Quinazolincs and 1,4-benzodiazepines. LXXXVI. The synthesis of imidazothienodiazepines and imidazopyrazolodiazepines

The thieno[3,2-e][1,4]diazepin-2-one ( 1a ), the thieno[2,3-e] [ 1,4] diazepin-2-one ( 1b ), the pyrazolo[3,4-e][1,4]diazepin-2-one ( 1c ) and a chloro analog of 1b , compound 1d , were each converted to derivatives of the novel tricyclic ring systems 4H-imidazo[1,5-a]thieno[2,3-f] [1,4]-diazepine, 4Himidazo[1,5a]thieno[2,3f][1,4]diazepine and 4H-imidazo[ 1,5-a]pyrazolo[4,3-f]-[1,4]diazepine. Depending on the substituents desired on the imidazo ring, two different synthetic pathways were employed.

The synthesis of three novel polycyclic heterocyclic ring systems are reported via photocyclization. The specific final products in these ring systems are: phenanthro[9′,10′:4,5]thieno[2,3-c]quinoline ( 13 ), benzo[h]-phenanthro[9′,10′:4,5]thieno[2,3-c]quinoline ( 14 ), and benzo[f]phenanthro[9′,10′:4,5]thieno[2,3-c]quinoline ( 15 ).

The synthesis of two novel polycyclic heterocyclic ring systems via photocyclization are reported. These are benzo[f]thieno[2′,3′:4,5]thieno[2,3-c]quinoline and benzo[A]thieno-[2′,3′:4,5]thieno[2,3-c]quinoline. The total assignment of their ¹H- and ¹³C-nmr spectra was determined by utilizing two-dimensional nmr spectroscopic methods.     

Synthesis of 5H-Triazolo[1,5-d]- and 5H- Tetrazolo- [1,5-d]thieno[3,2-f]-1,4-diazepin-6(7H)-ones

5H-Triazolo[1,5-d]- and 5H-tetrazolo[1,5-d]thieno[3,2-f]-1,4-diazepin-6(7H)-ones have been obtained by the base catalysed ring expansion reaction of 5-chloromethyl-1,2,4-triazolo[1,5-c]- and 5-chloromethyltetrazolo- [1,5-c]thieno[3,2-e]pyrimidines. The required thienopyrimidine derivatives were synthesized from 2-amino-3-triazolyl- and 2-amino-3-tetrazolylthiophenes by acylation, followed by dehydrative cyclization.     

The synthesis of novel polycyclic heterocyclic ring systems via photocyclization. 19. Thieno[3′,2′:4,5]thieno[2,3-c]-naphtho[1,2-f]quinoline,thieno[3′,2′:4,5]thieno[2,3-c]naphtho-[1,2-f][1,2,4]triazolo[4,3-a]quinoline and thieno[3′,2′:4,5]-thieno[2,3-c]naphtho[1,2-f]tetrazolo[1,5-a]quinoline

Photocyclization of 3-chloro-N-(3-phenanthryl)thieno[2,3-b]thiophene-2-carboxamide ( 5 ) yielded only one of the two possible structural isomers, thieno[3′,2′:4,5]thieno[2,3-c]naphtho[1,2-f]quinolin-6(5H)-one ( 6 ), which was further elaborated to afford the unsubstituted ring system 10 , its triazole 11 and tetrazole 12 . The structural confirmation of 10 was achieved by the total assignment of its ¹H and ¹³C nmr spectra by the concerted utilization of two-dimensional nmr spectroscopic methods.     

Benzomorphan analogues: Synthesis of some thieno[2,3-f] morphans

A series of N-substituted 5,9-dimethylthieno[2,3-f]morphans and 5-phenylthieno[2,3-f] - morphans has been prepared starting from 3,4-dimethylpyridine and 4-phenylpyridine and 3-thenylbromide by an application of the Grewe morphinan synthesis. The thieno [2,3-f]morphans were tested for analgetic activity, but only weak analgetic activity relative to toxicity and no morphine antagonism activity was observed.     

