Multimodal imaging techniques have been demonstrated to be greatly advantageous in achieving accurate diagnosis and gained increasing attention in recent decades. Herein, we present a new strategy to integrate the complementary modalities of 19F magnetic resonance imaging (19F MRI) and fluorescence imaging (FI) into a polymer nanoprobe composed of hydrophobic fluorescent organic core and hydrophilic fluorinated polymer shell. The alkyne‐terminated fluorinated copolymer (Pn) of 2,2,2‐trifluoroethyl acrylate (TFEA) and poly(ethylene glycol) methyl ether acrylate (PEGA) was first prepared via atom transfer radical polymerization (ATRP). The PEGA plays an important role in both improving 19F signal and modulating the hydrophilicity of Pn. The alkynyl tail in Pn is readily conjugated with azide modified tetra‐phenylethylene (TPE) through click chemistry to form azo polymer (TPE‐azo‐Pn). The core‐shell nanoprobes (TPE‐P3N) with an average particle size of 57.2 ± 8.8 nm are obtained via self‐assembly with ultrasonication in aqueous solution. These nanoprobes demonstrate high water stability, good biocompatibility, strong fluorescence and good 19F MRI performance, which present great potentials for simultaneous fluorescence imaging and 19F–MR imaging.  相似文献

Boron has been employed in materials science as a marker for imaging specific structures by electron energy loss spectroscopy (EELS) or secondary ion mass spectrometry (SIMS). It has a strong potential in biological analyses as well; however, the specific coupling of a sufficient number of boron atoms to a biological structure has proven challenging. Herein, we synthesize tags containing closo‐1,2‐dicarbadodecaborane, coupled to soluble peptides, which were integrated in specific proteins by click chemistry in mammalian cells and were also coupled to nanobodies for use in immunocytochemistry experiments. The tags were fully functional in biological samples, as demonstrated by nanoSIMS imaging of cell cultures. The boron signal revealed the protein of interest, while other SIMS channels were used for imaging different positive ions, such as the cellular metal ions. This allows, for the first time, the simultaneous imaging of such ions with a protein of interest and will enable new biological applications in the SIMS field.  相似文献

A new click bioorthogonal reaction system was devised to enable the fast ligation (kON≈340 m ?1 s?1) of conjugatable derivatives of a rigid cyclic diol (nopoldiol) and a carefully optimized boronic acid partner, 2‐methyl‐5‐carboxymethylphenylboronic acid. Using NMR and fluorescence spectroscopy studies, the corresponding boronates were found to form reversibly within minutes at low micromolar concentration in water, providing submicromolar equilibrium constant (Keq≈105–106 m ?1). Efficient protein conjugation under physiological conditions was demonstrated with model proteins thioredoxin and albumin, and characterized by mass spectrometry and gel electrophoresis.  相似文献

