus oxychloride and dichloroacetic acid in dimethylformamide to yield β-lactam 4 , a dihydro-1,2,4-triazolo[4,3-a]pyrimidine-5(1H)-one 5, a 1,2,4-triazolo[1,5-a]pyrimidin-5(3H)-one 9 and the corresponding 1,2,4-triazolo[4,3-a]pyrimidine-5(1H)-one 10 derivatives was studied.     

Improved microwave-assisted catalyst-free synthesis of 9-aryl-5,9-dihydropyrimido[4,5-d][1,2,4]triazolo[1,5-a]pyrimidine-6,8(4H,7H)-dione derivatives

A green regioselective synthesis of some new and known 9-aryl-5,9-dihydropyrimido[4,5-d][1,2,4]triazolo[1,5-a]pyrimidine-6,8(4H,7H)-diones has been described via the microwave-assisted one-pot reaction of 3-amino-1H-1,2,4-triazoles, aromatic aldehydes and barbituric acids under solvent- and catalyst-free conditions. This operationally simple procedure is less laborious and provides a better scope than previously reported procedures.     

5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-硫乙酰腙类 衍生物的合成及生物活性 研究

以2-巯基-5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶为起始原料,设计合成了16种新型的-5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-硫乙酰腙类衍生物,其结构均经1^HNMR,MS及元素分析确证。通过核磁共振氢谱分析及分子力学优化,对该类化合物在溶液中的顺反异构体的化学位移进行了归属。初步生测结果表明,所合成的化合物均表现出不同程度的除草及杀菌活性,尤其是在50×^-6浓度下所有化合物都对水稻纹枯病表现出很好的抑制效果。对化合物4c和4o活体小株实验结果表明,在先接菌后施药的处理方式下,化合物在50×^-6和100×^-6浓度下对水稻纹枯病的防效均优于多菌灵和井冈霉素,在100×^-6浓度下的防效达到了80%以上。     

Recyclization of Condensed Carbethoxypyrimidines Accompanied by Substitution of a Carbon Atom into the Heterocycle

Condensation in ethanol of ethyl ethoxymethyleneacetoacetate with systems containing an amidine fragment (substituted 3-aminopyrazoles and 3-amino-1,2,4-triazole) gave 6-carbethoxy-7-methylpyrazolo[1,5-a]pyrimidines. Addition of base to solutions of the obtained bicyclic carbethoxy derivatives in the course of several minutes caused rearrangement to 6-acetyl-7-hydroxypyrazolo[1,5-a]pyrimidine and 6-acetyl-7-hydroxy-1,2,4-triazolo[1,5-a]pyrimidine respectively. A more prolonged refluxing in 15% aqueous alcohol solution of base caused 6-carbethoxy-7-methyl-2-phenylpyrazolo[1,5-a]pyrimidine and 6-acetyl-7-hydroxy-2-phenylpyrazolo[1,5-a]pyrimidine to recyclize to 7-methylpyrazolo[1,5-a]pyrimidine.__________Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 569–576, April, 2005.     

Reactions of 3-aroylacrylates with α-aminoazoles*

Dihydro derivatives of pyrazolo[3,4-b]pyridine-, pyrazolo[1,5-a]pyrimidine-, and [1,2,4]triazolo-[1,5-a]pyrimidinecarboxylates have been prepared by cyclocondensation of β-aroylacrylates with 5-aminopyrazoles and 3-amino-1,2,4-triazole. Heating dihydro[1,2,4]triazolo[1,5-a]pyrimidine-7-carboxylates with hydrazine hydrate led to recyclization of the pyrimidine ring to form 6-arylpyridazin-3(2H)-ones.     

