How to improve the therapeutic efficacy of cell delivery during mechanical injection has been a great challenge for tissue engineering. Here, we present a facile strategy based on dynamic chemistry to prepare injectable hydrogels for efficient stem cell delivery using hyaluronic acid (HA) and poly(γ-glutamic acid) (γ-PGA). The combination of the guest–host (GH) complexation and dynamic hydrazone bonds enable the HA/γ-PGA hydrogels with physical and chemical dual dynamic network and endow hydrogels a stable structure, rapid self-healing ability, and injectability. The mechanical properties, self-healing ability, and adaptability can be programmed by changing the ratio of GH network to hydrazine bond cross-linked network. Benefitting from the dynamic cross-linking networks, mild preparation process, and cytocompatibility of HA/γ-PGA hydrogels, bone marrow mesenchymal stem cells (BMSCs) show high cell viability in this system following mechanical injection. Moreover, HA/γ-PGA hydrogels can promote BMSC proliferation and upregulate the expression of cartilage-critical genes. Notably, in a rabbit auricular cartilage defect model, BMSC-laden HA/γ-PGA hydrogels can effectively promote cartilage regeneration. Together, we propose a general strategy to develop injectable self-healing HA/γ-PGA hydrogels for effective stem cell delivery in cartilage tissue engineering.  相似文献

Ionically cross-linked polyampholytic hydrogels were synthesized by redox copolymerization of acrylamide and an ionic complex of (N,N-diethylamino)ethyl methacrylate and acrylic acid (designated as PADA hydrogel). The swelling behavior of the hydrogels in water indicated that a minimal equilibrium swelling ratio is found when the molar ratio of anionic/cationic monomers was 1.55. In NaCl solution, the hydrogels exhibited the typical swelling behavior of conventional polyampholytic gels. Their equilibrium swelling ratios increased with an increase in the NaCl concentration. In solutions of multivalent ions (CaCl2 and trisodium citrate solutions), the equilibrium swelling ratios of the hydrogels increased first and were then followed by a decrease with an increase in salt concentration. Interestingly, an unexpected abrupt swelling phenomenon was observed when the fully swollen hydrogels in salt solution were transmitted to pure water. The unique swelling behavior of PADA hydrogels depends not only on the molar ratio of the anionic/cationic monomers but also on the valency of the ions.  相似文献

Based on a biodegradable cross-linker, N-maleyl chitosan (N-MACH), a series of Poly(N-isopropylacrylamide) (PNIPAAm) and Poly(N-isopropylacrylamide-co-acrylamide) [P(NIPAAm-co-Am)] hydrogels were prepared, and their lower critical solution temperature (LCST), swelling kinetics, equilibrium swelling ratio in NaCl solution, and enzymatic degradation behavior in simulated gastric fluids (SGF) were discussed. The LCST did not change with different cross-linker contents. By altering the NIPAAm/Am molar ratio of P(NIPAAm-co-Am) hydrogels, the LCST could be increased to 39°C. The LCST of the hydrogel was significantly influenced by the monomer ratio of the NIPAAm/Am but not by the cross-linker content. In the swelling kinetics, all the dry hydrogels exhibited fast swelling behavior, and the swelling ratios were influenced by the cross-linker content and NIPAAm/Am molar ratios. Equilibrium swelling ratio of all the hydrogels decreased with increasing NaCl solution concentration. In enzymatic degradation tests, the weight loss of hydrogels was dependent on the cross-linker contents and the enzyme concentration.   相似文献

Based on a biodegradable cross-linker, acryloyloxyethylaminopolysuccinimide (AEA-PSI), a series of looser cross-linked poly(N-isopropylacrylamide-co-acrylic acid) [P(NIPAAm-co-AAc)] hydrogels were prepared, and their water content, swelling/deswelling kinetics, and the morphology of the gels were investigated. The swelling behaviors of AEA-PSI-cross-linked P(NIPAAm/AAc) hydrogels were investigated in Dulbecco’s phosphate-buffered saline (pH = 7.4), in the distilled water, and in the simulated gastric fluids (pH = 1.2), respectively. The water contents of the hydrogels were controlled by the monomer molar ratio of NIPAAm/AAc, swelling media, and the temperature. In the swelling kinetics, all the dried hydrogels exhibited fast swelling behavior, and the swelling ratios were influenced significantly by the amounts of AEA-PSI and AAc content. The deswelling kinetics of the hydrogel were independent of the content of AAc and cross-linker. Lastly, the morphology of the hydrogels was estimated by the field scan electron microscopy.  相似文献

