A technique is introduced that monitors the depletion of intracellular ground state oxygen concentration ([3O2]) during photodynamic therapy of Mat‐LyLu cell monolayers and cell suspensions. The photosensitizer Pd(II) meso‐tetra(4‐carboxyphenyl)porphine (PdT790) is used to manipulate and indicate intracellular [3O2] in both of the in vitro models. The Stern–Volmer relationship for PdT790 phosphorescence was characterized in suspensions by flowing nitrogen over the suspension while short pulses of 405 nm light were used to excite the sensitizer. The bleaching of sensitizer and the oxygen consumption rate were also measured during continuous exposure of the cell suspension to the 405 nm laser. Photodynamic therapy (PDT) was conducted in both cell suspensions and in cell monolayers under different treatment conditions while the phosphorescence signal was acquired. The intracellular [3O2] during PDT was calculated by using the measured Stern–Volmer relationship and correcting for sensitizer photobleaching. In addition, the amount of oxygen that was consumed during the treatments was calculated. It was found that even at large oxygen consumption rates, cells remain well oxygenated during PDT of cell suspensions. For monolayer treatments, it was found that intracellular [3O2] is rapidly depleted over the course of PDT.  相似文献

While cyclometalated complexes have been extensively studied for optoelectronic applications, these compounds also represent a relatively new class of photosensitizers for the production of singlet oxygen. Thus far, singlet oxygen generation from cyclometalated Ir and Pt complexes has been studied in detail. In this review, photophysical data for singlet oxygen generation from these complexes are presented, and the mechanism of 1O2 generation is discussed, including evidence for singlet oxygen generation via an electron‐transfer mechanism for some of cyclometalated Ir complexes. The period from the first report of singlet oxygen generation by a cyclometalated Ir complex in 2002 through August 2013 is covered in this review. This new class of singlet oxygen photosensitizers may prove to be rather versatile due to the ease of substitution of ancillary ligands without loss of activity. Several cyclometalated complexes have been tethered to zeolites, polystyrene, or quantum dots. Applications for photooxygenation of organic molecules, including “traditional” singlet oxygen reactions (ene reaction, [4 + 2] and [2 + 2] cycloadditions) as well as oxidative coupling of amines are presented. Potential biomedical applications are also reviewed.  相似文献

It was found that irradiation of the solution of 4,4"-diazidodiphenyl in ethyl alcohol with UV-light at a wavelength longer than 280 nm in the presence of dissolved oxygen resulted in appearance of post-luminescence. The luminescence intensity increases with increasing the oxygen concentration; the luminescence virtually disappears after purging the solution with argon. The observed emission of light was attributed to chemiluminescence of the products of photooxidation of 4,4"-diazidodiphenyl. The rate dependence of the afterglow shows two maxima. The first is observed immediately after cessation of the UV irradiation of the solution and its transfer to a photometric cell. The intensity of the corresponding chemiluminescence decreases by an exponential law with e= 5 s. The second maximum appears after a certain time interval after the first one. Addition of a triplet sensitizer (Michler's ketone) to the reaction mixture results in quenching of the chemiluminescence from the photooxidation products, whereas the addition of substances that stabilize electrophilic species in the singlet state leads to an increase in the chemiluminescence intensity. It was suggested that the chemiluminescence resulted from reactions involving the singlet nitrene adduct with oxygen.  相似文献

Measurements of pigment triplet-triplet absorption, pigment phosphorescence and photosensitized singlet oxygen luminescence were carried out on solutions containing monomeric bacteriochlorophylls (Bchl) c and d, isolated from green photosynthetic bacteria, and their magnesium-free and farnesyl-free analogs. The energies of the pigment triplet states fell in the range 1.29-1.34 eV. The triplet lifetimes in aerobic solutions were 200-250 ns; they increased to 280 +/- 70 microseconds after nitrogen purging in liquid solutions and to 0.7-2.1 ms in a solid matrix at ambient or liquid nitrogen temperatures. Rate constants for quenching of the pigment triplet state by oxygen were (2.0-2.5) x 10(9) M-1 s-1, which is close to 1/9 of the rate constant for diffusion-controlled reactions. This quenching was accompanied by singlet oxygen formation. The quantum yields for the triplet state formation and singlet oxygen production were 55-75% in air-saturated solutions. Singlet oxygen quenching by ground-state pigment molecules was observed. Quenching was the most efficient for magnesium-containing pigments, kq = (0.31-1.2) x 10(9) M-1 s-1. It is caused mainly by a physical process of singlet oxygen (1O2) deactivation. Thus, Bchl c and d and their derivatives, as well as chlorophyll and Bchl a, combine a high efficiency of singlet oxygen production with the ability to protect photochemical and photobiological systems against damage by singlet oxygen.  相似文献