1,2,4-Oxadiazoles from cycloreversions of oxadiazabicyclo[3.2.0]heptenes: 1-azetines as thiocyanate equivalents

1,3-Dipolar cycloaddition of nitrile oxides to 4-aryl-2-alkylthio-1-azetines gave a series of oxadiazabicyclo[3.2.0]heptenes as single diastereoisomers. Heating these cycloadducts in toluene resulted in an overall [2+2]-cycloreversion to give 5-alkylthio-3-aryl-1,2,4-oxadiazoles. In this process, the 1-azetine behaves as a thiocyanate equivalent. When the nitrile oxide substituent was 2-azidobenzene, the azide could be converted into a 1,2,3-triazole giving a (1,2,4-oxadiazolo)-(1,2,3-triazolo)-1,2-disubstituted benzene. 1,2,4-Oxadiazoles are sought after in medicinal chemistry and materials sciences.     

Cyclocondensation of α,Β-unsaturated ketones with 3-amino-1,2,4-triazole

Dihydro-1,2,4-triazolo[1,5-a]pyrimidine derivatives were obtained by condensation of 3-amino-1,2,4-triazole with 1,3-diaryl-1-propen-3-ones. The structure of 5-phenyl-7-(4-methylphenyl)-4,7-dihydro-1,2,4-triazolo[1,5-a]pyrimidine was established by x-ray diffraction analysis.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 2, pp. 229–233, February, 1988.     

Interaction of aminoazoles with Meldrum’s acid and dialkyl ketones or cyclohexanone

Interaction of 3-amino-5-methylpyrazole, 3-amino-5-methylthio-, and 3,5-diamino-1,2,4-triazole with Meldrum’s acid, acetone, ethyl methyl ketone, and cyclohexanone leads to alkyl-substituted pyrazolo[3,4-b]pyridin-6-ones and 1,2,4-triazolo[1,5-a]pyrimidin-7-ones respectively. The structure of 5,5-dimethyl-2-methylthio-4,5,6,7-tetrahydro-1,2,4-triazolo[1,5-a]pyrimidin-7-one was demonstrated by an X-ray structural investigation. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 595–601, April, 2007.     

2-取代硫醚-5-(5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶基)-1,3,4-噁二唑/噻二唑类化合物的合成及生物活性

以5-氨基-1H-1,2,4-三唑-3-羧酸乙酯为起始原料, 设计合成了11个新型的2-取代硫醚-5-(5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶基)-1,3,4-噁二唑类化合物(5)和6个新型的2-取代硫醚-5-(5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶基)-1,3,4-噻二唑类化合物(8). 通过¹H NMR, MS和元素分析对所合成的化合物进行了结构表征. 初步的生物活性测试结果表明, 所合成的化合物均表现出不同程度的除草及杀菌活性, 其中化合物5k和8f的活性最好, 在50 mg/L浓度下对水稻纹枯病菌的抑制率达90%以上.     

Facile and efficient synthesis of carbohybrids as stereodivergent druglike small molecules

A facile and efficient one-pot synthesis of acyclic polyols fused with pyrazolo[1,5-a]pyrimidines and 1,2,4-triazolo[1,5-a]pyrimidines as novel carbohybrids with regioselectivity was achieved through the condensation of 2-C-formyl glycals with 3-aminopyrazoles and 3-amino-1,2,4-triazoles in excellent yields under the microwave irradiation.     

含1,2,4-三唑-5-硫酮席夫碱的新型1,2,4-三唑并[1,5-a]嘧啶衍生物的合成及生物活性研究

以2-苄硫基-5-甲基-7-羟基-1,2,4-三唑并[1,5-a]嘧啶为原料,经醚化、肼解、成盐、关环和席夫碱反应合成了20个新型的含1,2,4-三唑-5-硫酮席夫碱的1,2,4-三唑并[1,5-a]嘧啶类化合物,通过IR,1H NMR,MS和元素分析对所合成的化合物进行了结构表征.初步生物活性测试结果表明,部分化合物表现出一定的抑菌或较好的抗烟草花叶病毒(TMV)活性.在500μg/mL浓度下,化合物6b,6f和6p对TMV的抑制率分别为41%,43%和40%.     

Synthesis of 2,8-disubstituted 1,2,4-triazolo[5,1-i]purines

This paper describes the preparation of 2-aryl-8-fluorobenzyl-1,2,4-triazolo[5,1-i]purines 2 , starting from 2-aryl-7,8-diamino-1,2,4-triazolo[1,5-c]pyrimidines 9 . Our synthetic procedure represents the first example of a feasible approach for the synthesis of 2,8-disubstituted triazolo[5,1-i]purines. New syntheses of 5-amino-3-arylpyrimido[5,4-e]1,2,4-triazines 12 and 8-fluorobenzyl-6-hydrazinopurines 15 are also reported.     

