A solvent extraction—spectrophotometric method has been developed for determination of bismuth in the form of tetra-n-butylammonium tetraiodobismuthate(III). The effects of pH, the concentrations of tetra-n-butylammonium and iodide ions, and the nature and amount of the organic, solvent have been studied. Tetra-n-butylammonium was found to be the most useful cation for extraction of tetraiodobismuthate with chloroform, giving quantitative extraction. The optimum pH is <3, and the extracted species is stable for at least a day at room temperature. Bismuth can be determined at the 10−5M level in aqueous solution, the relative standard deviation being 1.7%.  相似文献

A simple reversed-phase high-performance liquid chromatographic method employing C-18 column has been developed for simultaneous analysis of three intermediates in the synthesis of S-duloxetine, the antidepressant drug, viz., 2-acetyl thiophene (AT), N,N-dimethyl-3-keto-(2-thienyl)-propanamine (DKTP) and (S)-N,N-dimethyl-3-hydroxy-(2-thienyl)-propanamine (DHTP). Good separations were achieved by employing an isocratic system using acetonitrile and 0.05 M phosphate buffer (pH 7.0) containing 0.02% diethylamine. The detection was carried out at 241 nm. The method was validated for linearity, range, accuracy and precision. The developed method was applied for monitoring the progress of chemical synthesis of DKTP from AT followed by the biocatalytic reduction of DKTP to DHTP as the disappearance of the substrate and formation of the product can be monitored simultaneously by the present method.  相似文献

A selective and sensitive ion chromatographic method has been developed for the determination of beryllium in a number of water samples at low-μg/l concentrations. The separation was performed on a 250×4.0 mm I.D. iminodiacetic acid functionalised silica gel column. Chromatographed Be(II) was detected using visible detection at 590 nm following post-column reaction with chrome azurol S (CAS). The optimum separation and derivatisation conditions were studied in detail. The optimum eluent conditions were found to be 0.4 M KNO3, adjusted to pH 2.5 using HNO3, with optimum post-column detection being achieved using a solution containing 0.26 mM CAS, 2% Triton X-100, 50 mM 2-(N-morpholino)ethanesulfonic acid, pH 6.0. Under the above conditions, the concentration detection limit for Be(II) was found to be 3 μg/l in a standard solution and 4 μg/l in a typical tap water sample, using a 250 μl injection. The method was linear over the investigated range of 10 μg/l to 10 mg/l and highly reproducible. The method was successfully applied to a number of water samples of varying matrix complexity, including simulated seawater, and also to a natural freshwater certified reference material NIST 1640.  相似文献

A microemulsion electrokinetic chromatographic (MEEKC) method was developed for the separation of six catechins, specific marker phytochemicals of Cistus species. The MEEKC method involved the use of sodium dodecyl sulfate (SDS) as surfactant, heptane as organic solvent and butan-1-ol as co-solvent. In order to have a better stability of the studied catechins, the separation was performed under acidic conditions (pH 2.5 phosphate buffer). The effects of SDS concentration and of the amount of organic solvent and co-solvent on the analyte resolution were evaluated. The optimized conditions (heptane 1.36% (w/v), SDS 2.31% (w/v), butan-1-ol 9.72% (w/v) and 50 mM sodium phosphate buffer (pH 2.5) 86.61% (w/v)) allowed a useful and reproducible separation of the studied analytes to be achieved. These conditions provided a different separation profile compared to that obtained under conventional micellar electrokinetic chromatography (MECK) using SDS. The method was validated and applied to the determination of catechin and gallocatechin in lyophilized extracts of Cistus incanus and Cistus monspeliensis.  相似文献