Polyacrylamide grafted poly(vinyl alcohol)/polyvinylpyrrolidone (PAM-g-PVA/PVP) semi-interpenetrating polymer network (semi-IPN) hydrogels were designed and prepared via a simple free radical polymerization route initiated by a PVA-(NH4)2Ce(NO3)6 redox reaction technique. The structure of the PAM-g-PVA/PVP hydrogels was characterized by a Fourier transform infrared spectroscope (FTIR), and the morphologies were observed by a scanning electron microscopy (SEM). The swelling kinetics investigations demonstrated that the equilibrium swelling (Q e ) of the (PAM-g-PVA/PVP) semi-IPN hydrogels depended on PVP compositional ratios and temperature. The Q e values were reduced with increasing the PVP contents, which was in agreement with theoretical water contents (S ) fitted by swelling kinetic data, and the swelling mechanism belonged to a non-Fickian mode for the PAM-g-PVA/PVP hydrogels. These hydrogels displayed thermosensitivities different from the common thermoresponsive gels that have a lower critical solution temperature. The swelling is enhanced with increasing the temperature of the media before 42°C, and later the equilibrium swelling is contrarily reduced. Therefore, the swelling behavior of the PAM-g-PVA/PVP hydrogels may be controlled and modulated by means of the compositional ratios of PVP to PAM-g-PVA and temperature.  相似文献

A series of macroporous poly(N-isopropylacrylamide-co-acrylic acid) (PNIPAm-AA) hydrogels with different composition were synthesized by free-radical copolymerization in the presence of silica particles as a pore generating agent. The equilibrium swelling ratio, half swelling time and dynamic swelling kinetics of the copolymers previously soaked in different acidic buffer solutions were investigated at pH 7 at 25°C. Experimental results revealed that the swelling rate of the macroporous hydrogels was greatly increased compared to conventional hydrogels due to existence of the macroporous structures. It was found that the swelling history of previously putting in acidic solutions copolymers had strong influence on their dynamic swelling kinetics especially for the samples ranging in composition between 30 and 70 mol % of NIPAAm, whereas the swelling history had little influence on the equilibrium swelling ratio of copolymers. The swelling pattern exhibits sigmoid swelling curves. This is explained by an autocatalytic mechanism. The hydrogen bonding dissociation plays an important role in the dynamic swelling behavior.  相似文献

Thermoresponsive hydrogels are of great importance as smart materials. They are usually composed of cross-linked polymers with a lower critical solution temperature (LCST). Although much is known about networks of poly(N-isopropylacrylamide), all other polymers are somewhat neglected. In this work, the temperature-dependent swelling behavior of differently cross-linked thermoresponsive poly(2-ethyl-2-oxazoline) (PEtOx) hydrogels were investigated with regard to varying parameters of the network composition. It was found that the degrees of swelling of the hydrogels converge for a certain polymer/solvent system at a distinct temperature independent of its degree of cross-linking. Furthermore, this temperature correlates with the LCST of the respective starting PEtOx. Its net chain molecular weight Mc only affects the maximum degree of swelling and thus, the swelling–deswelling rate of the hydrogel. The fundamental structure/property relations found in this study could be useful to predict the behavior of other thermoresponsive hydrogels.  相似文献

A series of novel hydrogels based on poly(N-isopropylacrylamide) (PNIPAAm) with pendant poly(N-(2-(dimethylamino) ethyl)-methacrylamide) (PDMAEMA) grafts were designed and synthesized. The influence of the pendant PDMAEMA grafts on the properties of the resulted hydrogels was examined in terms of morphology observed by scanning electron microscopy (SEM), thermal property characterized by differential scanning calorimetry (DSC) and shrinking/swelling kinetics upon external temperature changes. In comparison with the conventional PNIPAAm hydrogels, resulting hydrogels presented favorable pH sensitivity as well as improved thermosensitive properties, including enlarged water containing capability at room temperature and faster shrinking/swelling rate upon heating. In addition, fish DNA, used as a model drug, was loaded into the hydrogels, and the controlled release behavior of the drug-loaded hydrogels at different temperatures (22 and 37 °C) was further studied.  相似文献

