Embryogenic tissues of Dioscorea bulbifera were cryopreserved using the encapsulation-dehydration technique. Genetic stability of plants regenerated from cryopreserved embryogenic tissues was assessed using molecular, biochemical and morphological analysis. The random amplified polymorphic DNA (RAPD) analysis of 60 cryopreserved-derived and 20 in vitro grown (control) plantlets showed that 10 primers produced 62 clear reproducible DNA fragment profiles. The amplification products were monomorphic for all the plantlets except one. A total of 4960 DNA fragments were obtained from this study showing no variation in RAPD profiles. The diosgenin content of cryopreserved-derived plants, analyzed using HPLC, was similar to that of control plants. Morphology and the ability to form microtuber were also found to be unaltered in cryopreserved embryo-derived plantlets. Thus, the D. bulbifera plants regenerated from cryopreserved embryogenic tissues were genetically stable at the molecular, biochemical and morphological levels.  相似文献

In vitro shoot tips of Dioscorea deltoidea Wall., an endangered medicinal plant, were successfully cryopreserved using the vitrification and the encapsulation-dehydration techniques with subsequent high frequency plant regeneration. Using vitrification, post-liquid nitrogen (LN) shoot regeneration up to 83% was recorded when excised shoot tips were pretreated overnight on MS medium containing 0.3 M sucrose followed by loading with MS containing 2 M glycerol plus 0.4 M sucrose for 20 min at 25 degree C, dehydration with PVS2 for 90 min at 0 degree C and quenching in LN. After 1 h of storage in LN, the shoot tips were rewarmed in a water-bath at 40 degrees C, unloaded with 1.2 M sucrose solution for 20 min and cultured on recovery growth medium. While using encapsulation-dehydration, the highest regeneration frequency recorded was 76% when sucrose-pretreated shoot tips were encapsulated with 3% calcium alginate, precultured in 0.75 M sucrose for 3 days, dehydrated to 25% moisture content (FW basis) under the laminar air flow, stored in LN for 1h and rewarmed at 40 degree C. The cryopreserved shoot tips maintained their viability and an unaltered level of regeneration capability after up to one year of storage in LN.  相似文献

The optical properties of two liquid crystalline mesogens S4 (code name) and S5 (code name) have been studied as a function of temperature. Our interest is to compare the macroscopic properties of these two thermotropic mesogens and to study the effect of replacing the OC6H13 group in S4 by NCS group to produce S5.With this aim in view the thermal variations of birefringence have been determined for both the compounds. The variations of polarizabilities with temperature have also been evaluated from density variations and birefringence studies and the thermal dependence of the order parameters of the two compounds have thereby been determined and compared. The theoretical values of the order parameters have also been calculated using the McMillan and Maier–Saupe potentials and compared with the obtained experimental values.  相似文献

The difficulty of using magnetic resonance imaging (MRI) to support early diagnosis of multiple sclerosis (MS) stems from the subtle pathological changes in the central nervous system (CNS). In this study, texture analysis was performed on MR images of MS patients and normal controls and a combined set of texture features were explored in order to better discriminate tissues between MS lesions, normal appearing white matter (NAWM) and normal white matter (NWM). Features were extracted from gradient matrix, run-length (RL) matrix, gray level co-occurrence matrix (GLCM), autoregressive (AR) model and wavelet analysis, and were selected based on greatest difference between different tissue types. The results of the combined set of texture features were compared with our previous results of GLCM-based features alone. The results of this study demonstrated that (1) with the combined set of texture features, classification was perfect (100%) between MS lesions and NAWM (or NWM), less successful (88.89%) among the three tissue types and worst (58.33%) between NAWM and NWM; (2) compared with GLCM-based features, the combined set of texture features were better at discriminating MS lesions and NWM, equally good at discriminating MS lesions and NAWM and at all three tissue types, but less effective in classification between NAWM and NWM. This study suggested that texture analysis with the combined set of texture features may be equally good or more advantageous than the commonly used GLCM-based features alone in discriminating MS lesions and NWM/NAWM and in supporting early diagnosis of MS.  相似文献

