Well-defined amphiphilic block copolymers composed of hydrophilic and hydrophobic blocks linked through an acid-labile acetal bond were synthesized directly by RAFT polymerization using a new poly(ethylene glycol) (PEG) macroRAFT agent modified with an acid-labile group at its R-terminal. The new macroRAFT agent was used for polymerization of poly(t-butyl methacrylate) (PtBMA) or poly(cholesterol-methacrylate) (PCMA) to synthesize well-defined block copolymers with a PEG block sheddable under acidic conditions. The chain extension polymerization kinetics showed known traits of RAFT polymerization. The molecular weight distributions of the copolymers prepared using the new macroRAFT agent remained below 1.2 during the polymerizations and the molecular weight of the copolymers was linearly proportional to monomer conversions. The acid-catalyzed hydrolysis behavior of the PEG-macroRAFT agent and the PEG-b-PtBMA (Mn = 13,600 by GPC, PDI = 1.10) was studied by GPC, 1H NMR and UV–vis spectroscopy. The half-life of acid-hydrolysis was 70 min at pH 2.2 and 92 h at pH 4.0. The potential use of the pH-labile shedding behavior of the copolymers was demonstrated by conjugating a thiol-modified siRNA to ω-pyridyldisulfide modified PEG-b-PCMA. The resultant PEG-b-PCMA-b-siRNA triblock modular polymer released PCMA-b-siRNA segment in acidic and siRNA segment in reductive conditions, as confirmed by polyacrylamide gel electrophoresis.  相似文献

7-Ethyl-10-hydroxycamptothecin (SN-38) is an active metabolite of Irinotecan (CPT-11), an anticancer pro-drug. To support clinical pharmacokinetic studies for liposome based formulation of SN-38 (LE-SN38) in cancer patients, a rapid, simple and sensitive liquid chromatography tandem mass spectrometry (LC-MS/MS) method has been developed and validated for the quantification of total SN-38 in human plasma. Sample preparation was carried out by one-step protein precipitation using cold acetonitrile with 0.5% acetic acid (v/v). Camptothecin was used as an internal standard (IS). Chromatographic separation of SN-38 and IS was achieved using a Synergi Hydro-RP column (C(18), 50 x 2 mm, 4 micro m), with a gradient elution of acetonitrile and 0.1% acetic acid. After ionization in electrospray source (positive ions), the acquisition was performed in the multiple reactions monitoring mode. Quantitation was accomplished using the precursor-->product ion combinations of m/z 393.1-->349.2 for SN-38 and 349.1-->305.1 for IS. The quantification limit of 0.05 ng/mL was achieved by using much lower volume (0.2 mL) of plasma and in the presence of LE-SN38. The method was validated over the concentration range of 0.05-400 ng/mL. Accuracy was within +/-12% of nominal at all concentration levels. Inter-day and intra-day precisions expressed as percentage coefficient of variation (%CVs) for quality control (QC) samples were less than 14 and 5%, respectively.  相似文献

The synthesis, characterization, and postpolymerization functionalization of star copolymers by RAFT polymerization, using ethylene glycol dimethacrylate as the difunctional monomer for core formation via crosslinking, is presented in this work. The “arm first” approach was used for the synthesis of PDMAEMAnPOEGMAn double‐hydrophilic mikto‐arm stars and PDMAEMAxPLMAy amphiphilic miktoarm stars, while the “core first” approach was used for the synthesis of (PDMAEMA‐b‐POEGMA)n double‐hydrophilic star block copolymers. Methyl iodide was used as the quaternizing agent for the transformation of the star copolymers into strong cationic star polyelectrolytes, through reaction on the dimethylamino groups of PDMAEMA blocks. The stars were characterized at the molecular level by SEC and proton nuclear magnetic resonance. Preliminary light scattering experiments, using THF and H2O as the solvents, were performed in order to get information regarding the solution behavior of the novel star copolymers synthesized. © 2019 Wiley Periodicals, Inc. In this study, a new benzoin-based multi-arm star polymer was synthesized by using ATRP, and characterization was achieved by spectrophotometric and chromatographic methods. Obtained multiarm (Benzoin-PS)m-polyDVB star polymer was employed as a polymeric photoinitiator for polymerization of methacrylates and acrylates. Photophysical properties of this initiator were determined by fluorescence and phosphorescence measurements, the phosphorescence lifetime was calculated as 29 ms hence the lowest triplet state nature was n-π* character, and laser flash photolysis technique was additionally used to get more information about triplet state and triplet lifetime which was calculated as 1.34 ms. Photokinetics of difunctional acrylate such as HDDA was studied with a multi-arm (Benzoin-PS)m-polyDVB star polymeric initiator using Photo-DSC.  相似文献

