以天然大黄酸为原料合成了三种大黄酸金属配合物. 通过元素分析、摩尔电导、紫外光谱、红外光谱、热重差热、核磁共振氢谱和荧光分析测定配合物的组成和性质. 结果表明配合物组成为ML2•2H2O [M=Cu(II), Fe(II), Mn(II)|L=rhein(失去1-OH中质子)]. 并对配体和配合物进行了清除超氧自由基(O )、羟自由基(•OH)、DPPH•自由基的对比研究. 抗氧化试验结果表明, 金属配合物抗氧化活性均强于配体.  相似文献

利用木犀草素、硫酸和金属盐为原料,合成了4种木犀草素磺酸金属配合物,利用红外光谱、元素分析、核磁共振光谱和质谱等手段表征了配合物的结构.以DPPH清除率、抑菌环的大小检测配合物的抗氧化、抗菌活性,结果表明木犀草素磺酸金属配合物对DPPH的清除率和对金黄色葡萄球菌和大肠杆菌的抑制作用随浓度的增大而提高,且效果优于母体木犀草素。  相似文献

Hbbimp的金属配合物的合成、表征及与DNA的作用研究   总被引:1,自引:0,他引:1  
合成了四种2,6-N,N-二[二(2-苯并咪唑甲基)]氨甲基-4-甲基苯酚(Hbbimp)的金属(Zn2+、Mn2+、Eu3+、Nd3+)配合物,并用红外、此外、摩尔电导、元互分析表征了它们的结构。用凝胶电泳实验研究了它们与PBR322DNA的作用,发现在37℃时四种金属配合物与DNA的作用效果均不明显;在50℃,pH8.0时,仅两种稀土(Eu3+和Nd3+)配合物可将大部分超螺旋DNA(CCC带)转化为缺刻产物(OC带);在37℃,[H2O2]<1×10-3mol/L,[稀土配合物]:[H2O2]=1∶20时,配合物在较低浓度时即可将CCC带全部转化为OC带.  相似文献

以NBT/VB2/蛋氨酸为O*-2产生、检测体系,对自制低聚壳聚糖及其金属配合物进行了抗O*-2活性研究,结果显示低聚壳聚糖及其金属配合物对O*-2均具有明显的清除活性.质量浓度为0.5×10-2 g/mL时,壳聚糖对O*-2的清除率达80.3%,其与氯化镧、醋酸铜、醋酸钴的配合物对O*-2的清除率分别为98.9%、84.1%、78.4%;相同条件下,高分子壳聚糖和单糖对O*-2没有明显的清除作用,清除率仅为13%、9.5%.随着样品质量浓度降低,低聚壳聚糖及其金属配合物的清除活性逐渐下降.  相似文献

研究了4种D-氨基葡萄糖甘氨酸混配金属配合物的表面增强拉曼光谱(SERS),发现它们在银胶上的吸附方式基本相同,因而SERS光谱相似,用电子光谱、荧光光谱、表面增强拉曼光谱研究了它们与DNA的相互作用,发现这4种化合物与DNA的作用能力有很大的不同,其中GuGluG、Co(Ⅲ)GluG是值得进一步研究的可能抗癌的药物。  相似文献

本文合成了三种铂(Ⅱ)配合物PtLCl(HL1=2-(3′,5′-二甲基-吡唑-1′-基)-N-(2″-吡啶甲基)乙基胺;HL2=2-(3′,5′-二甲基-吡唑-1′-基)-N-(2″-吡啶乙基)乙基胺;HL3=2-(3′,5′-二甲基-吡唑-1′-基)-N-(喹啉-8″-基)乙基胺,通过元素分析和质谱进行结构表征。利用荧光和圆二色光谱研究了3种配合物与小牛胸腺DNA的相互作用,结果发现配体结构对配合物与DNA的作用方式及强度产生极大影响。PtL3Cl具有较大的共轭平面而易以嵌入模式与DNA结合,而PtL1Cl和PtL2Cl的空间位阻较小,易与DNA以共价模式结合。通过质谱跟踪发现,配合物PtL1Cl和PtL2Cl均能与5′-鸟苷酸(5′-GMP)发生共价结合,但是没有发现PtL3Cl与5′-GMP的加合物。3种配合物对人宫颈癌细胞的体外毒活性数据表明:PtL3Cl的细胞毒活性最强。  相似文献

以4-(1H-1,2,4-三唑-1-亚甲基)苯甲酸(HL)为配体,水热合成了三个过渡金属配合物[Cu0.5L]n1),{[Ni(L)2·(H2O)2]·(H2O)2}n2),{[Co(L)2·(H2O)2]·(H2O)2}n3);运用红外光谱、元素分析、单晶X射线衍射、热分析、紫外光谱和荧光光谱等手段对化合物进行了表征. 结构分析表明,配合物1 呈现一维(1D)的链状结构,而同构的配合物23 则通过链间的氢键连接为三维(3D)网状结构. 抑菌活性研究表明,配合物对镰刀菌、苹果腐烂病菌、苹果轮纹病菌、梨炭疽病菌和烟草赤星病菌均有良好的抑菌效果,特别是配合物1 的抑菌效果最为显著. 质粒DNA(pUC 18)裂解活性实验结果表明,相比较于化合物23,化合物1 能更有效地促进质粒DNA裂解. 进一步采用光谱法检测三种配合物对DNA的结合作用. 结果显示三种配合物均可插入DNA分子中,其中配合物1的插入作用最为强烈.  相似文献

