A sensitive and reliable method using pressurized liquid extraction (PLE) and liquid chromatography (LC)/electrospray ionization (ESI) tandem mass spectrometry with a triple quadrupole (QqQ) analyzer has been developed for the analysis of fumonisin B(1) (FB(1)), fumonisin B(2) (FB(2)) and fumonisin B(3) (FB(3)) in corn-based baby foods. Influence of several extraction parameters that affect PLE efficiency such as temperature, pressure, solvent extraction, number of cycles and dispersant/clean-up agents were studied. The selected PLE operating method was: 3g of sample was packed into 11 ml stainless-steel cell and fumonisins were extracted with methanol at 40 degrees C, 34 atm in one cycle of 5 min at 60% flush. The analytes were ionized in ESI operating with positive ion mode and identified by selecting two monitoring transitions, permitting quantification and confirmation in a single injection. Recoveries ranged from 68% to 83% at fortification levels of 200 microg kg(-1) with relative standard deviation (RSD) from 4% to 12%. The limits of quantification were from 2 microg kg(-1) for FB(1) and FB(2), and 5 microg kg(-1) for FB(3), which are below the maximum residue level established by the European Union legislation in infant formulas. The proposed method was successfully applied to the analysis of twenty seven samples of baby food products collected from different markets, and one positive sample with a content of 15.9 microg kg(-1) for FB(1), 9.2 microg kg(-1)for FB(2) and 5.8 microg kg(-1) for FB(3) was obtained. Given the simplicity and potential of the proposed procedure, its application for safety control is recommended.  相似文献

The development of a reversed phase high performance liquid chromatography fluorescence method for the determination of the mycotoxins fumonisin B(1) and fumonisin B(2) by using silica-based monolithic column is described. The samples were first extracted using acetonitrile:water (50:50, v/v) and purified by using a C(18) solid phase extraction-based clean-up column. Then, pre-column derivatization for the analyte using ortho-phthaldialdehyde in the presence of 2-mercaptoethanol was carried out. The developed method involved optimization of mobile phase composition using methanol and phosphate buffer, injection volume, temperature and flow rate. The liquid chromatographic separation was performed using a reversed phase Chromolith(?) RP-18e column (100 mm × 4.6 mm) at 30 °C and eluted with a mobile phase of a mixture of methanol and phosphate buffer pH 3.35 (78:22, v/v) at a flow rate of 1.0 mL min(-1). The fumonisins separation was achieved in about 4 min, compared to approximately 20 min by using a C(18) particle-packed column. The fluorescence excitation and emission were at 335 nm and 440 nm, respectively. The limits of detections were 0.01-0.04 μg g(-1) fumonisin B(1) and fumonisin B(2), respectively. Good recoveries were found for spiked samples (0.1, 0.5, 1.5 μg g(-1) fumonisins B(1) and B(2)), ranging from 84.0 to 106.0% for fumonisin B(1) and from 81.0 to 103.0% for fumonisin B(2). Fifty-three samples were analyzed including 39 food and feeds and 14 inoculated corn and rice. Results show that 12.8% of the food and feed samples were contaminated with fumonisin B(1) (range, 0.01-0.51 μg g(-1)) and fumonisin B(2) (0.05 μg g(-1)). The total fumonisins in these samples however, do not exceed the legal limits established by the European Union of 0.8 μg g(-1). Of the 14 inoculated samples, 57.1% contained fumonisin B(1) (0.16-41.0 μg g(-1)) and fumonisin B(2) (range, 0.22-50.0 μg g(-1)). Positive confirmation of selected samples was carried out using liquid chromatography-tandem mass spectrometry, using triple quadrupole analyzer and operated in the multiple reaction monitoring mode.  相似文献