Abstract— A variety of experimental tests have been applied to the methylene-blue-sensitized photooxidation of amino acids to distinguish between singlet oxygen and non-singlet oxidation mechanisms. Conventional flash photolysis and laser photolysis were used to measure the rate constants for the quenching of excited triplet sensitizer and singlet oxygen by the amino acids histidine. tryptophan and methionine and the nucleotide guanosine-5′-monophosphate. In the case of histidine, the rate constants alone rule out an oxidation mechanism involving direct reaction with excited dye. With the other amino acids, and with guanosine monophosphate, the oxidation rates might be accounted for by either mechanism. The inhibition of the photo-oxidation of both tryptophan and methionine as well as histidine by the singlet-oxygen quenchers N3? and tetramethylethylene suggests that these reactions occur via a singlet-oxygen mechanism. A newly developed test of singlet oxygen reactions involving a comparison of photooxidation rates in normal and perdeuterated solvents has been used to establish that the photooxidation of tryptophan proceeds primarily by a singlet-oxygen mechanism. These experiments appear to constitute the first proof that singlet oxygen is involved in the photooxidation of the three amino acids tryptophan, methionine and histidine.  相似文献

Although the sonophotodynamic method has an effective therapeutic outcome for anticancer treatment compared with the photodynamic method, there are not enough related studies in the literature and this study aims to contribute to the development of sonophotodynamic studies. For this purpose, the Schiff base substituted silicon phthalocyanines were designed and synthesized as effective sensitizer candidates and the photophysicochemical and sonophotochemical features of the phthalocyanines were examined to increase singlet oxygen efficiency. The calculated ΦΔ values indicate that the contribution of substituent groups improved the production of singlet oxygen compared with silicon (IV) phthalocyanine dichloride (SiPcCI2) and also the sonophotochemical applications increased the singlet oxygen yields. The ΦΔ values (ΦΔ = 0.76 for axially bis-{4-[(E)-(pyridin-3-ylimino)methyl]phenol} substituted silicon (IV) phthalocyanine ( 2a ), 0.68 for axially bis-4-[(E)-{[(pyridin-3-yl)methyl]imino}methyl]phenol substituted silicon (IV) phthalocyanine ( 2b ) in photochemical study) reached to ΦΔ = 0.98 for 2a , 0.94 for 2b in sonophotochemical study. This article will enrich the literature on increasing singlet oxygen yield.  相似文献

Abstract— The 9, lodicyanoanthracene-sensitized photooxygenation of 2-methyl-2-butene and (+)-limonene proceeds via the singlet oxygen pathway in carbon tetrachloride as well as in acetonitrile, although the fluorescence of the sensitizer in acetonitrile is quenched by these olefins in an electron transfer quenching mechanism. The 9, 10-dicyanoanthracene-sensitized photooxygenation of cis- and trans-ä, ä′-dimethylstilbenes occurs exclusively via the singlet oxygen pathway in carbon tetrachloride; in acetonitrile, however, singlet oxygen and electron transfer photooxygenation reactions compete with one another. Addition of tetra-n-butyl ammonium bromide and increasing oxygen concentrations favor the formation of the singlet oxygen product, whereas addition of anisole, increasing substrate concentrations and decreasing oxygen concentrations favor the electron transfer photooxygenation products. In carbon tetrachloride, exciplexes of the sensitizer and the dimethylstilbenes are formed which give rise to cidrrans-isomerization of the substrates. In acetonitrile, neither exciplex formation nor cisltrans-isomerization are observed. A mechanism is proposed which allows us to calculate product distributions of the competing singlet oxygen/electron transfer photooxygenation reactions and thus to determine the efficiencies with which encounters between the singlet excited sensitizer and the substrates finally result in electron transfer photooxygenation products. Using (I) these efficiencies, (2) the β-value obtained from singlet oxygen photooxygenation sensitized by rose bengal, and (3) the appropriate k-values determined from fluorescence quenching of 9, 10-dicyanoanthracene in MeCN by oxygen and the stilbene, allows the calculation of the quantum yield of oxygen consumption by this stilbene. The quantum yield thus calculated is strictly proportional to the rate of oxygen consumption experimentally obtained; this result is considered as convincing evidence for the mechanism proposed.  相似文献

