We report a simple assay for visual detection of single nucleotide polymorphisms (SNPs) with good sensitivity and selectivity. The selectivity is determined by Escherichia coli (E. coli) DNA ligase mediated circular formation upon recognition of the point mutation on DNA targets. Rolling cycle amplification (RCA) of the perfect-matched DNA target is then initiated using the in situ formed circular template in the presence of Phi29 enzyme. Due to amplification of the DNA target, the RCA product has a tandem-repeated sequence, which is significantly longer than that for the SNP strand. Direct addition of a cationic conjugated polymer of poly[9,9'-bis(6'-(N,N,N-trimethylammonium)hexyl)fluorene-co-9,9'-bis(2-(2-(2-(N,N,N-trimethylammonium)ethoxyl)-ethoxy)-ethyl)fluorene tetrabromide] containing 20 mol% 2,1,3-benzothiadiazole (PFBT(20)) into the RCA solution leads to blue-whitish fluorescent color for SNP strand and yellowish fluorescent color for amplified DNA, due to PFBT(20)/DNA complexation induced intrachain/interchain energy transfer. To further improve the contrast for visual detection, FAM-labeled peptide nucleic acid (PNA) was hybridized to each amplified sequence, which is followed by the addition of poly{2,7-[9,9-bis(6'-N,N,N-trimethylammoniumhexyl)]fluorene-co-2,5-difluoro-1,4-phenylene dibromide} (PFP). The PNA/DNA hybridization brings PFP and FAM-PNA into close proximity for energy transfer, and the solution fluorescent color appears green in the presence of target DNA with a detection limit of 1 nM, which is significantly improved as compared to that for most reported visual SNP assay.  相似文献

Novel polyfluorene copolymers with pendant hydroxyl groups, poly[2,7‐(9,9‐dihexylfluorene)‐2,7‐(9,9‐bis(6‐hydroxyhexyl)fluorene)‐co‐2,7‐(9,9‐dihexylfluorene)‐1,4‐phenylene] (PFP‐OH) and poly[2,7‐(9,9‐dihexylfluorene)‐2,7‐(9,9‐bis(6‐hydroxyhexyl)fluorene)‐co‐2,7‐(9,9‐dihexylfluorene)‐4,7‐(2,1,3‐benzothiadiazole)] (PFBT‐OH) were prepared. Acid‐catalyzed polycondensations of tetraethoxysilane were carried out in the presence of these polymers to obtain homogeneous hybrids. Photoluminescence spectra of these hybrids suggested the polymers were immobilized in silica matrix retaining their π‐conjugated structures. Further, hybrids of coat film were prepared utilizing perhydropolysilazane as a silica precursor. Their optical properties were examined. © 2010 Wiley Periodicals, Inc. Developing low-cost and efficient photocatalysts to convert CO2 into valuable fuels is desirable to realize a carbon-neutral society. In this work, we report that polymer dots (Pdots) of poly[(9,9′-dioctylfluorenyl-2,7-diyl)-co-(1,4-benzo-thiadiazole)] (PFBT), without adding any extra co-catalyst, can photocatalyze reduction of CO2 into CO in aqueous solution, rendering a CO production rate of 57 μmol g−1 h−1 with a detectable selectivity of up to 100 %. After 5 cycles of CO2 re-purging experiments, no distinct decline in CO amount and reaction rate was observed, indicating the promising photocatalytic stability of PFBT Pdots in the photocatalytic CO2 reduction reaction. A mechanistic study reveals that photoexcited PFBT Pdots are reduced by sacrificial donor first, then the reduced PFBT Pdots can bind CO2 and reduce it into CO via their intrinsic active sites. This work highlights the application of organic Pdots for CO2 reduction in aqueous solution, which therefore provides a strategy to develop highly efficient and environmentally friendly nanoparticulate photocatalysts for CO2 reduction.  相似文献

Conjugated polymers (CPs) have been used as highly responsive optical sensors for chemical and biological targets. The density functional theory (DFT), the time-dependent density functional theory (TDDFT) and the singly excited configuration interaction (CIS) approach were employed to simulate the absorption and emission spectra of a model monomer unit of the typical CP, poly[9,9′-bis-(6′-N,N,N-trimethylammonium)hexyl]fluorene-alt-4,7-(2,1,3-benzothiadiazole)] (PFBT). Various basis sets were applied in the calculations and their effects on the predictions for the optical properties of FBT are discussed. The results suggest that the TD-B3LYP method with basis sets larger than 6-311G(d,p) provides a suitable approach for investigations of the studied system.  相似文献