Abstract

A method is described for the determination of fumonisin B1 in corn. The method involves sample extraction with methanol:water (75:25) and partial purification using a solid phase extraction. Fumonisin B1 is reacted with naphthalene-2,3-dicarboxaldehyde (NDA) to produce a highly fluorescent derivative, 1-cyano-2-alkyl-benz[f]isoindole (CBI) and then separated from the sample matrix on a reverse phase C-18 column with a mobile phase of acetonitrile:water:acetic acid (55:45:1). The NDA-derivative is quantitated by fluorescence detection at 410 nm excitation and a 440 nm, long past emission filter. Recoveries of fumonisin B1 added to corn at levels of 0.25–20.0 μg/g averaged 88.1% with a coefficient of variation of 10.3%. Confirmation of fumonisin B1 in corn samples was accomplished by fast atom bombardment (FAB) spectroscopy.  相似文献

A sensitive liquid chromatography/tandem mass spectrometry (LC/MS/MS) method for determining the type B fumonisin mycotoxins in corn-based foodstuffs is described. Fumonisins FB1 and FB2 were extracted from a 1 g sample by homogenization with acetonitrile/water (75:25, v/v, 50 mmol/L formic acid, 25 mL final volume) and the extract was defatted on C18 phase. Volumes of 5 mL of crude extracts were cleaned up on Carbograph-4 cartridges. The final solution was analyzed by HPLC with electrospray ionization mass spectrometry in positive ion mode using multiple reaction monitoring with a QqQ linear ion trap mass spectrometer. Recoveries for spiked corn-based foodstuffs ranged from 91-105% (RSD% < or =8%), and method detection limits were < or =2 ng/g for FB1 and < or =1 ng/g for FB2. Two different spiking levels were tested (5000 and 100 ng/g for FB1, 1000 and 20 ng/g for FB2). Quantitation was achieved by an external calibration procedure using matrix-matched standards, with diclofenac added post-cleanup as internal standard for the LC/MS/MS analyses. Calibration curves showed linearity in the concentration range 0.005-5 ng/microL of final extract (0.992 < or = R2< or =0.995). Two other fumonisins, FB3 and FB4, were identified in naturally contaminated samples of corn meal using an information-dependent acquisition protocol that looped three experiments, including neutral loss scan, enhanced resolution scan, and enhanced product ion scan. FB3 and FB4 quantitation was estimated as peak area ratios relative to the FB2 response in view of the lack of both standards. This work also includes an application of the present LC/MS/MS method to some maize and maize-based product samples (corn meal, cornflakes and popcorn) collected from Italian stores. FB1 and FB2 contamination levels exceeding the European Union recommendation were found in 8 out of 15 corn meal samples.  相似文献

A sensitive and selective analytical method was developed for the quantitative determination of fumonisins B(1) and B(2) in maize-based foods for direct human consumption. The method, based on high-performance liquid chromatography and fluorescence detection, presents a rapid and automated on-line post-column derivatization, performed with o-phtalaldehyde and N,N-dimethyl-2-mercaptoethylamine. Several factors affecting the separation and detection of fumonisins were investigated, including mobile phase composition, column features, derivatization agent flow-rate and both the excitation and the emission wavelengths. Optimal fluorescence detection was obtained by using a lambda(exc) of 343 nm and a lambda(em) of 445 nm. Under the optimized experimental conditions, a complete separation of fumonisins was obtained in less than 13 min by using a C(18) column and a gradient elution at 0.8 mL/min with methanol and 0.1M phosphate buffer at pH 3.15. The limits of detection for FB(1) and FB(2) were 4 and 5 microg/L corresponding to 5 and 6 microg/kg in matrix. Each fumonisin was determined in the range 40-320 microg/L that correspond to 50-400 microg/kg in matrix. The necessary requirements for accuracy, reproducibility and sensitivity were fulfilled and recovery values ranged from 87 to 94% for FB(1) and from 70 to 75% for FB(2) in cornflake samples at three fortification levels in the range 100-300 microg/kg. The potential of this method, combined with a simple clean-up procedure, was assessed by the measurements of FB(1) and FB(2) in maize-based products, such as maize flour, "polenta", tortillas and cookies.  相似文献

