High-performance counter-current chromatography has been used for the separation of delphinidin-3-O-sambubioside, cyanidin-3-O-sambubioside and p-coumaric acid from crude extract of cranberry. The separation was performed with a two-phase solvent system composed of butanol/0.05% aqueous trifluoroacetic acid/methanol at a volume ratio of 4:5:1. The two-phase solvent system was selected following the determination of partition coefficients (K) in a range of solvent systems using a robotic solvent system selection method. Analytical scale CCC confirmed that this phase system separated the components from a crude cranberry extract (40 mg scale) with acceptable purities. Preparative CCC of 400 mg of crude yielded 4.2 mg of p-coumaric acid at a purity of over 98%, 3.6 mg of delphinidin-3-O-sambubioside at a purity of over 97% and 4.5 mg of cyanidin-3-O-sambubioside at a purity of 73%, which was further purified by preparative high-performance liquid chromatography to yield 3 mg cyanidin-3-O-sambubioside at 95% purity. The identification of delphinidin-3-O-sambubioside, cyanidin-3-O-sambubioside and p-coumaric acid was performed by ESI-MS, 1H-NMR and 13C-NMR spectra.  相似文献

Reversed-phase HPLC analysis of the methanol extract of the seeds of Centaurea montana afforded a flavanone, montanoside (4), six epoxylignans, berchemol (7), berchemol 4′-O-β-d-glucoside (5), pinoresinol (10), pinoresinol 4-O-β-d-glucoside (8), pinoresinol 4,4′-di-O-β-d-glucoside (6), pinoresinol 4-O-apiose-(1→2)-β-d-glucoside (9), two quinic acid derivatives, trans-3-O-p-coumaroylquinic acid (1), cis-3-O-p-coumaroylquinic acid (2), and eight indole alkaloids, tryptamine (3), N-(4-hydroxycinnamoyl)-5-hydroxytryptamine (11), cis-N-(4-hydroxycinnamoyl)-5-hydroxytryptamine (12), centcyamine (16), cis-centcyamine (17), moschamine (13), cis-moschamine (14) and a dimeric indole alkaloid, montamine (15). While the structures of two new compounds, montanoside (4) and montamine (15), were established unequivocally by UV, IR, MS and a series of 1D and 2D NMR analyses, all known compounds were identified by comparison of their spectroscopic data with literature data. The antioxidant properties of these compounds were assessed by the DPPH assay, and their toxicity towards brine shrimps and cytotoxicity against CaCo-2 colon cancer cells were evaluated by the brine shrimp lethality and the MTT cytotoxicity assays, respectively. The novel dimer, montamine (15), showed significant in vitro anticolon cancer activity (IC50=43.9 μM) while that of the monomer, moschamine (13), was of a moderate level (IC50=81.0 μM).  相似文献

The phytochemical investigation of the methanol extract of the leaves of Castanea sativa Mill., source of the Italian PGI (Protected Geographical Indication) product ‘Marrone di Roccadaspide’ (Campania region) afforded as main compounds crenatin (1), chestanin (2), gallic acid (3), cretanin (4), 5-O-p-coumaroylquinic acid (5), p-methylgallic acid (6) and quercetin-3-O-glucoside (7). To quantify the isolated compounds a LC-ESI(QqQ)MS method working with a very sensitive and selective mass tandem experiment called Multiple Reaction Monitoring (MRM) has been developed. Moreover the antioxidant capacity by TEAC assay and the ability of compounds 17 to protect HaCaT human keratinocytes from UVB-induced damage has been investigated.  相似文献

Acyl glucoses are a group of specialized metabolites produced by Solanaceae. Solanum pennellii, a wild-type tomato plant, produces acyl glucoses in its hair-like epidermal structures known as trichomes. These compounds have been found to be herbicides, microbial growth inhibitors, or allelopathic compounds. However, there are a few reports regarding isolation and investigation of biological activities of acyl glucoses in its pure form due to the difficulty of isolation. Here, we report a new acyl glucose, pennelliiside D, isolated and identified from S. pennellii. Its structure was determined by 1D NMR and 2D NMR, together with FD-MS analysis. To clarify the absolute configuration of the acyl moiety of 2-methylbutyryl in the natural compound, two possible isomers were synthesized starting from β-D-glucose pentaacetate. By comparing the spectroscopic data of natural and synthesized compounds of isomers, the structure of pennelliiside D was confirmed to be 3,4-O-diisobutyryl-2-O-((S)-2-methylbutyryl)-D-glucose. Pennelliiside D and its constituent fatty acid moiety, (S)-2-methylbutanoic acid, did not show root growth-inhibitory activity. Additionally, in this study, chemical synthesis pathways toward pennelliisides A and B were adapted to give 1,6-O-dibenzylpennelliisides A and B.  相似文献

