Identification of xanthones and new arabinosides of flavone C-glucosides from Swertia perennis L. – Seven tetraoxygenated xanthones [1,3,7,8-tetrahydroxy-xanthone ( 1 ); 1,8-dihydroxy-3,7-dimethoxy-xanthone ( 2 ); 1,7-dihydroxy-3,8-dimethoxy-xanthone ( 3 ); 1-hydroxy-3,7,8-trimethoxy-xanthone ( 4 ); 3,7,8-trihydroxy-xanthone-xanthone-1-0-β-glucoside ( 5 ); 3,7,8-trimethoxy-xanthone-1-0-primeveroside ( 6 ); 8-hydroxy-3,7-dimethoxy-xanthone-1-0-primeveroside ( 7 )] have been isolated chromatographically, using polyamide columns, from roots of Swertia perennis L . From leaves and stems of the same plant, six xanthones [1,5,8-trihydroxy-3-methoxy-xanthone ( 8 ); 1,5-dihydroxy-3-methoxy-xanthone-8-0-β-glucoside ( 9 ); mangiferin ( 10 ); 1 ; 4 ; 5 ] and four flavone C-glycosides [iso-orientin ( 11 ); isovitexin ( 12 ); iso-orientin-6″-arabinoside ( 13 ); isovitexin-6″-arabinoside ( 14 )] have also been isolated. Among these compounds, 7 , 13 and 14 were not encountered before in nature. In the last two compounds, the position of arabinose on the C-glucoside moiety of the flavone was established by 13C-NMR. spectroscopy.  相似文献

Reinvestigation of the plant, Swertia decussata resulted in the isolation of four antioxidant xanthones, which were characterized as 1-hydroxy-3,7,8-trimethoxy, 1,8-dihydroxy-3,7-dimethoxy, 1,7-dihydroxy-3,8-dimethoxy and 1,7,8-trihydroxy-3-methoxyxanthones. The structural ambiguity of the first compound was settled by ASIS-NMR and 2D NOESY spectroscopy.  相似文献

We investigated the use of photochemistry to make novel derivatives of pentoxifylline. Under conditions that favour singlet excited states, we obtained 1-allyl-3,7-dimethyl-3,7-dihydro-1H-purine-2,6-dione, (R,R)-(±)-1-{[2-hydroxy-2-methylcyclobutyl]methyl}-3,7-dimethyl-3,7-dihydro-1H-purine-2,6-dione and 1-(5-hydroxyhexyl)-3,7-dimethyl-3,7-dihydro-1H-purine-2,6-dione. Naphthalene or molecular oxygen increases the yields and triplet sensitisers (acetophenone, benzophenone and acetone) decrease the yields. Efficient intramolecular triplet energy transfer from the carbonyl to the xanthine moiety allows the carbonyl moiety to react from a singlet excited state only. In solvents with an α-hydroxyalkyl hydrogen under conditions that favour triplet excited states, we obtained 8-substituted pentoxifylline derivatives: 8-(1-hydroxy-1-methylethyl)-3,7-dimethyl-1-(5-oxohexyl)-3,7-dihydro-1H-purine-2,6-dione in isopropanol, 8-(1-hydroxymethyl)-3,7-dimethyl-1-(5-oxo-hexyl)-3,7-dihydro-1H-purine-2,6-dione in methanol and 8-(1-hydroxyethyl)-3,7-dimethyl-1-(5-oxohexyl)-3,7-dihydro-1H-purine-2,6-dione in ethanol. The xanthine moiety reacts from a triplet state via a radical mechanism and yields are considerably improved by the addition of catalytic amounts of di-tert-butyl peroxide.  相似文献

Summary A series of 8-proparglyoxy-3,7-dimethyl-2,6-octadienyl and 8-propargyloxy-3,7-dimethyl-6-octenyl ethers were prepared from 8-hydroxygeranyl and 8-hydroxycitronellyl ethers, respectively. Almost all compounds showed high toxicity toCulex quinquefaciatus larvae at 1 mgl–1 dose level.

Fast alle Verbindungen zeigten hoch Toxizität gegenüber Larven vonCulex quinquefaciatus in einer Dosierung von 1 mgl–1.

The cyclodimerization of p-toluenesulfonamide and 3-chloro-2-(chloromethyl)-1-propene to prepare N,N'-bis(p-toluenesulfonyl)-3,7-bis(methylene)-1,5-diazacyclooctane (1a) and its ozonation to the corresponding 3,7-dione 2a are reported. Unusual transannular cyclizations initiated by lithium aluminum hydride treatment or bromination of 1a and oxidative coupling of the dioxime derived from 2a are described. These reactions lead, respectively, to the following derivatives of the little-studied 3,7-diazabicyclo[3.3.0]octane ring system: 1,5-dimethyl-3,7-diazabicyclo[3.3.0]octane (5), N,N'-bis(p-toluenesulfonyl)-1,5-bis(bromomethyl)-3,7-diazabicyclo[3.3.0]octane (8), and N,N'-bis(p-toluenesulfonyl)-1,5-dinitro-3,7-diazabicyclo[3.3.0]octane, (12). Acid-catalyzed hydration of 1a, in contrast, gives the expected 5-methyl-3,7-diazabicyclo[3.3.1]nonan-1-ol (10). Reaction of the dibromide 8 with the nucleophiles, sodium sulfide, sodium oxide, and sodium p-toluenesulfonamide conveniently delivers the corresponding novel 3,7,10-triheterocyclic [3.3.3]propellanes.  相似文献