A simple, fast, and robust method was developed for the determination of fumonisin B1 (FB1), fumonisin B2 (FB2), and fumonisin B3 (FB3) in corn-based human food and animal feed (cornmeal). The method involves a single extraction step followed by centrifugation and filtration before analysis by ultra-performance liquid chromatographylelectrospray ionization (UPLC/ESI)-MS/MS. The LC/MS/MS method developed here represents the fastest and simplest procedure (<30 min) among both conventional HPLC methods and other LC/MS methods using SPE cleanup. The potential for high throughput analysis makes the method particularly beneficial for regulatory agencies and analytical laboratories with a high sample volume. A single-laboratory validation was conducted by testing three different spiking levels (200, 500, and 1000 ng/g for FB1 and FB2; 100, 250, and 500 ng/g for FB3) for accuracy and precision. Recoveries of FB1 ranged from 93 to 98% with RSD values of 3-8%. Recoveries of FB2 ranged from 104 to 108%, with RSD values of 2-6%. Recoveries of FB3 ranged from 94 to 108%, with RSD values of 2-5%.  相似文献

A simple, sensitive, and reliable analytical method is developed for the rapid determination of fumonisin B(1) and fumonisin B(2) in corn by high-performance liquid chromatography-positive electrospray ionization tandem mass spectrometry (LC-ESI-MS-MS). Fumonisin B(1) and fumonisin B(2) are extracted from corn with methanol-water (3:1, v/v) by means of ultrasonic extraction, and directly injected into an LC-MS-MS system after centrifugation. Fumonisin B(1) and fumonisin B(2) are separated on a Zorbax Eclipse XDB-C(18) column with a solution of methanol-water-formic acid as the mobile phase. The method is validated with respect to linearity, accuracy, precision, specificity, and stability. Moreover, the method was applied to real samples and demonstrated to be suitable for the determination of fumonisin B(1) and fumonisin B(2) in corn. The total time required for the analysis of one sample was ~30 min.  相似文献

A liquid chromatographic method for the determination of fumonisins B1 (FB1) and B2 (FB2) in corn-based foods for infants and young children was subjected to an interlaboratory validation study involving 11 laboratories. Five blind duplicate sample pairs of each matrix were analyzed to establish the accuracy, repeatability, and reproducibility of the method. Mass fractions in the baby food samples ranged from 89.1 to 384.4 microg/kg FB1 and from 22.5 to 73.6 microg/kg FB2. The method involved a warm extraction with citrate phosphate buffer-methanol-acetonitrile (50 + 25 + 25, v/v/v), a cleanup through an immunoaffinity column, and an end-determination of fumonisins by LC after automated precolumn derivatization with o-phthaldialdehyde reagent. RSDs for within-laboratory repeatability (RSDr) ranged from 6.8 to 23.5% for FB1 and 7.6 to 22.9% for FB2. RSDs for between-laboratory reproducibility (RSDR) ranged from 15.4 to 26.2% for FB1 and 21.6 to 36.3% for FB2. Mean FB1 recoveries from baby foods spiked at 100.0 and 250.0 microg/kg were 89 and 96%, respectively; for FB2 spiked foods at 25.0 and 62.5 microg/kg recoveries were 90 and 85%, respectively. HorRat values ranged from 0.8 to 1.2 for FB1, whereas for FB2 they ranged from 0.9 to 1.4 when calculated according to Horwitz, and from 1.0 to 1.7 when calculated according to Thompson, indicating an acceptable among-laboratory precision for all matrixes (HorRat values <2).  相似文献