A novel optically active amphiphilic diblock copolymer bearing quinine pendants poly(ethylene oxide)‐b‐poly(glycidyl triazolyl‐L ‐quinine) (MPEO‐b‐PGTQ) was synthesized by “click” reaction of alkyne‐modified diblock copolymer poly(ethylene oxide)‐b‐poly(glycidyl propargyl ether) (MPEO‐b‐PGPE) and 9‐N3‐quinine. The structure and composition of copolymers were characterized by gel permeation chromatography, 1H nuclear magnetic resonance spectroscopy (1H NMR), elemental analysis and optical rotation measurements, which showed that the synthetic route could provide the copolymer with well‐defined composition and with similar optical activity compared to its parent quinine. The micellization behavior of this chiral copolymer was investigated in different solvent systems. The results from fluorescence spectroscopy, UV spectroscopy, dynamic light scattering, transmission electron microscopy, 1H NMR and circular dichroism (CD) spectroscopy indicated that the MPEO‐b‐PGTQ could form regular chiral spherical micelles in H2O and Tetrahydrofuran‐H2O (10:90, V/V) systems, and the state of aggregated chiral micelles depended on the nature of the medium. © 2009 Wiley Periodicals, Inc. Copolymer containing new thermally reversible click chemistry‐assisted triazole‐substituted azobenzene and fulgimide units in the pendant F‐co‐A was prepared by free‐radical solution addition polymerization technique. The F and A were also prepared for comparison. The DSC analysis of F indicates that the polymer possessing the C‐form of fulgimide unit exhibited higher Tm than that of E‐form of the same polymer and revealed that the C‐form of fulgimide unit in F is highly ordered. The cis‐trans back isomerization behavior of the click chemistry‐assisted triazole‐substituted azobenzene unit in film A has thermal irreversibility, while in F‐co‐A it exhibited thermal reversibility. The UV‐exposed film of F‐co‐A heated around Tg leads to cis‐trans back isomerization of azobenzene unit and thermally stable C‐form of fulgimide which retains its conjugated structure where both the photochromic units are converted into planar conformations and exhibit high fluorescence properties. The fluorescence maxima of C‐form in F‐co‐A red shifted compared with F , because the substituted triazole ring in the azobenzene unit stabilized the C‐form of fulgimide unit. © 2008 Wiley Periodicals, Inc. To better understand the range of cellular interactions of PtII‐based chemotherapeutics, robust and efficient methods to track and analyze Pt targets are needed. A powerful approach is to functionalize PtII compounds with alkyne or azide moieties for post‐treatment conjugation through the azide–alkyne cycloaddition (click) reaction. Herein, we report an alkyne‐appended cis‐diamine PtII compound, cis‐[Pt(2‐(5‐hexynyl)amido‐1,3‐propanediamine)Cl2] ( 1 ), the X‐ray crystal structure of which exhibits a combination of unusual radially distributed CH/π(CC) interactions, Pt Pt bonding, and NH:O/NH:Cl hydrogen bonds. In solution, 1 exhibits no Pt alkyne interactions and binds readily to DNA. Subsequent click reactivity with nonfluorescent dansyl azide results in a 70‐fold fluorescence increase. This result demonstrates the potential for this new class of alkyne‐modified Pt compound for the comprehensive detection and isolation of Pt‐bound biomolecules.  相似文献