Abstract

Bi- and trifunctional monomers with 2,3-dihydrofuranyl moieties were synthesized. The polymerizable dihydrofuranyl groups were connected to each other with ester and ether linkages. Alkyl-, alkenyl- and arylspacers were used. The synthesized monomers were copolymerized with 2,3-dihydrofuran by photoinduced cationic polymerization. As photo acid (η5-cyclopentadienyl)-Fe(II)-(η6-isopropylbenzene) hexafluorophosphate was used. The thermal stability of the crosslinked poly(2,3-tetrahydrofurandiyl)s is dependent of the monomer structure. Higher stability was observed for poly(tetrahydrofurandiyl)s crosslinked with bis(2,3-dihydrofuranyl) compounds instead of tetraethyleneglycol bis-2-propenyl-ether.  相似文献

An electrostatic complex of water‐soluble conjugated polyelectrolytes (CPs) between anionic poly(9,9‐bis(4′‐sulfonatobutyl)fluorene‐co‐alt‐1,4‐phenylene) disodium salt (a‐PFP) and cationic poly(9,9‐bis((6′‐N,N,N,‐trimethylammonium)hexyl)fluorene‐co‐2,1,3‐bezothiadiazole) dibromide (85:15) (c‐PFB15) was tested as a fluorescence resonance energy transfer (FRET) donor to Texas Red (TR)‐labeled single‐stranded DNA (ssDNA‐TR) via two‐step FRET processes. Electrostatic complexation of a‐PFP and c‐PFB15 in water leads to aggregation of polymer chains, a concomitant reduction of intersegment distances, and energy transfer to the benzothiadiazole (BT) segments. The following complexation with ssDNA‐TR leads to energy transfer from BT to TR via two‐step FRET processes. This detection schematic shows an FRET‐induced signal amplification, which can be achieved by adjusting the charge ratio in the cationic/anionic CP complex and controlling the number density of the binding CPs around the acceptor, resulting in enhanced antenna effects and sensitivity in CP‐based FRET DNA detection assays.

Three novel fluorene‐containing poly(arylene ethynylene)s with amino‐functionalized side groups were synthesized through the Sonogashira reaction. They were poly{9,9‐bis[6′‐(N,N‐diethylamino)hexyl]‐2,7‐fluorenylene ethynylene}‐altco‐{2,5‐bis[3′‐(N,N‐diethylamino)‐1′‐oxapropyl]‐1,4‐phenylene} ( P1 ), poly{9,9‐bis[6′‐(N,N‐diethylamino)hexyl]‐2,7‐fluorenylene ethynylene} ( P2 ), and poly({9,9‐bis[6′‐(N,N‐diethylamino)hexyl]‐2,7‐fluorenylene ethynylene}‐altco‐(1,4‐phenylene)) ( P3 ). Through the postquaternization treatment of P1 – P3 with methyl iodide, we obtained their cationic water‐soluble conjugated polyelectrolytes (WSCPs): P1′ – P3′ . The water solubility was gradually improved from P3′ to P1′ with increasing contents of hydrophilic side chains. After examining the ultraviolet–visible absorption and photoluminescence (PL) spectra, fluorescence lifetimes, and dynamic light scattering data, we propose that with the reduction of the water solubility from P1′ to P3′ , they exhibited a gradually increased degree of aggregation in H2O. The PL quantum yields of P1′ – P3′ in H2O displayed a decreasing tendency consistent with the increased degree of aggregation, suggesting that the pronounced degree of aggregation was an important reason for the low PL quantum yields of WSCPs in H2O. Two structurally analogous water‐soluble trimers of P2′ and P3′ , model compounds 2,7‐bis(9″,9″‐bis{6‴‐[(N,N‐diethyl)‐N‐methylammonium] hexyl}‐2″‐fluorenylethynyl)‐9,9‐bis{6′‐[(N,N‐diethyl)‐N‐methylammonium]hexyl}fluorene hexaiodide and 1,4‐bis(9′,9′‐bis{6″‐[(N,N‐diethyl)‐N‐methylammonium]hexyl}‐2′‐fluorenylethynyl)benzene tetraiodide, were synthesized. The amplified fluorescence quenching of these WSCPs by Fe(CN)64− in H2O was studied by comparison with a corresponding analogous trimer. We outline a theory for the optical absorption of the β‐phase of poly(9,9‐dioctylfluorene) (PFO) that is based on the Frenkel exciton model. The absorption peak at 435 nm is attributed to polymer segments having torsion angles equal to π that are weakly perturbed by the presence of random monomer junctions with torsion angles equal to 0. The broad band below 435 nm is associated with disordered segments having a broad distribution of random torsion angles. The effects of small random deviations from π in the torsion angles are discussed. The calculations support the interpretation that the β‐phase is characterized by alternating segments of highly ordered and strongly disordered regions. PFO is a widely studied fluorene‐based polymer with interesting and potentially useful photophysical properties. In this work, Frenkel exciton states in the β‐phase of PFO are studied, and a two‐region model—weak torsional disorder and strong torsional disorder—is presented. The peak in the optical absorption at 435 nm is associated with the weakly disordered regions. Abstract