A new xanthone-O-glycoside, the 1,7-dihydroxy-3-methoxyxanthone-8-O-β-D-glucopyranoside ( 7 ), has been isolated from the leaves of Gentiana verna L. by means of column chromatography on polyamid. Six known xanthones: 1-hydroxy-3,7,8-trimethoxyxanthone-1-O-primerveroside ( 2 ); 1,7,8-trihydroxy-3-methoxyxanthone ( 3 ); 7-hydroxy-3,8-dimethoxyxanthone-1-O-primeveroside ( 4 ); 7,8-dihydroxy-3-methoxyxanthone-1-O-primeveroside ( 5 ); mangiferin 6 and the flavone C-glycoside isoorientin 8 have also been isolated and identified.  相似文献

From Ligularia atroviolacea, four new eremophilendiolides, 81$-hydroxy-eremophil-3,7 (11)-dien-12,8α(14,6α)-diolide (1), 8β-methoxy-eremophil-3,7(11)-dien-12,8α(14,6α)-diolide (2), 8α-hydroxy-eremophil-3,7(11)-dien-12,8lβ(14,6α)-diolide (3) and eremophil-3,7(11),8-trien-12,8 (14,6α)-diolide (4), as well as a known diolide (5) were isolated. The fate of linalool, geraniol and nerol and their acetates in aqueous citric and hydrochloric acids has been investigated. Linalool and linalyl acetate yield predominantly α-terpineol and 3,7-dimethyloct-1-en-3,7-diol (6). Geraniol and nerol afford α-terpineol, linalool and the isomeric 3,7-dimethyloct-2-en-1,7-diols (7 and 8). While both neryl and geranyl acetate give α-terpineol and linalool, the former affords Z-1-acetoxy-3,7-dimethyloct-2-en-7-ol (8a), and the latter the E-isomer 7a and 2β-acetoxymethyl-1α,3,3-trimethylcyclohexanol (13).  相似文献

In our present study, two groups of xanthones isomers (1‐hydroxy‐3,5,8‐trimethoxyxanthone and 1‐hydroxy‐3,7,8‐trimethoxyxanthone; 1,8‐dihydroxy‐3,7‐dimethoxyxanthone and 1,8‐dihydroxy‐3,5‐dimethanolxanthone) and other two xanthones (3‐methoxy‐1,5,8‐trihydroxyxanthone and 3,5‐dimethoxy‐1‐hydroxyxanthone) were separated from Swertia franchetiana . First, a solvent system composed of petroleum ether/methanol/water (2:1:0.6, v/v) was developed for the liquid–liquid extraction of these xanthones from the crude extract. Then, an efficient method was established for the one‐step separation of these six xanthones by high‐speed countercurrent chromatography using n‐hexane/ethyl acetate/methanol/ethanol/water (HEMEW; 6:4:4:2:4, v/v) as the solvent system. The results showed that liquid–liquid extraction could be well developed for efficient enrichment of target compounds. Additionally, high‐speed countercurrent chromatography could be a powerful technology for separation xanthones isomers. It was found ethanol could be a good methanol substitute when the HEMEW system could not provide good separation factors.  相似文献

The protection of the 7-NH group in 8-bromo-3-methyl-3,7-dihydro-1H-pyrine-2,6-dione during the synthesis of 7-ubsubstituted 1-alkyl-8-bromo-3-methyl-3,7-dihydro-1H-pyrine-2,6-diones with a thienyl group was applied that was introduced by the reaction with 2-chloromethylthiirane. The thietanyl protection was removed by treating with sodium alcoholate after the oxidation with hydrogen peroxide to thietane 1,1-dioxide group.  相似文献

We report the crystal structures of two galactosylacetylenes: 3,7-anhydro-4,5,6,8-tetra-O-benzyl-1,1,2,2-tetradehydro-1,2-dideoxy-1-C-(trimethylsilyl)-D-glycero-L-mannooctitol (7) and 3,7-anhydro-4,5,6,8-tetra-O-benzyl-1,1,2,2-tetradehydro-1,2-D-glycero-L-mannooctitol (8). Four new xanthones, 1‐methoxy‐3,7,8‐trihydroxyxanthone (1), 1‐methoxy‐4,7,8‐trihydroxyxanthone (2), 1‐methoxy‐4,7‐dihydroxyxanthone (3), and 1,4‐dimethoxy‐2,7‐dihydroxyxanthone (4) were isolated from the stems of Cratoxylum cochinchinense along with four known xanthones (5–8). Heating 1,5-dinitro-3,7-di(tert-butyl)-3,7-diazabicyclo[3.3.1]-nonane with concentrated hydrobromic acid gives 1,5-dinitro-3,7-diazabicyclo[3.3.1]nonane. Cyclization of the latter with various aldehydes and ketones gave a series of 2,2-disubstituted 5,7-dinitro-1,3-diazaadamantanes. A new purine 3,7-dimethylguanine (1) has been isolated from the marine sponge Zyzzya fuliginosa, along with the known metabolites, makaluvamines A, C, K (2--4), 4-hydroxyphenylacetic acid (5), methyl ester of 4-hydroxyphenylacetic acid (6), 4-hydroxyphenethyl alcohol (7), L-phenylalanine (8) and L-tryptophan (9). The structure of 3,7-dimethylguanine (1) was elucidated by analysis of 1D and 2D (one- and two-dimensional) NMR [HMQC (heteronuclear multiple quantum coherence), gHMBC (heteronuclear multiple bond connectivity), 1H-15N gHMBC] data, mass spectroscopy data, and by comparison with 3,7-dimethylisoguanine (10).  相似文献