In order to generate a reliable and long‐lasting stable isotope ratio standard for CO2 in samples of clean air, CO2 is liberated from well‐characterized carbonate material and mixed with CO2‐free air. For this purpose a dedicated acid reaction and air mixing system (ARAMIS) was designed. In the system, CO2 is generated by a conventional acid digestion of powdered carbonate. Evolved CO2 gas is mixed and equilibrated with a prefabricated gas comprised of N2, O2, Ar, and N2O at close to ambient air concentrations. Distribution into glass flasks is made stepwise in a highly controlled fashion. The isotopic composition, established on automated extraction/measurement systems, varied within very small margins of error appropriate for high‐precision air‐CO2 work (about ±0.015‰ for δ13C and ±0.025‰ for δ18O). To establish a valid δ18O relation to the VPDB scale, the temperature dependence of the reaction between 25 and 47°C has been determined with a high level of precision. Using identical procedures, CO2‐in‐air mixtures were generated from a selection of reference materials; (1) the material defining the VPDB isotope scale (NBS 19, δ13C = +1.95‰ and δ18O = ?2.2‰ exactly); (2) a local calcite similar in isotopic composition to NBS 19 (‘MAR‐J1’, δ13C = +1.97‰ and δ18O = ?2.02‰), and (3) a natural calcite with isotopic compositions closer to atmospheric values (‘OMC‐J1’, δ13C = ?4.24‰ and δ18O = ?8.71‰). To quantitatively control the extent of isotope‐scale contraction in the system during mass spectrometric measurement other available international and local carbonate reference materials (L‐SVEC, IAEA‐CO‐1, IAEA‐CO‐8, CAL‐1 and CAL‐2) were also processed. As a further control pure CO2 reference gases (Narcis I and II, NIST‐RM 8563, GS19 and GS20) were mixed with CO2‐free synthetic air. Independently, the pure CO2 gases were measured on the dual inlet systems of the same mass spectrometers. The isotopic record of a large number of independent batches prepared over the course of several months is presented. In addition, the relationship with other implementations of the VPDB‐scale for CO2‐in‐air (e.g. CG‐99, based on calibration of pure CO2 gas) has been carefully established. The systematic high‐precision comparison of secondary carbonate and CO2 reference materials covering a wide range in isotopic composition revealed that assigned δ‐values may be (slightly) in error. Measurements in this work deviate systematically from assigned values, roughly scaling with isotopic distance from NBS 19. This finding indicates that a scale contraction effect could have biased the consensus results. The observation also underlines the importance of cross‐contamination errors for high‐precision isotope ratio measurements. As a result of the experiments, a new standard reference material (SRM), which consists of two 5‐L glass flasks containing air at 1.6 bar and the CO2 evolved from two different carbonate materials, is available for distribution. Time‐of‐flight secondary ion mass spectrometry (ToF‐SIMS) was previously used to characterize lignocellulosic materials, including woody biomass. ToF‐SIMS can acquire both rapid spectral and spatial information about a sample's surface composition. In the present study, ToF‐SIMS was used to characterize the cell walls of stem tissue from the plant model organism, Arabidopsis thaliana. Using principal component analyses, ToF‐SIMS spectra from A. thaliana wild‐type (Col‐0), cellulose mutant (irx3), and lignin mutant (fah1) stem tissues were distinguished using ToF‐SIMS peaks annotated for wood‐derived lignocellulose, where spectra from the irx3 and fah1 were characterized by comparatively low polysaccharide and syringyl lignin content, respectively. Spatial analyses using ToF‐SIMS imaging furthermore differentiated interfascicular fiber and xylem vessels based on differences in the lignin content of corresponding cell walls. These new data support the applicability of ToF‐SIMS peak annotations based on woody biomass for herbaceous plants, including model plant systems like arabidopsis. Fluorine NMR paramagnetic relaxation enhancement was evaluated as a versatile approach for extracting distance information in selectively F‐labeled proteins. Proof of concept and initial applications are presented for the HIV‐inactivating lectin cyanovirin‐N. Single F atoms were introduced at the 4‐, 5‐, 6‐ or 7 positions of Trp49 and the 4‐position of Phe4, Phe54, and Phe80. The paramagnetic nitroxide spin label was attached to Cys residues that were placed into the protein at positions 50 or 52. 19F‐T2 NMR spectra with different relaxation delays were recorded and the transverse 19F‐PRE rate, 19F‐Γ2, was used to determine the average distance between the F nucleus and the paramagnetic center. Our data show that experimental 19F PRE‐based distances correspond to 0.93 of the 1HN‐PRE distances, in perfect agreement with the gyromagnetic γ19F/γ1H ratio, thereby demonstrating that 19F PREs are excellent alternative parameters for quantitative distance measurements in selectively F‐labeled proteins.  相似文献

The characterization of protein–ligand interaction modes becomes recalcitrant in the NMR intermediate exchange regime as the interface resonances are broadened beyond detection. Here, we determined the 19F low‐populated bound‐state pseudocontact shifts (PCSs) of mono‐ and di‐fluorinated inhibitors of the BRM bromodomain using a highly skewed protein/ligand ratio. The bound‐state 19F PCSs were retrieved from 19F chemical exchange saturation transfer (CEST) in the presence of the lanthanide‐labeled protein, which was termed the 19F PCS‐CEST approach. These PCSs enriched in spatial information enabled the identification of best‐fitting poses, which agree well with the crystal structure of a more soluble analog in complex with the BRM bromodomain. This approach fills the gap of the NMR structural characterization of lead‐like inhibitors with moderate affinities to target proteins, which are essential for structure‐guided hit‐to‐lead evolution.  相似文献

