The interaction of a N-methylated diaminotriphenylmethane dye, malachite green, with lysozyme was investigated by fluorescence spectroscopic techniques under physiological conditions. The binding parameters have been evaluated by fluorescence quenching methods. The results revealed that malachite green caused the fluorescence quenching of lysozyme through a static quenching procedure. The thermodynamic parameters like ΔH and ΔS were calculated to be −15.33 kJ mol−1 and 19.47 J mol−1 K−1 according to van’t Hoff equation, respectively, which proves main interaction between malachite green and lysozyme is hydrophobic forces and hydrogen bond contact. The distance r between donor (lysozyme) and acceptor (malachite green) was obtained to be 3.82 nm according to Fӧrster’s theory. The results of synchronous fluorescence, UV/vis and three-dimensional fluorescence spectra showed that binding of malachite green with lysozyme can induce conformational changes in lysozyme. In addition, the effects of common ions on the constants of lysozyme-malachite green complex were also discussed.  相似文献

ABSTRACT

Azoreductase, a flavin mononucleotide-containing oxidoreductase from Escherichia coli, can catalyze the reduction of azo dyes to form aromatic amine compounds. Few spectroscopic studies have explored the binding mode of azo dyes or the role of the arginine at site 59 in Azoreductase. In this article, protein engineering strategy has been used to construct one mutant in which the arginine residue at site 59 was mutated to glycine. Fluorescence spectroscopic data showed that the addition of Methyl Red and Methyl Orange resulted in the fluorescence quenching of the cofactor flavin mononucleotide bound to Azoreductase. The association constant was fitted using the standard binding equation instead of the Stern-Volmer equation. The results showed that the mutation from the arginine to glycine at site 59 weakened the association constant from 2.21?×?105?L.mol?1 to 4.55?×?104?L.mol?1 at 25°C. A similar phenomenon was also observed when Methyl Orange was used as a substrate. In each case, the association constant tended to decrease as the temperature increased from 25°C to 37°C. Thermodynamic parameter analysis revealed that the interaction type changed from a van der Waals interaction (between Azoreductase and the dyes) to a hydrogen bonding interaction (between the mutant and the dyes). Moleculcar docking was also performed in this work to provide some support for the binding mode and binding stability between Azoreductase/mutant and azo dyes.  相似文献

Abstract

Under simulated physiological conditions (pH?=?7.40), the interaction between cefpiramide sodium and hen egg white lysozyme was studied with multi-spectroscopy and molecular docking. The results showed that cefpiramide sodium quenched the fluorescence of hen egg white lysozyme by static quenching, and the number of binding site n was about 1. The binding distance (r) between cefpiramide sodium and hen egg white lysozyme was obtained based on the Förster nonradioactive resonance energy transfer and r was less than 7?nm, which indicated that there was a non-radiative energy transition in the system. The thermodynamic parameters were obtained from the van't Hoff equation, and the Gibbs free energy ΔG?H?S?>?0, indicating hydrophobic interaction played a major role in forming the cefpiramide sodium-hen egg white lysozyme complex. Synchronous spectra, circular dichroism spectra and UV-Vis spectra showed that cefpiramide sodium changed the conformation of hen egg white lysozyme. The molecular docking results showed that the binding position of cefpiramide sodium was close to the active center composed of Asp52 and Glu35 residues, suggesting that cefpiramide sodium could change the microenvironment of amino acid residues at the catalytic active center of hen egg white lysozyme.  相似文献

ABSTRACT

The interactions between rutin and trypsin were investigated by UV-Vis absorption, CD, fluorescence, resonance light-scattering spectra, synchronous fluorescence, and three-dimensional fluorescence spectra techniques under physiological pH 7.40. Rutin effectively quenched the intrinsic fluorescence of trypsin via static quenching. The enthalpy change and entropy change were estimated to be ?8.23 kJ·mol?1 and 53.66 J·mol?1·K?1 according to the van't Hoff equation. The process of binding rutin to trypsin was a spontaneous molecular interaction procedure. This result indicates that hydrophobic and electrostatic interactions played a major role in stabilizing the complex. The conformation of trypsin was discussed by CD, synchronous, and three-dimensional fluorescence techniques.  相似文献

