Ethylene glycol (EG) initiated, hydroxyl‐telechelic poly(L ‐lactide) (PLLA) was employed as a macroinitiator in the presence of a stannous octoate catalyst in the ring‐opening polymerization of 5‐methyl‐5‐benzyloxycarbonyl‐1,3‐dioxan‐2‐one (MBC) with the goal of creating A–B–A‐type block copolymers having polycarbonate outer blocks and a polyester center block. Because of transesterification reactions involving the PLLA block, multiblock copolymers of the A–(B–A)n–B–A type were actually obtained, where A is poly(5‐methyl‐5‐benzyloxycarbonyl‐1,3‐dioxan‐2‐one), B is PLLA, and n is greater than 0. 1H and 13C NMR spectroscopy of the product copolymers yielded evidence of the multiblock structure and provided the lactide sequence length. For a PLLA macroinitiator with a number‐average molecular weight of 2500 g/mol, the product block copolymer had an n value of 0.8 and an average lactide sequence length (consecutive C6H8O4 units uninterrupted by either an EG or MBC unit) of 6.1. For a PLLA macroinitiator with a number‐average molecular weight of 14,400 g/mol, n was 18, and the average lactide sequence length was 5.0. Additional evidence of the block copolymer architecture was revealed through the retention of PLLA crystallinity as measured by differential scanning calorimetry and wide‐angle X‐ray diffraction. Multiblock copolymers with PLLA crystallinity could be achieved only with isolated PLLA macroinitiators; sequential addition of MBC to high‐conversion L ‐lactide polymerizations resulted in excessive randomization, presumably because of residual L ‐lactide monomer. © 2006 Wiley Periodicals, Inc. This article reports the synthesis and copolymerization of 6‐hydroxynon‐8‐enoic acid lactone. The ring‐opening polymerization of this lactone‐type monomer bearing a pendant allyl group led to new homopolymers and random copolymers with ε‐caprolactone and L ,L ‐lactide. The copolymerizations were carried out at 110 °C with Sn(Oct)2 as a catalyst. The introduction of unsaturations into the aliphatic polyester permitted us to carry out different chemical transformations on this family of polymers. For example, this article reports the bromination, epoxidation, and hydrosylilation of the allyl group in the new polyester copolymers. © 2000 John Wiley & Sons, Inc. In this study, synthesis and characterization of magnetic nanocarriers are reported for drug delivery based on the amphiphilic di‐block and tri‐block copolymers of poly(ethylene glycol) (PEG) and poly(ε‐caprolactone) (PCL) with surface modified super‐paramagnetite Fe3O4 nanoparticles (magnetic nanoparticles (MNPs)). The synthesized block copolymers (methoxy poly(ethylene glycol) (mPEG)–PCL and PCL–PEG–PCL) were characterized by Fourier transform infrared (FT‐IR), 1H nuclear magnetic resonance (1H NMR), gel permeation chromatography (GPC), scanning electron microscopy (SEM), and differential scanning calorimetry (DSC), and their properties such as critical micelle concentration, hydrophilicity to lipophilicity balance, and hydrolytic degradation were investigated. The block copolymers were functionalized with terminal azide groups (mPEG–PCL(N3) and (N3)PCL–PEG–PCL(N3)), and magnetic Fe3O4 nanoparticles were surface modified with poly(acrylic acid) (PAA) and propargyl alcohol (MNP–PAA–C≡CH). Magnetic nanocarriers were synthesized by click reaction between azide‐terminated block copolymers and MNP–PAA–C≡CH and characterized by FT‐IR, thermogravimetric analysis (TGA), dynamic light scattering (DLS), vibrating sample magnetometer (VSM), and transmission electron microscopy (TEM), and cytotoxicity was investigated by methyl thiazolyl tetrazolium assay. In vitro drug loading and release and release kinetics were investigated. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Amphiphilic BuO‐(PEO‐stat‐PPO)‐block‐PLA‐OH diblock and MeO‐PEO‐block‐(PEO‐stat‐PPO)‐block‐PLA‐OH triblock copolymers incorporating thermoresponsive poly(ethylene oxide‐stat‐propylene oxide) (PEO‐stat‐PPO) blocks were prepared by ring‐opening polymerization of lactide (LA) initiated by macroinitiators formed from treating BuO‐(PEO‐stat‐PPO)‐OH and MeO‐PEO‐block‐(PEO‐stat‐PPO)‐OH with AlEt3. MeO‐PEO‐block‐(PEO‐stat‐PPO)‐OH was prepared by coupling MeO‐PEO‐OH and HO‐(PEO‐stat‐PPO)‐OH, followed by chromatographic purification. The cloud points of 0.2% aqueous solutions are between 36 and 46 °C for the diblock copolymers that contain a 50 wt % EO thermoresponsive block and 78 °C for the triblock copolymer that contains a 75 wt % EO thermoresponsive block. Variable temperature 1H NMR spectra recorded on D2O solutions of the diblock copolymers display no PLA resonances below the cloud point and fairly sharp PLA resonances above the cloud point, suggesting that desolvation of the thermoresponsive block increases the miscibility of the two blocks. Preliminary characterization of the micelles formed in aqueous solutions of BuO‐(PEO‐stat‐PPO)‐block‐PLA‐OH conducted using laser scanning confocal microscopy and pulsed gradient spin echo NMR point to significant changes in the size of the micellar aggregates as a function of temperature. © 2005 Wiley Periodicals, Inc. Cationic copolymerization of racemic‐β‐butyrolactone (β‐BL) with l,l ‐lactide (LA) initiated by alcohol and catalyzed by trifluoromethanesulfonic acid proceeding by activated monomer (AM) mechanism was investigated. Although both comonomers were present from the beginning in the reaction mixture, polymerization proceeded in sequential manner, with poly‐BL formed at the first stage acting as a macroinitiator for the subsequent polymerization of LA. Such course of copolymerization was confirmed by following the consumption of both comonomers throughout the process as well as by observing the changes of growing chain‐end structure using 1H NMR. 13C NMR analysis and thermogravimetry revealed the block structure of resulting copolymers. The proposed mechanism of copolymerization was confirmed by the studies of changes of 1H NMR chemical shift of acidic proton in the course of copolymerization, providing an indication that indeed protonated species and hydroxyl groups are present throughout the process, as required for AM mechanism. © 2013 Wiley Periodicals, Inc. Four families of hyperbranched amphiphilic block copolymers of styrene (Sty, less polar monomer) and 2‐vinylpyridine (2VPy, one of the two more polar monomers) or 4‐vinylpyridine (4VPy, the other polar monomer) were prepared via self‐condensing vinyl reversible addition‐fragmentation chain transfer polymerization (SCVP‐RAFT). Two families contained 4VPy as the more polar monomer, one of which possessing a Sty‐b‐4VPy architecture, and the other possessing the reverse block architecture. The other two families bore 2VPy as the more polar monomer and had either a 2VPy‐b‐Sty or a Sty‐b‐2VPy architecture. Characterization of the hyperbranched block copolymers in terms of their molecular weights and compositions indicated better control when the VPy monomers were polymerized first. Control over the molecular weights of the hyperbranched copolymers was also confirmed with the aminolysis of the dithioester moiety at the branching points to produce linear polymers with number‐average molecular weights slightly greater than the theoretically expected ones, due to recombination of the resulting thiol‐terminated linear polymers. The amphiphilicity of the hyperbranched copolymers led to their self‐assembly in selective solvents, which was probed using atomic force microscopy and dynamic light scattering, which indicated the formation of large spherical micelles of uniform diameter. © 2015 Wiley Periodicals, Inc. Rare‐earth (Nd, Y) ptert‐butylcalix[n]arene (n = 4, 6, and 8) complexes without coligands were synthesized from rare‐earth isopropoxides in toluene. The products were characterized as the following structures: [C4(OH)O3 · CH3C6H5]Nd ( 4 ), [C6(OH)2O4 · CH3C6H5]3Ln4 [Ln = Nd ( 5 ), Y ( 6 )], and [C8(OH)2O6 · CH3C6H5]Nd2 ( 7 ). 