Acylation of N,O-bistrimethylsilyl-3-aminobenzoic acid with 3-acetoxybenzoylchloride yielded the trimethylsilylester of N-(3′-acetoxybenzoyl)-3-aminobenzoic acid, which was polycondensed in situ at 260 or 280°C. Cocondensation with acetylated tetraphenols yielded four-arm star copolymers with a random or preferentially alternating sequence of 3-hydroxy and 3-aminobenzoyl units. Due to ester-amide exchange detected by 1H- and 13C-NMR spectroscopy, the sequences were never perfectly alternating. Methyl groups attached to the star centers allowed the determination of degrees of polymerization by 1H-NMR spectroscopy. Acylation of N,O-bistrimethylsilyl-3-amino benzoic acid with 3,5-bisacetoxybenzoylchloride yielded a trifunctional monomer, the polycondensation of which yielded a hyperbranched poly(ester-amide). By cocondensation of the trifunctional monomer with acetylated tetraphenyl, star-shaped poly(ester-amide)s with four hyperbranched star arms were obtained. All these poly(ester-amide)s are amorphous materials with glass-transition temperatures in the 190–200°C range and good solubility in polar organic solvents.  相似文献

To open out new aspects of 9,9‐diarylfluorene (DAF)‐based polymers with high performances, 9,9‐(4‐hydroxyphenyl)‐4,5‐diazafluorene ( N‐BPF ) was designed as a new cardo structure and the properties of poly(ether ketone)s ( N‐PEKs ) containing N‐BPF skeletons were examined in detail. N‐PEKs were synthesized in high yields via polycondensation of N‐BPF with difluoroarenes. N‐PEKs showed cardo polymer‐specific properties such as high thermal stability and high solubility in organic solvents. The addition of p‐toluenesulfonic acid (TsOH) to N‐PEK resulted in the formation of network polymer based on interchain hydrogen bonds. It turned out that the films of network polymer are flexible and transparent and exhibit high refractive index and low birefringence. We have developed a new intermediate monomer, 2,7‐[bis(4,4,5,5‐tetramethyl‐1,3,2‐dioxaborolan‐2‐yl)‐9,9‐bis(3‐(tert‐butyl propanoate))]fluorene, that allows the easy synthesis of water‐soluble carboxylated polyfluorenes. As an example, poly[9,9′‐bis(3′′‐propanoate)fluoren‐2,7‐yl] sodium salt was synthesized by the Suzuki coupling reaction, and the properties of the polymer were studied in aqueous solutions of different pH. Fluorescence quenching of the polymer by different cationic quenchers (MV2+, MV4+, and NO2MV2+; MV=methyl viologen) was studied, and the quenching constants were found to be dependent on the charge and electron affinity of the quencher molecule and the pH of the medium. The largest quenching constant was observed to be 1.39×108 M ?1 for NO2MV2+ at pH 7. The change in polymer fluorescence upon interaction with different proteins was also studied. Strong fluorescence quenching of the polymer was observed in the presence of cytochrome c, whereas weak quenching was observed in the presence of myoglobin and bovine serum albumin. Lysozyme quenched the polymer emission at low protein concentrations, and the quenching became saturated at high protein concentrations. Under similar experimental conditions, the polymer showed improved quenching efficiencies toward cationic quenchers and a more selective response to proteins relative to other carboxylated conjugated polymers.  相似文献