Here we report the preparation and characterization of nanostructured thermo-responsive poly(acrylamide) (PAM)-based hydrogels. The addition of slightly crosslinked poly(N-isopropylacrylamide) (PNIPA) nanogels to AM reactive aqueous solution produces nanostructured hydrogels that exhibit a volume phase transition temperature (TVPT). Their swelling kinetics, TVPT's and mechanical properties at the equilibrium-swollen state (Heq) are investigated as a function of the concentration of PNIPA nanogels in the nanostructured hydrogels. Nanostructured hydrogels with PNIPA nanogels/AM mass ratios of 20/80 and above exhibit higher Heq and longer time to reach the equilibrium swelling than those of the conventional PAM hydrogels. However, the PNIPA nanogels possess thermo-responsive character missing in conventional PAM hydrogels. The TVPT of nanostructured hydrogels depends on PNIPA nanogel content but their elastic and Young moduli are larger than those of conventional hydrogels at similar swelling ratios. Swelling kinetics, TVPT, and mechanical properties are explained in terms of the controlled in-homogeneities introduced by the PNIPA nanogels during the polymerization.  相似文献

The hydrogels prepared by the crosslinking of partially saponified poly(vinyl alcohol) (PVA) which has low critical solution temperature (LCST) in water showed characteristics of swelling at low temperature and shrinkage at high temperature. The hydrogels showed repeatable swelling–shrinking behavior. The hydrogels containing release substances such as cyanocobalamin, p‐acetamidophenol, insulin and ovalbumin were prepared by dipping these aqueous solutions and the release substances were studied. Insulin and ovalbumin were not absorbed by the hydrogels when the use of partially PVA with the degrees of polymerization (DPs) of 540 were prepared, but absorbed by the hydrogels with DPs of 1860. The size of the polymer network prepared with a higher DP was suitable for the absorption of insulin and ovalbumin. In accordance with release substances, the release patterns were different. Novel interpenetrating network (IPN) hydrogels (PNIPAAm/clay/PAAm hydrogels) based on poly(N‐isopropylacrylamide) (PNIPAAm) crosslinked by inorganic clay and poly(acrylamide) (PAAm) crosslinked by organic crosslinker were prepared in situ by ultraviolet (UV) irradiation polymerization. The effects of clay content on temperature dependence of equilibrium swelling ratio, deswelling behavior, thermal behavior, and the interior morphology of resultant IPN hydrogels were investigated with the help of Fourier transform infrared spectroscopy, differential scanning calorimeter (DSC), scanning electron microscope (SEM). Study on temperature dependence of equilibrium swelling ratio showed that all IPN hydrogels exhibited temperature‐sensitivity. DSC further revealed that the temperature‐sensitivity was weakened with increasing amount of clay. Study on deswelling behavior revealed that IPN hydrogels had much faster response rate when comparing with PNIPAAm/clay hydrogels, and the response rate of IPN hydrogels could be controlled by clay content. SEM revealed that there existed difference in the interior morphology of IPN hydrogels between 20 [below lower critical solution temperature (LCST)] and 50 °C (above LCST), and this difference would become obvious with a decrease in clay content. For the standpoint of applications, oscillating swelling/deswelling behavior was investigated to determine whether properties of IPN hydrogels would be stable for potential applications. Dual thermo‐ and pH‐sensitive network‐grafted hydrogels made of poly(N,N‐dimethylaminoethyl methacrylate) (PDMAEMA) network and poly(N‐isopropylacrylamide) (PNIPAM) grafting chains were successfully synthesized by the combination of atom transfer radical polymerization (ATRP), reversible addition‐fragmentation chain transfer (RAFT) polymerization, and click chemistry. PNIPAM having two azide groups at one chain end [PNIPAM‐(N3)2] was prepared with an azide‐capped ATRP initiator of N,N‐di(β‐azidoethyl) 2‐chloropropionylamide. Alkyne‐pending poly(N,N‐dimethylaminoethyl methacrylate‐co‐propargyl acrylate) [P(DMAEMA‐co‐ProA)] was obtained through RAFT copolymerization using dibenzyltrithiocarbonate as chain transfer agent. The subsequent click reaction led to the formation of the network‐grafted hydrogels. The influences of the chemical composition of P(DMAEMA‐co‐ProA) on the properties of the hydrogels were investigated in terms of morphology and swelling/deswelling kinetics. The dual stimulus‐sensitive hydrogels exhibited fast response, high swelling ratio, and reproducible swelling/deswelling cycles under different temperatures and pH values. The uptake and release of ceftriaxone sodium by these hydrogels showed both thermal and pH dependence, suggesting the feasibility of these hydrogels as thermo‐ and pH‐dependent drug release devices. © 2011 Wiley Periodicals, Inc. 采用接枝共聚合成了羧甲基纤维素钠、丙烯酸与N,N′-亚甲基双丙烯酰胺的交联凝胶, 研究了这类凝胶在不同pH值的缓冲溶液中的溶胀行为, 发现在酸性介质中凝胶的溶胀动力学行为表现出过溶胀平衡特性(overshooting effect), 即凝胶先发生溶胀到最大值, 然后再逐渐消溶胀到平衡. 这种现象可归因于凝胶溶胀过程中羧基之间通过氢键所产生的协同物理交联. 较之凝胶的组成, 缓冲溶液的pH值对过溶胀平衡现象的影响更为显著. 前者是因为凝胶羧基的总摩尔分数并不随两组分结构单元摩尔数的改变而改变, 羧基之间通过氢键形成的物理交联程度在交联剂摩尔分数接近的条件下变化不大; 后者是由于溶液的pH值显著影响凝胶羧基的质子化程度, 进而影响羧基之间通过氢键形成的物理交联程度. 凝胶在酸性介质中的溶胀过程符合E. Díez-Peña等提出的溶胀动力学定量模型, 理论曲线与实验数据有较好的相关性. 凝胶在pH≥5.0的缓冲溶液中的溶胀不产生过溶胀平衡现象, 这一现象归因于完全离子化的羧基之间不能形成物理交联. 凝胶的溶胀过程遵循Schott二级溶胀动力学.  相似文献