The binding behavior of antibacterial drug sulfadiazine (SDZ) with water soluble globular proteins like bovine as well as human serum albumin (BSA and HSA, respectively) and lysozyme (LYS) was monitored by fluorescence titration and molecular docking calculations. The experimental data reveal that the quenching of the intrinsic protein fluorescence in presence of SDZ is due to the strong interaction in the drug binding site of the respective proteins. The Stern-Volmer plot shows positive deviation at higher quencher concentration for all the proteins and was explained in terms of a sphere of action model. The calculated fluorophore-quencher distances vary within 4?~?11 Å in different cases. Fluorescence experiments at different temperature indicate thermodynamically favorable binding of SDZ with the proteins with apparently strong association constant (~104–105 M?1) and negative free energy of interaction within the range of ?26.0?~??36.8 kJ mol?1. The experimental findings are in good agreement with the respective parameters obtained from best energy ranked molecular docking calculation results of SDZ with all the three proteins.  相似文献

According to our theoretical studies, the anomeric effect, an stereoelectronic interaction between lone pair and a vicinal antibonding orbital, has shown to contribute decisively for the conformational isomerism of 1‐fluoro‐N,N‐dimethylmethanamine ( 1 ) and of its corresponding P, As and Sb analogues ( 2 – 4 ). C? X bonds in 2 – 4 are larger than in the parent compound 1 , thus providing a LPX/C? F* interaction progressively weaker on going from 1 to 4 . However, such hyperconjugation contributed by more than 1.3 kcal mol?1 for the stabilization of anti conformer in 4 (θLP? X? C? F = 180°), increasing to 24.1 kcal mol?1 in 1 . We have investigated conformational fluctuations in a new green fluorescent protein(GFP)-like protein rb-Akane found in a red-brown-colored octocoral, Scleronephthya gracillima (Kuekenthal)), with high pressure fluorescence spectroscopy at 0.1–700?MPa. Besides the green fluorescence at 510?nm, two red fluorescence peaks are observed at 590 and 629?nm, the relative intensity of which varies reversibly with pressure. The phenomenon is interpreted as representing the cis–trans isomerization of the chromophore accompanied by the conformational transition between two sub-states of the red fluorescence form of rb-Akane. The two sub-states are separated only marginally in free energy (ΔG0?=?1.9?±?0.4?kJ?mol?1), but significantly in partial molar volume (ΔV0?=??19.8?±?1.4?ml?mol?1) at 0.1?MPa (pH 7.5, 25°C). Above 500?MPa, the fluorescence at λmax 629?nm undergoes another reversible change with pressure, showing the onset of unfolding.  相似文献

ABSTRACT

The interaction of isoquercitrin and bovine serum albumin (BSA) was investigated by means of fluorescence spectroscopy (FS), resonance light scattering spectroscopy (RLS), and ultraviolet spectroscopy (UV). The apparent binding constants (K a) between isoquercitrin and BSA were 5.37 × 105 L mol?1 (293.15 K) and 2.34 × 105 L mol?1 (303.15 K), and the binding site values (n) were 1.18 ± 0.03. According to the Förster theory of non-radiation energy transfer, the binding distances (r) between isoquercitrin and BSA were 1.94 and 1.95 nm at 293.15 K and 303.15 K, respectively. The experimental results showed that the isoquercitrin could be inserted into the BSA, quenching the inner fluorescence by forming the isoquercitrin–BSA complex. The addition of increasing isoquercitrin to BSA solution leads to the gradual enhancement in RLS intensity, exhibiting the formation of the aggregate in solution. It was found that both static quenching and non-radiation energy transfer were the main reasons for the fluorescence quenching. The entropy change and enthalpy change were negative, which indicated that the interaction of isoquercitrin and BSA was driven mainly by van der Waals interactions and hydrogen bonds. The process of binding was a spontaneous process in which Gibbs free energy change was negative.  相似文献