2,2‐Dimethyl trimethylene carbonate (DTC) can be polymerized with complexes 4 – 7 alone as the initiator. PolyDTC (weight‐average molecular weight: 5700, polydispersity index: 1.11, measured by gel permeation chromatography) initiated by complex 5 was obtained with a conversion of 69.1% within 6 h in toluene at 80 °C. The thermal behavior of polyDTC has been compared with the published data. The DTC ring is opened via acyl‐oxygen bond cleavage with end‐group examination. NMR analyses of the polymerization reaction mixture indicated that the polymerization proceeds via a coordination‐insertion mechanism. © 2003 Wiley Periodicals, Inc. The ring‐opening polymerization (ROP) of cyclic esters, such as ε‐caprolactone, 1,5‐dioxepan‐2‐one, and racemic lactide using the combination of 3‐phenyl‐1‐propanol as the initiator and triflimide (HNTf2) as the catalyst at room temperature with the [monomer]0/[initiator]0 ratio of 50/1 was investigated. The polymerizations homogeneously proceeded to afford poly(ε‐caprolactone) (PCL), poly(1,5‐dioxepan‐2‐one) (PDXO), and polylactide (PLA) with controlled molecular weights and narrow polydispersity indices. The molecular weight determined from an 1H NMR analysis (PCL, Mn,NMR = 5380; PDXO, Mn,NMR = 5820; PLA, Mn,NMR = 6490) showed good agreement with the calculated values. The 1H NMR and matrix‐assisted laser desorption ionization time‐of‐flight mass spectrometry analyses strongly indicated that the obtained compounds were the desired polyesters. The kinetic measurements confirmed the controlled/living nature for the HNTf2‐catalyzed ROP of cyclic esters. A series of functional alcohols, such as propargyl alcohol, 6‐azido‐1‐hexanol, N‐(2‐hydroxyethyl)maleimide, 5‐hexen‐1‐ol, and 2‐hydroxyethyl methacrylate, successfully produced end‐functionalized polyesters. In addition, poly(ethylene glycol)‐block‐polyester, poly(δ‐valerolactone)‐block‐poly(ε‐caprolactone), and poly(ε‐caprolactone)‐block‐polylactide were synthesized using the HNTf2‐catalyzed ROP. © 2013 Wiley Periodicals, Inc. The thermal ring‐opening polymerization of 5‐benzyloxy‐trimethylene carbonate (BTMC) in bulk in the absence of any catalyst resulted in high molecular weight poly(BTMC) ( = 80 300) and subsequent catalytic hydrogenolysis resulted in functional poly(5‐hydroxyl‐trimethylene carbonate) (PHTMC). Similar spontaneous polymerization of BTMC in the presence of PEG ( = 2 000) as a macroinitiator can provide amphiphilic block polymers. The results revealed that the thermal non‐catalyst (co)polymerization of BTMC is a highly attractive preparative method because of the lack of usage of toxic initiators or catalysts. Furthermore, an evaluation of the degradation and cytotoxicity of PHTMC demonstrated enhanced degradability compared to poly(trimethylene carbonate) and similar toxicity compared to PLGA, showing PHTMC to be a promising biomaterial.

The ring‐opening polymerization (ROP) of β‐butyrolactone (β‐BL) has been studied using the organocatalysts of diphenyl phosphate (DPP) and bis(4‐nitrophenyl) phosphate (BNPP). The controlled ROP of β‐BL was achieved using BNPP, whereas that of using DPP was insufficient because of its low acidity. For the BNPP‐catalyzed ROP of β‐BL, the dual activation property for β‐BL and the chain‐end models of poly(β‐butyrolactone) (PBL) were confirmed by NMR measurements. The optimized polymerization condition for the ROP of β‐BL proceeded through an O‐acyl cleavage to produce the well‐defined PBLs with molecular weights up to 10,650 g mol?1 and relatively narrow polydispersities of 1.19–1.39. Functional initiators were utilized for producing the end‐functionalized PBLs with the ethynyl, maleimide, pentafluorophenyl, methacryloyl, and styryl groups. Additionally, the