Deuterium isotope effects on swelling kinetics and volume phase transition in typical polymer hydrogels (poly(N-isopropylacrylamide) and polyacrylamide gels) are discussed. Deuterium substitutions affect on the swelling kinetics and volume phase transition of the polymer hydrogels. The slower swelling kinetics of hydrogels in D2O than in H2O arises mainly from the high viscosity of the medium. The deuterium isotope effect on the swelling-shrinking curve of hydrogels would come from the different polymer-solvent interaction. The microenvironments of hydrogels studied by solvatochromic fluorescence probe are compared with the bulk state. The zipper-type hydrogen-bonding inter-polymer complexes (poly(acrylic acid)-polyacrylamide and poly(acrylic acid)-poly(N-acryloylglycineamide)) are also investigated and show the huge isotope effect on the phase separation temperature.  相似文献

Sodium alginate (SA ) was combined with poly(N ‐isopropylacrylamide) (PNIPAAm ) to prepare thermosensitive hydrogels through semi‐interpenetrating polymer network (semi‐IPN ) and fully interpenetrating polymer network (full‐IPN ). The thermosensitive, swelling, mechanical, and thermal properties of pure PNIPAAm , SA /PNIPAAm semi‐IPN , and Ca‐alginate/PNIPAAm full‐IPN hydrogels were investigated. The formation of semi‐IPN and full‐IPN significantly improved the hydrogels’ swelling capability and mechanical properties without altering their thermosensitivity. 5‐Fluorouracil (5‐Fu) was selected as a model drug to study the release behaviors of the hydrogels. It was found that in vitro controlled drug release from semi‐IPN hydrogels showed an initial release burst, followed by a slower and sustained release, before reaching equilibrium. Full‐IPN hydrogels showed slow and sustained release during the whole process. Temperature and pH were found to affect the rate of drug release. Ca‐alginate/PNIPAAm full‐IPN hydrogels have potential application as drug delivery matrices in controlled drug release.  相似文献

A new kind of pH- and temperature-responsive semi-interpenetrating polymer network hydrogel based on linear carboxymethylchitosan (CMCS) and poly (N-isopropylacrylamide) (PNIPA) crosslinked by inorganic clay was prepared. The pH-and temperature-responsive behaviors, the deswelling kinetics, and the mechanical properties of the hydrogel were investigated. The hydrogels exhibited a volume phase transition temperature around 33 °C with no significant deviation from the conventional PNIPA hydrogels. The results of the influence of pH value on the swelling behaviors showed that the minimum swelling ratios of the hydrogels appeared near the isoelectric point (IEP) of CMCS, and when pH deviated from the IEP, the hydrogels behaved as polycations or polyanions. The novel hydrogels had much higher response rate than the conventional CMCS/PNIPA hydrogels. Moreover, the semi-IPN hydrogels crosslinked by clay could be elongated to more than 800% and the elongation could be recovered almost completely and instantaneously.  相似文献