A high-performance liquid chromatographic method was developed for the determination of the non-glycosidic anthraquinones alizarin (1,2-dihydroxy-9,10-anthracenedione), emodin (1,3,8-trihydroxy-6-methyl-9,10-anthracenedione) and anthraquinone (9,10-anthracenedione). The anthraquinones were separated by isocratic elution on a 125 × 4.6 mm I.D. column containing ODS Hypersil 5 reversed-phase material using methanol-5% acetic acid (pH 3.0) (70:30) as the mobile phase. Free alizarin was determined in plant cell suspension cultures of Rubia tinctorum and free emodin in mushrooms (Dermocybe sanguinea). The effective extraction of anthraquinones from plant cells was achieved with 80% (v/v) ethanol after incubation for 10 h at 80°C. Prepurification and concentration of anthraquinones in the plant cell and mushroom extracts were effected by a solid-phase technique using C8 cartridges.  相似文献

A capillary electrophoretic method using an uncoated capillary was developed to resolve potential impurities in FPL 67085XX, a novel phosphonate analogue of adenosine 5′-triphosphate. The effects of buffers, buffer concentration, eleunt pH and methanol were investigated. Optimum resolution of the impurities was achieved using an eluent consisting of 7% (v/v) methanol in 25 mM phosphate buffer-2 mM EDTA adjusted to pH 6.6.  相似文献

建立了粮食作物中三氯甲基吡啶及其代谢物6-氯吡啶甲酸的衍生化-气相色谱-三重四极杆质谱(GC-MS/MS)检测方法。选取高粱、小麦、玉米及爆谷等典型的粮食作物,采用酸性乙腈提取,以浓硫酸作为三氯甲基吡啶代谢物的衍生化试剂,优化了衍生化反应的最佳反应条件,并使用GC-MS/MS分析,以内标法对三氯甲基吡啶及其代谢物进行定量检测。结果表明,在0.025~0.4 mg/L范围内,线性关系良好,相关系数(r2)均大于0.995,在3个不同添加水平下的平均回收率在80.4%~98.4%之间,相对标准偏差(RSD)在1.0%~10.1%之间。该方法操作简单、高效,灵敏度高,抗干扰能力强,回收率和重复性良好,能够满足粮食作物中三氯甲基吡啶及其代谢产物残留量的检测要求。  相似文献

An analytical method for the determination of norethisterone acetate (NETA) in human plasma by capillary gas chromatography-mass-selective detection (GC-MS), with testosterone acetate as internal standard, was developed and validated. After addition of the internal standard, the compounds were extracted from plasma at basic pH into diethyl ether-dichloromethane (3:2 v/v), which was then evaporated to dryness. The compounds were converted into their pentafluoropropionyl derivatives which were determined by gas chromatography using a mass selective detector at m/z 486 for NETA and m/z 476 for the internal standard. Intra-day and inter-day accuracy and precision were found suitable over the range of concentrations between 0.10 to 10 ng/ml. The method was applied to clinical samples.  相似文献

A high-performance liquid chromatographic method with electrochemical detection has been developed for the determination of six 1,4-dihydropyridines: nifedipine, nimodipine, nisoldipine, nicardipine, felodipine and lacidipine. The chromatographic separation was performed using a Supelcosil LC-ABZ+Plus C18 column. A mobile phase of methanol–water (70:30), containing 2 mM CH3COOH–CH3COONa at a flow-rate of 1 ml/min and a pH of 5.0, was used. The temperature was optimized at 30±0.2°C. The amperometric detector, equipped with a glassy carbon electrode, was operated at 1000 mV versus Ag/AgCl in the direct current mode. The method was applied to the determination of these compounds at ng/ml concentrations, obtaining intra-day reproducibilities of lower than 5.0% in terms of relative standard deviations and detection limits ranging from 16 to 44 ng/ml. The method was applied to the screening of 1,4-dihydropyridines in spiked plasma samples, with a total elution time of lower than 18 min, obtaining the best recoveries for nimodipine and felodipine (91 and 88%, respectively). These recoveries together with the low detection limits achieved allow its application to the analysis of these drugs in human plasma.  相似文献

