An amphiphilic block copolymer, poly(ethylene glycol)‐block‐poly(L ‐lactide‐co‐2‐methyl‐2‐benzoxycarbonyl‐propylene carbonate) [PEG‐b‐P(LA‐co‐MBC)], was synthesized in bulk by the ring‐opening polymerization of L ‐lactide with 2‐methyl‐2‐benzoxycarbonyl‐propylene carbonate (MBC) in the presence of poly(ethylene glycol) as a macroinitiator with diethyl zinc as a catalyst. The subsequent catalytic hydrogenation of PEG‐b‐P(LA‐co‐MBC) with palladium hydroxide on activated charcoal (20%) as a catalyst was carried out to obtain the corresponding linear copolymer poly(ethyleneglycol)‐block‐poly(L ‐lactide‐co‐2‐methyl‐2‐carboxyl‐propylenecarbonate) [PEG‐b‐P(LA‐co‐MCC)] with pendant carboxyl groups. DSC analysis indicated that the glass‐transition temperature (Tg) of PEG‐b‐P(LA‐co‐MBC) decreased with increasing MBC content in the copolymer, and Tg of PEG‐b‐P(LA‐co‐MCC) was higher than that of the corresponding PEG‐b‐P(LA‐co‐MBC). The in vitro degradation rate of PEG‐b‐P(LA‐co‐MCC) in the presence of proteinase K was faster than that of PEG‐b‐P(LA‐co‐MBC), and the cytotoxicity of PEG‐b‐P(LA‐co‐MCC) to chondrocytes from human fetal arthrosis was lower than that of poly(L ‐lactide). © 2005 Wiley Periodicals, Inc. A series of poly(N‐isopropylacrylamide‐coN‐hydroxymethylacrylamide) P(NIPAM‐co‐NHMA) copolymers were firstly synthesized via free radical polymerization. Then, the hydrophobic, photosensitive 2‐diazo‐1,2‐naphthoquinone (DNQ) molecules were partially and randomly grafted onto P(NIPAM‐co‐NHMA) backbone through esterification to obtain a triple‐stimuli (photo/pH/thermo) responsive copolymers of P(NIPAM‐co‐NHMA‐co‐DNQMA). UV‐vis spectra showed that the lower critical solution temperature (LCST) of P(NIPAM‐co‐NHMA) ascended with increasing hydrophilic comonomer NHMA molar fraction and can be tailored by pH variation as well. The LCST of the P(NIPAM‐co‐NHMA) went down firstly after DNQ modification and subsequently shifted to higher value after UV irradiation. Meanwhile, the phase transition profile of P(NIPAM‐co‐NHMA‐co‐DNQMA) could be triggered by pH and UV light as expected. Thus, a triple‐stimuli responsive copolymer whose solution properties could be, respectively, modulated by temperature, light, and pH, has been achieved. Polycaprolactone‐graft‐Poly(2‐(dimethylamino)ethyl methacrylate‐co‐methoxy polyethylene glycol monomethacrylate) (PCL‐graft‐P(DMAEMA‐co‐mPEGMMA)) was synthesized by combination of ring‐opening polymerization (ROP) and atom transfer radical polymerization (ATRP). PCL‐graft‐P(DMAEMA‐co‐mPEGMMA) was characterized by FTIR, 1H NMR, and GPC. PCL‐graft‐P(DMAEMA‐co‐mPEGMMA) with expected composition and structure was achieved. pH‐ and thermo‐sensitive properties of the PCL‐graft‐P(DMAEMA‐co‐mPEGMMA) nanoparticles prepared by the nanoprecipitation method were investigated by TEM and DLS. With increase in the temperature, the size of PCL‐graft‐P(DMAEMA‐co‐mPEGMMA) nanoparticles is decreased under base environment. Furthermore, in vitro transfection and toxicity assays were tested in 293T cells. The results indicate that PCL‐graft‐P(DMAEMA‐co‐PEGMMA) has lower cytotoxicity at N/P ratios less than 10 with transfection efficiency concomitantly reducing at N/P ratios less than 20 compared to PCL‐graft‐PDMAEMA as the control. However, PCL‐graft‐P(DMAEMA‐co‐PEGMMA) presents higher transfection efficiency at N/P ratios more than 20 compared to PCL‐graft‐PDMAEMA. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