Five fluorene-based conjugated copolymers were studied to explore the effect of acceptor on the electronic and optical properties. Their ground-state, excited-state electronic structures and the tunable optical properties were theoretically investigated using density functional theory (DFT) and time-dependent density functional theory (TDDFT) methods. The acceptors including quinoxaline (Q), 2,1,3-benzothiadiazole (BT), thieno[3,4-b]pyrazine (TP), 2,1,3-benzooxadiazole (BO), and pyridopyrazine (PP) can significantly influence the copolymers’ electronic structures, molecular orbitals, geometric conformations, and optical properties. Calculations were made on systems containing one, two, three, and four oligomers in the neutral, cationic, and anionic structures, which can be extrapolated to infinite chain length polymers. The result indicated that the sequence of the band gap was on the reverse trend of emission wavelength. The strong electron-withdrawing strength of TP unit and coplanar backbone in poly[2,7-(9,9′-dihexylfluorene)-alt-2,3-dimethyl-5,7-dithien-2-yl-thieno[3,4-b] pyrazine] resulted in the enhanced degree of intramolecular charge transfer (ICT) and lowest band gap. The contribution of acceptors to IP was also found to follow the sequence of TP < Q < PP < BT < BO. The absorption and emission spectra exhibited red-shift with increasing the conjugation lengths. The present study suggested that the electronic and optical properties of donor–acceptor conjugated copolymers were affected by the acceptor structure.  相似文献

The cationic ring‐opening multibranching polymerization of 2‐hydroxymethyloxetane ( 1 ) as a novel latent AB2‐type monomer was carried out using trifluoromethane sulfonic acid or trifluoroboron diethyl etherate by a slow‐monomer‐addition (SMA) method. The polymer yield of poly‐1 ranged from ca. 58–88%, which increase with the increasing monomer addition time on the SMA method. The absolute molecular weights (Mw,MALLS) and the polydispersities of poly‐1 were in the range of 8,000–43,500 and 1.45–4.53, respectively, which also increased with the increasing monomer addition time. The Mark‐Houwink‐Sakurada exponents α in 0.2 M NaNO3 aq. were determined to be 0.02–0.25 for poly‐1 , indicating that poly‐1 has compact forms in the solution because of the highly branched structure. The degree of the branching value of poly‐1 , which was calculated by Frey's equation, ranged from ca. 0.50 to 0.58, which increased with the increasing monomer addition time. The steady shear flow of poly‐1 in aqueous solution exhibited a Newtonian behavior with steady shear viscosities independent of the shear rate. The results of the MALLS, NMR, and viscosity measurements indicated that poly‐1 is composed of a highly branched structure, i.e., the hyperbranched poly (2‐hydroxymethyloxetane). © 2011 Wiley Periodicals, Inc. The effect of a cationic monomer (N,N,N,N-trimethyl[methacryloxyethyl]ammonium methyl sulfate) on the formation, structure, and local dynamics of associates resulted from the interaction of the monomer with sodium dodecyl sulfate in aqueous solutions was studied by ESR spectroscopy. In the presence of the monomer, micelles are formed at concentrations much lower than the CMC of the pure surfactant with the monomer molecules that form a condensed layer of counterions around a micelle of sodium dodecyl sulfate. The binding of surfactant micelles with the cationic monomer causes a significant decrease in the local molecular mobility of dodecyl sulfate ions.  相似文献