Photophysical properties in dilute MeCN solution are reported for seven RuII complexes containing two 2,2′‐bipyridine (bpy) ligands and different third ligands, six of which contain a variety of 4,4′‐carboxamide‐disubstituted 2,2′‐bipyridines, for one complex containing no 2,2′‐bipyridine, but 2 of these different ligands, for three multinuclear RuII complexes containing 2 or 4 [Ru(bpy)2] moieties and also coordinated via 4,4′‐carboxamide‐disubstituted 2,2′‐bipyridine ligands, and for the complex [(Ru(bpy)2(L)]2+ where L is N,N′‐([2,2′‐bipyridine]‐4,4′‐diyl)bis[3‐methoxypropanamide]. Absorption maxima are red‐shifted with respect to [Ru(bpy)3]2+, as are phosphorescence maxima which vary from 622 to 656 nm. The lifetimes of the lowest excited triplet metal‐to‐ligand charge transfer states 3MLCT in de‐aerated MeCN are equal to or longer than for [Ru(bpy)3]2+ and vary considerably, i.e., from 0.86 to 1.71 μs. Rate constants kq for quenching by O2 of the 3MLCT states were measured and found to be well below diffusion‐controlled, ranging from 1.2 to 2.0⋅109 dm3 mol−1 s−1. The efficiencies f of singlet‐oxygen formation during oxygen quenching of these 3MLCT states are relatively high, namely 0.53 – 0.89. The product of kq and f gives the net rate constant k for quenching due to energy transfer to produce singlet oxygen, and kqk equals k, the net rate constant for quenching due to energy dissipation of the excited 3MLCT states without energy transfer. The quenching rate constants were both found to correlate with ΔGCT, the free‐energy change for charge transfer from the excited Ru complex to oxygen, and the relative and absolute values of these rate constants are discussed.  相似文献

Singlet molecular oxygen, a1Delta(g), can be detected from a single cell by its weak 1270 nm phosphorescence (a1Delta(g)-->X3Sigma(g)-) upon irradiation of the photosensitizer 5,10,15,20-tetrakis(N-methyl-4-pyridyl)-21H,23H-porphine (TMPyP) incorporated into the cell. The behavior of this sensitizer in a cell, and hence the behavior of the associated singlet oxygen phosphorescence signal, depends on the conditions under which the sample is exposed to light. Upon irradiation of a neuron freshly incubated with TMPyP, the intensity of TMPyP fluorescence initially increases and there is a concomitant increase in the singlet oxygen phosphorescence intensity from the cell. These results appear to reflect a photoinduced release of TMPyP bound to DNA in the nucleus of the cell, where TMPyP tends to localize, and the subsequent relocalization of TMPyP to a different microenvironment in the cell. Upon prolonged irradiation of the cell, TMPyP photobleaches and there is a corresponding decrease in the singlet oxygen phosphorescence intensity from the cell. The data reported herein provide insight into key factors that can influence photosensitized singlet oxygen experiments performed on biological samples.  相似文献

Rate constants for the quenching of singlet oxygen (1O2, 1δg) for a series of piperidines, piperidine-N-oxyl free radicals and some commercially used hindered amine light stabilisers (HALS) have been measured by a laser flash photolysis method. Quenching rate constants are in the order: piperidine-N-oxyl free radicals ≤ secondary piperidines < tertiary piperidines. For some commercial HALS, 1O2 quenching rate constants and the light protective effect towards polypropylene photo-oxidation have been compared. No correlation has been found between the stabilizing action and the quenching efficiency towards 1O2. The data obtained point to little contribution of singlet oxygen to the key steps of polyolefin photo-oxidation.  相似文献

Time-resolved fluorescence and phosphorescence study of hypericin (Hyp) in complex with low-density lipoproteins (LDL) as well as the evolution of singlet oxygen formation and annihilation after illumination of Hyp/LDL complexes at room temperature are presented in this work. The observed shortening of the fluorescence lifetime of Hyp at high Hyp/LDL molar ratios (>25:1) proves the self-quenching of the excited singlet state of monomeric Hyp at these concentration ratios. The very short lifetime (∼0.5 ns) of Hyp fluorescence at very high Hyp/LDL ratios (>150:1) suggests that at high local Hyp concentration inside LDL molecules fast and ultrafast nonradiative decay processes from excited singlet state of Hyp become more important. Contrary to the lifetime of the singlet excited state, the lifetime (its shorter component) of Hyp phosphorescence is not dependent on Hyp/LDL ratio in the studied concentration range. The amount of singlet oxygen produced as well as the integral intensity of Hyp phosphorescence after illumination of Hyp/LDL complexes resemble the dependence of the concentration of molecules of Hyp in monomeric state on Hyp/LDL until a concentration ratio of 60:1. This fact confirms that only monomeric Hyp is able to produce the excited triplet state of Hyp, which in aerobic conditions leads to singlet oxygen production. The value of singlet oxygen lifetime (∼8 μs) after its formation from the excited triplet state of Hyp in LDL proves that molecules of singlet oxygen remain for a certain period of time inside LDL particles and are not immediately released to the aqueous surrounding. That Hyp exists in the complex with LDL in the monodeprotonated state is also demonstrated.  相似文献