The presented work investigates the differences in magnetic properties of free and supported clusters via ab-initio calculations. The electronic structure of the clusters was calculated using a spin polarized relativistic multiple-scattering Green’s function formalism. We focus on Fe clusters of 2–9 atoms, either free or supported by Ni(001), and on Co clusters of 2–7 atoms, either free or supported by Au(111). For the supported clusters, the spin and orbital magnetic moments depend on the cluster size nearly monotonically, while for the free clusters large quasi-oscillations of magnetic moments with the cluster size were observed. Similarly, for supported clusters, the local spin magnetic moments decrease nearly linearly with increasing coordination number, while for free clusters of the same size range the trend is much more complicated. These findings are consistent with the fact that the spectral distribution function contains much sharper features for free clusters than for supported clusters.   相似文献

The perceived negative influence of standard hearing protectors on communication is a common argument for not wearing them. Thus, "augmented" protectors have been developed to improve speech intelligibility. Nevertheless, their actual benefit remains a point of concern. In this paper, speech perception with active earplugs is compared to standard passive custom-made earplugs. The two types of active protectors included amplify the incoming sound with a fixed level or to a user selected fraction of the maximum safe level. For the latter type, minimal and maximal amplification are selected. To compare speech intelligibility, 20 different speech-in-noise fragments are presented to 60 normal-hearing subjects and speech recognition is scored. The background noise is selected from realistic industrial noise samples with different intensity, frequency, and temporal characteristics. Statistical analyses suggest that the protectors' performance strongly depends on the noise condition. The active protectors with minimal amplification outclass the others for the most difficult and the easiest situations, but they also limit binaural listening. In other conditions, the passive protectors clearly surpass their active counterparts. Subsequently, test fragments are analyzed acoustically to clarify the results. This provides useful information for developing prototypes, but also indicates that tests with human subjects remain essential.  相似文献

Using high-level ab initio calculations, the cooperativity effects between an aerogen-bonding and a pnicogen- or chalcogen-bonding interactions are studied in ternary Y···PH2CN···ZO3 and Y···SHCN···ZO3 complexes (Y?=?NH3, N2 and Z?=?Ar, Kr, Xe). A detailed analysis of the structures, interaction energies and bonding properties is performed on these systems. For each set of the complexes, a favourable cooperativity is observed between Z···N and P/S···N interactions, especially in complexes involving NH3 and XeO3 molecules. It is found that for a given Y or Z, the amount of cooperativity effects in Y···PH2CN···ZO3 complexes are important than Y···SHCN···ZO3 ones. For each ternary complex considered, the effect of a Z···N aerogen bond on a P/S···N bond is more pronounced than that of a P/S···N bond on a Z···N bond. The mechanism of the cooperativity effects in the ternary complexes is studied by electron density difference, quantum theory of atoms in molecules and natural bond orbital analyses. The solvent effects are also studied on the interaction energy and cooperativity of Z···N and P/S···N bonds in the ternary systems.  相似文献

A comparative study of 11 pneumonectomized patients was undertaken in order to evaluate the respective advantages and drawbacks of MRI and CT in post-operative follow-up. Nine patients were healthy at the time of the study and two presented with tumor recurrence. MR examination included 500/40 ms axial, and frontal 800/40-80 ms or 1300/60-120 ms nongated spin echo sequences. MRI was slightly more efficient in identifying vascular stump and main nodal stations, and detected better than CT tumoral and metastatic spread in cancer recurrences. It was as informative as CT in evaluating postpneumonectomy space and bronchial stump. It was noncontributory in the detection of calcifications. Although clips were visible with MRI, their location was less definite than with CT, a potential pitfall when radiotherapy is planned.  相似文献