Fluorescent dyes are commonly conjugated to nanomaterials for imaging applications using stochastic synthesis conditions that result in a Poisson distribution of dye/particle ratios and therefore a broad range of photophysical and biodistribution properties. We report the isolation and characterization of generation 5 poly(amidoamine) (G5 PAMAM) dendrimer samples containing 1, 2, 3, and 4 fluorescein (FC) or 6‐carboxytetramethylrhodamine succinimidyl ester (TAMRA) dyes per polymer particle. For the fluorescein case, this was achieved by stochastically functionalizing dendrimer with a cyclooctyne “click” ligand, separation into sample containing precisely defined “click” ligand/particle ratios using reverse‐phase high performance liquid chromatography (RP‐HPLC), followed by reaction with excess azide‐functionalized fluorescein dye. For the TAMRA samples, stochastically functionalized dendrimer was directly separated into precise dye/particle ratios using RP‐HPLC. These materials were characterized using 1H and 19F NMR spectroscopy, RP‐HPLC, UV/Vis and fluorescence spectroscopy, lifetime measurements, and MALDI.  相似文献

We present the access to [5‐19F, 5‐13C]‐uridine and ‐cytidine phosphoramidites for the production of site‐specifically modified RNAs up to 65 nucleotides (nts). The amidites were used to introduce [5‐19F, 5‐13C]‐pyrimidine labels into five RNAs—the 30 nt human immunodeficiency virus trans activation response (HIV TAR) 2 RNA, the 61 nt human hepatitis B virus ? (hHBV ?) RNA, the 49 nt SAM VI riboswitch aptamer domain from B. angulatum, the 29 nt apical stem loop of the pre‐microRNA (miRNA) 21 and the 59 nt full length pre‐miRNA 21. The main stimulus to introduce the aromatic 19F–13C‐spin topology into RNA comes from a work of Boeszoermenyi et al., in which the dipole‐dipole interaction and the chemical shift anisotropy relaxation mechanisms cancel each other leading to advantageous TROSY properties shown for aromatic protein sidechains. This aromatic 13C–19F labeling scheme is now transferred to RNA. We provide a protocol for the resonance assignment by solid phase synthesis based on diluted [5‐19F, 5‐13C]/[5‐19F] pyrimidine labeling. For the 61 nt hHBV ? we find a beneficial 19F–13C TROSY enhancement, which should be even more pronounced in larger RNAs and will facilitate the NMR studies of larger RNAs. The [19F, 13C]‐labeling of the SAM VI aptamer domain and the pre‐miRNA 21 further opens the possibility to use the biorthogonal stable isotope reporter nuclei in in vivo NMR to observe ligand binding and microRNA processing in a biological relevant setting.  相似文献

Multiple two‐dimensional nuclear magnetic resonance (2D‐NMR) techniques have been used to study the structures of Krytox® perfluoro(polyalkyl ether) and its mechanism of polymerization. Model compound K4, containing four Krytox® fluoropolymer repeat units, was analyzed to interpret the multiplet patterns in the NMR spectra from the polymer model. 19F {13C}‐Heteronuclear single‐quantum correlation experiments, performed with delays optimized for 1JCF and 2JCF, provided spectra that permitted identification of resonances from individual monomer units. Selective, 19F‐19F COSY 2D‐NMR experiments were performed with different excitation regions; these experiments were combined with selective inversion pulses to remove 19F‐19F J couplings in the f1 dimension. The resulting COSY spectra were greatly simplified compared with standard 19F‐19F COSY spectra, which are too complicated to interpret. They give information regarding the attachments of monomer units and also provide insights into the nature of the stereoisomers that might be present in the polymer. Both infrared and NMR spectra show peaks identifying chain end structures. With the help of these studies, resonances can be assigned, and the average number of repeat units in the polymer chain can be calculated based on the assignments obtained. Oligomers of (R)‐3‐hydroxybutanoate (OHB) have previously been shown to transport cations through lipid bilayers. The ion‐transport activity has been attributed to the formation of hydrophobic aggregates or pores, which have been identified by fluorescence‐microscopy measurements of membrane‐incorporated fluorescence‐labeled OHBs. To obtain more information about these aggregates, we describe here the synthesis of the specifically F‐labeled HB oligomers II – IV for structural investigation by means of solid‐state 19F‐NMR spectroscopic techniques.  相似文献