A series of new P‐methylphenyl P‐heterocycles are introduced. The para and ortho substituted 2,5‐dihydro‐1H‐phosphole oxides ( 1a and 1b ) were converted to the double‐bond isomers ( A and B ) of 1,2‐dihydrophosphinine oxides ( 3a and 3b ) via the corresponding phosphabicyclo[3.1.0]hexane oxides ( 2a or 2b ). Isomeric mixture ( A and B ) of the dihydrophosphinine oxides ( 3a and 3b ) gave, in turn, the isomers ( A and B ) of phosphabicyclo[2.2.2]oct‐5‐enes ( 4a and 4b ) or a phosphabicyclo[2.2.2]octa‐5,7‐diene ( 5 ) in Diels‐Alder reaction with dienophiles. The bridged P‐heterocycles ( 4 and 5 ) were useful in the photo‐ or thermoinduced fragmentation‐related phosphinylation of hydroxy compounds and amines. We synthesized two new alternating polymers, namely P(Tt‐FQx) and P(Tt‐DFQx) , incorporating electron rich tri‐thiophene and electron deficient 6‐fluoroquinoxaline or 6,7‐difluoroquinoxaline derivatives. Both polymers P(Tt‐FQx) and P(Tt‐DFQx) exhibited high thermal stabilities and the estimated 5% weight loss temperatures are 425 and 460 °C, respectively. Polymers P(Tt‐FQx) and P(Tt‐DFQx) displayed intense absorption band between 450 and 700 nm with an optical band gap (Eg) of 1.78 and 1.80 eV, respectively. The determined highest occupied/lowest unoccupied molecular orbital's (HOMO/LUMO) of P(Tt‐DFQx) (?5.48 eV/?3.68 eV) are slightly deeper than those of P(Tt‐FQx) ( ?5.32 eV/?3.54 eV). The polymer solar cells fabricated with a device structure of ITO/PEDOT:PSS/ P(Tt‐FQx) or P(Tt‐DFQx) :PC70BM (1:1.5 wt %) + 3 vol % DIO/Al offered a maximum power conversion efficiency (PCE) of 3.65% with an open‐circuit voltage (Voc) of 0.59 V, a short‐circuit current (Jsc) of 10.65 mA/cm2 and fill factor (FF) of 59% for P(Tt‐FQx) ‐based device and a PCE of 4.36% with an Voc of 0.69 V, a Jsc of 9.92 mA/cm2, and FF of 63% for P(Tt‐DFQx) ‐based device. © 2018 Wiley Periodicals, Inc. Phosphorylation of suitable piperidine precursors yielded a series of novel decalin‐type O,N,P‐heterocycles. The title compounds, P(3)‐axially and P(3)‐equatorially X‐substituted, cis‐ and trans‐configurated 2,4‐dioxa‐7‐aza‐, 2,4‐dioxa‐8‐aza‐, and 2,4‐dioxa‐9‐aza‐3‐phosphabicyclo[4.4.0]decane 3‐oxides (X=Cl, F, 4‐nitrophenoxy, and 2,4‐dinitrophenoxy), are configuratively fixed and conformationally constrained P‐analogues of acetylcholine and as such represent acetylcholine (7‐aza and 9‐aza isomers) or γ‐homo‐acetylcholine mimetics (8‐aza isomers). Being irreversible inhibitors of acetylcholinesterase (AChE), the compounds are considered to be suitable probes for the investigation of the stereochemical course of the inhibition reaction by 31P‐NMR spectroscopy. Moreover, the design of these mimetics will enable studies of molecular interactions with AChE, in particular, the recognition conformation of acetylcholine.  相似文献

The title compounds, the P(3)‐axially and P(3)‐equatorially substituted cis‐ and trans‐configured 8‐benzyl‐3‐fluoro‐2,4‐dioxa‐8‐aza‐3‐phosphadecalin 3‐oxides (=8‐benzyl‐3‐fluoro‐2,4‐dioxa‐8‐aza‐3‐phosphabicyclo[4.4.0]decane 3‐oxides=2‐fluorohexahydro‐6‐(phenylmethyl)‐4H‐1,3,2‐dioxaphosphorino[5,4‐c]pyridine 2‐oxides) were prepared (ee>98%) and fully characterized (Schemes 2 and 3). The absolute configurations were established from that of their precursors, the enantiomerically pure cis‐ and trans‐1‐benzyl‐4‐hydroxypiperidine‐3‐methanols which were unambiguously assigned. Being configuratively fixed and conformationally constrained phosphorus analogues of acetyl γ‐homocholine (=3‐(acetyloxy)‐N,N,N‐trimethylpropan‐1‐aminium), they are suitable probes for the investigation of molecular interactions with acetylcholinesterase. As determined by kinetic methods, all of the compounds are weak inhibitors of the enzyme.  相似文献