The Er:YAG and the CO2 laser are competitors in the field of hard tissue ablation. The use of Er:YAG lasers (2.94 μm, pulse length L of 100 to 200 μs) show smaller areas of thermal defects then ‘‘superpulsed’’ CO2 lasers with pulse lengths of approximately 100 μs. Only the development of a Q-switched CO2 laser (9.6 μm, τL=250 ns) allowed for similar results. In this paper new results for the Er:YAG and the Q-switched CO2 laser under the influence of water spray will be presented. Several parameters are of special interest for these investigations: the specific ablation energy, which shows a minimum for the CO2 laser at an energy density of 9 J/cm 2 and a broad shallow minimum in the range of 10 to 70 J/cm2 for the Er:YAG laser, and comparison of the cut-shape and depth. An understanding of high-temperature aging effects on the electrical properties of electrolytes is very important in selecting optimum compositions for practical applications. The aging behavior and mechanisms of doped zirconia ceramics have been extensively studied. However, little information is available regarding the aging behavior of ceria-based electrolytes. The present study has demonstrated that a high-temperature aging at 1000 °C has a significant effect on the ionic conductivity of the Y- or Gd-doped ceria (Ce1−xYxO2−δ and Ce1−xGdxO2−δ), especially in the case of the Gd doping. The aging behavior is characterized by a critical dopant concentration, above which the aging has a detrimental effect on the conductivity of the doped ceria ceramics. The aging behavior in the doped ceria cannot be explained using the aging mechanisms applied to the doped zirconia. Instead, the formation of the microdomains in the doped ceria has been acknowledged to be the main contribution to the aging behavior of the Y- or Gd-doped ceria ceramics. The formation ability of microdomains has been estimated to be in the order of La3+>Gd3+>Y3+, based on the degree of size mismatch between the dopant ion and Ce4+ ion. The critical dopant concentrations at which the microdomains start to form for La3+, Gd3+ and Y3+ in the doped ceria ceramics are x=0.15, 0.2 and 0.25, respectively. This critical dopant concentration is also an important indication: below which the conductivity is governed by only the association enthalpy, and above which the conductivity is dominated mainly by the microdomains rather than the association enthalpy.  相似文献

This article addresses the problem of parametric time-domain identification and dynamic analysis for time-varying (TV) mechanical structures under unobservable random excitation. The methods presented are based on time-dependent autoregressive moving average (TARMA) models, and are classified according to the mathematical structure imposed on the TV parameter evolution as unstructured parameter evolution, stochastic parameter evolution, and deterministic parameter evolution. The features and relative merits of each class are outlined. A representative method from each is then assessed through its application to the identification and dynamic analysis of a laboratory TV structure consisting of a beam with a mass moving on it. The results are mutually compared and contrasted to those obtained through “frozen-configuration” (multiple experiment) baseline identification.  相似文献

A number of single parameter variational approximations for single-mode graded-index fibers have been reported in the literature. One of these approximations was also proposed by us some time ago which describes the propagation characteristics of such fibers having power-law profile with a high degree of accuracy. Using these approximations we have computed the propagation constant and the normalized dispersion for single-mode fibers with smoothed-out and composite refractive index profiles which are of great current interest. We have shown that the single parameter approximation proposed by us earlier computes these propagation characteristics with a better degree of accuracy than all other single parameter approximations even for these special types of profiles.  相似文献

A comparative analysis of the effects induced by radiative corrections on the different sectors of the neutral current interactions at low momentum transfer (v?q, v?e ande?q interactions) and on the direct measurement of the vector boson masses is presented. In particular atomic parity violation is reviewed on the basis of new experimental data and more precise theoretical calculations. It is seen that the inclusion of radiative correction effects begins to become crucial in order to find an agreement between the parameters of the standard model extracted from the different sectors not only in the minimal version of the standard model, but also in two-parameter analysis.  相似文献