diblock copolymers consisting of the PBL segment with the polyester or polycarbonate segments were prepared by the BNPP‐catalyzed ROPs of ε‐caprolactone or trimethylene carbonate without quenching. © 2014 Wiley Periodicals, Inc. An amphiphilic block copolymer of silacyclobutane and methacrylic acid (MAA) was synthesized via a living anionic polymerization of 1,1‐diethylsilacylcobutane (EtSB). Sequential addition of 1,1‐diphenylethylene and t‐butyl methacrylate (tBMA) to living poly(EtSB) in the presence of lithium chloride gave poly(EtSB‐blocktBMA) with narrow molecular weight distributions. The t‐butyl ester groups in the obtained polymer were readily hydrolyzed via heating in 1,4‐dioxane in the presence of concentrated aqueous hydrochloric acid. The block copolymer with a short MAA segment was soluble in chloroform and insoluble in methanol and basic water, whereas the block copolymer with a long MAA segment was soluble in methanol and basic water and insoluble in chloroform. The block polymer (EtSB/tBMA = 45/60) formed a monolayer film on the water surface; this was confirmed by surface pressure measurement. © 2000 John Wiley & Sons, Inc. Summary: A novel type of glycerol‐derived, water‐soluble polycarbonate with pendant, primary hydroxyl groups was prepared from 2‐(2‐benzyloxyethoxy)trimethylene carbonate (BETC). Ring‐opening polymerization of BETC and 2,2‐dimethyltrimethylene carbonate (DTC) gave narrow distribution of homopolymers or random copolymers with high molecular weights. The protecting benzyl groups were removed by catalyzed hydrogenation at atmosphere H2 pressure to give hydroxyl polycarbonates without observable changes on the polymer backbone and molecular weight distribution. The hydrophilicity of the copolymers increases with the increase in the hydrophilic glycerol‐derived carbonate content.

Multiblock copolymers containing periodically spaced side‐chain carboxyl groups were obtained by a two‐step synthesis involving the preparation of ABA triblock prepolymers of ε‐caprolactone (A blocks) and ethylene glycol (B block) followed by chain extension to ABAn multiblock copolymers by reaction with pyromellitic dianhydride (PMDA). NMR analysis demonstrated the incorporation of PMDA in polymer chain and revealed the possibility of PMDA units to exist in two isomers, cisoid and transoid forms. Chain extension resulted in the incorporation of free carboxylic groups in polymer backbone and in an almost twofold increase of molecular weight. Thermal analysis indicated that the presence of PMDA residues interferes with the formation of crystalline phases by the chain‐extended polymers. The polymers were found to slowly degrade in buffer solution at 37 °C. Further, the carboxylated polymers can be processed into nanoparticulates by nanoprecipitation. Depending on the selected organic solvent, the particulate was constituted by either a monodispersed nanoparticles with average size of 150 nm or a bimodal distribution centered at about 100 nm and 7 μm. © 2013 Wiley Periodicals, Inc. Aliphatic polyesters and polyphosphoesters (PPEs) have received much interest in medical applications due to their favorable biocompatibility and biodegradability. In this work, novel amphiphilic triblock copolymers of PPE and poly(L ‐lactic acid) (PLLA) with various compositions were synthesized and characterized. The blocky structure was confirmed by GPC analyses. These triblock copolymers formed micelles composed of hydrophobic PLLA core and hydrophilic PPE shell in aqueous solution. Critical micellization concentrations of these triblock copolymers were related to the polymer compositions. Incubation of micelles at neutral pH followed by GPC analyses revealed that these polymer micelles were hydrolysized and resulted in decreased molecular weights and small oligomers, whereas its degradation in basic and acid mediums was accelerated. MTT assay