The conformational landscape and aggregation behaviour of tetrahydro-2-furoic acid (THFA) were investigated by using matrix isolation-vibrational circular dichroism (MI-VCD). The well-resolved experimental MI-IR and MI-VCD features in an argon matrix at 10 K allow one to identify two dominant monomeric conformations as trans-THFA where the hydroxyl and carbonyl groups of COOH are at opposite sides, as well as one cis-conformer. At 24 K and 30 K deposition temperatures, the experimental IR and VCD spectral features reveal further growth of the binary THFA aggregates. Systematic conformational searches identified three vastly different binary binding topologies, resulting in a few hundred stable (THFA)2 conformers. Interestingly, the main binary structures observed correspond to an unusual type of structure which is made of two trans-THFA subunits, in contrast to the usual double H-bonded ring binary structures, identified in a previous solution study. The present work showcases the power of MI-VCD spectroscopy in revealing unusual structures formed in a cold rare gas matrix.  相似文献

利用荧光光谱法和圆二色谱法研究了重组内皮抑素与中药有效成分芦荟大黄素的相互作用机制,求得两者在25℃和37℃的结合常数Ka为6.623×104和3.796×104。研究发现,Zn2+等5种金属离子能够竞争性降低重组内皮抑制与芦荟大黄素的结合常数,下降幅度为35%~55%,并且其影响程度随温度升高而增强。鸡胚尿囊膜血管生成实验结果表明,芦荟大黄素能增强重组内皮抑素的抗血管生成活性,使其对新生血管的抑制作用提高50%。  相似文献

The induced circular dichroism (ICD) spectra of the inclusion complexes of maleimide, phthalimide, and naphthalene‐2,3‐dicarboximide with β‐ or γ‐cyclodextrin (CDx) have been measured. The structure of the CDx inclusion complexes are interpreted by the signs and shapes of ICD spectra compared with the results of PPP calculations. In maleimide and naphthalene‐2,3‐dicarboximide, the ICD spectra of the β‐CDx inclusion complex are very similar to those of the γ‐CDx inclusion complex in spite of the differences in dimensions between the cavity of β‐CDx and that of γ‐CDx. In phthalimide, the ICD spectra of the β‐CDx inclusion complex are very different from those of the γ‐CDx inclusion complex. The split‐type ICD bands at 220–235 nm show that the dimer of phthalimide is formed in the presence of β‐CDx.  相似文献

Abstract

A new 2-arylbenzofuran, spathobenzofuran (1), together with ten known compounds including a 2-arylbenzofuran, three pterocarpans and six isoflavones were isolated from the acetone crude extract of the stems of Spatholobus parviflorus. All compounds were characterised by spectroscopic methods. Compound 4 was active (MIC 8?µg/mL) against Gram-negative Pseudomonas aeruginosa TISTR 781 while compound 2 had modest activity against Gram-positive Staphylococcus aureus TISTR 1466 with a MIC value of 16?µg/mL. All isolated compounds showed no cytotoxicity against Vero and KB cells.  相似文献

The fluorescence detected circular dichroism (FDCD) appearance of some exciton coupling compounds is reviewed by a newly developed ellipsoidal mirror system (FDCD465). The FDCD465 achieves both the complete elimination of the polarization artifact and a dramatic enhancement of detection sensitivity expanding the applicability of exciton coupling fluorophores. A variety of chromophores that possess large extinction coefficients and high fluorescence quantum yields are available for exciton coupled FDCD, regardless of the degree of fluorescence polarization. Different types of FDCD devices can regulate FDCD polarization artifacts and the photoselection effect, enabling effective surveys of FDCD potential.  相似文献

Preliminary reports of the nature of the vibrational circular dichroism (VCD) peak at around 1145 cm?1, which is characteristic of axial glycosidic sugars and is called the glycoside band (J. Am. Chem. Soc. 2004 , 126, 9496), have been throughly examined. Through systematic carbohydrate measurements, it was found that the sign of the glycoside band reflects not only the anomeric configuration but also the pyranose conformation. Isotope and theoretical studies characterized its vibrational mode as C1–H1 deformation coupled with C1–O1 stretching, which indicates its applicability to more‐complicated glycoconjugates. In this study, for the first time, carbohydrate VCD spectra were reliably predicted by means of density functional theory (DFT) calculations. The VCD technique was applied to glycopeptides, and simultaneous analysis of both the carbohydrate and aglycan parts was carried out.  相似文献

In the present investigation, an attempt has been made to study the interaction of phenosafranin (PSF), a cationic phenazinium dye with the transport proteins, bovine serum albumin (BSA) and human serum albumin (HSA), employing steady-state and time-resolved fluorometric and circular dichroism (CD) techniques. The photophysical properties of the dye are altered on binding with the serum proteins. An explicit study with respect to the modification of the fluorescence and fluorescence anisotropy upon binding, effect of denaturant, fluorescence lifetime and CD measurements reveal that the dye binds to both BSA and HSA with almost the same affinity. Far-UV CD spectra indicate a decrease in the percentage of α-helicity only for BSA upon binding with the probe. Near-UV CD responses indicate an alteration in the tertiary structure of both the transport proteins because of binding.  相似文献

Abstract

The use of fluorescence detected circular dichroism (FDCD) for monitoring the quenching of chiral molecules is described. This technique combines the specificity for chiral centers of circular dichroism (CD), the sensitivity and selectivity of the fluorescence measurement with the additional chemical information provided by fluorescence quenching. Spectra are acquired using a multidimensional FDCD spectrometer which obtains a FDCD matrix of spectral data as a function of multiple excitation wavelengths (CD information) and multiple fluorescence emission wavelengths. This combination of FDCD and fluorescence quenching produces a very selective monitor for chiral fluorophores.  相似文献

Although almost 200‐years‐old, several unknown aspects remain to be explored of colchicine, the unique available drug for acute flares of gout. In this article, we report density‐functional theory (DFT) studies of geometry, energy, and NMR; 1H‐, 13C‐, and 15N‐NMR chemical shifts and some spin‐spin coupling constants, including the complete analysis of the saturated part (ring B); the assignment of both enantiomers by NMR using a chiral solvating agent; solid‐state NMR experiments of the different forms of natural and racemic colchicine, and IR and vibrational circular dichroism (VCD) studies of these same forms.  相似文献

Summary. In collaboration with Jasco Corporation we have recently developed an FDCD (fluorescence detected circular dichroic) instrument J-465, which eliminates the photoselection artifacts and efficiently collects the emitted light from the sample solution based on the ideal ellipsoidal mirror principle. Using the J-465 we have investigated a variety of fluorophores with/without polarization for exciton-coupled FDCD stereochemical analysis. The following three cases extend the FDCD methodology to new challenging areas beyond the limits of conventional CD: (1) substrates containing C=C double bonds, (2) molecules with sterically hindered hydroxyls, and (3) substrates bearing remote stereogenic centers. The pico- to nano-level FDCD analysis described in this paper leads to promising opportunities for the stereochemical analysis of a wide range of natural products with minuscule amounts of sample available.  相似文献