The influence of the following surfactants on the peak of copper in 0.2M EDTA at pH 4.5 was investigated: polyoxyethylated alkylphenols having an average of 3 and 9.5 ethylene oxide units; polyoxyethylene alcohols having 4 and 7 ethylene oxide units; poly(ethylene glycols) having M.W. 4000, 9000 and 20000; hexadecyltributylphosphonium bromide (HDTBPB), tetraphenylphosphonium bromide (TPPB),N,N,N,N,N',N',N-examethylhexamethylenediammonium bromide (HMB), benzyl(di-isobutylphenoxyethoxy) dimethylammonium chloride (Hyamine 1622), hexadecyltrimethylammonium bromide (HDTMAB), hexadecyldimethylbenzylammonium chloride (HDDMBAC) and tetrabutylammonium chloride (TBAC). HDDMBAC, as well as all the substances examined which contained an ethylene oxide chain, completely suppressed the copper peak. HDTBPB and TPPB partially suppressed the peak, whereas HDTMAB, HMB and Hyamine 1622 enhanced it. TBAC was without effect. In 0.2M EDTA at pH 4.5 containing TBAC at 0.01M concentration and 10 ppm of Rokafenol N-3, Cu(II), Pb(II) and Bi(III) can be tolerated at concentrations of up to 0.05M, the height of the thallium peak being unaffected. The precision of the determination (3–10%) and the recovery are satisfactory. A 103-fold ratio of Fe(III) to Tl(I) does not interfere with the determination.  相似文献

The adsorption of W (VI) on different metal oxides (TiO2, ZrO2), different crystal form of TiO2 (rutile, anatase) with high surface areas was studied and compared under different pH. A novel method for preconcentration of W (VI) with nanometer size titanium dioxide (rutile) and determination by spectrophotometry has been developed. W (VI) was selective adsorbed on 100 mg TiO2 from 250 ml solution at pH 3.0, then eluted by 2 ml 9 mol l−1 sodium hydroxide solution. The eluent was adjusted to 5 ml pH 0 solution, added 0.5 ml 12 mol l−1 HCl, 0.3 ml 3% TiCl3, 0.3 ml 50% NH4SCN, stirred for 20 min, used for the analysis of W (VI) by measuring the absorbance at 402 nm with spectrophotometry, based on the chromogenic reaction between the W (VI) and the mixture of TiCl3 and NH4SCN. This method gave a concentration enhancement of 50 for 250 ml sample, eliminated the sizable interferences on direct determination with spectrophotometry. Detection limits (3σ, n=11) of 1.2 ng ml−1, relative standard deviation of 2.3% at 10 ng ml−1 level were obtained. The method was applied to determine the W (VI) in hot spring water, river water, tap water and stream sediment. Analytical recoveries of W (VI) added to samples were 98–101%.  相似文献

A simple and sensitive flow injection method with fluorimetry and 5,5-dimethylcyclohexane-1,3-dione (dimedone) was developed for the determination of formaldehyde. Formaldehyde reacted with dimedone in the presence of ammonium acetate to form a fluorescence compound, which has an excitation wavelength at 395 nm and an emission wavelength at 463 nm. A two-channel flow system was assembled. Distilled water and 0.3% dimedone buffered at pH 5.5 were delivered at 0.7 ml min−1 and 100 μl of sample was injected into the carrier stream. The reaction was done in the reaction system designed newly, which consists of heating and cooling devices. The chemical reactivity with formaldehyde was excellent in the reaction system and selective. The calibration graphs were linear in the range of 25–100 and 5–10 ppb. RSDs (n=10) for 50 and 10 ppb formaldehyde were 0.6 and 3.4% and the LOD (S/N=3) was 0.9 ppb. The sample throughput was 20 h−1. The method was applied to the determination of formaldehyde in gas sample evolved from adhesive agents and in living environmental indoor. The sensitive and selective method is useful for monitoring trace of formaldehyde in the environmental atmosphere.  相似文献