A group of rhenium (I) complexes including in their structure ligands such as CF(3)SO(3)-, CH(3)CO(2)-, CO, 2,2'-bipyridine, dipyridil[3,2-a:2'3'-c]phenazine, naphthalene-2-carboxylate, anthracene-9-carboxylate, pyrene-1-carboxylate and 1,10-phenanthroline have been studied for the first time by mass spectrometry. The probe electrospray ionization (PESI) is a technique based on electrospray ionization (ESI) that generates electrospray from the tip of a solid metal needle. In this work, mass spectra for organometallic complexes obtained by PESI were compared with those obtained by classical ESI and high flow rate electrospray ionization assisted by corona discharge (HF-ESI-CD), an ideal method to avoid decomposition of the complexes and to induce their oxidation to yield intact molecular cation radicals in gas state [M](+·) and to produce their reduction yielding the gas species [M](-·). It was found that both techniques showed in general the intact molecular ions of the organometallics studied and provided additional structure characteristic diagnostic fragments. As the rhenium complexes studied in the present work showed strong absorption in the UV-visible region, particularly at 355?nm, laser desorption ionization (LDI) mass spectrometry experiments could be conducted. Although intact molecular ions could be detected in a few cases, LDI mass spectra showed diagnostic fragments for characterization of the complexes structure. Furthermore, matrix-assisted laser desorption ionization (MALDI) mass spectra were obtained. Nor-harmane, a compound with basic character, was used as matrix, and the intact molecular ions were detected in two examples, in negative ion mode as the [M](-·) species. Results obtained with 2-[(2E)-3-(4-tert-buthylphenyl)-2-methylprop-2-enylidene] malononitrile (DCTB) as matrix are also described. LDI experiments provided more information about the rhenium complex structures than did the MALDI ones.  相似文献

This paper investigates the importance of loading and treatment with a vitrification solution on the survival of Citrus madurensis embryonic axes cryopreserved using a vitrification protocol. Among the seven different loading solutions tested, the solution containing 2 M glycerol + 0.4 M sucrose was the most efficient. Of the six vitrification solutions tested, the PVS2 vitrification solution, applied for 20 min at 25 degree C or for 60 min at 0 degree C, ensured the highest survival. A three-step vitrification protocol, involving the treatment of embryonic axes at 0 degree C with half strength PVS2 solution for 20 min then with full strength PVS2 for an additional 40 min was more efficient than a two-step protocol that involved treatment of axes directly with full strength PVS2 solution for 60 min. After rapid immersion in liquid nitrogen, rapid rewarming, unloading in a 1.2 M sucrose solution for 20 min, culture on solid medium with 0.3 M sucrose for 1 day and growth recovery for 4 weeks on standard medium, survival of C. madurensis embryonic axes reached 85 % following the three-step process, compared with 70 % for the two-step process.  相似文献

In the present paper the structural and electronic properties of polonides of lead, barium and calcium polonide (PbPo, BaPo and CaPo) have been reported. These properties have been studied using first principles calculations as well as the interionic potential model modified with the covalency effect. Gibbs free energy and enthalpy calculations show that these compounds undergo a structural phase transition from NaCl-type structure to CsCl-type structure. The electronic band structure and density of states of the PbPo, BaPo and CaPo have also been reported. The calculated equilibrium structural parameters are in good agreement with the available experimental results.  相似文献

A comparative study, ultrasound (US) versus microwave (MW) versus conventional thermal heating (TH), for synthesis of isoindolo-1,2-diazine is described. The reaction pathway is fast, efficient and straight applicable, involving a Huisgen [3+2] dipolar cycloaddition of cycloimmonium ylides to 1,4-naphthoquinone. A feasible reaction mechanism for the obtaining of the fully aromatized tetra- and penta- cyclic isoindolo-1,2-diazine is presented. Under US irradiation the yields are much higher (sometimes substantially, by almost double), the reaction time decreases substantially, the reaction conditions are milder. The use of a generator with a higher nominal power induces higher yields and short reaction times. Overall the use of US it proved to be more efficient than MW or TH. A feasible explication for US efficiency is presented.  相似文献