also demonstrated the biocompatibility against HEK293 cells. These biodegradable polymers are potential as drug carriers for biomedical application. © 2008 Wiley Periodicals, Inc. The synthesis, characterization, and ring‐opening polymerization of a new cyclic carbonate monomer containing an allyl ester moiety, 5‐methyl‐5‐allyloxycarbonyl‐1,3‐dioxan‐2‐one (MAC), was performed for the first time. MAC was synthesized in five steps in good yield beginning from the starting material, 2,2‐bis(hydroxymethyl)propionic acid. Subsequent polymerization and copolymerizations of the new cyclic carbonate with rac‐lactide (rac‐LA) and ?‐caprolactone (CL) were attempted. Rac‐LA copolymerized well with MAC, but CL copolymerizations produced insoluble products. Oligomeric macroinitiators of MAC and rac‐LA were synthesized from stannous ethoxide, and both macroinitiators were used for the controlled ring‐opening polymerization of rac‐LA. The polymerization kinetics were examined by monitoring the disappearance of the characteristic C? O ring stretch of the monomer at 1240 cm?1 with real‐time in situ Fourier transform infrared spectroscopy. Postpolymerization oxidation reactions were conducted to epoxidize the unsaturated bonds of the MAC‐functionalized polymers. Epoxide‐containing polymers may allow further organic transformations with various nucleophiles, such as amines, alcohols, and carboxylic acids. NMR was used for microstructure identification of the polymers, and size exclusion chromatography and differential scanning calorimetry were used to characterize the new functionalized poly(ester‐carbonates). © 2003 Wiley Periodicals, Inc. A series of efficient catalysts, based on zinc alkoxides coordinated with NNO‐tridentate Schiff‐base ligands (L1H‐L6H), for ring opening polymerization of L ‐lactide have been prepared. The reactions of diethyl zinc (ZnEt2) with L1H‐L6H yielded [(μ‐L)ZnEt]2 ( 1a–6a ), respectively. Further reaction of compounds 1a–6a with benzyl alcohol (BnOH) produced the corresponding compounds of [LZn(μ‐OBn)]2 ( 1b–6b), respectively. X‐ray crystal structural studies reveal that all of these compounds 1a–6a are dimeric bridging through the phenolato oxygen atoms of the Schiff‐base ligand. However, the molecular structures of 1b–6b show a dimeric character bridging through the benzylalkoxy oxygen atoms. Ring‐opening polymerization of L ‐lactide, initiated by 1b–6b , proceeds rapidly with good molecular weight control and yields polymer with a very narrow molecular weight distribution. Experimental results show that the substituents on the imine carbon of the NNO‐ligand affect the reactivity of zinc complexes dramatically. © 2008 Wiley Periodicals, Inc. Polyesters and poly(ester carbonates) were synthesized via ring‐opening polymerization with new tin(II) macroinitiator adducts containing oligomeric L ‐lactide (LLA), rac‐lactide (rac‐LA), and ?‐caprolactone (CL). The novel initiating species were synthesized by the reaction of LLA, rac‐LA, or CL with Sn(OEt)2 (monomer concentration/initiator concentration ≤20) and then were dissolved in methylene chloride or toluene and stored in a stoppered flask for the subsequent ring‐opening polymerization of cyclic esters and carbonates. The soluble tin alkoxide macroinitiators yielded predictable and quantitative initiation of polymerization for up to 1 month of storage time at room temperature. The resulting polymers displayed low polydispersity (≤1.5), and a high monomer conversion (>95%) was obtained within relatively short polymerization times (≤2 h). Adjusting the monomer/macroinitiator ratio effectively controlled the molecular weights of the polymers. NMR was used to characterize the initiating species and polymer microstructure, and size exclusion chromatography was used to determine the molecular weight properties of the polymers. © 2002 Wiley Periodicals, Inc. The synthesis of amphiphilic