Tautomerism of dihydro-1,2,4-triazolo[1,5-α]pyrimidines containing 2-hydroxy- and 4-dimethylaminoaryl substituents

We have studied the effect of structural factors and the nature of the solvent on the tautomeric composition of aromatic substituted dihydro-1,2,4-triazolo[1,5-a]pyrimidines. The kinetics of the tautomeric exchange of 5-(2-hydroxyphenyl)-7-phenyldihydro-1,2,4-triazolo[1,5-a]pyrimidines has also been investigated.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 2, pp. 245–249, February, 1991.     

Convenient Synthesis of Some 1,2,4-triazolo[3,4-b]-1,3,4-thiadiazoles and 1,2,4-triazolo[3,4-b]-1,3,4-thiadiazines Bearing 3-Methylthio{7H-1,2,4-triazolo[1,5-d]tetrazol-6-yl}moiety as Possible Antimicrobial Activity

4-Amino-4H-3-methylthio{7H-1,2,4-triazolo[1,5-d]tetrazol-6-yl}-1,2,4-triazole-5thi-ol (1) was condensed with various substituted aromatic aldehydes or acid chlorides to yield a series of arylideneamines 2 or aroylamines 3. These were easily cyclized to the corresponding 6-aryl-3-methylthio7H-1,2,4-triazolo[1,5-d]tetrazol-6-yl-1,2,4-triazolo [3,4-b]-1,3,4-thiadiazoles (4) on treatment with palladium-on-charcoal or phosphorus oxychloride. Compounds 4 were also prepared directly via the reaction of 1 with aromatic carboxylic acids in the presence of phosphorus oxychloride. Cyclocondensation of 1 with a variety of two-carbon cyclizing reagents, namely chloroacetone, pyruvic acid, benzoylformic acid, and benzoin led to the formation of 3-methylthio{7H-1,2,4-triazolo[1,5-d]tetrazol-6-yl}-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazines (5?8). Some of the newly synthesized compounds were tested for their antibacterial and antifungal activity against a variety of microorganisms.     

Dinuclear silver(I) complexes for the design of metal-ligand networks based on triazolopyrimidines

Silver(I) coordination complexes with the versatile and biomimetic ligands 1,2,4-triazolo[1,5-a]pyrimidine (tp), 5,7-dimethyl-1,2,4-triazolo[1,5-a]pyrimidine (dmtp) and 7-amine-1,2,4-triazolo[1,5-a]pyrimidine (7atp) all feature dinuclear [Ag(2)(μ-tp)(2)](2+) building units (where tp is a triazolopyrimidine derivative), which are the preferred motif, independently of the counter-anion used. According to AIM (atoms in molecules) and ELF (electron localization function) analyses, this fact is due to the great stability of these dinuclear species. The complexes structures range from the dinuclear entities [Ag(2)(μ-tp)(2)(CH(3)CN)(4)](BF(4))(2) (1), [Ag(2)(μ-tp)(2)(CH(3)CN)(4)](ClO(4))(2) (2), [Ag(2)(μ-7atp)(2)](ClO(4))(2) (3) and [Ag(2)(μ-dmtp)(2)(CH(3)CN)](PF(6))(ClO(4)) (4) over the 1D polymer chain [Ag(2)(μ-CF(3)SO(3))(2)(μ-dmtp)(2)](n) (5) to the 3D net {[Ag(2)(μ(3)-tp)(2)](PF(6))(2)·～6H(2)O}(n) (6) with NbO topology.     

Synthesis of 1,2,4-triazolo[1,5-a]pyrimidine-2-sulfonamides

The 1,2,4-triazolo[1,5-a]pyrimidine-2-sulfonamides are a new class of highly active herbicides. A novel cyclization method for the synthesis of these compounds is discussed.