Nature 358(6383):209–215 1992] triazolo [1,5-c]pyrimidine-2-sulfonanilide) and bovine serum albumin (BSA) was investigated by fluorescence, ultraviolet absorption (UV) and Far-UV circular dichroism (CD) spectrometries. A strong fluorescence quenching was observed and the quenching mechanism was considered as static quenching. The binding constant of FU with BSA at 299 and 309 K were obtained as 1.5?×?104 and 7.1?×?103 l mol?1, respectively. There was one binding site between FU and BSA. The thermodynamic parameters enthalpy change (ΔH) and entropy change (ΔS) were calculated as ?57.89 kJ mol?1 and ?113.6 J mol?1 K?1, respectively, which indicated that the acting force between FU and BSA was mainly hydrogen bond and Van der Waals force. According to the Förster non-radiation energy transfer theory, the average binding distance between donor (BSA) and acceptor (FU) was obtained (r?=?1.59 nm). The investigations of the UV/Vis and CD spectra of the system showed that the conformation of BSA was changed in presence of FU.  相似文献

The interaction of lysozyme with bromophenol blue (BPB) in acetate buffer (pH 6.0) was studied by fluorescence quenching method for the first time. It was found that BPB could conspicuously quench the fluorescence of lysozyme by the static quenching process, possibly due to the binding on the active site near Trp62. The binding parameters including the binding constant and the number of binding site were calculated. The thermodynamic parameters ΔH°, ΔS° and ΔG° at different temperatures were obtained. The formation of lysozyme–BPB complex depended on the cooperation of the hydrophobic and electrostatic forces. And the binding average distance between lysozyme and BPB was determined. The binding of quercetin to lysozyme (LYSO) in aqueous solution was investigated by fluorescence spectroscopy, UV-vis absorption spectroscopy and molecular simulation at pH 7.4. The fluorescence quenching of LYSO by addition of quercetin is due to static quenching, the binding constants, K a , were 3.63 × 104, 3.31 × 104 and 2.85 × 104 L·mol−1 at 288, 298 and 308 K, respectively. The thermodynamic parameters, enthalpy change, ∆H, and entropy change, ∆S, were noted to be −7.56 kJ·mol−1 and 61.07 J·mol−1·K−1. The results indicated that hydrophobic interaction may play a major role in the binding process. The distance r between the donor (LYSO) and acceptor (quercetin) was determined as 3.34 nm by the fluorescence resonance energy transfer. The synchronous fluorescence spectroscopy showed the polarity around the tryptophan residues increased and the hydrophobicity decreased. Furthermore, the study of molecular simulation indicated that quercetin could bind to the active site (a pocket made up of 24 amino-acid residues) of LYSO mainly via hydrophobic interactions and that there were hydrogen interactions between the residues (Gln 57, Ile 98) of LYSO and quercetin. The accessible surface area (ASA) calculation verified the important roles of tryptophan (Trp) residues during the binding process.  相似文献

Abstract

With the widespread application of neonicotinoid insecticides, Clothianidin has received much attention due to the potential harm to human health and ecological environment. However, the mechanism of Clothianidin's underlying toxicity to organisms remains unclear. In this work, the interaction between Clothianidin and human serum albumin was investigated and the intrinsic fluorescence of human serum albumin got quenched via static mechanisms upon the addition of Clothianidin. The binding constants between Clothianidin and human serum albumin at three different temperature were obtained to be 3.543?×?104, 2.995?×?104, and 2.490?×?104 M?1, respectively. Based on the van't Hoff equation, the thermodynamic parameters, ΔH0 and ΔS0 were estimated to be ?53.885?KJ mol?1 and ?110.535?J mol?1K?1, respectively. A single binding site was predicted from the binding constants at different temperatures by multiple spectroscopic techniques and the negative values of ΔH0 and ΔS0 indicated the binding of human serum albumin with Clothianidin was driven by hydrogen bonds or van der Waals forces. Furthermore, the loose and unfolded secondary structure of human serum albumin along with the addition of clothianidin had been observed through ultraviolet-visible absorption and circular dichroism spectra. In addition, it was also found that Clothianidin had polar effects of structural microenviroment not only on Trp but also Tyr residues from synchronous fluorescence analysis. This study illuminates the molecular mechanism of the interaction between human serum albumin and clothianidin for the first time and helps to construct a specific pesticide biosensor system of human health.  相似文献