N‐(Substituted aryl/cyclohexyl)‐N'‐[5‐bromo‐5‐nitro‐2‐oxido‐1,3,2‐dioxaphosphorinane‐2‐yl]ureas RR'P(O)NHC(O)NHR' (5) were synthesized by the reactions of 2‐bromo‐2‐nitro‐1,3‐propanediol (4) with chlorides of aryl/cyclohexyl carbamidophosphoric acids (3) in the presence of triethylamine at room temperature. A series of amphiphilic graft copolymers of poly (vinylidene fluoride‐co‐chlorotrifluoroethylene)‐g‐poly(2‐vinyl pyridine), P (VDF‐co‐CTFE)‐g‐P2VP, with different degrees of P2VP grafting (from 26.3 to 45.6 wt%) was synthesized via one‐pot atom transfer radical polymerization (ATRP). The amphiphilic properties of P (VDF‐co‐CTFE)‐g‐P2VP graft copolymers allowed itself to self‐assemble into nanoscale structures. P (VDF‐co‐CTFE)‐g‐P2VP graft copolymers were introduced into neat P (VDF‐co‐CTFE) as additives to form blending membranes. When two different solvents, N‐methyl‐2‐pyrrolidone (NMP) and dimethylformamide (DMF), were used, specific organized crystalline structures were observed only in the NMP systems. P (VDF‐co‐CTFE)‐g‐P2VP played a pivotal role in controlling the morphology and pore structure of membranes. The water flux of the membranes increased from 57.2 to 310.1 L m?2 h?1 bar?1 with an increase in the PVDF‐co‐CTFE‐g‐P2VP loading (from 0 to 30 wt%) due to increased porosity and hydrophilicity. The flux recovery ratio (FRR) increased from 67.03% to 87.18%, and the irreversible fouling (Rir) decreased from 32.97% to 12.82%. Moreover, the pure gas permeance of the membranes with respect to N2 was as high as 6.2 × 104 GPU (1 GPU = 10–6 cm3[STP]/[s cm2 cmHg]), indicating their possible use as a porous polymer support for gas separation applications.  相似文献

The irreversible inhibition of δ‐chymotrypsin with the enantiomerically pure, P(3)‐axially and P(3)‐equatorially X‐substituted cis‐ and trans‐configurated 2,4‐dioxa‐3‐phospha(1,5,5‐2H3)bicyclo[4.4.0]decane 3‐oxides (X=F, 2,4‐dinitrophenoxy) was monitored by 31P‐NMR spectroscopy. 1H‐Correlated 31P{2H}‐NMR spectra enabled the direct observation of the vicinal coupling (3J) between the P‐atom of the inhibitor and the CH2O moiety of Ser195 (=‘Ser195’(CH2O)), thus establishing the covalent nature of the ‘Ser195’(CH2O? P) bond in the inhibited enzyme. The stereochemical course of the phosphorylation is dependent on the structure of the inhibitor, and neat inversion, both inversion and retention, as well as neat retention of the configuration at the P‐atom was found.  相似文献

The postmodification of poly[9‐(2‐hexyldecyl)‐9H‐carbazole‐2,7‐diyl] ( P1 ) upon its reaction with N‐bromosuccinimide affords exclusive and full bromination of the 3,6‐positions of the carbazole repeat units to yield poly[3,6‐dibromo‐9‐(2‐hexyldecyl)‐9H‐carbazole‐2,7‐diyl] ( P2 ). Brominated polymer P2 can be used as a precursor for further functionalization at the 3,6‐positions with the desired functional group to afford other useful polymers. Polymer P2 has hence been reacted with copper(I) cyanide to afford poly[3,6‐dicyano‐9‐(2‐hexyldecyl)‐9H‐carbazole‐2,7‐diyl] ( P3 ). Full substitution of the bromide groups with nitrile‐functional groups has been achieved. The preparation and structural characterization of polymers P2 and P3 are presented together with studies on their electronic conjugation and photoluminescence properties. Cyclic voltammetry studies on polymer P3 indicate that