Simultaneously recovered absorption and scattering images that separate these optical property features within the female breast are demonstrated from frequency-domain measurements. A study of known absorbing and scattering objects is presented as a foundation for interpreting these in vivo images once the contrast space has been fully characterized. No measurable influence of absorbing-object contrast appears in the scattering images, whereas localized scattering contrast enhances the corresponding region within the absorption image by approximately 30% (e.g., a 2:1 scatterer also reconstructs as an approximately 1.3:1 absorber). Scattering and absorption images of a female volunteer with a 3.4-cm fibroadenoma show a clear 2:1 localized increase in absorption coefficient with little or no evidence of scattering enhancement in the lesion.  相似文献

Abstract

The application of high hydrostatic pressure is one of the most promising novel minimal processing methods for food preservation. Under optimal process conditions the natural fresh properties of foods are retained. Pressure is an important thermodynamic factor as is temperature. Both pressure and temperature may influence the position of a chemical reaction equilibrium and the reaction rate. Examples of pressure effects on food quality related reactions are presented The effect of pressure on reactions resulting in flavour or colour defects or the formation of undesirable substances and the effect of pressure on the stability of valuable constituents like vitamin A or sulforaphane.  相似文献

An ultrahigh-speed spectral domain optical coherence tomography (SD-OCT) system is presented that achieves acquisition rates of 29,300 depth profiles/s. The sensitivity of SD-OCT and time domain OCT (TD-OCT) are experimentally compared, demonstrating a 21.7-dB improvement of SD-OCT over TD-OCT. In vivo images of the human retina are presented, demonstrating the ability to acquire high-quality structural images with an axial resolution of 6 microm at ultrahigh speed and with an ocular exposure level of less than 600 microW.  相似文献

The objective of this study was to investigate MRI methods for monitoring focused ultrasound surgery (FUS) of breast tumors. To this end, the mammary glands of sheep were used as tissue model. The tissue was treated in vivo with numerous single sonications which covered extended target volumes by employing a scanning technique. The ultrasound focus position was controlled by online temperature mapping based on the temperature dependence of the relaxation time T(1). This approach proved to be reliable and offers thus an alternative to proton resonance frequency methods, whose application is hampered in fatty tissues. FUS-induced tissue changes were visible on T(2)- as well as on pre- and post-contrast T(1)-weighted images. According to our initial experience, noninvasive MRI-guided FUS of breast tumors is feasible.  相似文献

Traumatic brain injury (TBI) is a prevalent disease, and many TBI patients experience disturbed cerebral blood flow (CBF) after injury. Moreover, TBI is difficult to quantify with conventional imaging modalities. In this paper, we utilized susceptibility weighted imaging (SWI) as a means to monitor functional blood oxygenation changes and to quantify CBF changes in animals after trauma. In this study using six rats, brain trauma was induced by a weight drop model and the brain was scanned over four time points: pre trauma, and 4 h, 24 h and 48 h post trauma. Five rats survived and one died after trauma. A blood phase analysis using filtered SWI phase images suggested that three rats recovered after 48 h and two rats deteriorated. SWI also suggested that CBF decreased by up to 26%. The CBF change is in agreement with the results of arterial spin labeling methods conducted in this study and with previously published results. Furthermore, SWI revealed an enlargement of the major venous vasculature in deep brain structures, in accordance with the location of diffuse axonal injury. Compared with the traditional, invasive, clinical monitoring of cerebral vascular damage and reduction in blood flow, this method offers a novel, safe and noninvasive approach to quantify changes in oxygen saturation and CBF and to visualize structural changes in blood vasculature after TBI.  相似文献