Pseudo‐meta and pseudo‐para diamino‐octafluoro[2.2]paracyclophanes are challenging to separate either by chromatography or recrystallization, but through the use of a mixture of the two isomers, the 1H, 19F, and 13C NMR spectra of these compounds have been fully and unambiguously assigned using 1H COSY, 1H‐19F HOESY, 1H‐13C HSQC, 1H‐13C HMBC, and 19F‐13C HSQC techniques. This permits the easy identification of either of the individual isomers. In addition, the 13C spectrum of the pseudo‐ortho analogue is reported and assigned for the first time. The gem shift effect in this series of bridge‐fluorinated paracyclophanes serves to deshield 1H resonances and shield 13C. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

Time‐of‐flight secondary ion mass spectrometry (ToF‐SIMS) equipped with a bismuth imaging source and an argon gas cluster ion beam (GCIB) was used to image polished cross‐sections of four automotive multilayer paint samples. Secondary ion mass spectrometry chemical imaging of the individual layers was possible after a GCIB sputter ion dose of (7 × 1015) ions/cm2 was applied for the removal of polishing residue, at which point the chemical composition of the individual clear coats could be distinguished using principal components analysis. For the differentiation of the four clear coat chemistries, only four secondary ion peaks were necessary; C2H5O+ (m/z 45.04), C9H9NO2+ (m/z 163.09), and C10H11NO2+ (m/z 177.10) that appeared to be fragments of the carbamate‐based clear coat, and C7H11+ (m/z 95.09) that was strongly associated with the polyurethane‐based clear coat. Clear identification of the four paint samples based on this short peak list highlights the strength of the SIMS technique as a potential forensic approach to discriminate automotive paints and suggests that many more variables could be included in the multivariate and statistical analysis to differentiate a wider range of clear coat chemistries.  相似文献

 Secondary ion mass spectrometry (SIMS) was optimised for characterisation of uranium- and plutonium-containing particles in soils, swipes and forensic samples. This was done by analysing in-house produced spherical UO2-particles. Screening techniques as α-autoradiography together with SIMS analysis were employed to detect UO2-particles in a soil sample from Chernobyl. The use of SIMS was exploited for the identification of uranium- and plutonium-containing particles and for the determination of their isotopic composition. The particles collected on swipe samples were transferred to a special adhesive support for the analysis by SIMS. Particles containing highly enriched uranium with diameters up to 10 μm were also detected in a forensic sample. For the measurements of the isotopic ratios a mass resolution of 1000 was used. At this resolution flat-top peaks were obtained which greatly improve the accuracy of the measurement. The isotopic composition of the particles was measured with a typical accuracy and precision of 0.5%. Statistically meaningful results can be obtained, for instance, from a specimen containing as few as 1010 atoms/μm3 of uranium in particles of UO2 weighing a few picograms.  相似文献

We report through‐space (TS) 19F–19F coupling for ortho‐fluoro‐substituted Z ‐azobenzenes. The magnitude of the TS‐coupling constant (TSJFF) ranged from 2.2–5.9 Hz. Using empirical formulas reported in the literature, these coupling constants correspond to non‐bonded F–F distances (dFF) of 3.0–3.5 Å. These non‐bonded distances are significantly smaller than those determined by X‐ray crystallography or density functional theory, which argues that simple models of 19F–19F TS spin–spin coupling solely based dFF are not applicable. 1H, 13C and 19F data are reported for both the E and Z isomers of ten fluorinated azobenzenes. Density functional theory [B3YLP/6‐311++G(d,p)] was used to calculate 19F chemical shifts, and the calculated values deviated 0.3–10.0 ppm compared with experimental values.