A conformationally rigid chiral molecule LB-I with Lewis basic site has been designed and synthesized in racemic form from ferrocene via Lewis acid mediated diastereoselective cyclization of hydroxy lactam. Both isomers were successfully obtained in enantiomerically pure form through classical resolution using dibenzoyl-d-tartaric acid as the chiral resolving agent in acetone. The nature of the diastereomeric salt formed in the resolution process was investigated by single crystal X-ray crystallographic studies. The absolute configuration of (+)-LB-I was unambiguously assigned as (S,Rp) by single crystal analysis of the salt I obtained from precipitate fraction containing (+)-LB-I and dibenzoyl-d-tartaric acid.  相似文献

Blue sepal-color of Hydrangea macrophylla might be due to a supramolecular metal-complex pigment consisting of delphinidin 3-glucoside (1), co-pigments (5-O-caffeoylquinic acid (2), and/or 5-O-p-coumaroylquinic acid (3)) and Al3+ in an aqueous solution around pH 4.0. To clarify the mechanism of blue sepal-color development of hydrangea, we tried to reproduce the blue color in vitro by mixing 1 with designed synthetic co-pigments in the presence of Al3+ at pH 4.0. We at first succeeded in clarifying the essential functional structure in the co-pigment that could form the stable blue solution. Here, we present the structure of the blue pigment caused by an Al-complex coordinating of 1 at ortho-dihydroxyl groups of the B-ring, 1-hydroxy, 1-carboxylic acid, and the carbonyl residue in the ester at 5-position of 2 and/or 3. The hydrophobic interaction between the aromatic acyl residue at 5-position and the nucleus of 1 may also contribute to stabilize the complex.  相似文献

A 1,4 OO silyl migration followed by nucleophilic substitution with phthalimide was observed under Mitsunobu reaction conditions. This one step secondary alcohol protection and primary alcohol substitution with N-nucleophiles was extended to a variety of 2-hydroxyethyl trialkylsilylether derivatives. A possible mechanism has been postulated based on the pKa values of the alcohol and nucleophile. The present one-pot silyl migration and substitution reaction might find application in the stereoselective synthesis of novel iminosugar derived anti diabetic agents.  相似文献

The enantioseparation of three hydroxyphenylpropionic acid isomers via diastereomeric salt formation with (1R,2S)-2-amino-1,2-diphenylethanol has been demonstrated. The racemates of all three acid isomers were successfully separated with high efficiency (0.56–0.84) after single crystallization. For 2-hydroxy-3-phenylpropionic acid 4, the configuration of the less-soluble salt was controlled by the crystallization solvent: the (R)-4 salt was crystallized from water, while 2-propanol afforded the (S)-4 salt. The chiral recognition mechanism of the three acids was discussed based on the crystal structures of the diastereomeric salts.  相似文献

The stereoselective total syntheses of botcinic acid, botcinic acid methyl ester, 3-O-acetylbotcinic acid methyl ester, botcineric acid, and botcinin E were achieved. The structures of these compounds have been unequivocally determined through their total synthesis, and they are identified with the revised structures of botcinolide, 4-O-methylbotcinolide, 3-O-acetyl-5-O-methylbotcinolide, homobotcinolide, and 2-epibotcinolide, respectively.  相似文献

Acid-promoted α-stereoselective O-glycosidation of 1-exo-methylenesugars was successfully applied to the synthesis of a PI-88 analogue. By using methanesulfonic acid as a promoter, 1′-C-methyl-α-disaccharides with p-methoxybenzyl protection were obtained in high yield. The synthesis of 2′-O,3′-O bicyclic adenosine derivatives is presented as the first examples of a new family of 13-membered ring bicyclic nucleoside analogues. From the leaves of Schefflera sessiliflora De P. V., one new C20-gibberellin diterpene 2β,12β-dihydroxygibberellin (12β-hydroxy-GA110 or 2β-hydroxy-GA112) (1), together with three known compounds, trans-tiliroside (2), kaempferol 3-O-β-d-glucuronopyranoside (3), 5-p-trans-coumaroylquinic acid (4), was isolated for the first time from the genus Schefflera by various chromatography methods. Their structures were elucidated by IR, UV, HR-ESI-MS, NMR 1D and 2D experiments and comparison with previous reported data. The α-glucosidase inhibitory activity of all compounds was measured. The isolates (2, 3) showed better α-glucosidase inhibitory activity (IC50 = 134.60, 147.10 μM, respectively) than the standard drug acarbose (IC50 = 214.50 μM).  相似文献