Assignments of the 1H and 13C NMR spectra of pseudo-symmetric heterocyles using the HMQC-TOCSY experiment to differentiate overlapping spin systems

The ¹H nmr spectra of phenanthro[9′,10′:4,5]thieno[2,3-c]quinoline, benzo[f]phenanthro-[9′,10′:4,5]thieno[2,3-c]quinoline and benzo[h]phenanthro[9′,10′:4,5]thieno[2,3-c]quinoline are highly congested. For each compound, all protons abide in an aromatic environment complicated by pseudo-symmetric regions which result in multiple overlap of the different spin systems these molecules contain. We illustrate here the utility of the HMQC-TOCSY experiment to identify spin systems when the proton spectrum is highly congested. To complete the assignment of the ¹H and ¹³C nmr spectra of each compound the HMBC experiment is used to assign the quaternary carbons.     

Studies in sulphur heterocyles. 2—The 13C NMR spectra of thienocoumarins

¹³C NMR spectra of 3- substituted thieno[2,3-h][1]benzopyran-2-ones and 8-substituted thieno[3,2-f]-[1]benzopyran-7-ones are reported and the substituent dependence of certain ¹³C chemical shifts is noted.     

Convenient synthesis of fused pyrano[3,2-h]- and furo[3,2-h]benzo[f]coumarins from naphthalene-2,3-diol

The treatment of 8-propargyloxy-benzo[f]coumarin with boron trifluoride diethyl etherate in N,N-dimethylformamide under reflux or MW irradiation resulted in pyrano[3,2-h]benzo[f]coumarin, while the furo[3,2-h]benzo[f]coumarin is received from the treatment with N-methylformamide under MW irradiation.     

Tetracyclic systems: Synthesis of isoindolo[1,2-b]thieno-[2,3(3,2 or 3,4)-e][1,3]thiazocines and Isoindolo[2,1-a]thieno-[2,3(3,2 or 3,4)-f][1,4] and [1,5]diazocines

Cyclization of thioglycolic acids derivatives 3a-d gave isoindolo[1,2-b]thieno[2,3(3,2 or 3,4)-e][1,3]-thiazocines 4a-d . Isoindolo[2,1-a]thieno[2,3(3,2 or 3,4)-f][1,4] or [1,5]diazocines 10b or 11a-c were synthesized from Beckmann or Schmidt rearrangement of the ketones 7a-c .     

Benzomorphan related compounds. VII. Synthesis of benzothienomorphans

The synthesis of some benzo [b] thieno [2,3-f] - and [3,2-f] morphans by the Grewe method is described. Spectroscopic data of these compounds and of the intermediate isomeric tetrahydropyridines are included. A side-product isolated from the aluminum bromide cyclization of the 2-(2-benzothienylmethyl)tetrahydropyridine IVd is reported and a mechanism for its formation is proposed.     

Intramolecular cyclization of amido acids into pyrrolidinothieno(or [1]benzothieno)[3]azepinediones

The synthesis of pyrrolidino[2,1-b]thieno[3,2(2,3)-f][3]azepinediones 7a,b and pyrrolidino[2,1-b]-[1]benzothieno[3,2(2,3)-f][3]azepinediones 7c,d are described starting from thiophenes or [1]benzothio-phenes acetic acids. Their selective reduction using triethylsilane led to the corresponding azepinones 17a-d.     

Intramolecular 4+2 cycloaddition of thieno[2,3-e][1,2,4]triazines: routes towards condensed thieno[2,3-b]pyridines

Synthetic approaches towards new condensed thienopyridine ring systems including furo[2,3-b]thieno[3,2-e]pyridines, bisthieno[2,3-b:3′,2′-e]pyridines, 5H-chromeno[2,3-b]thieno[3,2-e]pyridines, 5H-benzo(f)chromeno[2,3-b]thieno[3,2-e]pyridines have been achieved by application of intramolecular 4+2 cycloaddition reactions of suitably designed thieno[2,3-e][1,2,4]triazines tethered with alkene or alkyne terminals.     

Synthesis of 2-(3-coumarinyl)pyrrolo[2,1-<Emphasis Type="Italic">a</Emphasis>]isoquinoline derivatives by the chichibabin reaction

2-(3-Coumarinyl)-5,5-dimethyl-5,6-dihydropyrrolo[2,1-a]isoquinolines were synthesized by the Chichibabin reaction of 3,3-dimethyl-1,2,3,4-tetrahydroisoquinoline with 3-bromoacetylcoumarin. Derivatives of benzo[f]isoquinoline and benzo[f]coumarin were obtained similarly.     