The values of the enthalpy (53.3; 51.3; 20.0 kJ mol?1), entropy (?106; ?122; ?144 J mol?1K?1), and volume of activation (?29.1; ?31.0; ?cm3 mol?1), the reaction volume (?25.0; ?26.6; ?cm3 mol?1) and reaction enthalpy (?155.9; ?158.2; ?150.2 kJ mol?1) have been obtained for the first time for the ene reactions of 4‐phenyl‐1,2,4‐triazoline‐3,5‐dione 1 , with cyclohexene 4 , 1‐hexene 6 , and with 2,3‐dimethyl‐2‐butene 8 , respectively. The ratio of the values of the activation volume to the reaction volume (?VcorrVr ? n) in the ene reactions under study, 1 + 4 → 5 and 1 + 6 → 7 , appeared to be the same, namely 1.16. The large negative values of the entropy and the volume of activation of studied reactions 1 + 4 → 5 and 1 + 6 → 7 better correspond to the cyclic structure of the activated complex at the stage determining the reaction rate. The equilibrium constants of these ene reactions can be estimated as exceeding 1018 L mol?1, and these reactions can be considered irreversible. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

It was first found that the intrinsic fluorescence of lysozyme at 340 nm can be quenched by cephalosporin analogues through the static quenching and non-radiative energy transferring procedure. In the acetate buffer solution with pH 7.0 and 298 K, the quenching fluorescence intensity was in a good linearity over the concentration of drugs in the range of 1–100 μmol L−1, 0.1–100 μmol L−1, 0.5–100 μmol L−1 and 0.05–100 μmol L−1 for cefradine, cefuroxime, cefotaxime and ceftriaxone, respectively. The quenching ability or the binding ability of the studied drugs followed the pattern: ceftriaxone > cefotaxime > cefuroxime > cefradine, which was close to the order of their antibacterial ability. The binding parameters including the association constant and the number of binding potential point were calculated at different temperatures (288, 298 and 308 K), and thermodynamic parameters ΔH°, ΔS° and ΔG° were given. The binding mode of lysozyme with cephalosporins showed that the hydrophobic effect might play a major role. The binding distance between cephalosporin and tryptophan residue in lysozyme was obtained. The results provided the quantitative information for the binding of cephalosporin to lysozyme, and it was suggested that the drugs probably bound to the active site near Trp62 in lysozyme.  相似文献

Ball milled nanocrystalline AlMg4.8 powder was investigated in terms of hardening and thermal stability. The validity of the Hall–Petch relation was confirmed down to the minimum grain size of ~44 nm. Prolonged milling in the range of the minimum grain size still increased the hardness. This development is discussed in terms of contamination effects and the influence of full and partial dislocations. Concerning thermal stability, recovery processes occur in the range of 100–230°C, whereas substantial grain growth starts at a temperature of ~250°C. The enthalpy release for recovery was detected to be ~39 J mol?1 and ~208 J mol?1 for grain growth. Dynamic strain ageing was indicated by an activation energy for recovery of Q?~?120 kJ mol?1. The activation energy of grain growth was calculated by means of the Kissinger theory (Q?=?200–210 kJ mol?1) and using the results of static grain growth (Q?=?204 kJ mol?1).  相似文献

Amitriptyline.HCl (AMI) and clomipramine.HCl (CMI) react with eosin Y (EY) in pH 3.8 NaAc-AcH buffer solution to form ion association complex which results in quenching of fluorescence of EY and appearance of a new resonance Rayleigh scattering (RSS) spectrum at 620 nm. The spectral characteristics of absorption, fluorescence and RSS spectra have been investigated. The factors influencing the reaction were studied and optimum conditions for the reaction have been determined. Based on fluorescence quenching, a simple and sensitive spectrofluorimetric method for determination of AMI and CMI has been developed. The fluorescence quenching intensity was measured at 550 nm using an excitation wavelength of 310 nm. The calibration graph was found to be rectilinear in the range 0.08–2.0 μg?mL?1 with detection limit of 0.017 μg?mL?1 for AMI and 0.06–2.0 μg?mL?1 with detection limit of 0.015 μg?mL?1 for CMI. The method can be satisfactorily applied to the determination of AMI and CMI in tablets without interference from commonly occurring exicipients. The recovery and RSD values obtained indicate good accuracy and precision of the method. The mechanism of the reaction and fluorescence quenching has also been discussed.  相似文献