Gynura bicolor (Compositae) is a popular vegetable in Asia and believed to confer a wide range of benefits including anti-cancer. Our previous findings showed that the ethyl acetate extract of G. bicolor possessed cytotoxicity and induced apoptotic and necrotic cell death in human colon carcinoma cells (HCT 116). A combination of column chromatography had been used to purify chemical constituents from the ethyl acetate and water extract of G. bicolor leaves. Eight chemical constituents 5-p-trans-coumaroylquinic acid (I), 4-hydroxybenzoic acid (II), rutin (III), kampferol-3-O-rutinoside (IV), 3,5-dicaffeoylquinic acid (V), kampferol-3-O-glucoside (VI), guanosine (VII) and chlorogenic acid (VIII) were isolated from G. bicolor grown in Malaysia. To our best knowledge, all chemical constituents were isolated for the first time from G. bicolor leaves except rutin (III). 3,5-dicaffeoylquinic acid (V), guanosine (VII) and chlorogenic acid (VIII) demonstrated selective cytotoxicity (selective index>3) against HCT 116 cancer cells compared to CCD-18Co human normal colon cells.  相似文献

Butterfly pea flower have great sensory attraction, but they have not yet been used widely in Vietnam. Extracts of butterfly pea flowers can be used conveniently as a natural blue colorant for food products. In this study, the identification of anthocyanin compounds in butterfly pea flowers was performed by UPLC coupled with a UV and Mass spectrometer instrument. Positive and negative ion electrospray MS/MS chromatograms and spectra of the anthocyanin compounds were determined. By analyzing the chromatograms and spectra for each ion, five anthocyanins were identified in the butterfly pea flower extract; these were delphinidin-3-(6″-p-coumaroyl)-rutinoside, cyanidin 3-(6″-p-coumaroyl)-rutinoside, delphinidin-3-(p-coumaroyl) glucose in both cis- and trans- isomers, cyanidin-3-(p-coumaroyl-glucoside) and delphinidin-3-pyranoside. Additionally, based on their intensity, it was determined that cyanidin-3-(p-coumaroyl-glucoside) was the most abundant anthocyanin, followed by cyanidin 3-(6″-p-coumaroyl)-rutinoside, delphinidin-3-(p-coumaroyl-glucoside), delphinidin-3-(6″-p-coumaroyl)-rutinoside and delphinidin-3-pyranoside. In this study, cyanidin derivatives were discovered in butterfly pea flower extract, where these compounds had not been detected in previous studies.  相似文献

Purification of seven regio-isomers of O-decanoyl sucrose, 2-O-, 3-O-, 4-O-, 6-O-, 3′-O-, 4′-O- and 6′-O-decanoyl sucrose, were performed by LC followed by preparative RP-HPLC with ELSD. Using an optimized gradient of acetonitrile in water 2-O-, 3-O-, 6-O- and 3′-O-decanoyl sucrose were purified in yields (w/w) of 52.5%, 34.7%, 45.0% and 36.9%, respectively. In the purified preparations of the 2-O- and 3′-O-decanoyl sucrose respectively, acyl migration was observed as a result of the drying process. Lyophilization resulted in the highest purities (w/w) of 96% and 100% for the 2-O- and the 3′-O-decanoyl sucrose, respectively.  相似文献

Four novel isomers of norlignan glycoside were isolated from Cephalotaxus oliveri Mast.. Their structures were elucidated as 3S-4″-O-β-d-glucopyranosylnyasol 1, 3S-4′-O-β-d-glucopyranosylnyasol 2, 3S-4″-O-β-d-glucopyranosylhinokiresinol 3, 3S-4′-O-β-d-glucopyranosylhinokiresinol 4 by extensive spectroscopic methods including 1D and 2D NMR experiments (1H, 13C, DEPT, 1H–1H COSY, HSQC, HMBC, ROESY) along with HR-ESIMS and comparison to literature data. The trisaccharide 2-O-(α-L-fucopyranosyl)-3-O-(α-D-galactopyranosyl)-D-galactose has been synthesised stereospecifically using the imidate procedure. Allyl 3-O-benzoyl-4,6-O-benzylidene-β-D-galactopyranoside was first α-L-fucosylated by 1-O-(N-methyl)-acetimidyl-2,3,4-tri-O-benzyl-β-L-fucopyranose then, after O-debenzoylation, α-D-galactosylated by 1-O-(N-methyl)-acetimidyl 2,3,4,6-tetra-O-benzyl-β-D-galactopyranose. The resulting tri-saccharide has also been obtained from allyl 2-O-benzoyl-4,6-O-benzylidene-β-D-galactopyranoside after α-D-galactosylation, O-debenzoylation and α-L-fucosylation. The glycosylations were performed at room temperature in nitromethane in the presence of p-toluenesulfonic acid. Deallylation followed by catalytic hydrogenolysis gave the B blood-group antigenic determinant. The allyl group was also selectively transformed into hydroxyethyl group.  相似文献

The potential of an N-Troc-protected sialic acid donor, equipped with phenylsulfenyl functionality as a leaving group, has been explored. As a result, the entitled donor was proven to be highly reactive and to have broad applicability toward the synthesis of variant sialo-glycans, which have N-glycolyl, de-N-acetyl, 1,5-lactam and 8-O-sulfo sialic acid analogs.  相似文献