the new polymer is easier to reduce (n‐dope) but more difficult to oxidize than its unsubstituted counterpart ( P1 ) as a result of the introduction of the electron‐withdrawing nitrile‐functional groups at the 3,6‐positions on the carbazole repeat units on the polymer chains. © 2006 Wiley Periodicals, Inc. With the aim of accessing colloidally stable, fiberlike, π‐conjugated nanostructures of controlled length, we have studied the solution self‐assembly of two asymmetric crystalline–coil, regioregular poly(3‐hexylthiophene)‐b‐poly(2‐vinylpyridine) (P3HT‐b‐P2VP) diblock copolymers, P3HT23b‐P2VP115 (block ratio=1:5) and P3HT44b‐P2VP115 (block ratio=ca. 1:3). The self‐assembly studies were performed under a variety of solvent conditions that were selective for the P2VP block. The block copolymers were prepared by using Cu‐catalyzed azide–alkyne cycloaddition reactions of azide‐terminated P2VP and alkyne end‐functionalized P3HT homopolymers. When the block copolymers were self‐assembled in a solution of a 50 % (v/v) mixture of THF (a good solvent for both blocks) and an alcohol (a selective solvent for the P2VP block) by means of the slow evaporation of the common solvent; fiberlike micelles with a P3HT core and a P2VP corona were observed by transmission electron microscopy (TEM). The average lengths of the micelles were found to increase as the length of the hydrocarbon chain increased in the P2VP‐selective alcoholic solvent (MeOH<iPrOH<nBuOH). Very long (>3 μm) fiberlike micelles were prepared by the dialysis of solutions of the block copolymers in THF against iPrOH. Furthermore the widths of the fibers were dependent on the degree of polymerization of the chain‐extended P3HT blocks. The crystallinity and π‐conjugated nature of the P3HT core in the fiberlike micelles was confirmed by a combination of UV/Vis spectroscopy, photoluminescence (PL) measurements, and wide‐angle X‐ray scattering (WAXS). Intense sonication (iPrOH, 1 h, 0 °C) of the fiberlike micelles formed by P3HT23b‐P2VP115 resulted in small (ca. 25 nm long) stublike fragments that were subsequently used as initiators in seeded growth experiments. Addition of P3HT23b‐P2VP115 unimers to the seeds allowed the preparation of fiberlike micelles with narrow length distributions (Lw/Ln <1.11) and lengths from about 100‐300 nm, that were dependent on the unimer‐to‐seed micelle ratio.  相似文献

Being configuratively fixed and conformationally constrained phosphorus analogues of acetylcholine, they mimic rotamers of acetylcholine and are suitable probes for the investigation of molecular interactions with acetylcholinesterase. Novel thermoresponsive double‐hydrophilic fluorinated block copolymers were successfully synthesized by reversible addition‐fragmentation chain transfer (RAFT) polymerization. Poly[N‐(2,2‐difluoroethyl)acrylamide] (P2F) was synthesized via RAFT polymerization of N‐(2,2‐difluoroethyl)acrylamide (M2F) using 2‐dodecylsulfanylthiocarbonylsulfanyl‐2‐methylpropionic acid (DMP) as the chain transfer agent (CTA) and 2,2′‐azobisisobutyronitrile (AIBN) as the initiator. The resulting P2F macroCTA was further chain extended with N‐(2‐fluoroethyl)acrylamide (M1F) to yield poly{[N‐(2,2‐difluoroethyl)acrylamide]‐b‐[N‐(2‐fluoroethyl)acrylamide]} (P2F‐b‐P1F) block copolymers with different lengths of the P1F block. Molecular weight and molecular weight distribution were determined by gel permeation chromatography. The average molecular weight (Mn) of the resulting polymers ranged from 2.9 × 104 to 5.8 × 104 depending on