A novel tetradentate dianionic Schiff base ligand, N ,N ′‐bis(2‐carboxyphenylimine)‐2,5‐thiophenedicarboxaldhyde (H2L) and some first row d‐transition metal chelates (Co(II), Cu(II), Ni(II) and Zn(II)) were synthesized and characterized using various physicochemical and spectroscopic methods. The spectroscopic data suggested that the parent Schiff base ligand coordinates through both deprotonated carboxylic oxygen and imine nitrogen atoms. The free Schiff base and its metal chelates were screened for their antimicrobial activities for various pathogenic bacteria and fungi using the agar well diffusion method. The antibacterial and antifungal activities of all the newly synthesized compounds are significant compared to the standard drugs ciprofloxacin and nystatin. The antioxidant activities of the compounds were determined by reduction of 1,1‐diphenyl‐2‐picrylhydrazyl and compared with that of vitamin C as a standard. DNA binding ability of the novel Schiff base and its complexes was investigated using absorption spectroscopy, fluorescence spectroscopy, viscosity measurements and thermal denaturation. The obtained results clearly demonstrate that the binding affinity with calf thymus DNA follows the order: Cu(II) complex > Ni(II) complex > Zn(II) complex > Co(II) complex >H2L. Furthermore, the DNA cleavage activity of the newly synthesized ligand and its metal complexes was investigated using supercoiled plasmid DNA (pUC18) gel electrophoresis.  相似文献

The coordination of ruthenium(II) complexes to anionic oxygen-based donors are very rare. This study describes a simple, one-pot method for obtaining [ruthenium(II)(trithiacyclononane)(curcumin)(S-DMSO)]Cl (1) in 37% yield. The structural characterization of complex 1 by elemental analysis, FT-IR, 1-D and 2-D NMR, ESI+-MS as well as UV–vis and fluorescence spectroscopies are presented. The DNA-melting temperature (Tm) assay shows that salmon sperm DNA (smDNA) in the presence of complex 1 has a higher melting temperature, with ΔTm = 7.4 °C, while in the presence of curcumin the melting temperature remains unaltered. The in vitro cytotoxic activities of curcumin and complex 1 were investigated using the tumor human prostate cell line, PC-3, and the healthy cell line, PNT-2. Complex 1 is innocuous toward normal prostate epithelial cells and, whereas curcumin is toxic, with inhibition rates of ca. 35 and 65% at 50 and 80 μM, respectively. On the tumor cell line PC-3, complex 1 did not cause viability changes, whereas curcumin exhibited dose-dependent inhibition, with ca. 73% inhibition at the highest concentration tested, i.e. 80 μM. This study suggests that coordination with the trithiacyclononane ruthenium(II) scaffold stabilizes the photochemical properties of curcumin and strongly changes its biologic activity.  相似文献

Abstract

A one pot procedure was used to synthesize two new derivatives of α-aminophosphonates. Novel copper(II) complexes of α-aminophosphonates were synthesized by coordinating different copper salts with the newly synthesized α-aminophosphonates. Their structures were characterized by different spectral and analytical techniques. Evaluation of the metal-free ligands HL1, HL2, and their Cu(II) complexes against human colon carcinoma HT-29 cell lines was performed, using cisplatin as a reference drug. The results indicated that the complexes of the ligand HL1 exhibited enhanced anticancer activity, while ligand HL2 complexes showed decreased anticancer activity.  相似文献

[Pd(L1)(C,N)]CF3SO3 and [Pd(L2)(C,N)]CF3SO3 (L1 = 2,2′ ‐bipyridine, L2 = 1,10‐phenanthroline and C,N = benzylamine) novel orthopalladated complexes have been synthesized and characterized using various techniques. The binding of the complexes with native calf thymus DNA (CT‐DNA) was monitored using UV–visible absorption spectrophotometry, fluorescence spectroscopy and thermal denaturation studies. Our results indicate that these complexes can strongly bind to CT‐DNA via partial intercalative mode. In addition, fluorescence spectrometry of bovine serum albumin (BSA) with the complexes shows that the fluorescence quenching mechanism of BSA is a static process. The results of site‐competitive replacement experiments with specific site markers clearly indicate that the complexes bind to site I of BSA. Notably, the complexes exhibit significant in vitro cytotoxicity against two human cancer cell lines (Jurkat and MCF‐7) with IC50 values varying from 37 to 53 μM. Finally, a molecular docking experiment effectively proves the binding of the Pd(II) complexes to DNA and BSA.  相似文献