9,9-Bis[4-(p-aminophenoxy)phenyl]fluorene ( II ) was used as a monomer with various aromatic dicarboxylic acids and tetracarboxylic dianhydrides to synthesize polyamides and polyimides, respectively. The diamine II was derived by a nucleophilic substitution of 9,9-bis(4-hydroxyphenyl)fluorene with p-chloronitrobenzene in the presence of K2CO3 and then hydro-reduced. Polyamides IV a-g having inherent viscosities of 0.73–1.39 dL/g were prepared by the direct polycondensation of the diamine II with various aromatic diacids using triphenyl phosphite and pyridine as condensing agents. All the aromatic polyamides were amorphous and readily soluble in various polar solvents such as N,N-dimethylacetamide, N,N-dimethylformamide, dimethylsulfoxide, and N-methyl-2-pyrrolidone. Transparent and flexible films of these polymers could be cast from the DMAc solutions. These aromatic polyamides had glass transition temperatures in the range of 283–309°C and 10% weight loss occurred up to 460°C. The polyimides were synthesized from diamine II and various aromatic dianhydrides via the two-stage procedure that included ring-opening poly-addition in DMAc to give poly(amic acid)s, followed by thermal or chemical conversion to polyimides. The poly(amic acid)s had inherent viscosities of 0.62–1.78 dL/g, depending on the dianhydrides. Most of the aromatic polyimides obtained by chemical cyclization were found to be soluble in NMP. These polyimides showed almost no weight loss up to 500°C in air or nitrogen atmosphere. © 1993 John Wiley & Sons, Inc.  相似文献

Rh‐catalyzed polymerization of a new N‐propargylamide monomer having a galactose residue was investigated. The polymerization proceeded in a mixed solvent of THF and water to produce the galactose‐carrying poly(N‐propargylamide) consisting of cis‐isomer. Summary: By the Suzuki coupling reaction of 9,9-dioctyl-2,7-bis(1,3,2-dioxaborinan-2-yl)fluorene ( I ) and 3,5-di-tert-butylphenyl 2,5-dibromobenzenesulfonate ( II ) the alternating poly{[9,9-dioctylfluoren-2,7-diyl]-alt-[2-(3,5-di-tert-butyl-phenoxysulfonyl)-1,4-phenylene]} ( III ) was synthesized. Alkaline hydrolysis of III gave a conjugated polyelectrolyte carrying sulfonic acid groups ( IV ). Monomers 2,5-dibromo-3-[2-(pyren-1-yl)vinyl]thiophene and 2,5-dibromo-3-[2-(quinolin-4-yl)vinyl)thiophene were prepared and copolymerized with I to afford poly{[9,9-dioctylfluoren-2,7-diyl]-alt-[3-(2-(pyren-1-yl)vinyl)thiophen-2,5-diyl]} ( V ) and poly{[9,9-dioctylfluoren-2,7-diyl]-alt-[3-(2-(quinolin-4-yl)-vinyl)thiophen-2,5-diyl] ( VI ), respectively. Conjugated backbone of V contains the conjugated pyrene unit in the side chain. Similarly the side chain of VI contains the conjugated quinoline structure unit which can be for instance protonated. By the Suzuki polycondensation reaction of I and of the prepared methyl 3-(2,7-dibromocarbazole-9-yl)propionate ( VII ) the new poly{[9,9-dioctylfluorene-2,7-diyl]-alt-[9-(2-methoxycarbonylethyl)carbazole-2,7-diyl]} ( VIII ) was synthesized and characterized.  相似文献

High molecular weight trans‐poly(9,9‐di‐n‐octylfluorene‐2,7‐vinylene) was prepared under reduced pressure in the presence of a well‐defined Schrock‐type catalyst, Mo(CHCMe2Ph)(N‐2,6‐Me2C6H3)[OCMe(CF3)2]2, in toluene. For the first time, organic semiconducting polymer dots (Pdots) based on poly[(9,9′‐dioctylfluorenyl‐2,7‐diyl)‐co‐(1,4‐benzo‐{2,1′,3} thiadiazole)] (PFBT) and polystyrene grafting with carboxyl‐group‐functionalized ethylene oxide (PS‐PEG‐COOH) are introduced as a photocatalyst towards visible‐light‐driven hydrogen generation in a completely organic solvent‐free system. With these organic Pdots as the photocatalyst, an impressive initial rate constant of 8.3 mmol h?1 g?1 was obtained for visible‐light‐driven hydrogen production, which is 5‐orders of magnitude higher than that of pristine PFBT polymer under the same catalytic conditions. Detailed kinetics studies suggest that the productive electron transfer quench of the excited state of Pdots by an electron donor is about 40 %. More importantly, we also found that the Pdots can tolerate oxygen during catalysis, which is crucial for further application of this material for light‐driven water splitting.  相似文献