the length of the P1F block. The molecular weight distribution was low (Mw/Mn = 1.11–1.19). Turbidimetry by UV‐Visble (UV‐Vis) spectroscopy, dynamic light scattering, and in situ temperature‐dependent 1H NMR measurements demonstrated that the P2F block underwent a thermal transition from hydrophilic to hydrophobic, which in turn induced self‐assembly from unimers to aggregates. Transmission electron microscopy studies demonstrated that polymeric aggregates formed from an aqueous solution of P2F‐b‐P1F at 60 °C were disrupted by cooling down to 20 °C and regenerated by heating to 60 °C. Poly[isobutyl methacrylate‐co‐butanediol dimethacrylate‐co‐3‐methacrylylpropylheptaisobutyl‐T8‐polyhedral oligomeric silsesquioxane] [P(iBMA‐co‐BDMA‐co‐MA‐POSS)] nanocomposites with different crosslink densities and different polyhedral oligomeric silsesquioxane (MA‐POSS) percentages (5, 10, 15, 20, and 30 wt %) were synthesized by radical‐initiated terpolymerization. Linear [P(iBMA‐co‐MA‐POSS)] copolymers were also prepared. The viscoelastic properties and morphologies were studied by dynamic mechanical thermal analysis, confocal microscopy, and transmission electron microscopy (TEM). The viscoelastic properties depended on the crosslink density. The dependence of viscoelastic properties on MA‐POSS content at a low BDMA loading (1 wt %) was similar to that of linear P(iBMA‐co‐MA‐POSS) copolymers. P(iBMA‐co‐1 wt % BDMA‐co‐10 wt % MA‐POSS) exhibited the highest dynamic storage modulus (E′) values in the rubbery region of this series. The 30 wt % MA‐POSS nanocomposites with 1 wt % BDMA exhibited the lowest E′. However, the E′ values in the rubbery region for P(iBMA‐co‐3 wt % BDMA‐co‐MA‐POSS) nanocomposites with 15 and 30 wt % MA‐POSS were higher than those of the parent P(iBMA‐co‐3 wt % BDMA) resin. MA‐POSS raised the E′ values of all P(iBMA‐co‐ 5 wt % BDMA‐co‐MA‐POSS) nanocomposites in the rubbery region above those of P(iBMA‐co‐5 wt % BDMA), but MA‐POSS loadings < 15 wt % had little influence on glass‐transition temperatures (Tg's) and slightly reduced Tg values with 20 or 30 wt % POSS. Heating history had little influence on viscoelastic properties. No POSS aggregates were observed for the P(iBMA‐co‐1 wt % BDMA‐co‐MA‐POSS) nanocomposites by TEM. POSS‐rich particles with diameters of several micrometers were present in the nanocomposites with 3 or 5 wt % BDMA. © 2004 Wiley Periodicals, Inc. Two new and a known N‐(O‐carbamoylglucopyranosyl)ansamitocins were isolated from Actinosynnema pretiosum ssp. auranticum ATCC 31565. The known N‐(4‐O‐carbamoyl‐β‐D ‐glucopyranosyl)‐N‐demethylansamitocin P 2 (=ACGP‐2; 1 ) was assigned according to 1D‐ and 2D‐NMR data, and the two new compounds were identified as N‐(6‐O‐carbamoyl‐β‐D ‐glucopyranosyl)‐N‐demethylansamitocin P 2 (=ACGP‐2′; 2 ) and N‐(4‐O‐carbamoyl‐β‐D ‐glucopyranosyl)‐N‐demethylansamitocin P 1 (=ACGP‐1; 3 ) on the basis of spectroscopic data interpretation including 2D‐NMR and tandem MS analysis.  相似文献

As a part of our ongoing research program in developing new anticancer agents, herein, we report the synthesis of a series of 2‐mercapto‐2,3‐dihydrobenzoxazole‐2‐methyl‐, ‐2‐thioethyl‐, and/or 2‐spirothiophene‐phosphonates. Five of these new compounds that combine three bioactive moieties: oxazole, thiol, and phosphonate motifs, exhibited remarkable growth inhibition against 13 carcinoma cell lines. The results of the experimentally determined log K and clog P values are in agreement with Lipinski Rule >5 and showed a strong correlation with the pharmacological results; nevertheless, it is poorly consisted with calculated log P /Clog P and log P /ACD.  相似文献