The synthesis and chemical trapping of a highly pyramidalized pentacyclo[6.4.0.0(2,10).0(3,7).0(4,9)]dodec-8-ene containing ketal functions is reported. The synthesis of new difunctionalized 2,6-dioxatricyclo[3.3.1.03,7]nonanes is described. This type of structure is an interesting synthetic building block for potential bioactive molecules and it was prepared from 8-oxabicyclo[3.2.1]oct-6-en-3-one having a NHBoc function on C-1. This precursor was obtained by a [4+3] cycloaddition reaction of 2-tert-butoxycarbonylaminofuran and the oxyallyl cation generated in situ from 2,4-dibromo-3-pentanone. Reduction of the carbonyl group at C-3 was accomplished in high yield and stereoselective manner to afford the corresponding axial alcohol at C-3 as a major product. Further intramolecular haloetherification of this type of alcohols with NBS and I(py)2BF4 led to the corresponding bromo and iodo-derivatives at C-8 of the 2,6-dioxatricyclo[3.3.1.03,7]nonane framework, in high yield. Epoxidation of 8-oxabicyclo[3.2.1]oct-6-en-3-ol followed by treatment with NaCN, NaN3, and/or NaOH in MeOH afforded 8-hydroxy-2,6-dioxatricyclo[3.3.1.03,7]nonanes in high yield via a transannular hydroxycyclization mediated by a base and through an alkoxide intermediate. The new 2,6-dioxatricyclo[3.3.1.03,7]nonanes were tested for biological activity against HIV-1 virus and MT-4 lymphoid cell line, showing a low anti-HIV activity and a high degree of cytotoxicity.  相似文献

Summary Alkylation of 8-vinyl-1,3-dimethyl-3,7-dihydro-1H-purine-2,6-dione (1) with alkyl halides inDMF in the presence of potassium carbonate, or alternatively, alkylation of its sodium salt (2) with alkyl iodide or hydroxyalkyl iodide afforded the 7-alkyl- or 7-(-hydroxyalkyl)-8-vinyl-1,3-dimethyl-3,7-dihydro-1H-purine-2,6-diones3. 2-Substituted oxiranes reacted with1 catalyzed by2 or Triton B to yield 7-(2-hydroxyalkyl)-8-vinyl-1,3-dimethyl-3,7-dihydro-1H-purine-2,6-diones4. Compounds3 and4 were tested for broncholytic activity. The most effective derivatives were3c and4b.

Zusammenfassung Alkylierung von 8-Vinyl-1,3-dimethyl-3,7-dihydro-1H-purin-2,6-dion (1) mit Halogenalkanen in Gegenwart von Kaliumkarbonat inDMF oder Alkylierung seines Natrium-Salzes (2) mit Iodalkanen beziehungsweise Iodalkanolen führte zu den 7-Alkyl- bzw. 7-(-Hydroxyalkyl)-8-vinyl-1,3-dimethyl-3,7-dihydro-1H-purin-2,6-dionen3. 2-Substituierte Oxirane reagierten mit Verbindung1 unter Katalyse durch2 oder Triton B zu den entsprechenden 7-(2-Hydroxyalkyl)-derivaten4. Die Substanzen3 und4 wurden auf ihre broncholytische Wirkung geprüft. Die höchste Aktivität wurde bei den Derivaten3c und4b festgestellt.

Simple preparation methods of modified guanidines have been explored as potential chiral superbases. Thus, 3,7,8-trisubstituted and 3,6,7, 8-tetrasubstituted 1,4,6-triazabicyclooctene systems were prepared from (1S,2S)-1,2-diphenylethylenediamine through stepwise 2-chloro-1, 3-dimethylimidazolinium chloride (DMC)-induced cyclizations of protected thioureas to the corresponding 2-iminoimidazolidines and then of 2-(2-hydroxyethylimino)imidazolidines to the bicyclic systems. Linear guanidines with a 2-hydroxyethyl functional group were prepared by the reaction of carbodiimides with 2-amino alcohols. Reaction of linear-type guanidines with DMC followed by base treatment afforded 1,4-disubstitued 2-iminoimidazolidines. Furthermore, another type of 1,4,6-triazabicyclooctene was also prepared through double DMC-induced cyclization of guanidines with two 2-hydroxyethyl substituents.  相似文献