Dual temperature‐ and pH‐sensitive hydrogels composed of N‐isopropylacrylamide (NIPAM) and 2‐acrylamido‐2‐methyl‐propanosulfonic acid (AMPS) were prepared by free‐radical crosslinking copolymerization in aqueous solution at 22 °C. The mole percent of AMPS in the comonomer feed was varied between 0.0 and 7.5, while the crosslinker ratio was fixed at 5.0/100. The effect of AMPS content on thermo‐ and pH‐ induced phase transitions as well as equilibrium swelling/deswelling, interior morphology and network structure was investigated. The volume phase transition temperature (VPT‐T) was determined by both swelling/deswelling measurements and differential scanning calorimetry (DSC) technique. In addition, the volume phase transition pH (VPT‐pH) was detected from the derivative of the curves of the swelling ratio (dQv/dpH) versus pH. The polymer‐solvent interaction parameter (χ) and the average molecular mass between crosslinks ( ) of hydrogels were calculated from swelling ratios in buffer solutions at various pHs. The enthalpy (ΔH) and entropy (ΔS) changes appearing in the χ parameter of hydrogels were also determined by using the modified Flory–Rehner equation. The negative values for ΔH and ΔS indicated that the hydrogels had a negative temperature‐sensitive property in water, that is, swelling at a lower temperature and shrinking at a higher temperature. It was observed that the experimental swelling data of hydrogels at different temperature agreed with the modified Flory‐Rehner approach based on the affine network model. © 2008 Wiley Periodicals, Inc. The paper presents the experimental studies regarding synthesis and characterization of hydrogels based on gellan (Gel)/chitosan (CS) and collagen (Col), obtained by crosslinking with glutaraldehyde (GLA). The influence of the polysaccharide content and GLA ratio on the final composition and swelling characteristics was evaluated. Hydrogels swelling analysis, in distilled water and phosphate buffer (PBS, pH 7.2) has shown higher swelling degrees at increased concentration of polysaccharide into hydrogels. In vitro release of pilocarpine has demonstrated the possibility to use gellan-collagen and chitosan-collagen hydrogels as ophthalmic drug delivery matrix.  相似文献

This study includes the synthesis of graft copolymer (GG-g-PAPA) from N-Acryloyl-L-phenylalanine (APA) and guar gum through free radical polymerization. Then, the novel pH sensitive GG-g-PAPA-cl-(PHEA-co-PAMPS) [GGAH] polymeric hydrogels were synthesized by employing free radical cross-linking polymerization using graft copolymer, 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS), and 2-hydroxy ethyl acrylate (HEA). The GGAH hydrogels were characterized using FTIR and SEM. Swelling studies of GGAH hydrogels were performed in distilled water, pH 1.2, and pH 7.4 solutions. The network and swelling kinetic parameters of GGAH hydrogels are also calculated.  相似文献

Temperature-sensitive poly(N-tert-butylacrylamide-co-acrylamide) [P(NTBA-co-AAm)] hydrogels were synthesized by free-radical copolymerization in a water–methanol mixture using three types of crosslinkers: 1,2-ethyleneglycol dimethacrylate, N,N-methylenebisacrylamide, and 1,3-butandiol dimethacrylate. These thermosensitive hydrogels were swollen to equilibrium in water at 20°C and examined by gravimetric measurements. The influence of type and content of crosslinkers on the swelling ratio, the polymer–solvent interaction parameter (χ), the average molecular mass between crosslinks and the effective crosslinking density (ν E) of the hydrogels were reported and discussed. The swelling process in water was found to be non-Fickian diffusion. The enthalpy (ΔH) and entropy (ΔS) changes appearing in the χ parameter for the hydrogels were determined by using the Flory–Rehner theory based on the phantom network model of swelling equilibrium. Negative values for ΔH and ΔS indicated that the hydrogels had a negative temperature-sensitive property in water; that is, swelling at a lower temperature and shrinking at a higher temperature. The temperature-reversibility and on–off switching properties of the P(NTBA-co-AAm) hydrogels may be considered as good candidates for designing novel drug-delivery systems.  相似文献