A molecularly imprinted polymer prepared using 1-allylpiperazine (1-ALPP) as the functional monomer, trimethyltrimethacrylate (TRIM) as the crosslinker and the zearalenone (ZON)-mimicking template cyclododecanyl-2,4-dihydroxybenzoate (CDHB) has been applied to the clean-up and preconcentration of this mycotoxin (zearalenone) and a related metabolite, alpha-zearalenol (alpha-ZOL), from cereal and swine feed sample extracts. The extraction of ZON and alpha-ZOL from the food samples was accomplished using pressurized liquid extraction (PLE) with MeOH/ACN (50:50, v/v) as the extraction solvent, at 50 degrees C and 1500 psi. The extracted samples were cleaned up and preconcentrated through the MIP cartridge and analyzed using HPLC with fluorescence detection (lambda (exc)=271/ lambda (em)=452 nm). The stationary phase was a polar endcapped C18 column, and ACN/MeOH/water 10/55/35 (v/v/v, 15 mM ammonium acetate) at a flow rate of 1.0 mL min(-1) was used as the mobile phase. The method was applied to the analysis of ZON and alpha-ZOL in wheat, corn, barley, rye, rice and swine feed samples fortified with 50, 100 and 400 ng g(-1) of both mycotoxins, and it gave recoveries of between 85 and 97% (RSD 2.1-6.7%, n=3) and 87-97% (RSD 2.3-5.6%, n=3) for alpha-ZOL and ZON, respectively. 建立了婴幼儿谷基辅助食品中黄曲霉毒素B1、B2、G1、G2、赭曲霉毒素A、玉米赤霉烯酮、T-2毒素、脱氧雪腐镰刀菌烯醇、伏马毒素B1共9种真菌毒素的快速测定方法。试样用改良的QuEChERS方法进行提取,无需进一步净化,直接用液相色谱-串联质谱仪进行测定,基质外标法定量。在较宽的线性范围内,9种毒素的线性相关系数(r2)均不小于0.98,检出限为0.1~15.8 μg/kg,在3个不同添加水平下的加标回收率为77.6%~105.7%,RSD为2.5%~13.7%。采用建立的方法对市面上销售的41批次婴幼儿谷基辅助食品中的9种真菌毒素进行了筛查,数批产品检出不同含量的毒素。该方法准确、灵敏,可适用于婴幼儿谷基辅助食品中多种真菌毒素的快速分析。  相似文献

This study aimed to develop and optimize a method for the extraction and analysis of moenomycin antibiotics (a.k.a. flavomycin) in corn-based feed premix and in chicken litter. Moenomycin-A was isolated from chicken litter using pressurized liquid extraction followed by a solid-phase extraction (SPE) clean-up step. The highly lipophilic nature of moenomycin necessitated the use of the less hydrophobic sorbent, C(4)-based SPE cartridge, and a higher temperature elution solvent, methanol at 50 degrees C, in order to obtain satisfactory percent recoveries. After clean-up, the sample was analyzed by liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS). Various reversed-phase columns were examined, including C(18), CN, perfluorinated C(6), and a porous graphitic carbon. The set of conditions that gave the highest separation efficiency while still maintaining symmetrical peak shape was the C(18) column using the H(2)O+0.3% HCOOH and acetonitrile mobile phase.  相似文献

Various analytical methods used in the analysis of type B trichothecenes (deoxynivalenol, nivalenol, 3- and 15-acetyldeoxynivalenol) in cereals were compared and optimised in this work. These methods use either GC-electron-capture detection (ECD) of trimethylsilyl, trifluoroacetyl and heptafluorobutyryl derivatives or HPLC with UV or photodiode array detection of analytes. A new HPLC procedure using fluorescence detection prior derivatisation with coumarin-3-carbonyl chloride has been also tested. Five extraction solvents and two solid-phase extraction cartridges (silica, Florisil) plus a especial clean-up column (MycoSep 225) were compared in order to obtain the best recovery of the mycotoxins with minimal presence of coextractives in the chromatograms. The chosen extraction solvent was a mixture of acetonitrile-water (84:16, v/v). The MycoSep 225 column was chosen as the best alternative for clean-up of grain samples. For GC-ECD analysis, derivatisation of analytes with heptafluorobutyric anhydride prior the final determination was chosen as the most suitable procedure. HPLC-photodiode array (at 221 nm) analysis was more suitable for determination of type B trichothecenes than HPLC of the fluorescent coumarin-3-carbonyl derivatives. Recoveries obtained in spiked corn, rice and wheat are reported. The utility of the proposed methodology was assayed in cereal cultures of various Fusarium strains.  相似文献