Geometrical structure of free-base porphin (H2P) and Mg- and Zn-porphyrins together with their vibrational frequencies and vibronic intensities in phosphorescence are investigated by density functions theory (DFT) with the standard B3LYP functional. These molecules have a closed-shell singlet ground state (S0) and low-lying triplet (T1) excited states of ππ* type. The S0–T1 transition probability and radiative lifetime of phosphorescence (τp) of these molecules are calculated by time-dependent DFT utilizing quadratic response functions for account of spin–orbit coupling (SOC) and electric-dipole transition moments including displacements along active vibrational modes. The infrared and Raman spectra in the ground singlet and first excited triplet states are also studied for proper assignment of vibronic patterns. The long radiative lifetime of free-base porphin phosphorescence (τp  360 s at low temperature limit, 4.2 K) gets considerably shorter for the metalloporphyrins. An order of magnitude reduction of τp is predicted for Mg-porphyrin but no change of phosphorescence polarization is found. A forty times enhancement of the radiative phosphorescence rate constant is obtained for Zn-porphyrin in comparison with the H2P molecule which is accompanied by a strong change of polarization and spin-sublevel radiative activity. A strong vibronic activity of free-base porphin phosphorescence is found for the b2g mode at 430 cm−1, while the 679 and 715 cm−1 vibronic bands of b3g symmetry are less active. These and other out-of-plane vibrations produce considerable changes in the radiative constants of different spin sublevels of the triplet state; they also promote the S1  T1 intersystem crossing. Among the in-plane vibrations the ag mode at 1614 cm−1 is found very active; it produces a long progression in the phosphorescence spectrum. The time-dependent DFT calculations explain the effects of the transition metal atom on phosphorescence of porphyrins and reproduce differences in their phosphorescence and EPR spectra.  相似文献

The azide anion is often used as a physical quencher of singlet oxygen, the important active intermediate in photosensitized oxidation. An observed effect of azide on the rate of a reaction is considered an indication to the involvement of singlet oxygen. In most biological photosensitizations, the light‐absorbing sensitizer is located in a membrane or in an intracellular organelle, whereas azide is water soluble. The quenching it causes relies on a physical encounter with singlet oxygen during the latter's short lifetime. This can happen either if azide penetrates into the membrane's lipid phase or if singlet oxygen is intercepted when diffusing in the aqueous phase. We demonstrate in this article the difference, in liposomes’ suspension, between the effect of azide when using a water‐soluble and membrane‐bound chemical targets of singlet oxygen, whereas this difference does not exist when micelles are used. We explain the difference on the population of sensitizer and target in the liposome vs micelle. We also show the effect that exists on azide quenching of singlet oxygen by electrically charged lipids in liposomes. This is a result of the accumulation or dilution of azide in the debye layer near the membranes’ surface, due to the surface Gouy–Chapman potential.  相似文献

The reactions of singlet molecular oxygen (O21Δg) with a series of organic compounds have been studied in the gas phase at 298°K. The concentration of singlet molecular oxygen was determined by titration with 2,5-dimethylfuran. The titration technique was checked using a photoionization technique. Absolute rate constants were measured on the basis of the loss of organic reactant and, in some cases, of singlet molecular oxygen. It was found that the usual method of producing singlet molecular oxygen in the gas phase can also, under some conditions, allow reactive species other than singlet molecular oxygen to enter the reactor, leading to serious errors in the determination of rate constants. This problem was eliminated by carrying out the rate measurements in the presence of a small amount of nitrogen dioxide a radical scavenger.  相似文献

The absorption and emission spectroscopic behaviour of cyclometalated fac-tris(2-phenylpyridine) iridium(III) [Ir(ppy)3] is studied at room temperature. Liquid solutions, doped films, and neat films are investigated. The absorption cross-section spectra including singlet–triplet absorption, the triplet–singlet stimulated emission cross-section spectra, the phosphorescence quantum distributions, the phosphorescence quantum yields and the phosphorescence signal decays are determined. In neat films fluorescence self-quenching occurs, in diluted solid solution (polystyrene and dicarbazole-biphenyl films) as well as deaerated liquid solution (toluene) high phosphorescence quantum yields are obtained, and in air-saturated liquid solutions (chloroform, toluene, tetrahydrofuran) the phosphorescence efficiency is reduced by triplet oxygen quenching. At intense short-pulse laser excitation the phosphorescence lifetime is shortened by triplet–triplet annihilation. No amplification of spontaneous emission in the phosphorescence spectral region was observed indicating higher excited-state absorption than stimulated emission.  相似文献