In this paper, the toxic influence of copper ions (II) on bovine hemoglobin was investigated by the combination of ultraviolet-visible absorption, fluorescence, time-resolved fluorescence, synchronous fluorescence, and circular dichroism spectra. Driven by hydrophobic and electrostatic forces, copper ions (II) could interact with bovine hemoglobin to form bovine hemoglobin-copper ions (II) complex with one binding site. The binding constant (K) was 1.57?×?104, 1.89?×?104 and 2.11?×?104?L/mol at 298, 304, and 310?K, respectively. The binding distance (r) was 4.24?nm. Fluorescence and time-resolved fluorescence spectra showed that bovine hemoglobin quenched by copper ions (II) was a static quenching process. Results of synchronous fluorescence spectra revealed that the microenvironment and the conformation of bovine hemoglobin were changed during the binding reaction. Data of circular dichroism spectra suggested that with the increasing concentration of copper ions (II), the secondary structure of bovine hemoglobin underwent a decrease in α-helix and alteration in backbone microenvironment. Copper ions (II) was thus evidenced to have a certain toxic effect on physical bodies.  相似文献

Abstract

In order to explore the interaction between meloxicam and trypsin, the interaction mechanism between meloxicam and trypsin was studied by fluorescence spectrum, UV-vis absorption spectrum, circular dichroism spectrum, and molecular docking simulation under the experimental condition of pH = 7.40. The results of spectral experiments showed that meloxicam could effectively quench the internal fluorescence of trypsin in the form of static quenching, formed a stable complex at 1:1, and changed the conformation of trypsin. The results of thermodynamic constant showed that ΔG?H?S?>?0 indicates that the main force type of the binding system was hydrophobic interaction and hydrogen bonding. Molecular docking technique showed that the best binding site between meloxicam and trypsin was near the catalytic active center of trypsin, and the interaction between them changed the microenvironment of amino acid residues in the catalytic active center of trypsin. The mathematical model of drug and protein showed that when the concentration ratio of meloxicam to trypsin was 1:1, the protein binding rate of the binding system was 5.15%. The concentration ratio of meloxicam to trypsin was 30: 1, and the protein binding rate was 45.4%. The results showed that when the drug concentration was high, the binding effect of the system had a great influence on the concentration of free trypsin.  相似文献

The gas‐phase elimination kinetics of selected ethyl esters of 2‐oxo‐carboxylic acid have been studied over the temperature range of 270–415 °C and pressures of 37–114 Torr. The reactions are homogeneous, unimolecular, and follow a first‐order rate law in a seasoned static reaction vessel, with an added free radical suppressor toluene. The supramolecular interaction of ofloxacin (Oflo) and methyl β-cyclodextrin (Mβ-CD) has been examined by UV–vis, IR and fluorescence spectroscopy. The formation of inclusion complex has been confirmed based on the changes of the spectral properties. The results showed that Mβ-CD reacted with Oflo to form an inclusion complex. The Oflo and Mβ-CD complex formed a host-guest complex in 1:1 stoichiometry and inclusion constant (K?=?7.8?×?10?3 L mol?1) was ascertained by the typical double reciprocal plots. Furthermore, the thermodynamic parameters (?H°, ?S° and ?G°) associated with the inclusion process were also determined. In addition, solid inclusion complex was synthesized. Based on the significant enhancement of the fluorescence intensity of Oflo produced through complex formation, a simple, accurate, rapid and highly sensitive spectrofluorometric method for the determination of Oflo in pharmaceutical formulation was developed. The measurement of relative fluorescence intensity was carried out at 497 nm with excitation at 296 nm. The factors affecting the inclusion complex formation were studied and optimized. Under the optimum reaction conditions, linear relationships with good correlation coefficients (0.9995) were in the concentration range of 50–350 ng/mL for spectrofluorimetry. The limit of detection (LOD) was 11.5 ng/mL. The proposed method was successfully applied to the analysis of Oflo in pharmaceutical preparation.  相似文献