1-Anthroylnitrile (1-AN) has been shown to be an efficient labelling reagent for the determination of T-2 toxin (T-2) by high-performance liquid chromatography (HPLC)-fluorescence detection. This reaction has been used to develop a sensitive, reproducible and accurate method for the determination of T-2 in wheat, corn, barley, oats, rice and sorghum. The method uses immunoaffinity columns containing antibodies specific for T-2 for extract clean-up, pre-column derivatization with 1-AN and HPLC with fluorescence detection for toxin determination. Ground cereal samples were extracted with methanol-water (80:20, v/v), the extracts were purified by immunoaffinity columns and the toxin was quantified by reversed-phase HPLC with fluorometric detection (excitation wavelength 381 nm, emission wavelength 470 nm) after derivatization with 1-AN. Recoveries from the different cereals spiked with T-2 at levels ranging from 0.05 to 1.5 microg/g were from 80 to 99%, with relative standard deviations of less than 6%. The limit of detection was 0.005 microg/g, based on a signal-to-noise ratio of 3:1.  相似文献

Rapid determination of aflatoxins in corn and peanuts   总被引:1,自引:0,他引:1  
A rapid and simple method using ultra-high-pressure liquid chromatography with UV detection for the determination of aflatoxins B1, B2, G1 and G2 in corn and peanuts has been developed. In this method, aflatoxins were extracted with a mixture of acetonitrile and water (86:14) and then purified by solid-phase clean-up with a MycoSep#226 AflaZon(+) column. The toxins were determined by UPLC-UV without derivatizing aflatoxins in real samples, which has not been used in other studies. The mean recoveries of aflatoxins from non-infected peanut and corn samples spiked with aflatoxins B1, B2, G1 and G2 at concentrations from 0.22 to 5 microg/kg were between 83.4% and 94.7%. The detection limits (S/N=3) for B1, B2, G1 and G2 were 0.32, 0.19, 0.32 and 0.19 microg/kg, and the corresponding quantification limits (S/N=10) were 1.07, 0.63, 1.07 and 0.63 microg/kg, respectively. We also applied this method on real samples. Among 16 peanut samples, 2 (12.5% incidence) were contaminated with aflatoxin; among 18 corn samples, 4 (22% incidence) were contaminated. The proposed method is rapid, simple and accurate for monitoring aflatoxins in corn and peanuts.  相似文献

Anti-fumonisin B1 polyclonal antibodies were isolated from the serum of rabbits, immobilized onto the surface of glutaraldehyde-activated silica or Sepharose CL-4B particles, and placed into empty small plastic solid-phase extraction cartridges. The immobilized antibodies were evaluated for their ability to retain fumonisin B1 and fumonisin B2. Cartridge capacity and elution conditions were determined, and the results were compared to those obtained with a commercially available cartridge. The cartridges, which were tested for their effectiveness to isolate the fumonisins from extracts of corn flour and nacho chips, detected fumonisins down to levels of about 20 ng/g. However, additional cleanup was required for detection at lower concentrations. With the use of a strong anion-exchange cartridge as a preliminary cleanup before immunoaffinity chromatography, the detection limit reached 2-5 ng/g in the products tested. The silica sorbent material exhibited strong interactions with the fumonisins, requiring acidified ethanol-water mixtures for elution and resulting in an additional degree of selectivity in isolating fumonisins from sample extracts. The silica-based immunoaffinity cartridges were successfully reused more than 10 times; the Sepharose-based cartridges were less robust. Liquid chromatography with fluorescence detection was used after prechromatographic derivatization with o-phthaldialdehyde-mercaptoethanol.  相似文献

 394-397 -------------------------------------------------------------------------------- 建立了液-液萃取后高效液相色谱法(HPLC)测定银杏叶提取物(EGb)中萜类内酯含量的快速方法。EGb样品溶于体积分数为30%的乙醇溶液后用乙醚萃取,有机相浓缩后的残留物以HPLC分析,组分的分离采用C18柱,以甲醇-水-磷酸(体积比为25∶75∶0.1)为流动相,示差检测器检测。结果表明,样品净化程序较已报道的方法更简便,选择性强、快速(少于20 min)。该方法回收率大于99.0%、相对标准偏差小于2.0%、重现性好,可有效地用于银杏叶提取物的产品质量评价。  相似文献