A novel Schiff base ligand, namely 2,2′‐((1E,1′E)‐(1,3‐phenylenebis(azanylylidene))bis(methanylylidene))diphenol (H2L), was synthesized by condensation of m‐phenylenediamine and 2‐hydroxybenzaldehyde (in 1:2 ratio). Series of complexes were obtained from the reaction of La(III), Er(III) and Yb(III) chlorides with H2L. The ligand and complexes were characterized using elemental analysis, infrared, 1H NMR, UV–visible and mass spectroscopies, magnetic susceptibility and conductivity measurements and thermal analysis. Infrared and 1H NMR spectra indicated the coordination of the azomethine nitrogens and deprotonated phenolic oxygen atoms in a tetradentate manner (ONNO). The thermal behaviour of the complexes was studied from ambient temperature to 1000°C. The complexes were found to have water molecules of hydration and coordinated water molecules. The complexes were found to possess high biological activities against various organisms compared to the free ligand (Gram‐positive bacteria Staphylococcus aureus and Bacillus subtilis, Gram‐negative bacteria Salmonella sp., Escherichia coli and Pseudomonas aeruginosa and fungi Aspergillus fumigatus and Candida albicans). The more effective and probable binding modes between H2L with different active sites of colon cancer (PDB code: 2hq6) and lung cancer (PDB code: 1x2j) receptors were investigated using molecular docking studies.  相似文献

Bis(N‐ethylbenzimidazol‐2‐ylmethyl)aniline (Etbba) and its transition metal complexes, [Cu(Etbba)(Cl)2]?DMF ( 1 ), [Mn(Etbba)(Cl)2] ( 2 ) and [Ni(Etbba)(Cl)2] ( 3 ), have been synthesized and characterized on the basis of elemental analysis, molar conductivity, UV–visible, infrared and NMR spectroscopies and X‐ray crystallography. The coordination environment of complex 1 can be described as distorted square‐based pyramidal, while complexes 2 and 3 each have a distorted trigonal bipyramidal geometry. Cyclic voltammograms of complex 1 indicate an electrochemically quasi‐reversible Cu2+/Cu+ couple. In addition, the antioxidant activities of the free ligand and its complexes were investigated using the superoxide and hydroxyl radical scavenging methods in vitro. Complexes 1 , 2 , 3 are found to possess potent hydroxyl radical scavenging activity and to be better than standard antioxidants like vitamin C and mannitol. Mononuclear transition metal(II) complexes of the type M(L)2?2H2O (where M = Co, Ni, Cu, Zn) have been synthesized from uninegative Schiff base ligands (HL1–HL4) designed by condensation of 4‐fluorobenzylamine with 2‐hydroxy‐1‐naphthaldehyde/3,5‐dichlorosalicylaldehyde/3,5‐dibromosalicylaldehyde/3‐bromo‐5‐chlorosalicylaldehyde. The compounds were successfully characterized using spectroscopic and physiochemical methods together with elemental analysis. Spectroscopic elucidation indicates a monobasic bidentate nature of ligands coordinated via deprotonated phenolic oxygen and azomethine nitrogen atom which suggests an octahedral geometry around the central metal ions. The complexes and ligands were screened for their in vitro antimicrobial activity against bacterial and fungal strains, the zinc(II) complexes being more active against the tested microbial strains. Further, the metal complexes were found to be more active than the uncomplexed ligands due to chelation process and, moreover, the complexes were more active against fungal strains than bacterial strains. Cytotoxic activities of all compounds were evaluated towards human alveolar adenocarcinoma epithelial cell line (A549), human breast adenocarcinoma cell line (MCF7), human prostate cancer cell line (DU145) and one normal human lung cell line (MRC‐5) using MTT colorimetric assay with doxorubicin as a standard. The zinc complexes were most active against the cancer cell lines and also found to be less toxic against MRC‐5 normal cell line than standard doxorubicin.  相似文献

A novel bi‐nucleating Schiff base ligand, 6,6′‐(((1E,1′E)‐thiophene‐2,5‐diylbis (methaneylylidene))bis (azaneylylidene))bis (3,4‐dimethylaniline), and five binuclear M (II) complexes were synthesized. The bi‐nucleating Schiff base ligand and its metal complexes were characterized using various physicochemical techniques, e.g. elemental analyses, spectroscopic methods, conductivity and magnetic moment measurements. The low molar conductance of the complexes in dimethylsulfoxide shows their non‐electrolytic nature. The antibacterial activities were screened against pathogenic bacteria (Staphylococcus aureus, Escherichia coli, Pseudomonas putida and Bacillus subtilis). The antifungal activity was screened against Aspergillus niger, Aspergillus flavus and Rhizoctonia bataicola. The antimicrobial activity data showed that the metal complexes are more potent than the parent Schiff base ligand against microorganisms. The antioxidant activities of the synthesized compounds were investigated through scavenging activity against 2,2‐diphenyl‐2‐picrylhydrazyl, superoxide anion, hydroxyl and 2,2′‐ azinobis (3‐ethylbenzothiazoline‐6‐sulfonic acid) radicals. The complexes have superior radical scavenging activity than the free ligand and the scavenging effects of the Cu (II) complex are stronger than those of the other complexes. DNA binding studies were performed using electronic spectroscopy, fluorometric competition studies and viscosity measurements. The data indicated that there is a marked enhancement in biocidal activity of the ligand under similar experimental conditions because of coordination with metal ions.  相似文献