PurposeHepatic thermal ablation therapy can result in c-Met-mediated off-target stimulation of distal tumor growth. The purpose of this study was to determine if a similar effect on tumor metabolism could be detected in vivo with hyperpolarized 13C MRI.Materials and methodsIn this prospective study, female Fisher rats (n = 28, 120–150 g) were implanted with R3230 rat breast adenocarcinoma cells and assigned to either: sham surgery, hepatic radiofrequency ablation (RFA), or hepatic RFA + adjuvant c-Met inhibition with PHA-665752 (RFA + PHA). PHA-665752 was administered at 0.83 mg/kg at 24 h post-RFA. Tumor growth was measured daily. MRI was performed 24 h before and 72 h after treatment on 14 rats, and the conversion of 13C-pyruvate into 13C-lactate within each tumor was quantified as lactate:pyruvate ratio (LPR). Comparisons of tumor growth and LPR were performed using paired and unpaired t-tests.ResultsHepatic RFA alone resulted in increased growth of the distant tumor compared to sham treatment (0.50 ± 0.13 mm/day versus 0.11 ± 0.07 mm/day; p < 0.001), whereas RFA + PHA (0.06 ± 0.13 mm/day) resulted in no significant change from sham treatment (p = 0.28). A significant increase in LPR was seen following hepatic RFA (+0.016 ± 0.010, p = 0.02), while LPR was unchanged for sham treatment (−0.048 ± 0.051, p = 0.10) or RFA + PHA (0.003 ± 0.041, p = 0.90).ConclusionIn vivo hyperpolarized 13C MRI can detect hepatic RFA-induced increase in lactate flux within a distant R3230 tumor, which correlates with increased tumor growth. Adjuvant inhibition of c-Met suppresses these off-target effects, supporting a role for the HGF/c-Met signaling axis in these tumorigenic responses.  相似文献

We report experimental evidence of correlation-induced spectral changes in biological tissues. The overall spectral shift in our transmission measurements is to the red and the mean wavelength of the original spectrum is up 10% larger. These results indicate that the spectral changes due to elastic scattering are significant and likely to hinder all spectroscopic measurements based on the inelastic (i.e., emission and absorption) interaction between light and tissues. Thus, simultaneous morphology and spectral measurements are required for accurate measurements spectroscopic information.  相似文献

Magnetic resonance imaging is very sensitive to magnetic field variations. This inherent sensitivity can be exploited to measure small electric currents flowing in the human body. This report describes an experiment in which the magnetic fields produced by small currents applied to the forearm of a living subject have been detected in the tissue. It shows how such measurements have been used to measure current density. The suggested technique is used to measure one component of a current density in a saline solution in vitro.  相似文献

The application of high pressure (HP) provides an opportunity for the non-thermal preservation of high-quality foods, whereas highly resistant bacterial endospores play an important role. It is known that the germination of spores can be initiated by the application of HP. Moreover, the resistance properties of spores are highly dependent on their physiological states, which are passed through during the germination. To distinguish between different physiological states and to detect the amount of germinated spores after HP treatments, two fluorescence-based methods were applied. A flow cytometric method using a double staining with SYTO 16 as an indicator for germination and propidium iodide as an indicator for membrane damage was used to detect different physiological states of the spores. During the first step of germination, the spore-specific dipicolinic acid (DPA) is released [P. Setlow, Spore germination, Curr. Opin. Microbiol. 6 (2003), pp. 550–556]. DPA reacts with added terbium to form a distinctive fluorescent complex. After measuring the fluorescence intensity at 270 nm excitation wavelength in a fluorescence spectrophotometer, the amount of germinated spores can be determined. Spores of Bacillus subtilis were treated at pressures from 150 to 600 MPa and temperatures from 37 °C to 60 °C in 0.05 M ACES buffer solution (pH 7) for dwell times of up to 2 h. During the HP treatments, inactivation up to 2log 10 cycles and thermal sensitive populations up to 4log 10 cycles could be detected by plate counts. With an increasing number of thermal sensitive spores, an increased proportion of spores in germinated states was detected by flow cytometry. Also the released amount of DPA increased during the dwell times. Moreover, a clear pressure-temperature-time-dependency was shown by screening different conditions. The fluorescence-based measurement of the released DPA can provide the opportunity of an online monitoring of the germination of spores under HP inside the HP vessel. Implementation can be done using diamond anvil cells, units with inspection glasses or by inserting an optical fiber into the HP vessel. The analytical methods used can help to understand the complex mechanism of germination and inactivation of bacterial spores. Due to its universal, process-independent character, the application of these methods is feasible for established and emerging technologies.  相似文献

A biopsy is a well-known medical test used to evaluate tissue abnormality. Biopsy specimens are invasively taken from part of a lesion and visualized by microscope after chemical treatment. However, diagnosis by means of biopsy is not only variable due to depth and location of specimen but may also damage the specimen. In addition, only a limited number of specimens can be obtained, thus, the entire tissue morphology cannot be observed. We introduce a three-dimensional (3-D) endoscopic optical biopsy via optical coherence tomography employing a dual-axis microelectromechanical system scanning mirror. Since this technique provides high-resolution, noninvasive, direct, and multiple visualization of tissue, it could function as a clinical biopsy with advanced performance. The device was integrated with a conventional endoscope and utilized to generate in vivo 3-D clinical images in humans and animals.  相似文献