A new condensed system,benzothienofurazanothiepine trioxide

The dimetallation of phenyl thienyl sulfones was used to obtain derivatives of benzo[b]thieno[3,2-f]fura2ano[3,4-d]thiepine-1(3),7,7-trioxide, which is anew condensed system.Translated from Izvestiya Akademii Nauk SSSR, Seriya Khimicheskaya, No. 4, pp. 913–916, April, 1990.     

Pyrrolothieno[1,4]diazepines. Part V. Study of their chemical reactivity and first synthesis of oxazino[4,3-c]pyrrolo[1,2-a]thieno[2,3-f] [1,4]diazepines

The study of the chemical reactivity of methyloxopyrrolothienodiazepinones led to new derivatives such as oximes, methyloximes, hydrazones and alcohols and also, according to an intramolecular cyclization, to new oxazino[4,3-c]pyrrolo[1,2-a]thieno[3,2-f][1,4]diazepines.     

Synthesis and transformations of 3-(1H-pyrrol-1-yl)thieno[2,3-b]pyridines

Reactions of 2,5-dimethoxytetrahydrofuran with 3-aminothieno[2,3-b]pyridines afford a number of substituted 3-(1H-pyrrol-1-yl)thieno[2,3-b]pyridines. The possibility of the reaction and the yield of the product are determined by the character of a substituent in position 2 of thieno[2,3-b]pyridine. The Curtius rearrangement of 2-acylazido-3(1H-pyrrol-1-yl)thieno[2,3-b]pyridines yields 4,5-dihydropyrido[3",2":4,5]thieno[2,3-e]pyrrolo[1,2-a]pyrazin-4-ones. The molecular and crystal structures of ethyl 4-methoxymethyl-6-methyl-3-(1H-pyrrol-1-yl)thieno[2,3-b]pyridine-2-carboxylate were determined by X-ray diffraction analysis.     

Chemistry of thienopyridines. XXXVIII. Diels-Alder reactions of thienopyridine sulfones. Part 2

Thieno[3,2-b]pyridine 1,1-dioxide ( 2 ) undergoes Diels-Alder condensation with the dienophiles cyclopentadiene, anthracene, and naphthacene in a manner analogous to its isomer thieno[2,3-b]pyridine 1,1-dioxide ( 1 ). Compound 2 dimerizes in refluxing xylene with the loss of sulfur dioxide plus either the loss or transfer of hydrogen to give a small yield (ca. 2%) of pyrido[2′,3′:4,5]thieno[3,2-f]quinoline 7,7-dioxide ( 7 ) and its 5,6-dihydro derivative 12 . Formation of 7 and 12 are compared and contrasted with products reported from dimerization of 1 and of benzo[b]thiophene 1,1-dioxide and its derivatives.     

New access to 4-phenyl-6H-pyrrolo[1,2-a]thieno[3,2-f]-[1,4]diazepines

Pyrrolo[1,2-a]thieno[3,2-f][1,4]diazepines were obtained in an original one step synthesis by treatment of imines 1 with paraformaldehyde in refluxing ethanol. The intermediate Mannich bases were also converted into the thienooxadiazocines 12 and the diazepine N-oxide 13 .     

Synthese und Eigenschaften der Furo- und Thieno-Analogen von PQQ-Triester

Synthesis and Characterization of the Furo and Thieno Analogues of the Triester of PQQ We report here the synthesis and properties of the furo and thieno analogues of 4,5-dihydro-4,5-dioxo-1H-pyrrolo[2,3-f]quinoline-2,7,9- tricarboxylic acid ( = PQQ), i.e. the furo- and thieno[2,3-f]quinoline-4,5-quinone (FQQ and TQQ, resp.) derivatives B and C , obtained as triesters. The triester of PQQ derivative A is much more stable than the triesters of B or C , and only the triester of A shows strong activity in nonenzymatic catalytic oxidations.