One-pot, spontaneous, and in-situ incorporation of Ru(II) complexes into a microgel (solubilized nanometer-scale network) has been achieved in near quantitative efficiency by a polymer-linking reaction of linear living poly(methyl methacrylate) (PMMA) with a bifunctional methacrylate (ethylene glycol dimethacrylate or bisphenol A dimethacrylate; linking agent) and a phosphine-ligand monomer [diphenyl-4-styryl-phosphine ( 3 ); i.e., CH2CH C6H4p-PPh2] in the RuCl2(PPh3)3-catalyzed living radical polymerization. The products were Ru-bearing. PMMA-armed star polymers with a microgel-core that consisted of a copolymer network of the linking agent and 3 . Upon the network formation, the phosphine ligands efficiently encapsulated RuCl2(PPh3)3, thus achieving a polymer catalyst directly from a polymerization catalyst. Colored dark brown-red, the star polymers exhibited UV-vis absorptions originating from the entrapped complex (3.1–7.4 × 10−5 mol Ru/g of polymer), the incorporation efficiency being close to 100% with respect to the original polymerization-catalyst. Detailed spectroscopic characterization showed the following: an absolute molecular weight of 1.7 × 105 to 1.7 × 106, an arm number of 11–92 arms/polymer, and a radius of gyration of 8–19 nm (in DMF). Direct observation of the individual star molecules in solid state was achieved by transmission electron microscopy (unstained; 2–3 nm dark dots for the core) and atomic force microscopy (semi-circular images). © 2006 Wiley Periodicals, Inc. Poly(ethylene glycol) (PEG) is widely used as a water soluble carrier for polymer-drug conjugates. Herein, we report degradable linear PEG analogs (DPEGs) carrying multifunctional groups. The DPEGs were synthesized by a Michael addition based condensation polymerization of dithiols and PEG diacrylates (PEGDA) or dimethacrylates (PEGDMA). They were stable at pH 7.4 but quickly degraded at pH 6.0 and 5.0. Thus, DPEGs could be used as drug carriers without concern for their retention in the body. DPEGs could be made to carry such functional groups as terminal thiol or (meth)acrylate and pendant hydroxyl groups. The functional groups were used for conjugation of drugs and targeting groups. This new type of PEG analog will be useful for drug delivery and the PEGylation of biomolecules and colloidal particles.  相似文献

Well-defined polystyrenes and poly(n-butyl acrylate)s of the two ends being functionalized with terpyridine groups were synthesized via addition-fragmentation chain transfer (RAFT) polymerization using a symmetric bisterpyridine-functionalized trithiocarbonate as a chain transfer agent (CTA). Kinetic studies on RAFT mediated thermal polymerization of styrene indicated the controlled polymerization. Corresponding triblock copolymers of styrene and n-butyl acrylate were obtained by utilizing the bisterpyridine-functionalized homopolymers as the macro-CTAs. Supramolecular metallo-polystyrenes with different repeat blocks were prepared by the chelating interaction between the terpyridine ends and Ru(II) ions. The formation of the metallo-polymers was proven by UV-vis spectra and dynamic light scattering (DLS).  相似文献

The reversible addition-fragmentation chain transfer polymerization of diisopropyl fumarate (DiPF) was carried out using ethyl 2-[[(dodecylthio)thioxymethyl]thio]-2-methylpropionate (T1) and 1,1′-(1,2-ethanediyl) bis[2-[[(dodecylthio)thioxymethyl]thio]-2-methylpropionate] (T2) as the monofunctional and difunctional chain transfer agents (CTAs) to synthesize poly(diisopropyl fumarate) (PDiPF) with a rigid chain conformation. The obtained PDiPF had a well-controlled molecular weight, molecular weight distribution, and structure of the chain ends. Size exclusion chromatography and NMR measurements revealed an excellent introduction efficiency (84–98%) of the terminal trithiocarbonate group into the polymer chain end. They were available as the monofunctional and difunctional macro-CTAs to synthesize the AB and ABA block copolymers, respectively. While the well-controlled block copolymers were solely obtained by the polymerization of 2-ethylhexyl acrylate as the second monomer in the presence of PDiPF as the macro-CTA, the block copolymerization of DiPF using poly(2-ethylhexyl acrylate) as the macro-CTA failed. The trithiocarbonate group at the chain end was completely removed by the reaction with n-butylamine and it was valid for the improvement of the coloration and other optical properties of the transparent polymers. © 2018 Wiley Periodicals, Inc. 用自组装技术在金(纯金和经阳极氧化的金)表面上获得了新型两亲聚合物PAMC_(16)S的有序膜。用接触角测试,XPS谱和电化学分析等方法对自组装膜进行了表征。根据膜表面的润湿性,金表面的自组装膜是疏水的,亲水的磺酸基团连于金表面,而疏水的碳氢链从表面伸展出。XPS实验结果支持金表面上单层膜的疏水结构。聚合物单层膜复盖的金电极起到含有针孔缺陷的阻膈型电极的作用。单层膜在法拉第反应中显示很强的吸附稳定性,说明聚合物LB膜在潜在应用中有其特有的特点。  相似文献