The difficulty in ion-chromatographic determination of nitrite in aqueous solutions containing a high concentration of chloride arises mainly from incomplete resolution of the peaks for these anions on the separation column whose efficiency is not high. A photometric measurement of iodine formed by a reaction of nitrite with iodide has been found to make it possible to determine, chromatographically, trace amounts of nitrite without any interference from chloride; chloride does not oxidize iodide to produce iodine. The proposed method was based on the separation of nitrite from matrix anions on a silica-based anion-exchange column with a 1.5·10−3 M phthalate eluent (pH 5.0), followed by photometric measurement of the iodine (as triiodide) formed via a post-column reaction of the separated nitrite with iodide. The optimal conditions for the post-column reaction were established by varying the concentrations of iodide, copper(II) and nitric acid in a post-column-reaction solution and the length of a reaction tube. A calibration graph for nitrite, plotted as peak heights versus concentrations, was linear up to 1.50·10−5 M (690 ppb). The detection limit, defined at S/N=3, was 1.00·10−7 M (4.60 ppb) nitrite. The presence of chloride ions up to 0.01 M did not give any interference to the determination of nitrite. This method was successfully applied to the determination of nitrite in lake water, river water, sewage works water and snow samples without any pretreatment.  相似文献

We have developed a method for the determination of histamine (His), tyramine (Tyr) and cadaverine (Cad) using high-performance capillary electrophoresis with fluorescence detection and an on-line mode in-capillary derivatization with o-phthalaldehyde (OPA)/N-acetylcysteine (NAC) as derivatization reagent. HPCE separation of His, Tyr, Cad and Spermidine (Spd) was influenced by sodium dodecyl sulfate (SDS) and phosphate–borate buffer (pH 10) concentration. After optimization of the method, a 4-component amine solution containing His, Tyr, Cad and Spd could be separated and detected by using 2 mM OPA/NAC–20 mM SDS–20 mM phosphate–borate buffer (pH 10) as a run buffer at an applied voltage of 25 kV, with detection at 340 nm. Although a practical sensitivity level can be obtained by using fluorescence detection (λex=340 nm, λem=450 nm) instead of ultraviolet–visible detection, Spd was not detected at all. The precision (relative standard deviation; n=15) of this method for within- and between-days is less than 2.9% (peak area) and 1.3% (migration time), respectively. Linearity for these analytes, except for Spd, was established over a concentration range of 0.02 to 1.00 μmol/ml and detection limits (S/N=3) range from 1 nmol/ml for His and Tyr to 5 nmol/ml for Cad. The determination of His and some amines in aging raw fish meat samples (room temperature, 48 h) was carried out using the described method with fluorescence detection.  相似文献

基于四氮杂杯[2]芳烃[2]三嗪键合硅胶吸附剂(NC-Si),构建了固相萃取-高效液相色谱法同时测定河水中3种硝基苯酚和己烯雌酚的新方法。考察并获得了固相萃取和液相色谱分离的优化条件:将样品溶液pH调至5,以5 mL/min上样,经自制固相萃取柱净化,2 mL氨水-甲醇(2:98,v/v)洗脱;在C8柱上以甲醇-0.1%磷酸溶液为流动相进行梯度洗脱。4种目标分析物的检出限(LOD,S/N=3)为0.03~0.3 μg/L,定量限(LOQ,S/N=10)为0.1~1.0 μg/L;加标回收率为75.5%~104.2%,相对标准偏差(RSD,n=5)小于6.3%。该方法准确、可靠,可用于河水中硝基苯酚及己烯雌酚的灵敏检测。  相似文献

利用原子转移自由基聚合(ATRP)法和连续ATRP法合成了温度敏感型聚合物和pH/温度双重敏感型聚合物。用紫外光谱考察聚合物在水溶液中的温敏行为,发现聚合物的低临界溶解温度(LCST)可以通过单体的比例进行调控,而且聚合物的温度响应行为非常敏感且具有可逆性。pH/温度双重敏感型聚合物还具有非常灵敏的pH响应行为,且不受单体比例的影响。最后,对聚合物胶束的体外释药动力学进行了研究,结果表明聚合物胶束的环境敏感性决定了药物的释放行为。  相似文献