The synthesis, one‐ and two‐photon absorption (TPA) and emission properties of two novel 2,6‐anthracenevinylene‐based copolymers, poly[9,10‐bis(3,4‐bis(2‐ethylhexyloxy)phenyl)‐2,6‐anthracenevinylene‐alt‐N‐octyl‐3,6‐carbazolevinyl‐ene] ( P1 ) and poly[9,10‐bis(3,4‐bis(2‐ethylhexyloxy)phenyl)‐2,6‐anthracenevinyl‐ene‐alt‐N‐octyl‐2,7‐carbazolevinylene] ( P2 ) were reported. The as‐synthesized polymers have the number‐average molecular weights of 1.56 × 104 for P1 and 1.85 × 104 g mol?1 for P2 and are readily soluble in common organic solvents. They emit strong bluish‐green one‐ and two‐photon excitation fluorescence in dilute toluene solution (? P1 = 0.85, ? P2 = 0.78, λem( P1 ) = 491 nm, λem( P2 ) = 483 nm). The maximal TPA cross‐sections of P1 and P2 measured by the two‐photon‐induced fluorescence method using femtosecond laser pulses in toluene are 840 and 490 GM per repeating unit, respectively, which are obviously larger than that (210 GM) of poly[9,10‐bis‐(3,4‐bis(2‐ethylhexyloxy) phenyl)‐2,6‐anthracenevinylene], indicating that the poly(2,6‐anthracenevinylene) derivatives with large TPA cross‐sections can be obtained by inserting electron‐donating moieties into the polymer backbone. © 2009 Wiley Periodicals, Inc. The novel fungicidal agents, (E )‐5‐[1‐(2‐oxo‐1‐oxaspiro[4,5]dec/non‐3‐en‐3‐yl)ethylidene]‐2‐aminoimidazolin‐ 4‐one derivatives, were designed and synthesized in moderate to excellent yields in four steps using α ‐hydroxyketone and diketene as raw materials and characterized by HR‐ESI‐MS , 1H NMR and X‐ray diffraction. The preliminary bioassay showed that some of these compounds, such as 5e , 6a , 6e , and 7 h exhibit 87.8%, 91.3%, 89.9% and 87.8% inhibition rates against Sclerotinia scleotiorum , 3b , 3c , 4c and 7 h exhibit 96.4%, 92.5%, 90.3% and 76.9% inhibition rates against Phytophthora capsici at the concentration of 50 µg/mL , respectively. These compounds exhibited significant fungicidal activities against S. scleotiorum and P. capsici with EC50 values of 2.56–11.60 µg/mL , and compounds 6e and 7 h exhibited weak inhibition against the spore germination of S. scleotiorum , while the spore germination of P. capsici was strongly inhibited by compound 7 h solution. Scanning electron microscopy (SEM ) and transmission electron microscopy (TEM ) observation indicated that compound 7 h had a significant impact on the structure and function of the hyphal cell wall of P. capsici mycelium.  相似文献

The title compounds, the P(3)‐axially and P(3)‐equatorially substituted cis‐ and trans‐configured 9‐benzyl‐3‐fluoro‐2,4‐dioxa‐9‐aza‐3‐phosphadecalin 3‐oxides (=9‐benzyl‐3‐fluoro‐2,4‐dioxa‐9‐aza‐3‐phosphabicyclo[4.4.0]decane 3‐oxides=7‐benzyl‐2‐fluorohexahydro‐4H‐1,3,2‐dioxaphosphorino[4,5‐c]pyridine 2‐oxides) were prepared (ee >99%) and fully characterized (Schemes 2 and 4). The absolute configurations were deduced from that of their precursors, the enantiomerically pure ethyl 1‐benzyl‐3‐hydroxypiperidine‐4‐carboxylates and 1‐benzyl‐3‐hydroxypiperidine‐4‐methanols which were unambiguously assigned. Being configuratively fixed and conformationally constrained phosphorus analogues of acetylcholine, the title compounds represent acetylcholine mimetics and are suitable probes for the investigation of molecular interactions with acetylcholinesterase. As determined by kinetic methods, all of the compounds are moderate irreversible inhibitors of the enzyme.  相似文献