Solvent extraction, sonication, and microwave-assisted extractions in the presence of extraction agents (thioacetic acid, citric acid, cysteine, 2-mercaptoethanol, HCl + NaCl, etc.) were tested for the isolation of mercury species. A mixture of 6 M HCl and 0.1 M NaCl was selected as the most suitable extraction agent. The extraction efficiency was about 10% higher and the RSD below 3.3% when microwave-assisted extraction was applied instead of sonication. The liquid chromatography-cold vapour atomic fluorescence spectrometry (LC/CV-AFS) method was optimised and used for separation and determination of inorganic mercury cations and alkylated and arylated mercury species. Isocratic elution at a flow rate of 0.15 mL/min (with a mobile phase containing 0.05% 2-mercaptoethanol (pH = 5) and 7% methanol and with a stepwise increase of methanol content up to 100% MeOH in the 15th min) was used for separation of mercury species on a Hypersil BDS C18 RP column. The limits of detection of the LC/CV-AFS system were estimated as 0.2 microg/L (3%) for MeHg+, 0.07 microg/L (5.3%) for inorganic Hg, 0.06 microg/L (3.4%) for PhHg+, and 0.12 microg/L (4.4%) for EtHg with the corresponding RSDs at 5 microg/L (n = 10) given in parentheses. The concentrations (2-10 mg/kg fresh weight) of total mercury and methylmercury (90-99% of the total mercury) in selected fish obtained by HPLC/CV-AFS were in good agreement (absolute deviations 0.05 mg/kg) but more precise (RSDs <5.4% at 5 mg/L, n = 10) than those determined by GC coupled to an electron capture detector. The RSDs (3.1-8.2% and 4.1-9.0%) of the overall analytical procedure for the determination of total mercury (AMA 254) and methylmercury (HPLC/CV-AFS) were determined for intra-day and inter-day assays, respectively.  相似文献

Two detection methods are described for the determination of dexamethasone in feed, which may be used as screening and confirmatory methods. The procedures were developed after studying different extraction and clean-up procedures: feed extraction was optimized using methylene chloride and hexane followed by a tandem solid-phase clean-up step with silica and C18 cartridges. The confirmatory method presented is based on normal-phase high-performance liquid chromatography on a Diol 5 microns column, with hexane-propan-2-ol (90:10 v/v) as mobile phase, followed by diode array detection and confirmation. All parameters are discussed, especially the extraction and clean-up steps. Recovery studies after application of the proposed method to different kinds of feed samples spiked with dexamethasone at levels between 120 and 1600 ng g-1 yielded a mean value of 55.4% with an overall standard deviation of 15.6%. The proposed procedure allows the determination of dexamethasone in feed at levels lower than 50 ng g-1.  相似文献

With polyamide( PA)as an efficient sorbent for solid phase extraction( SPE)of Sudan dyes II,III and Red 7B from saffron and urine,their determination by HPLC was performed. The optimum conditions for SPE were achieved using 7 mL methanol/water( 1:9,v/v,pH 7)as the washing solvent and 3 mL tetrahydrofu-ran for elution. Good clean-up and high( above 90%)recoveries were observed for all the analytes. The opti-mized mobile phase composition for HPLC analysis of these compounds was methanol-water( 70:30,v/v). The SPE parameters,such as the maximum loading capacity and breakthrough volume,were also determined for each analyte. The limits of detection( LODs),limits of quantification( LOQs),linear ranges and recoveries for the analytes were 4. 6-6. 6 μg/L,13. 0-19. 8 μg/L,13. 0-5 000 μg/L( r2> 0. 99)and 92. 5% -113. 4%,respec-tively. The precisions( RSDs)of the overall analytical procedure,estimated by five replicate measurements for Sudan II,III and Red 7B in saffron and urine samples were 2. 3%,1. 8% and 3. 6%,respectively. This study examines the application of coupled-column LC-photochemically induced fluorimetry-fluorescence detection (LC-LC-PIF-FD), demonstrating its potential for the quantitative and selective detection of six herbicides, including propanil and the phenylureas monuron, monolinuron, chlorotoluron, diuron and neburon in groundwater samples. An AQUASIL C18 50 x 4.6 mm(2) id column coupled to an AQUASIL C18 150 x 4.6 mm(2) id column for analyte clean-up and determination were used, respectively. A simple SPE with Cl8 cartridges was carried out, yielding average recoveries between 80 and 112% (n = 6) with RSDs between 0.5 and 9%.