We describe a pointsource sensitizer‐tipped microoptic device for the eradication of glioma U87 cells. The device has a mesoporous fluorinated silica tip which emits singlet oxygen molecules and small quantities of pheophorbide sensitizer for additional production of singlet oxygen in the immediate vicinity. The results show that the device surges in sensitizer release and photokilling with higher rates about midway through the reaction. This was attributed to a self‐amplified autocatalytic reaction where released sensitizer in the extracellular matrix provides positive feedback to assist in the release of additional sensitizer. The photokilling of the glioma cells was analyzed by global toxicity and live/dead assays, where a killing radius around the tip with ~0.3 mm precision was achieved. The implication of these results for a new PDT tool of hard‐to‐resect tumors, e.g. in the brain, is discussed.  相似文献

The spectroscopy and photophysics of the photosensitizer hypericin when in homogeneous solutions and when bound to liposomes were studied. Hypericin was found to partition efficiently into DMPC liposomes, with a binding constant of 58 (mg lipid/mL)?1. In these liposomes the singlet oxygen production quantum yield was 0.43 ± 0.09. To determine the deactivation constant of singlet oxygen in lipid bilayers for the first time, we calculated extrapolated values from its quenching by DMPC and lecithin in homogeneous solutions and obtained decay times of 36.4 and 12.2 μs, respectively. We also measured the quenching of singlet oxygen, sensitized by hypericin in DMPC liposomes, by NaN3, diphenyl isobenzofuran and H2,O: D2O mixtures and explained the results on the basis of singlet oxygen diffusing rapidly out of the lipid bilayer into the aqueous medium. The observed temperature effect on the lifetime of singlet oxygen of about 50% over a 15°C range in liposome suspension contrasts with a 3% change in a homogeneous solution in 1-nonanol and is explained by the temperature effect on the diffusion out of the liposome. A strong pH effect was observed, indicating that the deprotonated species formed above about pH 10 is a much weaker photosensitizer of singlet oxygen than the native, protonated species.  相似文献

Abstract Rose Bengal is shown to photosensitize free-radical production and oxygen consumption in solutions of melanin from autooxidation of 3,4-dihydroxyphenylalanine (DOPA). In anaerobic solutions the sensitizer enhances rates of free-radical production by up to a factor of 20. In aerobic solutions, rates of oxygen consumption can be increased by a factor of several hundred. The reactions appear to involve the triplet state of the sensitizer. The effect of the sensitizer in increasing oxygen consumption is quenched by low concentrations of azide and enhanced by D2O, suggesting that a singlet oxygen mechanism is involved.  相似文献

Intense phosphorescence is observed when an alcohol is introduced to an aqueous solution containing 1-bromonaphthalene and a cyclodextrin complex. The enhancement of the phosphorescence intensity of 1-bromonaphalene is related closely with the alcohol’s effectiveness in shielding the photo-excited lumophore from dissolved oxygen. The phosphorescence intensity decreases and the phosphorescence decay rate increases as the dissolved oxygen concentration increases. In this paper, we measure the phosphorescence intensity and decay rate of the complex using different alcohols and explain their respective advantages. By relating the concentration of oxygen to the intensity and the lifetime of the phosphorescence, the dissolved oxygen concentration can be measured. The sensitivities and measurement ranges with the complexes containing different alcohols are compared. Currently, this technique can measure dissolved oxygen concentrations from 0 to 40.2 mM (31,400% saturation). The range of the measurement is 40 times larger than previous studies.  相似文献

The near-IR phosphorescence of singlet delta oxygen, O2(a1Δg), has provided a wealth of information since it was first observed in solution-phase systems. The techniques employed and the quality of the data obtained have unproved significantly over the years that, in turn, presently makes it possible to address a wide variety of problems using both steady-state and time-resolved measurements. The development of spectroscopic methods to monitor other transitions in oxygen, specifically those that involve the singlet sigma state, O2(b1σg+), and the incorporation of high-level computational methods provides access to an even broader range of fundamental issues. The expertise presently available to monitor radiative transitions in oxygen, coupled with the current understanding of the effect of solvent on these transitions as achieved through state-of-the-art theoretical modeling makes it possible to consider the next step forward: the incorporation of spatial resolution and the construction of the singlet oxygen microscope.  相似文献