A simple and rapid method for the simultaneous determination of six ephedrine alkaloids (ephedrine, pseudoephedrine, norephedrine, norpseudoephedrine, methylephedrine and methylpseudoephedrine) in Ephedrae herba by capillary electrophoresis was developed. A buffer solution that contained 0.005 M barium hydroxide and 0.02 M isoleucine and adjusted to pH 10.0 with ammonia solution was found to be the most suitable electrolyte for this separation. The contents of the six alkaloids in the crude drug of Ephedrae herba could be easily determined.  相似文献

Changes in fresh weight, total protein amounts (Bradford’s method), cadmium concentration (DPASV) and glutathione content (HPLC/MS) were studied in maize kernels cultivated for 5 days at three different cadmium concentrations (0, 10 and 100 μmol l−1 CdCl2). A highly sensitive HPLC/MS method was used for the determination of glutathione on a reversed-phase Atlantis dC18 chromatographic column (150 mm×2.1 mm, 3 μm particle size). An isocratic mode with acetonitrile–0.01% TFA (5:95, flow rate 0.1 ml min−1 and 30 °C) was applied. The m/z spectra and the data for the selected ion monitoring (SIM) mode were recorded at m/z for glutathione 308→179. Cadmium concentration was measured by a differential pulse adsorptive stripping voltammetry (DPASV) after deposition on a hanging mercury drop electrode (HMDE) at potential −0.7 V (accumulation time 180 s, acetate buffer of pH 3.6, 22 °C). An AUTOLAB with a VA-Stand 663 and a three-electrode system consisting of the HMDE as a working electrode with area 0.4 mm2, an Ag/AgCl/3 mol l−1 KCl as a reference electrode and a Pt-wire as an auxiliary electrode was employed. The maize kernels exposed to the highest cadmium concentration (100 μmol l−1) germinated formerly and much better. A rapid increase of the fresh weight probably relates with more intensive uptake of water in order to decrease cadmium concentration. An intensive preservation of homeostasis of Cd2+ ions in the germinating plants by defending mechanisms might explain differences of uptake rate of cadmium. The linear increase of GSH content with the exposure time at all studied concentration suggests the defending mechanisms might be triggered by concentrations of a heavy metal.  相似文献

A direct sensing method for monitoring meat quality was developed. The sensor is composed of an Ag/AgCl electrode and a platinum electrode on which putrescine oxidase or xanthine oxidase were immobilized to estimate bacterial spoilage or the progress of aging, respectively. A potential-step chronoamperometric method was applied in which the potential was stepped from 300 mV to 600 mV. A linear relationship was obtained between 5 and 60 nmol g−1 for putrescine (Put) and 0.05 and 1.0 μmol g−1 for hypoxanthine (Hx). The coefficient of variation was 0.75% for 20 nmol ml−1 Put solution and 2.2 for a meat sample using the putrescine sensor, and 1.09% for 0.25 μmol ml−1 Hx solution and 2.6% for a meat sample using the xanthine sensor. The pH requirements and substrate selectivity were suitable for the direct measurement of substrates on the surface of meat. From the results of practical experiments, the direct sensing method was indicated to be useful with some modifications for the estimation of meat quality during aging.  相似文献

An isotachophoretic (ITP) method for the determination of fluoride in feed mixtures was developed. A sample of feed mixture, after extraction with 1 M HCl, was analysed using a ZKI 02 column-coupling isotachopherograph. Leading electrolytes for presentation and analytical capillaries consisted of 0.008 M HCl-0.022 M -aminocaproic acid (EACA)-0.001 M CaCl2-0.05% hydroxypropylmethyl cellulose (HPMC) and 0.002 M HCl-0.005 M EACA-0.05% HPMC, respectively. The terminating electrolyte was 0.01 M tartaric acid. The fluoride released from samples by microdiffusion in 25% perchloric acid was determined using an Ionosep 900.1 single capillary isotachopherograph with 0.002 M HCl-0.005 M EACA-0.05% HPMC as the leading electrolyte and 0.002 M tartaric acid as the terminating electrolyte. The detection limit, depending on the sample treatment, was as low as 4 μg/g as fluoride. A comparison of the developed ITP method with ion- selective electrode method was carried out.  相似文献