Since the inception of the laser, pulse duration has been continuously decreased by the use of a variety of techniques: Q-switching, mode-locking, and pulse compression. We would like to present a new technique based on the addition of laser sub-harmonic, ALS, that allows the production of a single-cycle pulse with single-femtosecond pulse duration. This simple technique takes advantage of the recent progress made in the generation of a few optical cycles and in optical parametric amplification. An in vivo flow cytometer is developed that allows the real-time detection and quantification of circulating fluorescently labeled cells in live animals. A signal from a cell population of interest is recorded as the cells pass through a slit of light focused across a blood vessel. Confocal detection of the excited fluorescence allows continuous monitoring of labeled cells in the upper layers of scattering tissue, such as the skin. The device is used to characterize the in vivo kinetics of red and white blood cells circulating in the vasculatúre of the mouse ear. Potential applications in biology and medicine are discussed.  相似文献

An auditory enhancement effect occurs when one component of a harmonic series is omitted for a few hundred milliseconds and then reintroduced: The reintroduced harmonic stands out perceptually. Three experiments are reported that studied a version of this effect in which several components of a harmonic series are enhanced to define the formants of a vowel. Using the accuracy of vowel identification to measure the prominence of the formant peaks in the effective auditory representation, forms of the effect were identified that are qualitatively similar to the incremental and decremental responses seen in primary auditory-nerve fibers. These results are compatible with an origin for the enhancement effect in peripheral auditory adaptation. However, an additional mechanism is required to account for the demonstration [Viemeister and Bacon, J. Acoust. Soc. Am. 71, 1502-1507 (1982)] that enhancement can involve a true gain in the frequency region of the reintroduced component. These effects demonstrate one way in which the auditory system may attenuate the prominence of background noises while preserving the ability to represent changes in spectral amplitude produced by newly arriving signals.  相似文献

在探讨近红外光谱快速检测丙氨酸氨基转移酶的可行性过程中,首先对不同厚度(0.5,1,2和4mm)血液样品的近红外透射光谱进行了分析。发现全血样品0.5 mm厚时的近红外透射光谱更适合于进行光谱分析。进而采集了176个全血样品0.5 mm厚时的近红外光谱。对采集的光谱进行多元散射校正、二阶微分法光谱预处理后,采用逐步多元线性回归和偏最小二乘回归方法建立定量分析模型,预测了全血丙氨酸氨基转移酶的含量。结果表明:利用近红外光谱法测定丙氨酸氨基转移酶时,采用偏最小二乘回归方法建立的定标模型预测效果最好,定标相关系数、定标标准差和预测标准差的值分别为:0.98,2.42和7.22。  相似文献

Brillouin scattering measurements of a silicate glass, carried out at high pressures in the diamond anvil cell, show a dramatic increase in the pressure dependence of longitudinal velocity, and a discontinuity in the compressibility of the glass at about 6 GPa. While a first-order phase transition has been documented under pressure within amorphous ice, we demonstrate that an apparent second-order transition to a new, structurally distinct amorphous phase can occur via the abrupt onset of a new compressional mechanism, which may be triggered by a shift in polymerization of the glass or an onset of a change in coordination of silicon, within pressurized amorphous silicates.  相似文献

A single-shot multiple quantum filtering method is developed that uses two double-band frequency selective pulses for enhanced spectral selectivity in combination with a slice-selective 90 degrees, a slice-selective universal rotator 90 degrees, and a spectral-spatial pulse composed of two slice-selective universal rotator 45 degrees pulses for single-shot three-dimensional localization. The use of this selective multiple quantum filtering method for C(3) and C(4) methylene protons of GABA resulted in improved spectral selectivity for GABA and effective suppression of overlapping signals such as creatine and glutathione in each single scan, providing reliable measurements of the GABA doublet in all subjects. The concentration of GABA was measured to be 0.7 +/- 0.2 micromol/g (means +/- SD, n = 15) in the fronto-parietal region of the human brain in vivo.  相似文献