linear‐hyperbranched graft‐copolymers in a grafting‐from approach is reported. The linear polyethylene with terminated hydroxyls, prepared by copolymerization of ethylene and 10‐undecen‐1‐ol, was used as macroinitiator for ring‐opening multibranching polymerization of glycidol by a typical slow monomer addition approach. Successful attachment of the hyperbranched grafts to the linear polyethylene backbone was confirmed by 1H/13C NMR, GPC, and TGA. The degree of polymerization and Mw/Mn of hyperbranched grafts were efficiently controlled by temperature, deprotonation ratio as well as the molar ratio of glycidol/hydroxyl (Nglycidol/NOH). The complicated microstructures caused by unsymmetric glycidol structure were analyzed by DEPT and 2D HSQC spectra, the degree of branching of 0.63–0.66 were calculated, indicating the extent of branch is close to theoretical values. The thermal analysis of linear‐hyperbranched copolymers via TGA and DSC is also presented. To our knowledge, this is the first report of a linear‐hyperbranched graft‐copolymer with a crystalline and nonpolar linear‐polyethylene segment. © 2014 Wiley Periodicals, Inc. J. Polym. Novel and well‐defined amphiphilic dendrimer‐star copolymer poly(ε‐caprolactone)‐block‐(poly(2‐(2‐methoxyethoxy)ethylmethacrylate‐co‐oligo(ethylene glycol) methacrylate))2 with Y‐shaped arms were synthesized by the combination of ring‐opening polymerization (ROP) and atom transfer radical polymerization (ATRP). The investigation of thermal properties and the analysis of crystalline morphology indicate that the high‐branched structure of dendrimer‐star copolymers with Y‐shaped arms and the presence of amorphous P(MEO2MA‐co‐OEGMA) segments together led to the complete destruction of crystallinity of the PCL segments in the dendrimer‐star copolymer. In addition, the hydrophilicity–hydrophobicity transition of the dendrimer‐star copolymer film can be achieved by altering the external temperatures. The amphiphilic copolymers can self‐assemble into spherical nanomicelles in water. Because the lower critical solution temperature of the copolymers can be adjusted by varying the ratio of MEO2MA and OEGMA, the tunable thermosensitive properties can be observed by transmittance, dynamic laser light scattering, and transmission electron microscopy (TEM). The release rate of model drug chlorambucil from the micelles can be effectively controlled by changing the external temperatures, which indicates that these unique high‐branched amphiphilic copolymers have the potential applications in biomedical field. © 2011 Wiley Periodicals, Inc. Ring‐opening polymerizations of trimethylene carbonate (TC) and 2,2‐dimethyltrimethylene carbonate (DTC) are initiated from hydroxyl‐terminated polyethylene glycol (PEG) and PEG‐based surfactants (Triton X‐100 or Triton X‐405) in the absence of any catalysts. The metal‐ and solvent‐free polymerizations proceed under melt at 150 °C, affording Triton X‐100‐block‐poly(TC) with Mn of 1400–5200 and Triton X‐100‐block‐poly(DTC) with Mn of 1800–7100 in excellent yields. The molecular weights and the comonomer composition of the resulting copolymers are controlled by the feed ratios of the monomers to the initiators, confirmed by gel permeation chromatography and 1H NMR spectroscopy. The solubilities of the block copolymers composed of hydrophilic PEG segment and hydrophobic poly(TC) or poly(DTC) segment depend on both the compositions and the components. For example, Triton X‐100‐block‐poly (TC) (TC/EG = 9.5/9.5) and Triton X‐405‐block‐poly(TC) (TC/EG = 28/40, 46/40) milky suspend in water, while Triton X‐405‐block‐poly(TC) (TC/EG = 9.7/40) dissolves in water. A dynamic light scattering study reveals that the particle distribution of a copolymer, Triton X‐405‐block‐poly(TC) (TC/EG = 9.7/40) in water, has a monodisperse unimodal pattern ranging from 92 to 368 nm. © 2006 Wiley Periodicals, Inc.
